Sjögren's and non-Sjögren's sicca share a similar symptom burden but with a distinct symptom-associated proteomic signature.

OBJECTIVES: Given the similarity in symptoms between primary Sjogren's syndrome (SjS) and non-SjS sicca syndrome (sicca), we sought to characterise clinical and proteomic predictors of symptoms in both groups in order to better understand disease mechanisms and help guide development of immunomodulatory treatments. These have not, to date, unequivocally improved symptoms in SjS clinical trials. METHODS: Serum proteomics was performed using O-link inflammation and cardiovascular II panels. SjS (n=53) fulfilled 2016 ACR/European Alliance of Associations for Rheumatology (EULAR) criteria whereas

Comparative omics reveals conserved extracellular matrix signatures during mammary tumor progression.

The extracellular matrix (ECM) plays a critical role in tumor progression by modulating cell adhesion, migration, and signaling; however, its contribution to metastatic progression in spontaneous mammary tumors remains poorly understood. Mammary tumors are among the most common neoplasms in female dogs and share histopathological and molecular similarities with human breast cancer, supporting their use as a comparative oncology model. To investigate ECM remodeling during tumor progression, we analyzed normal, non-metastatic, and metastatic canine mammary tissues using histological approaches a

Reelin in the extracellular matrix and dendritic spines of the cortex and hippocampus: a comparison between wild type and heterozygous reeler mice by immunoelectron microscopy.

Reelin is a glycoprotein ( approximately 400 kDa) secreted by GABAergic neurons into the extracellular matrix of the neocortex and hippocampus as well as other areas of adult rodent and nonhuman primate brains. Recent findings indicate that the heterozygote reeler mouse (haploinsufficient for the reeler gene) shares several neurochemical and behavioral abnormalities with schizophrenia and bipolar disorder with mania. These include (1) a downregulation of both reelin mRNA and the translated proteins, (2) a decrease in the number of dendritic spines in cortical and hippocampal neurons, (3) a con

The Phenotypic Responses of Vascular Smooth Muscle Cells Exposed to Mechanical Cues.

During the development of atherosclerosis and other vascular diseases, vascular smooth muscle cells (SMCs) located in the intima and media of blood vessels shift from a contractile state towards other phenotypes that differ substantially from differentiated SMCs. In addition, these cells acquire new functions, such as the production of alternative extracellular matrix (ECM) proteins and signal molecules. A similar shift in cell phenotype is observed when SMCs are removed from their native environment and placed in a culture, presumably due to the absence of the physiological signals that maint

Glycoproteomic Analysis of the Aortic Extracellular Matrix in Marfan Patients.

OBJECTIVE: Marfan syndrome (MFS) is caused by mutations in FBN1 (fibrillin-1), an extracellular matrix (ECM) component, which is modified post-translationally by glycosylation. This study aimed to characterize the glycoproteome of the aortic ECM from patients with MFS and relate it to aortopathy. Approach and Results: ECM extracts of aneurysmal ascending aortic tissue from patients with and without MFS were enriched for glycopeptides. Direct N-glycopeptide analysis by mass spectrometry identified 141 glycoforms from 47 glycosites within 35 glycoproteins in the human aortic ECM. Notably, MFAP4

Decreased extracellular deposition of fibrillin and decorin in neonatal Marfan syndrome fibroblasts.

Abnormalities of the microfibrillar protein fibrillin (Fib) have been reported in Marfan syndrome (MFS). The so-called neonatal Marfan syndrome (nMFS) is a lethal phenotype displaying features that are not seen in classical MFS. We have therefore studied the biosynthesis and extracellular deposition of Fib and decorin in fibroblasts from a patient with nMFS and controls. Immunofluorescence of the patient's cell cultures showed an almost complete absence of Fib and a marked reduction of decorin in the extracellular matrix (ECM). The nMFS skin revealed Fib on subbasal microfibrillar bundles in t

Tissue and molecular events in human conjunctival scarring in ocular cicatricial pemphigoid.

Detailed histomorphometric analysis of human conjunctival biopsy specimens has convincingly demonstrated that tissue remodeling of the extracellular matrix (ECM) is an essential and dynamic process associated with conjunctival scarring in ocular cicatricial pemphigoid (OCP). The conjunctival scarring often eventually results in impaired vision and/or blindness. The molecular mechanisms of conjunctival scarring are not completely understood. Accumulating evidence indicates that the early phase of conjunctival fibrosis is linked with an immuno-inflammatory process mediated by cytokines released

Cellular communications and cell-matrix interactions in the pathogenesis of fibroproliferative diseases: liver fibrosis as a paradigm.

