Global genetic structure and demographic history of Haemaphysalis longicornis revealed by mitochondrial cytochrome c oxidase subunit I sequences.

BACKGROUND: Haemaphysalis longicornis is an important vector of multiple tick-borne pathogens and exhibits both parthenogenetic and bisexual reproductive modes. Among them, the parthenogenetic strain has spread to North America and Oceania. However, a systematic comparison of the genetic structure, dispersal patterns, and demographic history between the parthenogenetic and bisexual strains is still lacking. METHODS: Global mitochondrial cytochrome c oxidase subunit I (COI) sequences of H. longicornis were retrieved from GenBank. Phylogenetic reconstruction, Bayesian genetic clustering, haplot

Bothriocestus claviceps (Cestoda: Bothriocephalidae) in pike in southwestern France (Teleostei: Esocidae): molecular evidence.

Bothriocestus claviceps (Goeze, 1782) is a cestode parasite infecting several fish species. One of its principal definitive hosts is the European eel Anguilla anguilla. The parasite is widely distributed across Europe and has been reported from numerous countries, such as Poland, Czechia, and France. In southern France, the European eel co-occurs with the Aquitanian pike, Esox aquitanicus, an endemic species. This pike is threatened by human activities and hybridization with the introduced Northern pike Esox lucius and is therefore classified as "vulnerable" on the French Red List of the Inter

Morphological and molecular description of a novel species of Eimeria (Apicomplexa) that infects extraintestinal tissues of kiwi (Aves: Apteryx spp.).

Coccidia (Apiconmplexa) are naturally occurring and occasionally detrimental parasites of kiwi (Apteryx spp.), a unique, flightless bird species dependent upon conservation efforts for survival. Using morphological and molecular data, a new coccidia species, Eimeria koka n. sp., isolated from two closely related but geographically isolated kiwi host species, Apteryx rowi Tennyson et al. (rowi) and Apteryx mantelli Bartlett (North Island brown kiwi), is described. Oocysts are oval (20.8 × 15.9 μm) with a mean L/W ratio of 1.3, and a distinctive rough, crenellated brown oocyst wall (mean 1.2 μm)

Development of a Real-Time PCR Assay for Noninvasive Detection of Miamiensis avidus in Olive Flounder Aquaculture.

Miamiensis avidus (syn. Philasterides dicentrarchi) leads to high mortality and economic losses in olive flounder (Paralichthys olivaceus) aquaculture. In this study, we developed a real-time PCR assay targeting the cytochrome c oxidase subunit I (cox1) gene of M. avidus to sensitively detect and quantify the parasite in seawater. The assay showed a strong linear correlation between the log copy number of the standard plasmid and Ct values (R2 = 0.9985), achieving an amplification efficiency of 96.26%. The cox1 gene copy number was estimated at 6017 ± 2794 copies per cell. The assay specifical

A morphological and molecular study of phlebotomine sand flies of Taiwan reveals the record of six species.

BACKGROUND: Phlebotomine sand flies, the key vectors in the transmission of Leishmania parasites, pose a global health challenge. Although leishmaniasis has been reported in the indigenous Taiwanese population, the last sand fly survey, based on morphological features, was conducted over two decades ago. Thus, updated information on the phlebotomine sand fly fauna and disease transmission risk in Taiwan is required. In this study, a nationwide survey was conducted using molecular methods to ascertain the current sand fly status and disease transmission risk in Taiwan. METHODS: A total of 1292

Galactose-1-phosphate inhibits cytochrome c oxidase and causes mitochondrial dysfunction in classic galactosemia.

Classic galactosemia is an inborn error of metabolism caused by mutations in the GALT gene resulting in the diminished activity of the galactose-1-phosphate uridyltransferase enzyme. This reduced GALT activity leads to the buildup of the toxic intermediate galactose-1-phosphate and a decrease in ATP levels upon exposure to galactose. In this work, we focused our attention on mitochondrial oxidative phosphorylation in the context of this metabolic disorder. We observed that galactose-1-phosphate accumulation reduced respiratory rates in vivo and changed mitochondrial function and morphology in

Physical activity as a promoter of stress resilience: An analysis of behavioral effects and brain connectivity with cytochrome c-oxidase activity in adult male Wistar rats.

Physical activity (PA) is very beneficial for physical and mental health. This study aims to examine the resilience-inducting effect of PA in adult male Wistar rats exposed to unpredictable chronic mild stress (UCMS). Furthermore, we analyzed the influence of PA on behavioral tasks and functional brain connectivity with cytochrome c oxidase technique. The cytochrome c oxidase (CCO) is a mitochondrial enzyme involved in oxidative phosphorylation and ATP generation. For this analysis, we included five groups: Basal (n = 10, to determine the basal level of brain activity), Behav (n = 15, subjecte

A novel m.5906G > a variant in MT-CO1 causes MELAS/Leigh overlap syndrome.

The MELAS/Leigh overlap syndrome manifests with a blend of clinical and radiographic traits from both MELAS and LS. However, the association of MELAS/Leigh overlap syndrome with MT-CO1 gene variants has not been previously reported. In this study, we report a patient diagnosed with MELAS/Leigh overlap syndrome harboring the m.5906G > A variant in MT-CO1, with biochemical evidence supporting the pathogenicity of the variant. The variant m.5906G > A that led to a synonymous variant in the start codon of MT-CO1 was filtered as the candidate disease-causing variant of the patient. Patient-derived

In vitro galactose impairs energy metabolism in the brain of young rats: protective role of antioxidants.

We, herein, investigated the in vitro effects of galactose on the activity of pyruvate kinase, succinate dehydrogenase (SDH), complex II and IV (cytochrome c oxidase) of the respiratory chain and Na+K+-ATPase in the cerebral cortex, cerebellum and hippocampus of 30-day-old rats. We also determined the influence of the antioxidants, trolox, ascorbic acid and glutathione, on the effects elicited by galactose. Galactose was added to the assay at concentrations of 0.1, 3.0, 5.0 and 10.0 mM. Control experiments were performed without galactose. Galactose, at 3.0, 5.0 and 10.0 mM, decreased pyruvate

Assessing the diversity and distribution of potential intermediate hosts snails for urogenital schistosomiasis: Bulinus spp. (Gastropoda: Planorbidae) of Lake Victoria.

BACKGROUND: The Lake Victoria basin is one of the most persistent hotspots of schistosomiasis in Africa, the intestinal form of the disease being studied more often than the urogenital form. Most schistosomiasis studies have been directed to Schistosoma mansoni and their corresponding intermediate snail hosts of the genus Biomphalaria, while neglecting S. haematobium and their intermediate snail hosts of the genus Bulinus. In the present study, we used DNA sequences from part of the cytochrome c oxidase subunit 1 (cox1) gene and the internal transcribed spacer 2 (ITS2) region to investigate Bu

Cannabidiol Prevents the Expression of the Locomotor Sensitization and the Metabolic Changes in the Nucleus Accumbens and Prefrontal Cortex Elicited by the Combined Administration of Cocaine and Caffeine in Rats.

In the last years, clinical and preclinical researchers have increased their interest in non-psychotomimetic cannabinoids, like cannabidiol (CBD), as a strategy for treating psychostimulant use disorders. However, there are discrepancies in the pharmacological effects and brain targets of CBD. We evaluated if CBD was able to prevent the locomotor sensitization elicited by cocaine and caffeine co-administration. The effect of CBD on putative alterations in the metabolic activity of the medial prefrontal cortex (mPFC) and nucleus accumbens (NAc), and its respective subregions (cingulated, prelim
