Porcine intestinal epithelial barrier disruption by the Fusarium mycotoxins deoxynivalenol and T-2 toxin promotes transepithelial passage of doxycycline and paromomycin.

BACKGROUND: The gastrointestinal tract is the first target for the potentially harmful effects of mycotoxins after intake of mycotoxin contaminated food or feed. With deoxynivalenol (DON), T-2 toxin (T-2), fumonisin B1 (FB1) and zearalenone (ZEA) being important Fusarium toxins in the northern hemisphere, this study aimed to investigate in vitro the toxic effect of these mycotoxins on intestinal porcine epithelial cells derived from the jejunum (IPEC-J2 cells). Viability of IPEC-J2 cells as well as the proportion of apoptotic and necrotic IPEC-J2 cells was determined by flow cytometry after 72…

Genome-wide identification of ATP-binding cassette (ABC) transporters and functional characterization of FpYOR1 in multidrug sensitivity and virulence of Fusarium pseudograminearum.

ATP-binding cassette (ABC) transporters hydrolyze ATP to drive the transmembrane transport of diverse substrates and thereby participate in a wide range of biological processes. However, the genome-wide landscape and functional contributions of ABC transporters in Fusarium pseudograminearum remain insufficiently characterized. Here, we systematically identified 61 putative ABC transporters and classified them into nine subfamily groups, with ABCB and ABCG subfamily genes representing the major expansions. We further functionally characterized an ABCC transporter gene, named FpYOR1 in this stud…

Interactive effects of temperature and water activity on growth, mycotoxin production, and transcriptomic profiles of Aspergillus flavus, Aspergillus parasiticus, and Fusarium verticillioides.

Maize is frequently contaminated with aflatoxins (AFs) and fumonisins (FBs), posing a significant global food safety threat. Understanding how environmental factors regulate fungal toxigenesis under global warming is crucial. This study systematically investigated the in vitro stress responses of Aspergillus flavus, Aspergillus parasiticus, and Fusarium verticillioides (strains isolated from contaminated maize) across broad gradients of temperature (15-40 °C) and water activity (0.93-0.995 aw). Phenotypically, primary and secondary metabolism exhibited striking divergence: A. flavus grew optim…

FgNup2 regulates nuclear import of the histone H2B monoubiquitination enzyme by stabilizing the FgImpα-FgBre1 complex to mediate pathogenicity in Fusarium graminearum.

Fusarium head blight caused by Fusarium graminearum threatens global wheat production. The pathogenicity of this fungus depends on histone H2B monoubiquitination (H2Bub1), yet the nuclear import mechanism of its key enzyme, FgBre1, remains unclear. In this study, we identified 24 nucleoporins in F. graminearum and found that FgNup2 is essential for fungal growth, toxin biosynthesis, and virulence. FgNup2 positively regulates H2Bub1 levels by facilitating the nuclear import of FgBre1, thereby influencing pathogenic development. The nuclear import of FgBre1 relies on the importin receptor FgImpα…

The Conserved Guanyl-Specific Ribonuclease T1 Effector EC19 Is Required for Full Virulence of the Vascular Wilt Fungus Fusarium oxysporum on Tomato.

Plant fungal pathogens secrete a plethora of effectors into host cells to facilitate their infection by interfering with the normal physiological processes of the host plant. However, the role of guanyl-specific ribonuclease T1 effector protein in the virulence of fungal pathogens remains largely unexplored. Here, we show that an effector protein EC19, secreted by the soil-borne fungus Fusarium oxysporum f. sp. lycopersici (Fol) that causes tomato wilt disease, contributes to the full virulence of Fol. In-locus knockout of EC19 significantly reduced the virulence of Fol, while complementing EC…

Enhanced Inhibition of Fusarium graminearum Growth and Deoxynivalenol Production by Quinolizidine Alkaloids Combined with Berberine: In Laboratory Culture Media and Pig Feed.

Phytochemicals with antifungal activity against Fusarium graminearum (F. graminearum) are considered potential in-feed anti-mycotoxigenic agents that may complement conventional post-harvest control strategies. Therefore, we screened fourteen phytochemicals, including seven quinolizidine alkaloids (QAs), for anti-F. graminearum activity, evaluated their individual and combined efficacy in inhibiting mycelial growth, spore germination, and deoxynivalenol (DON) production, and explored the underlying mechanisms via transcriptomic analysis. Matrine (MT), sophoridine (SR), and sophocarpine (SC) ex…

The Effect of Combined Exposure of Fusarium Mycotoxins on Lipid Peroxidation, Antioxidant Defense, Fatty Acid Profile, and Histopathology in Laying Hens' Liver.

