Og4C3 circulating antigen: a marker of infection and adult worm burden in Wuchereria bancrofti filariasis.

Og4C3 circulating filarial antigen was detected in the sera of 94.5% (259/274) of microfilaremic patients, 32% (239/751) of persons with presumption of filariasis, and 23% (11/48) of chronic filariasis patients. The antigen level was correlated with the microfilariae (Mf) density and patient age (P < .01). It remained stable in patients treated with microfilaricidal drugs. Og4C3 antigen, undetectable in Mf culture media, was demonstrated to be a rare somatic Mf antigen. It appears to be an excreted or secreted antigen from adult filaria. It could be used as a marker of infection and an indicat

The resurgence of lymphatic filariasis in the Nile delta.

A study of 325,000 residents of 314 villages in six governorates of the Nile delta area of Egypt revealed that the prevalence of lymphatic filariasis increased from < 1% in 1965 to > 20% in 1991, especially in the governorates of Qalyubiya, Monufiya, Dakhaliya, and Giza. The distribution of the communites with endemic filariasis is focal. Clusters of villages with high prevalences are surrounded by others in which the disease is absent, although their environmental, social, and agricultural features appear similar. The article analyses why the significant decline in filariasis between 1945 and

Bancroftian filarial pleural effusion.

This paper describes a case of filarial pleural effusion, the fifth such to be reported. Microfilariae of Wuchereria bancrofti were detected in the pleural fluid on cytological examination. There was a prompt and complete response to treatment with diethylcarbamazine. There were, however, no symptoms and signs of tropical pulmonary eosinophilia nor any peripheral eosinophilia.

On the infectivity of early third-stage Brugia larvae isolated from the abdomen of Aedes aegypti.

Third-stage larvae (L3) of Wuchereria bancrofti, Brugia malayi and B. pahangi are known to be recovered not only from the head and thorax but also from the abdomen of dissected vector mosquitoes. For epidemiological reasons, was of interest to determine whether L3 larvae from the abdomen of the vector would be infectious for the final host. Early abdominal L3 larvae of B. pahangi isolated as early as on day 7 post-infection (p.i.) from Aedes aegypti were injected s.c. into five male Meriones unguiculatus. Four of the five jirds were microfilaria-positive after 67 days.

Wuchereria bancrofti microfilarial density of autochthonous cases and natural Culex infectivity rates in northeast Brazil.

Autochthonous Wuchereria bancrofti microfilaraemic cases from three neighbouring cities of Northeast Brazil (Recife, Jaboatao and Olinda) were selected from thick smear or filtration positive individuals and classified by their microfilarial (mf) densities through polycarbonate membrane blood filtration. Individuals were subdivided into two groups: autochthonous cases without any previous specific drug administrations (group A) and those with one or more previous DEC treatments (group B). In Recife, unlike Jaboatao and Olinda (P less than 0.05), the majority of autochthonous cases were over 20

Diethylcarbamazine treatment of bancroftian and malayan filariasis with emphasis on side effects.

A total of 1015 filarial carriers were hospitalized and treated with DEC either in a long course with light doses (6, 7, 8, 9 or 10 mg kg-1 b.w. divided into three doses daily for seven days) or in a short course with heavy doses (15 mg kg-1 b.w. once daily for one, two or three days). The efficacy of the long course (85%) was much higher than that of the short course (59%); but the reaction rate following the heavy dose (87%) was higher than that following the light dose (72%). For Brugia malayi infections the overall efficacy was 92% and the reaction rate 82%, for mixed infections the respec

Factors affecting transmission of Wuchereria bancrofti by anopheline mosquitoes. 4. Facilitation, limitation, proportionality and their epidemiological significance.

Quantitative understanding of the transmission dynamics of lymphatic filarial parasites is essential for the rational planning of control strategies. One of the most important determinants of transmission dynamics is the relationship between parasite yield, the success rate of ingested microfilariae (mf) becoming infective larvae in a mosquito vector, and mf density in the source of the human blood meal. Three types of relationship have been recognized in human filaria/mosquito couples--limitation, facilitation and proportionality; facilitation has hitherto been observed only in the couple Wuc

The significance of low density microfilaraemia in the transmission of Wuchereria bancrofti by Culex (Culex) quinquefasciatus Say in Sri Lanka.

