Influence of coexisting spiramycin contaminant on the harm of Microcystis aeruginosa at different nitrogen levels.

The influence of nitrogen on the effects of the antibiotic contaminant spiramycin on Microcystis aeruginosa was studied through a 7-day exposure test. At current contamination levels of 0.5-100 mg L(-1) for nitrogen and 0.1-0.4 μg L(-1) for spiramycin, the two factors significantly interacted with each other (p<0.05) on the synthesis of chlorophyll-a and protein, the production and release of microcystins as well as the expression of ntcA gene and mcyB gene in M. aeruginosa. Nitrogen significantly affected the toxicity of spiramycin on algal growth (p<0.05) via regulation of protein synthesis.

The effect of iron availability on transcription of the Neisseria meningitidis fHbp gene varies among clonal complexes.

Factor H binding protein (fHbp) is a major antigenic component of novel vaccines designed to protect against meningococcal disease. Prediction of the potential coverage of these vaccines is difficult, as fHbp is antigenically variable and levels of expression differ among isolates. Transcriptional regulation of the fHbp gene is poorly understood, although evidence suggests that oxygen availability is involved. In this study iron accessibility was found to affect fHbp transcription. However, regulation differed among meningococcal clonal complexes (ccs). For the majority of isolates, increased

Global transcriptome analysis reveals small RNAs affecting Neisseria meningitidis bacteremia.

Most bacterial small RNAs (sRNAs) are post-transcriptional regulators involved in adaptive responses, controlling gene expression by modulating translation or stability of their target mRNAs often in concert with the RNA chaperone Hfq. Neisseria meningitides, the leading cause of bacterial meningitis, is able to adapt to different host niches during human infection. However, only a few sRNAs and their functions have been fully described to date. Recently, transcriptional expression profiling of N. meningitides in human blood ex vivo revealed 91 differentially expressed putative sRNAs. Here we

Sporulation kinetics of Bacillus subtilis under dynamic temperature conditions: impact on gene expression and spore properties.

Bacillus subtilis forms highly resistant spores that contribute to food spoilage but are also valuable for biotechnological applications. While temperature strongly affects sporulation and spore properties, industrial temperature profiles are often dynamic and remain poorly accounted for in predictive models. In this study, growth and sporulation were monitored under four temperature scenarios. A static 37.0 °C condition was compared with three dynamic profiles: after 16 h at 37.0 °C, cultures were cooled to 10.0 °C, cooled to 10.0 °C for 24 h, then returned to 37.0 °C, or cooled to 20.0 °C. The phosphate specific transport system protein PhoU modulates persister cell formation of Klebsiella pneumoniae.

The indiscriminate use of antibiotics has exacerbated the clinical challenge posed by Klebsiella pneumoniae, with persisters often evading antibiotic treatment. To investigate the mechanisms underlying persister formation, we identified the phosphate transporter gene phoU as differentially expressed during persister emergence and recovery. Using CRISPR-Cas9 gene editing, we created a ∆phoU knockout strain in K. pneumoniae ATCC 700603, along with its complemented (CphoU) and various overexpression strains (OE phoU, OE metE, and OE phoA). While growth rates and antibiotic susceptibility remained

(P)ppGpp synthase Rsh promotes persister cells formation by regulating sulfur metabolism mediated by sulfate reductase CysI in Brucella abortus.

Persister cells constitute a subpopulation of dormant cells that are transiently tolerant to antibiotics and associated with chronic infection. We have known (p)ppGpp synthetase Rsh promoted persister cells formation during rifampicin exposure in Brucella, but the metabolic regulation mechanism by which Rsh promotes persister cells formation is unknown. In this study, we firstly characterized the proteome and metabolome of persister cells formation during rifampicin exposure between B. abortus A19 (WT) and Δrsh strains. We found that sulfur metabolism-related protein (NAD(P)/FAD-dependent sulf

MclX modulates the process of mother cell lysis by SpoIIID in Bacillus thuringiensis.

UNLABELLED: Cell differentiation is essential for the survival and adaptation of organisms. In Bacillus species, sporulation is a tightly coordinated program that culminates in mother cell lysis, a critical step for spore release. Despite its importance, the mechanisms modulating the process remain poorly understood. Here, we show that MclX regulates mother cell lysis by inhibiting SpoIIID, a sporulation-specific transcription factor that represses cwlC, which encodes a key hydrolase required for lysis. SpoIIID directly binds to the cwlC promoter, and its overexpression during sporulation mark

Biological and clinical significance of iron in Pseudomonas aeruginosa biofilms.

