Black shank disease, caused by Phytophthora parasitica var. Nicotianae (P. nicotianae), severely affects the quality and yield of Nicotiana plants. Although resistance genes from some wild Nicotiana plant species, such as Nicotiana plumbaginifolia, were transferred to cultivated Nicotiana plant long ago, the underlying molecular mechanisms remain unclear. In previous work, NpSKP1-1A was found to interact with NpPP2-B10, which positively regulates Nicotiana plants resistance to P. nicotianae. In this study, we investigated the roles of NpSKP1-1A in resistance to P. nicotianae in Nicotiana plant

Iron (Fe), zinc (Zn), manganese (Mn), and copper (Cu) are essential micronutrients for plant growth and development, but their low bioavailability in soil limits crop growth and yield. The ZIP (ZRT/IRT-like protein) family plays critical roles in the uptake and distribution of these divalent metal ions. Here, we performed a genome-wide identification of the ZIP family in tomato (Solanum lycopersicum) and analyzed their expression profiles under various metal deficiencies. A total of 14 SlZIP genes were identified and classified into three clades based on phylogenetic analysis. Conserved motif

Plants sense a diverse array of small molecules and macromolecules derived from their natural environment, including diverse microbe-associated molecular patterns (MAMPs) that can trigger defense responses. Several MAMPs have been shown to prime plants for enhanced defense, providing extended protection against pathogens with minimal fitness costs. However, the extent to which conserved small molecule signatures of other phyla contribute to priming of plant defenses is unclear. Here, we demonstrate that exposure of seeds or plants to the ascaroside ascr#18, a pheromone secreted by plant-parasi

Previous studies have suggested that Dense and Erect Panicle 1 (DEP1) interacts with Lose Plant Architecture 1 (LPA1) to regulate auxin transport by which DEP1-LPA1 modulates rice sheath blight (ShB) resistance. In this study, we identified that dep1 and lpa1 exhibited semi-dwarfism and dep1/lpa1 was shorter than the single mutant. LPA1 OX displayed higher height, whereas DEP1 OX exhibited similar height with wild-type. The gibberellic acid (GA)-dependent shoot growth was inhibited in dep1 and lpa1 while activated in LPA1 OX, suggesting that LPA1 may play a role in GA signaling transduction. Y

Melatonin (MT) plays a critical role in regulating rice responses to heavy metal stress. However, the mechanism by which MT alleviates copper (Cu) toxicity in rice remains insufficiently understood. In this study, excessive Cu increased endogenous MT level in rice roots, accompanied by enhanced gene expression involved in MT biosynthesis, suggesting its role in Cu stress responses. Exogenous MT reduced Cu accumulation and restored root growth in rice under Cu stress, thereby mitigating Cu-induced phytotoxicity. Mechanistically, MT increased the root pectin content and its Cu retention, thereby

Selenium-binding proteins (SBPs) represent a ubiquitous and conserved protein family with yet unclear biochemical and molecular functions. The importance of the human homolog has been extensively studied as it is implicated in many cancer types and other diseases. On the other hand, little is known regarding plant homologs. In plants, there is evidence that SBP participates in developmental procedures, oxidative stress responses, selenium and cadmium binding, and pathogenic tolerance. Moreover, recent studies have revealed that SBP is a methanethiol oxidase (MTO) catalyzing the conversion of m

The conquest of land by plants was concomitant with, and possibly enabled by, the evolution of three-dimensional (3D) growth. The moss Physcomitrium patens provides a model system for elucidating molecular mechanisms in the initiation of 3D growth. Here, we investigate whether the phytohormone ethylene, which is believed to have been a signal before land plant emergence, plays a role in 3D growth regulation in P. patens. We report ethylene controls 3D gametophore formation, based on results from exogenously applied ethylene and genetic manipulation of PpEIN2, which is a central component in th

Plant activators have emerged as promising alternatives to conventional crop protection chemicals for managing crop diseases due to their unique mode of action. By priming the plant's innate immune system, these compounds can induce disease resistance against a broad spectrum of pathogens without directly inhibiting their proliferation. Key advantages of plant activators include prolonged defense activity, lower effective dosages, and negligible risk of pathogen resistance development. Among the various defensive pathways targeted, the salicylic acid (SA) signaling cascade has been extensively

Plant-specific lateral organ boundaries domain (LBD) proteins play important roles in plant growth and development. Foxtail millet (Setaria italica) is one new C4 model crop. However, the functions of foxtail millet LBD genes are unknown. In this study, a genome-wide identification of foxtail millet LBD genes and a systematical analysis were conducted. A total of 33 SiLBD genes were identified. They are unevenly distributed on nine chromosomes. Among these SiLBD genes, six segmental duplication pairs were detected. The thirty-three encoded SiLBD proteins could be classified into two classes an