Laboratory-bred Culex quinquefasciatus were fed on carriers with low and moderate densities of microfilariae (mf) of Wuchereria bancrofti. In the first series of experiments, mosquitoes were dissected 12 d after feeding. The percentage of infected mosquitoes and the numbers of larvae per infected mosquito were directly proportional to the mf density at the time of feeding. There was an overall high infection rate and a wide distribution of larvae per individual mosquito at all levels, except for the 4 lowest counts. Of the 4 carriers with counts of 5 mf/ml and less, 3 were capable of infecting

Changes in circulating parasite antigen levels after treatment of bancroftian filariasis with diethylcarbamazine and ivermectin.

This study assessed changes in circulating parasite antigen levels after diethylcarbamazine (DEC) and ivermectin treatment of bancroftian filariasis to determine effects of these drugs on adult Wuchereria bancrofti in vivo. Thirty adult Haitians with microfilaremia were treated with 1 mg of ivermectin to reduce counts of microfilariae. Later, subjects were treated with either one or two 200 micrograms/kg doses of ivermectin or with 12 daily 6 mg/kg doses of DEC. Macrofilaricidal activity of these drugs was indirectly monitored by measuring circulating W. bancrofti antigen by EIA. Antigen level

Antifilarial activity of CGP 20376 against subperiodic Brugia malayi in the leaf-monkey Presbytis cristata.

CGP 20376, a 5-methoxyl-6-dithiocarbamic-S- (2-carboxy-ethyl) ester derivative of benzothiazole was evaluated for its antifilarial properties and shown to be extremely effective against subperiodic Brugia malayi in the leaf-monkey, Presbytis cristata at oral doses of 20-100 mg/kg. The compound and/or its metabolites had complete micro- and microfilaricidal activities even when given at a single dose of 20 mg/kg. Lower doses had incomplete filaricidal action.

Experimental infection of the leaf-monkeys, Presbytis cristata and Presbytis melalophos with subperiodic Brugia malayi.

The leaf-monkeys, Presbytis cristata and Presbytis melalophos, experimentally infected with subperiodic Brugia malayi, have been used for studies on the pathoimmunology of the infection and the screening of potential filaricides during the last 6-8 years, and considerable information on the pattern of microfilaraemia and adult worm recoveries have been obtained. The prepatent periods in 97 P. cristata and 45 P. melalophos, each infected with about 200 infective larvae, were similar, these being approximately 70 and 68 days respectively. Although all infected animals became microfilaraemic, the

Circadian periodicity in microfilarial counts.

We propose a second-order harmonic model to describe circadian periodicity in the 24-h cycle of microfilarial counts. Rhesus monkeys (Macaca mulatta) were infected with Brugia malayi. Definite evidence of circadian periodicity was found in the microfilarial counts. The estimates of the parameters, periodicity indices and peak hours were determined.

Detection of a new focus of Brugia malayi infection in Orissa.

526 people were surveyed in a village called Chudamani, in Balasore district of Orissa, for detection of asymptomatic microfilaria (mf) carriers. Of these 36 (6.8 per cent) were cases found to harbour mf; 19 cases had Brugia malayi, 4 Wuchereria bancrofti and 5 cases had mixed infection. In 8 cases, species could not be ascertained. For the first time after 1955, a focus of B. malayi has been detected in Orissa.

Microfilariae in fine needle aspirates from epididymal lesions.

Microfilariae of Wuchereria bancrofti were observed in fine needle aspiration smears from three epididymal nodules, and degenerating microfilariae suggestive of Brugia malayi were found in the smears from a fourth case. The smears in all four cases showed a polymorphonuclear inflammatory cell component as well as epithelioid cell granulomata. While blood eosinophilia was present in all four cases, eosinophilia was present in the aspiration smears in only one case. Microfilariae could be demonstrated in the peripheral blood in only one case.
