GABAB receptors in glial cells: from physiological regulation to pathological implications.

The GABAB receptor, a metabotropic receptor for the inhibitory neurotransmitter γ-aminobutyric acid, is essential for the proper functioning of inhibitory neurons. Previous studies have shown that GABAB receptors are primarily expressed at presynaptic or postsynaptic membranes of inhibitory or excitatory neurons, where they play a critical role in regulating synaptic signalling. Interestingly, a growing body of evidence indicates that GABAB receptors are also expressed in various types of glial cells and can significantly influence their functions, ultimately contributing to the regulation of

Electrophysiologically identified presynaptic mechanisms underlying amylinergic modulation of area postrema neuronal excitability in rat brain slices.

Amylin, which is co-secreted together with insulin by pancreatic beta cells, is considered to be an important peptide hormone involved in the control of feeding behavior and energy homeostasis. Although the area postrema has been implicated to be a primary target of amylin, there are no studies of the mechanisms by which amylin may alter the excitability of area postrema neurons. To investigate the mechanism for amylinergic modulation of neuronal excitability, we performed perforated patch-clamp recordings from area postrema neurons in rat brainstem slices. Amylin-induced changes in excitatory

AMPA Receptors in NG2 Glia Differently Affect Signal Transduction in the Hippocampus and Cerebellum.

Gray matter NG2 glia constitute a heterogeneous population of cells whose functions remain incompletely understood. In the hippocampus, Schaffer collaterals activate AMPA receptors (AMPARs) in NG2 glia, giving rise to small excitatory post-synaptic currents (EPSCs). Climbing fibers of the cerebellum also form synapses with NG2 glia, although producing much larger EPSCs. We aimed to identify mechanisms generating these regional differences in the efficacy of neuron-glia synapses. Combined patch-clamp and RT-PCR analyses allowed for determining structural and functional differences of AMPARs exp

Synaptic Plasticity of D1 and D2 Types of Neurons in the Central Amygdala After Sucrose and Cocaine Exposure.

Dopamine-sensitive neurons are organized in two classes of cells, expressing D1- or D2-type dopamine receptors, and often mediate opposing aspects of reward-oriented behaviors. Here, we focused on D1- and D2-type neurons in the central amygdala, a brain structure critically involved in processing emotion-related stimuli. We demonstrated the distinct composition of the central amygdala based on these receptors: the lateral part (CeL) consists almost exclusively of D2-type neurons, while the medial part (CeM) contains a mix of both D1- and D2-type neurons. Recording excitatory postsynaptic curre

Soluble form of Lingo2, an autism spectrum disorder-associated molecule, functions as an excitatory synapse organizer in neurons.

Autism Spectrum Disorder (ASD) is a developmental disorder characterized by impaired social communication and repetitive behaviors. In recent years, a pharmacological mouse model of ASD involving maternal administration of valproic acid (VPA) has become widely used. Newborn pups in this model show an abnormal balance between excitatory and inhibitory (E/I) signaling in neurons and exhibit ASD-like behavior. However, the molecular basis of this model and its implications for the pathogenesis of ASD in humans remain unknown. Using quantitative secretome analysis, we found that the level of leuci

Amyloid Beta-Mediated Changes in Synaptic Function and Spine Number of Neocortical Neurons Depend on NMDA Receptors.

Onset and progression of Alzheimer's disease (AD) pathophysiology differs between brain regions. The neocortex, for example, is a brain region that is affected very early during AD. NMDA receptors (NMDARs) are involved in mediating amyloid beta (Aβ) toxicity. NMDAR expression, on the other hand, can be affected by Aβ. We tested whether the high vulnerability of neocortical neurons for Aβ-toxicity may result from specific NMDAR expression profiles or from a particular regulation of NMDAR expression by Aβ. Electrophysiological analyses suggested that pyramidal cells of 6-months-old wildtype mice

The duration of withdrawal affects the muscarinic signaling in the nucleus accumbens after chronic morphine exposure in neonatal rats.

The infants experience withdrawal from opiates, and time-dependent adaptations in neuronal activity of nucleus accumbens (NAc) may be crucial for this process. A key adaptation is an increased release of acetylcholine. The present study investigates muscarinic acetylcholine receptors (mAChRs) functions in the NAc at short-term (SWT) and long-term (LWT) withdrawal time following chronic morphine exposure in neonatal rats. The inhibitory role of presynaptic mAChRs activation in spontaneous excitatory postsynaptic currents (sEPSCs) in medium spiny neurons was decreased at LWT but not at SWT. Wher

Electrophysiological Assessment of Serotonin and GABA Neuron Function in the Dorsal Raphe during the Third Trimester Equivalent Developmental Period in Mice.