Fumonisin B1, T-2 toxin, and deoxynivalenol are frequently detected in feed materials. The mycotoxins induce free radical formation and, thereby, lipid peroxidation. The effects of mycotoxin exposure at the EU recommended limit (T-2/HT-2 toxin: 0.25 mg/kg; DON = 3AcDON/15-AScDON: 5 mg/kg; fumonisin B1: 20 mg/kg) and double dose (T-2/HT-2 toxin: 0.5 mg/kg, DON/3-AcDON/15-AcDON: 10 mg, and FB1: 40 mg/kg feed) were investigated during short-term (3 days) per os exposure in the liver of laying hens. On day 1 higher while on day 3 lower MDA concentrations were found in the low-dose group compared t

Effects of Supplementation of a Mycotoxin Mitigation Feed Additive in Lactating Dairy Cows Fed Fusarium Mycotoxin-Contaminated Diet for an Extended Period.

Fusarium mycotoxins are inactivated by rumen flora; however, a certain amount can pass the rumen and reticulum or be converted into biological active metabolites. Limited scientific evidence is available on the impact and mitigation of Fusarium mycotoxins on dairy cows' performance and health, particularly when cows are exposed for an extended period (more than 2 months). The available information related to these mycotoxin effects on milk cheese-making parameters is also very poor. The objective of this study was to evaluate a commercially available mycotoxin mitigation product (MMP, i.e., TO…

Cyclic Lipopeptides of Bacillus amyloliquefaciens DHA6 Are the Determinants to Suppress Watermelon Fusarium Wilt by Direct Antifungal Activity and Host Defense Modulation.

Fusarium wilt, caused by Fusarium oxysporum f. sp. niveum (Fon), poses a serious threat to watermelon productivity. We previously characterized six antagonistic bacterial strains, including DHA6, capable of suppressing watermelon Fusarium wilt under greenhouse conditions. This study investigates the role of extracellular cyclic lipopeptides (CLPs) produced by strain DHA6 in Fusarium wilt suppression. Taxonomic analysis based on the 16S rRNA gene sequence categorized strain DHA6 as Bacillus amyloliquefaciens. MALDI-TOF mass spectrometry identified five families of CLPs, i.e., iturin, surfactin,…

First Report of the Occurrence of Fusarium oxysporum f. sp. niveum Race 2 in Commercial Watermelon Production Areas of Maryland and Delaware.

Resistance to race 1 of Fusarium oxysporum f. sp. niveum (E.F. Sm.) W.C. Snyder & H.N. Hans) is the most widely used tool for management of Fusarium wilt of watermelon (Citrullus lanatus (Thunb.) Matsum. and Nakai). However, this resistance is ineffective against the more aggressive F. oxysporum f. sp. niveum race 2. Race 2 was first identified in Israel in 1973 (2) and was subsequently reported in Texas (1981), Oklahoma (1988), and Florida (1989) (1). From July through September 2000 in Maryland and Delaware, 123 isolates of F. oxysporum f. sp. niveum were collected from wilted plants or plan…

Interaction between Trichoderma harzianum qt40003 and Bacillus velezensis BV25 Triggers Harzianopyridone Biosynthesis, Enhancing Biological Control Efficacy against Fusarium oxysporum f. sp. Conglutinans.

Cabbage Fusarium wilt, caused by Fusarium oxysporum f. sp. conglutinans (FOC), is a destructive soil-borne disease. Herein, we deciphered how coculture of the bacterium Bacillus velezensis BV25 with the fungus Trichoderma harzianum qt40003 activates expression of the harzianopyridone (HPY) biosynthetic gene cluster, thereby enhancing biocontrol against FOC. Greenhouse assays showed the coculture significantly enhanced biocontrol efficacy compared with single-strain treatments, reducing disease index from 88 to 26. Comparative transcriptomic analysis revealed that coculture specifically trigger…

Genetic mapping identifies QTL and candidate genes including Mendel's A gene controlling high levels of partial resistance to Fusarium avenaceum in Pea.