In vitro and in vivo studies suggest a strong cooperation between hepatocytes and mesenchymal cells and between the different non-parenchymal cell types in liver injury and fibrogenesis. The fibrogenic reaction might be described as a dynamic cascade of mechanisms beginning with hepatocyte necrosis and followed by inflammation, activation of macrophages, release of fibrogenic mediators, activation of fat-storing cells, stimulated extracellular matrix synthesis, and diminished matrix degradation resulting in a net matrix accumulation. In detail the initial events of hepatocyte injury and necros

Regulation of extracellular matrix remodeling and MMP-2 activation in cultured rat adrenal medullary endothelial cells.

We previously reported that short-term exposure of cultured rat adrenal medullary endothelial cells (RAMEC) to thrombin enhances the subendothelial deposition of extracellular matrix (ECM) proteins fibronectin, laminin, and collagen types I (C-I) and IV (C-IV) (Papadimitriou et al. 1997). In this work, we extended our previous studies on factors that effect ECM protein deposition to include agents that activate or inhibit some of the most common intracellular signals such as cAMP, protein kinase C (PKC), and calcium. Furthermore, we investigated the possible link between the observed alteratio

Basement membranes at a glance.

Basement membranes (BMs) underlie or surround most tissues. They are formed of secreted proteins that associate with cell surfaces and are the most ancient animal extracellular matrix. Laminin and collagen IV are core components that polymerize into self-associating networks, providing BMs with an organizing scaffold and tensile strength. In humans, BMs also contain over 150 other secreted proteins, such as structural matrix components, enzymes and growth factors, as well as over 50 cell-membrane adhesion and signalling receptors. From this toolbox, BMs are tailored for tissue-specific functio

Degradation of extracellular matrices propagates calcification during development and healing in bones and teeth.

BACKGROUND: Bone, dentin, and enamel are tissues formed through calcification, a process involving deposition of calcium phosphate minerals on extracellular organic matrices. Calcification, the underlying mechanism of which is unknown, is initiated with mineral deposition followed by advancing of the deposit and subsequent maturation of the mineral crystal. HIGHLIGHT: We have reviewed the current knowledge of how calcification proceeds during bone development, bone healing, and enamel and dentin development, based on reported studies. Previous studies reported by us and by other authors have

Dual-Metal Regulation of Cardiac Remodeling: Targeting the LOXL2-Ferroptosis Axis in Metal-Induced Cardiac Dysfunction.

Cardiac remodeling is driven by progressive extracellular matrix stiffening and cardiomyocyte loss. Cardiomyocyte death arising from the progression of fibrosis and ferroptosis, which are regulated by interconnected pathogenic pathways such as lysyl oxidase-like (LOXL), Yes-associated protein (YAP) transcriptional coactivator with PDZ-binding motif (TAZ), and transforming growth factor-beta, leads to cardiac remodeling. However, existing treatments are insufficient to restore heart health. There is mounting evidence that maintaining a healthy balance of metals, particularly copper and iron, ca

Methods for Defining the Role of ADAMTS1 in the Ovary.

The protease activity of ADAMTS1 is essential for ovarian development and function. It cleaves and remodels the extracellular matrix (ECM) to accommodate growing follicles and the vascular networks. While the loss of ADAMTS1 activity affects ovulation and leads to pathological conditions like polycystic ovary syndrome (PCOS), an increased activity may result in premature ovarian failure and ovarian cancers. These findings highlight the precise action of this enzyme in maintaining normal ovarian functions. This chapter describes the methods for detecting, localizing, and determining the activit

A spectrum of cysteine mutations in fibulin-3 highlight its disulfide bonding complexity and potential to induce ER stress.

Fibulin-3 (FBLN3), also known as EFEMP1, is a secreted extracellular matrix (ECM) glycoprotein that contains forty cysteine residues. These cysteines, which are distributed across one atypical and five canonical calcium-binding epidermal growth factor (EGF) domains, are important for regulating FBLN3 structure, secretion, and presumably function. As evidence of this importance, a rare homozygous p.C55R mutation in FBLN3 negates its function, alters disulfide bonding, and causes marfanoid syndrome. Additional studies suggest that heterozygous premature stop codon mutations in FBLN3 may also cau

Proteolysis: a key post-translational modification regulating proteoglycans.

Proteoglycans are composite molecules comprising a protein backbone, i.e., the core protein, with covalently attached glycosaminoglycan chains of distinct chemical types. Most proteoglycans are secreted or attached to the cell membrane. Their specialized structures, binding properties, and biophysical attributes underlie diverse biological roles, which include modulation of tissue mechanics, cell adhesion, and the sequestration and regulated release of morphogens, growth factors, and cytokines. As an irreversible post-translational modification, proteolysis has a profound impact on proteoglyca