Alterations in the development of the serotonin system can have prolonged effects, including depression and anxiety disorders later in life. Serotonin axonal projections from the dorsal raphe undergo extensive refinement during the first 2 weeks of postnatal life in rodents (equivalent to the third trimester of human pregnancy). However, little is known about the functional properties of serotonin and GABA neurons in the dorsal raphe during this critical developmental period. We assessed the functional properties and synaptic connectivity of putative serotoninergic neurons and GABAergic neuron

WIP is a negative regulator of neuronal maturation and synaptic activity.

Wiskott-Aldrich syndrome protein (WASP) -interacting protein (WIP) is an actin-binding protein involved in the regulation of actin polymerization in cells, such as fibroblasts and lymphocytes. Despite its recognized function in non-neuronal cells, the role of WIP in the central nervous system has not been examined previously. We used WIP-deficient mice to examine WIP function both in vivo and in vitro. We report here that WIP(-)(/-) hippocampal neurons exhibit enlargement of somas as well as overgrowth of neuritic and dendritic branches that are more evident in early developmental stages. Dend

Differential regulation of synaptic plasticity and cerebellar motor learning by the C-terminal PDZ-binding motif of GluRdelta2.

The delta2 glutamate receptor (GluRdelta2) is predominantly expressed in Purkinje cells and plays crucial roles in cerebellar functions: GluRdelta2-/- mice display ataxia and impaired motor learning. In addition, long-term depression (LTD) at parallel fiber (PF)-Purkinje cell synapses is abrogated, and synapse formation with PFs and climbing fibers (CFs) is severely disturbed in GluRdelta2-/- Purkinje cells. Recently, we demonstrated that abrogated LTD was restored in GluRdelta2-/- Purkinje cells by the virus-mediated expression of the wild-type GluRdelta2 transgene (Tg(wt)) but not by that of

Vasopressin reverses aluminum-induced impairment of synaptic plasticity in the rat dentate gyrus in vivo.

Aluminum (Al), an important neurotoxin, contributes to a variety of cognitive dysfunction and mental diseases. Previous studies have demonstrated that Al impairs hippocampal long-term potentiation (LTP) in vitro and in vivo. In the present study, both LTP and LTD (long-term depression) were recorded in the same animal to investigate the Al-induced impairment of synaptic plasticity. Another aim of the present research was to verify whether the impairment of synaptic plasticity induced by Al could be reversed by vasopressin (VP) treatment. Neonatal Wistar rats were exposed to Al from parturition

Gonadal steroids differentially modulate the actions of orphanin FQ/nociceptin at a physiologically relevant circuit controlling female sexual receptivity.

Orphanin FQ/nociceptin (OFQ/N) inhibits the activity of pro-opiomelanocortin (POMC) neurones located in the hypothalamic arcuate nucleus (ARH) that regulate female sexual behaviour and energy balance. We tested the hypothesis that gonadal steroids differentially modulate the ability of OFQ/N to inhibit these cells via presynaptic inhibition of transmitter release and postsynaptic activation of G protein-gated, inwardly-rectifying K(+) (GIRK)-1 channels. Whole-cell patch clamp recordings were performed in hypothalamic slices prepared from ovariectomised rats. OFQ/N (1 μm) decreased the frequenc

A biophysically grounded model of glutamatergic synaptic transmission integrating glutamate transport, receptor kinetics, and electrotonic effects.

Gluatamatergic synaptic transmission, critical for learning and memory, relies on precise regulation of extracellular glutamate levels by astrocytic transporters, particularly EAAT2. While existing models of AMPA and NMDA receptor kinetics often oversimplify glutamate dynamics or become computationally intractable, this study develops a balanced, biophysically grounded model that integrates glutamate transport, receptor sensitivity, and electrotonic effects. Using rat hippocampal slices, we recorded postsynaptic currents in CA1 pyramidal neurons under control conditions and during glutamate tr

Stoichiometry of the Heteromeric Nicotinic Receptors of the Renshaw Cell.

Neuronal nicotinic acetylcholine receptors (nAChRs) are pentamers built from a variety of subunits. Some are homomeric assemblies of α subunits, others heteromeric assemblies of α and β subunits which can adopt two stoichiometries (2α:3β or 3α:2β). There is evidence for the presence of heteromeric nAChRs with the two stoichiometries in the CNS, but it has not yet been possible to identify them at a given synapse. The 2α:3β receptors are highly sensitive to agonists, whereas the 3α:2β stoichiometric variants, initially described as low sensitivity receptors, are indeed activated by low and high

Activation of α1-adrenoceptors facilitates excitatory inputs to medullary airway vagal preganglionic neurons.

In mammals, the neural control of airway smooth muscle is dominated by a subset of airway vagal preganglionic neurons in the ventrolateral medulla. These neurons are physiologically modulated by adrenergic/noradrenergic projections, and weakened α₂-adrenergic inhibition of them is indicated to participate in the pathogenesis and exacerbation of asthma. This study tests whether these neurons are modulated by α₁-adrenoceptors, and if so, how. In anesthetized adult rats, microinjection of the α₁A-adrenoceptor agonist A61603 (1 pmol) unilaterally into the medullary region containing these neurons