Pseudomonas aeruginosa is a ubiquitous gram-negative bacterium that causes diverse opportunistic infections in vulnerable populations. P. aeruginosa is innately resistant to many therapeutic agents, and biofilm-mediated infections demonstrate enhanced recalcitrance to antimicrobials. It has been known for over 25 years that iron plays a critical role in P. aeruginosa biofilm physiology, and numerous studies have revealed mechanistic insights into how this metallonutrient affects the regulatory, structural, and nutritional requirements of biofilm communities. This mini-review presents the curre

Deciphering strain differences in codY regulation of Clostridioides difficiles sporulation.

Clostridioides difficile is an anaerobic, spore-forming pathogen that causes diarrhea, colitis, and even death. C. difficile grows and replicates in the intestine as a vegetative bacillus, but must transition into a dormant spore to survive and transmit in the environment. The transformation into a spore is a complex developmental process that is regulated in response to conditions within the host, most notably nutrient limitation. Nutrient availability is sensed by C. difficile through transcriptional regulators, such as CodY. CodY is a global nutritional gene regulator that controls gene exp

Adaptive Mechanisms of Mycobacterium tuberculosis: Role of fbiC Mutations in Dormancy and Survival.

This review examines the impact of F420 biosynthesis protein C (fbiC) mutations in Mycobacterium tuberculosis (Mtb) and their influence on the bacterium's dormancy mechanisms. The potential role of fbiC mutations and functional impairments in the persistence of Mtb is emphasized. Tuberculosis (TB) bacilli can enter a dormant state with minimal metabolic activity, allowing them to conserve resources and survive in low-nutrient, low-oxygen environments for extended periods. While the fbiC gene contributes to dormancy, Mtb can achieve this state through multiple genetic and metabolic pathways, su

Overlapping and Distinct Functions of the Paralogous PagR Regulators of Bacillus anthracis.

The Bacillus anthracis pagA gene, encoding the protective antigen component of anthrax toxin, is part of a bicistronic operon on pXO1 that codes for its own repressor, PagR1. In addition to the pagAR1 operon, PagR1 regulates sap and eag, two chromosome genes encoding components of the surface layer, a mounting structure for surface proteins involved in virulence. Genomic studies have revealed a PagR1 paralog, PagR2, encoded by a gene on pXO2. The amino acid sequences of the paralogues are 71% identical and show similarity to the ArsR family of transcription regulators. We determined that the e

Human Bile-Mediated Regulation of Salmonella Curli Fimbriae.

Typhoid fever is caused primarily by Salmonella enterica serovar Typhi. Approximately 3% to 5% of individuals infected with S Typhi become chronic carriers with the gallbladder (GB) as the site of persistence, as gallstones within the GB are a platform on which the bacteria form a biofilm. S Typhi is a human-restricted pathogen; therefore, asymptomatic carriers represent a critical reservoir for further spread of disease. To examine the dynamics of the Salmonella biofilm during chronic carriage, the human gallstone (GS) environment was simulated by growing biofilms on cholesterol-coated surfac

Unraveling neutrophil- Yersinia interactions during tissue infection.

The human and animal pathogens Yersinia pestis, which causes bubonic and pneumonic plague, and Yersinia pseudotuberculosis and Yersinia enterocolitica, which cause gastroenteritis, share a type 3 secretion system which injects effector proteins, Yops, into host cells. This system is critical for virulence of all three pathogens in tissue infection. Neutrophils are rapidly recruited to infected sites and all three pathogens frequently interact with and inject Yops into these cells during tissue infection. Host receptors, serum factors, and bacterial adhesins appear to collaborate to promote neu

Attenuation of Pseudomonas aeruginosa biofilm by hordenine: a combinatorial study with aminoglycoside antibiotics.

