Quantum dot-enhanced tunable FRET nanosensor for Alzheimer's-associated dual miR-34 detection.

The microRNAs miR-34a and miR-34b are dysregulated in Alzheimer's disease (AD) and are promising diagnostic biomarkers. However, their low abundance, high sequence homology, and instability present severe challenges for accurate multiplex detection. Herein, we report a quantum dot-enhanced turn-up/down dual-miRNA FRET sensor (QDTF sensor), a platform that achieves simultaneous detection of two highly homologous miRNAs with low crosstalk via an unprecedented asymmetric dual-mode FRET strategy enhanceded by a single quantum dot 605. Among the two pairs of molecular beacons, one triggers a consid

Using Dark Dyes for Fast Super-Resolution Imaging: A Proof-of-Concept Study.

DNA-PAINT (Point Accumulation for Imaging in Nanoscale Topography) exploits the transient hybridization of a short fluorescent DNA imager strand to a complementary docking strand to achieve super-resolution imaging. However, its application is hampered by the background noise caused by unbound imager strands and the subsequent slow image acquisition. Several variants of DNA-PAINT have already been developed to successfully overcome these limitations. Here we design and validate a new alternative, based on intermolecular dark resonance energy transfer (DRET) and the use of a fluorenyl nucleobas…

Synthesis, fluorescence and theoretical insights into a novel FRET-based dansyl-rhodamine sensor for the in vitro detection of toxic bioaccumulated Hg(II) ions.

This work describes the successful design and synthesis of a new fluorescence resonance energy transfer (FRET)-based sensor, denoted as RD1. This sensor incorporates a robust dual-fluorophore design, which combines a rhodamine and a dansyl derivative, functionalized with a thiosemicarbazide group that acts as Hg(II) specific recognition site. A synthetic pathway was developed that allowed the efficient synthesis of RD1 with a remarkable overall yield of 44% over four steps, through microwave-assisted protocols. The influence of ethyl, benzyl and phenyl substituents of isothiocyanate in the pre…

Fluorescence lifetime FRET assay for live-cell high-throughput screening of the cardiac SERCA pump yields multiple classes of small-molecule allosteric modulators.

We have used FRET-based biosensors in live cells, in a robust high-throughput screening (HTS) platform, to identify small-molecules that alter the structure and activity of the cardiac sarco/endoplasmic reticulum calcium ATPase (SERCA2a). Our primary aim is to discover drug-like small-molecule activators that improve SERCA's function for the treatment of heart failure. We have previously demonstrated the use of an intramolecular FRET biosensor, based on human SERCA2a, by screening two different small validation libraries using novel microplate readers that detect the fluorescence lifetime or e

Advancements in a FRET Biosensor for Live-Cell Fluorescence-Lifetime High-Throughput Screening of Alpha-Synuclein.

There is a critical need for small molecules capable of rescuing pathophysiological phenotypes induced by alpha-synuclein (aSyn) misfolding and oligomerization. Building upon our previous aSyn cellular fluorescence lifetime (FLT)-Förster resonance energy transfer (FRET) biosensors, we have developed an inducible cell model incorporating the red-shifted mCyRFP1/mMaroon1 (OFP/MFP) FRET pair. This new aSyn FRET biosensor improves the signal-to-noise ratio, reduces nonspecific background FRET, and results in a 4-fold increase (transient transfection) and 2-fold increase (stable, inducible cell lin…

A dual "turn-on" biosensor based on AIE effect and FRET for in situ detection of miR-125b biomarker in early Alzheimer's disease.

MicroRNA-125b (miR-125b) is highly associated with synaptic dysfunction and tau hyperphosphorylation in the early pathogenesis of Alzheimer's disease (AD), making it a promising biomarker for early AD diagnosis. Hence, there is an urgent need for a reliable sensing platform to assist in situ miR-125b detection. In this work, we report a dual "turn-on" fluorescence biosensor based on the nanocomposite of aggregation-induced emission fluorogen (AIEgen)-labeled oligonucleotide (TPET-DNA) probes immobilized on the surface of cationic dextran modified molybdenum disulfide (TPET-DNA@Dex-MoS2). The Human Mercaptopyruvate Sulfurtransferase TUM1 Is Involved in Moco Biosynthesis, Cytosolic tRNA Thiolation and Cellular Bioenergetics in Human Embryonic Kidney Cells.

Sulfur is an important element that is incorporated into many biomolecules in humans. The incorporation and transfer of sulfur into biomolecules is, however, facilitated by a series of different sulfurtransferases. Among these sulfurtransferases is the human mercaptopyruvate sulfurtransferase (MPST) also designated as tRNA thiouridine modification protein (TUM1). The role of the human TUM1 protein has been suggested in a wide range of physiological processes in the cell among which are but not limited to involvement in Molybdenum cofactor (Moco) biosynthesis, cytosolic tRNA thiolation and gene

Graphene oxide-mediated fluorescence turn-on GO-FAM-FRET aptasensor for detection of sterigmatocystin.

Sterigmatocystin (STC) is a toxic fungal secondary metabolite recognized by the FAO and WHO as a genotoxic and carcinogenic substance. STC contaminates several foods and feed commodities, posing a health risk to humans. The present study proposes to develop a graphene oxide-mediated aptasensor platform for the one-step detection of STC. In this study, DNA aptamers were generated against STC by using a target immobilization-free graphene oxide (GO)-SELEX protocol. The champion aptamers were subjected to in silico maturation using a genetic algorithm to improve binding affinity. Further, MSA-C6

Mitochondria-targeted and FRET-based fluorescent probe for the imaging of endogenous SO2 in living cells.