Fav-Ps2.1 is the first major QTL reported for Fusarium avenaceum resistance in pea and co-localizes with Mendel's A gene and putative resistance genes. A novel moderate-effect QTL Fav-Ps5.1 and one epistatic locus, independent of the A gene, were also reported. Flavonoid compounds associated with resistance to this pathogen were identified. Fusarium avenaceum, a necrotrophic soil-borne pathogen, is a member of the root rot pathogen complex posing a major threat to pea yield and quality worldwide. Strong resistance to Fusarium root rot (FRR) in pea is associated with seed coat and flower pigmen…

Sensitivity of Meloidogyne incognita, Fusarium oxysporum f. sp. niveum, and Stagonosporopsis citrulli to Succinate Dehydrogenase Inhibitors Used for Control of Watermelon Diseases.

Watermelon is affected by diseases such as Fusarium wilt, gummy stem blight, and root-knot nematode (RKN). Succinate dehydrogenase inhibitors (SDHIs) with potential fungicide and nematicide activity provide the opportunity to control multiple diseases with one compound. In this study, we aimed to determine the sensitivity of Meloidogyne incognita race 4 (MI4), Fusarium oxysporum f. sp. niveum (FON), and Stagonosporopsis citrulli (SCIT) to existing SDHIs: benzovindiflupyr, fluopyram, cyclobutrifluram, and pydiflumetofen. All SDHIs had fungicidal activity against 19 SCIT isolates in mycelial gro…

Fusarium Mycotoxins Zearalenone and Deoxynivalenol Reduce Hepatocyte Innate Immune Response after the Listeria monocytogenes Infection by Inhibiting the TLR2/NFκB Signaling Pathway.

Zearalenone (ZEA) and deoxynivalenol (DON) are two common mycotoxins produced by the genus Fusarium and have potential immunotoxic effects that may lead to a weak immune response against bacterial infections. Listeria monocytogenes (L. monocytogenes), a food-borne pathogenic microorganism ubiquitous in the environment, actively multiplies in the liver, where hepatocytes are capable of resistance through mediated innate immune responses. At present, it is not clear if ZEA and DON affect hepatocyte immune responses to L. monocytogenes infection or the mechanisms involved. Therefore, in this stud…

A plant biostimulant made from the marine brown algae Ascophyllum nodosum and chitosan reduce Fusarium head blight and mycotoxin contamination in wheat.

Fusarium head blight (FHB) caused by Fusarium graminearum is a disease that results in yield loss and mycotoxin contamination in wheat globally. This study assessed the effect of a plant biostimulant prepared from a brown macroalga Ascophyllum nodosum (Liquid Seaweed Extract; LSE) alone and in combination with chitosan in controlling Fusarium. Wheat seedlings drenched with LSE and chitosan in combination showed reduced severity of F. graminearum infection on leaves as evidenced by a significant reduction in necrotic area and fewer number of conidia produced in the necrotic area. Gene expressio…

Differential gene expression to heat stress drives thermotolerance, pathogenicity, and reproduction in Fusarium head blight pathogens: from transcriptomic profiling to functional validation.

BACKGROUND: Fusarium head blight (FHB) is a destructive wheat disease caused primarily by Fusarium graminearum and F. asiaticum. Climate warming increases the frequency of high-temperature episodes during anthesis, yet the molecular mechanisms underlying thermal adaptation in FHB pathogens remain unclear. Here, we integrated ecological, transcriptomic, and functional approaches to investigate how differential gene expression contributes to heat adaptation and its interplay with virulence and reproduction. RESULTS: Field surveys in Sichuan Province identified naturally occurring strains capabl…

Unleashing antifungal power: Nanopriming with selenium boosts JA signaling, suppresses pathogen enzymes, and activates phytoalexins for enhanced Fusarium graminearum resistance in maize.

INTRODUCTION: Stalk rot caused by Fusarium graminearum (F. graminearum) poses a serious threat to maize production, further exacerbated by contamination with the mycotoxin deoxyninylenol (DON). Although selenium nanoparticle (SeNP) priming technology shows potential for enhancing crop resistance, the systematic regulatory mechanisms by which it reprograms plant defense responses against this pathogen remain unclear. OBJECTIVES: This study aimed to elucidate the molecular mechanisms through which SeNP priming regulates transcriptional and metabolic reprogramming in maize seedlings to resist F.…