Pseudomonas aeruginosa is a ubiquitous pathogen that is the leading cause of chronic infections. Bacterial biofilm formation facilitates CF development and restricts the anti-bacterial potential of many current antibiotics. The capacity of P. aeruginosa to form biofilms and resist antibiotics is closely correlated with quorum sensing (QS). Disrupting QS by QS inhibitors is a promising strategy for treating chronic infections. Here, we evaluated the effect of hordenine, a recently characterized QS inhibitor, on the susceptibility of aminoglycoside antibiotics against P. aeruginosa biofilms. Hor

Phase variation of Opa proteins of Neisseria meningitidis and the effects of bacterial transformation.

Opa proteins are major proteins involved in meningococcal colonization of the nasopharynx and immune interactions. Opa proteins undergo phase variation (PV) due to the presence of the 5'-CTCTT-3' coding repeat (CR) sequence. The dynamics of PV of meningococcal Opa proteins is unknown. Opa PV, including the effect of transformation on PV, was assessed using a panel of Opa-deficient strains of Neisseria meningitidis. Analysis of Opa expression from UK disease-causing isolates was undertaken. Different opa genes demonstrated variable rates of PV, between 6.4 × 10(-4) and 6.9 × 10(-3) per cell per

Detection and identification of Rickettsia species in Ixodes tick populations from Estonia.

A total of 1640 ticks collected in different geographical parts of Estonia were screened for the presence of Rickettsia species DNA by real-time PCR. DNA of Rickettsia was detected in 83 out of 1640 questing ticks with an overall prevalence of 5.1%. The majority of the ticks infected by rickettsiae were Ixodes ricinus (74 of 83), while 9 of the 83 positive ticks were Ixodes persulcatus. For rickettsial species identification, a part of the citrate synthase gltA gene was sequenced. The majority of the positive samples were identified as Rickettsia helvetica (81 out of 83) and two of the samples

OspC is potent plasminogen receptor on surface of Borrelia burgdorferi.

Host-derived proteases are crucial for the successful infection of vertebrates by several pathogens, including the Lyme disease spirochete bacterium, Borrelia burgdorferi. B. burgdorferi must traverse tissue barriers in the tick vector during transmission to the host and during dissemination within the host, and it must disrupt immune challenges to successfully complete its infectious cycle. It has been proposed that B. burgdorferi can accomplish these tasks without an endogenous extra-cytoplasmic protease by commandeering plasminogen, the highly abundant precursor of the vertebrate protease p

Staphylococccus pseudintermedius surface proteins SpsD and SpsO mediate adherence to ex vivo canine corneocytes.

The Gram-positive bacterium Staphylococcus pseudintermedius is regarded as the major cause of canine bacterial pyoderma. Despite its clinical importance, there is only very limited knowledge about the pathogenesis of S. pseudintermedius infection and the specific bacterial virulence factors involved in causing disease. Using a whole-genome approach, we have previously identified 18 predicted cell-wall-anchored surface proteins representing possible virulence factors in a clinical isolate of S. pseudintermedius (strain ED99). They were designated S. pseudintermedius surface proteins A-R (SpsA-S

Porin-mediated antibiotic resistance in Neisseria gonorrhoeae: ion, solute, and antibiotic permeation through PIB proteins with penB mutations.

Neisseria gonorrhoeae has two porins, PIA and PIB, whose genes (porA and porB, respectively) are alleles of a single por locus. We recently demonstrated that penB mutations at positions 120 and 121 in PIB, which are presumed to reside in loop 3 that forms the pore constriction zone, confer intermediate-level resistance to penicillin and tetracycline (M. Olesky, M. Hobbs, and R. A. Nicholas, Antimicrob. Agents Chemother. 46:2811-2820, 2002). In the present study, we investigated the electrophysiological properties as well as solute and antibiotic permeation rates of recombinant PIB proteins con

Profiling of BDQ-induced transcriptome suggests amino acid metabolism and stress responses as alternate mechanisms contributing to BDQ tolerance in Mycobacterium tuberculosis.

UNLABELLED: Under bedaquiline (BDQ) pressure, a temporary persistence period (24-96 h) has been observed, during which H37Rv undergoes metabolic rerouting. However, little is known of transcriptomic changes in BDQ-resistant Mycobacterium tuberculosis (Mtb) isolates during this period. We explored transcriptomic adaptations occurring under inhibitory concentrations of BDQ to delineate pathways supporting drug tolerance and contributing to BDQ resistance. We report overexpression of genes involved in the biosynthesis of L-arginine and L-cysteine in our study isolates. Among stress response genes