Sulfur dioxide (SO2) is an important signal molecule in living systems, and plays a wide range of physiological functions. Real-time and in situ detection of the dynamic balance of SO2 in mitochondria is of great significance to in-depth study its biological roles. Herein, we have developed a mitochondria-targeted fluorescent probe Nap-L based on the FRET mechanism to detect SO2 in living cells. The probe Nap-L employed naphthalimide and positively charged benzopyridine as the donor and acceptor in the FRET system, and emitted green and red fluorescence under excitation. In respond to SO2, the

The reaction mechanism of Zika virus NS2B/NS3 serine protease inhibition by dipeptidyl aldehyde: a QM/MM study.

Zika virus (ZIKV) infection has become a global public health problem, associated with microcephaly in newborns and Guillain-Barré syndrome in adults. Currently, there are no commercially available anti-ZIKV drugs. The viral protease NS2B/NS3, which is involved in viral replication and maturation, is a potential drug target. Peptidomimetic aldehyde inhibitors bind covalently to the catalytic S135 of the NS3 protease. Here, we apply hybrid quantum mechanics/molecular mechanics (QM/MM) free-energy simulations at the PDDG-PM3/ff14SB level to investigate the inhibition mechanism of the ZIKV protea

Monomeric Huntingtin Exon 1 Has Similar Overall Structural Features for Wild-Type and Pathological Polyglutamine Lengths.

Huntington's disease is caused by expansion of a polyglutamine (polyQ) domain within exon 1 of the huntingtin gene (Httex1). The prevailing hypothesis is that the monomeric Httex1 protein undergoes sharp conformational changes as the polyQ length exceeds a threshold of 36-37 residues. Here, we test this hypothesis by combining novel semi-synthesis strategies with state-of-the-art single-molecule Förster resonance energy transfer measurements on biologically relevant, monomeric Httex1 proteins of five different polyQ lengths. Our results, integrated with atomistic simulations, negate the hypoth…

One-step synthesis of DNA functionalized cadmium-free quantum dots and its application in FRET-based protein sensing.

DNA functionalized quantum dots (QDs) are promising nanoprobes for the fluorescence resonance energy transfer (FRET)-based biosensing. Herein, cadmium-free DNA functionalized Mn-doped ZnS (DNA-ZnS:Mn2+) QDs were successfully synthesized by one-step route. As-synthesized QDs show excellent photo-stability with the help of PAA and DNA. Then, we constructed a novel FRET model based on the QDs and WS2 nanosheets as the energy donor-acceptor pairs, which was successfully applied for the protein detection through the terminal protection of small molecule-linked DNA assay. This work not only explores…

Fluorescent CdS Quantum Dots: Synthesis, Characterization, Mechanism and Interaction with Gold Nanoparticles.

CdS quantum dot (QD) is a typical kind of II-IV nanoparticles, which plays an important role in the common type of core-shell QDs. It is of great practical significance to synthesize the water-soluble CdS QDs used in multicolor biomarkers and prepare core-shell QDs. In our case, we came up with a novel green method to manufacture CdS QDs with high quality, different size, and adopted UV-vis absorption, fluorescence, FTIR, XPS, HRTEM, SAED and STEM-EDX to discuss their growth mechanism. We successfully constructed fluorescence resonance energy transfer (FRET) system between CdS QDs and gold nan…

Topologically nontrivial narrow bands in ultrathin SnTe films with defect superstructures.

It is shown that a two-dimensional topological insulator can be realized and the band topology (equivalently, the edge states) may be further controlled by charge doping in an ultrathin SnTe film with a defect superstructure. Based on first-principles density functional theory (DFT), we predict that a Sn-Te bilayer, if exfoliated from three-dimensional bulk SnTe in the (1 1 1) direction, has a trivial band topology in its pristine form, but is made topologically nontrivial by introducing an appropriate array of defects. The emergence of the topological state is ascribed to the formation of top

Binding energy of D(0) and D(-) impurity centers in CdTe/ZnTe spherical quantum dot.

Binding energy of neutral (D(0)) and negatively charged (D(-)) hydrogenic impurity located at the center of the CdTe/ZnTe spherical quantum dot has been investigated. Calculations are performed under the effective mass approximation on the basis of exact solution of the Schrödinger and Poisson equations. Eigenfunctions are expressed in terms of the Whittaker and Coulomb wave functions. Calculated results show that D(0) and D(-) impurity binding energies of ground 1s, 1S(1s2) and 2p, 3P(1s2p) excited states strongly depend on CdTe/ZnTe QD size if QD radius is less than one effective Bohr radius

Comparison of the apparent mass of the seated human measured using random and sinusoidal vibration.

Exposure to whole-body vibration is generally accepted as being a risk factor for low back pain and therefore exposure to vibration should be minimised. The results of previous laboratory based research investigating the biomechanical response of the seated human to vibration has been used to develop models that can be used within tools that are capable of predicting the response of seats. Several studies in the literature have reported apparent masses of seated human subjects whilst exposed to either random or sinusoidal vibration. Although these studies have shown similar trends, there have

The wounding characteristics of spherical steel fragments in live tissues.

This paper reports the wounding effectiveness tests of four kinds of spherical steel fragments, striking against live tissues at various velocities. The most pronounced wound characteristic for the spherical fragments was that the entrances were larger than the exits; the higher the striking velocity, the larger the size of the wound. The skin near the entrance tended to be torn away when the striking velocity was above 1,200 m/s. As a general rule, most exits appeared to be circular with a diameter slightly bigger than that of the fragment. Shapes of the wound channels were conic in general.