Volatile compounds (VCs) of Trichoderma fungi trigger induced systemic resistance (ISR) in Arabidopsis that is effective against a broad spectrum of pathogens. The root-specific transcription factor MYB72 is an early regulator of ISR and also controls the activation of iron-deficiency responses. Nitric oxide (NO) is involved in the regulation of MYB72-dependent iron-deficiency responses in Arabidopsis roots, but the role of NO in the regulation of MYB72 and ISR by Trichoderma VCs remains unexplored. Using in vitro bioassays, we applied Trichoderma VCs to Arabidopsis seedlings. Plant perception

TaD11-2A affects grain size and root length and its natural variations are associated with significant differences in yield-related traits in wheat. Brassinosteroids (BRs) control many important agronomic traits and therefore the manipulation of BR components could improve crop productivity and performance. However, the potential effects of BR-related genes on yield-related traits and stress tolerance in wheat (Triticum aestivum L.) remain poorly understood. Here, we identified TaD11 genes in wheat (rice D11 orthologs) that encoded enzymes involved in BR biosynthesis. TaD11 genes were highly e

Male and female unisexual flowers evolved from hermaphroditic ancestors, and control of flower sex is useful for plant breeding. We isolated a female-to-male sex transition mutant in melon and identified the causal gene as the carpel identity gene <i>CRABS CLAW (CRC)</i>. We show that the master regulator of sex determination in cucurbits, the transcription factor <i>WIP1</i> whose expression orchestrates male flower development, recruits the corepressor TOPLESS to the <i>CRC</i> promoter to suppress its expression through histone deacetylation. BACKGROUND: Plant mitochondrial transcription termination factor (mTERF) family members play important roles in development and stress tolerance through regulation of organellar gene expression. However, their molecular functions have yet to be clearly defined.
RESULTS: Here an mTERF gene V14 was identified by fine mapping using a conditional albino mutant v14 that displayed albinism only in the first two true leaves, which was confirmed by transgenic complementation tests. Subcellular localization and real-time PCR analyses indicated that V14 encodes a chloroplastic protein ubiquitously expr

Thermosensitive cytoplasmic male sterile (TCMS) lines play an important role in wheat breeding, heterosis utilization, and germplasm innovation. MicroRNAs (miRNAs) can regulate the expression level of target genes by inhibiting the translation of these genes. YS3038 is a wheat TCMS line. In this study, the fertility conversion mechanism of YS3038 was studied by examining the abortion characteristics of YS3038, the regulation pattern of miRNAs and the target genes of miRNAs in YS3038. MiRNA-seq was performed on three important stages of YS3038 under sterile and fertile conditions. Then, the cle

Stomatal development is regulated by signaling pathways that function in multiple cellular programs, including cell fate and cell division. However, recent studies suggest that molecular signals are affected by CO2 concentration, light intensity, and water pressure deficit, thereby modifying distribution patterns and stomatic density and likely other foliar features as well. Here, we show that in addition to lacking chloroplasts, the albino somaclonal variants of Agave angustifolia Haw present an irregular epidermal development and morphological abnormalities of the stomatal complex, affecting

The copper chaperone ATX1 has been investigated previously in the herbaceous plants Arabidopsis and rice. However, the molecular mechanisms of ATX1 underlying copper transport and functional characteristics in the woody plant Populus are poorly understood. In this study, PnATX1 and PnATX2 of Populus simonii × P. nigra were identified and characterized. Sequence analysis showed that PnATXs contained the metal-binding motif MXCXXC in the N-terminus and a lysine-rich region. Phylogenetic analysis of ATX protein sequences revealed that PnATXs were clustered in the same group as AtATX1. PnATX prote

CG methylation (m CG) is essential for preserving genome stability in mammals, but this link remains obscure in plants. OsMET1-2, a major rice DNA methyltransferase, plays critical roles in maintaining m CG in rice. Null mutation of OsMET1-2 causes massive CG hypomethylation, rendering the mutant suitable to address the role of m CG in maintaining genome integrity in plants. Here, we analyzed m CG dynamics and genome stability in tissue cultures of OsMET1-2 homozygous (-/-) and heterozygous (+/-) mutants, and isogenic wild-type (WT). We found m CG levels in cultures of -/- were substantially l

Fatty acid desaturase (FAD) is a core catalytic enzyme in plants for the synthesis of unsaturated fatty acids, profoundly affecting plant growth, development, and adaptability to various environmental stresses. The walnut (Juglans regia L.) is an important woody oil tree species, and its kernel is rich in unsaturated fatty acids. Systematic identification of the walnut FAD gene family and analysis of its function are of great significance for revealing the molecular mechanisms underlying unsaturated fatty acid metabolism in the walnut. Based on walnut whole-genome data, this study used homolog

The toxic metal cadmium (Cd) constitutes one of the major inorganic contaminants in environments. microRNAs (miRNAs) are a class of endogenous non-coding small RNAs. miR395 is conserved and regulates sulfate assimilation and distribution in higher plants, but whether it is involved in detoxification of Cd in plants has not been described. In this study, transgenic rapeseed (Brassica napus) over-expressing miR395 was identified under Cd stress. miR395-over-expressing plants showed a lower degree of Cd-induced oxidative stress than wild type. By contrast, chlorophyll, glutathione and non-protein
