The intracellular pathogen Legionella pneumophila translocates >300 effector proteins into host cells, many of which are regulated at the transcriptional level. Here, we describe a novel L. pneumophila genomic island, which undergoes horizontal gene transfer within the Legionella genus. This island encodes two Icm/Dot effectors: LegK3 and a previously uncharacterized effector which we named CegK3, as well as a LuxR type regulator, which we named RegK3. Analysis of this island in different Legionella species revealed a conserved regulatory element located upstream to the effector-encoding genes

Gene therapy, the transfer of genetic material for therapeutic purposes, has undergone an explosive development in the last few years. Within this context, development of gene therapy approaches for the neuroendocrine system, while incipient, has already generated a core of results which emerge as a promising area of research in neuroendocrinology. The present review presents a brief description of the viral vector-based gene delivery systems being currently used in neuroendocrinology, namely the adenoviral and herpes simplex type-1 (HSV-1)-derived vector systems, as well as an updated account

A genomic library from one clinical isolate of Acinetobacter baumannii was obtained to find genes responsible for tetracycline resistance. Escherichia coli DH5alpha-MDR transformants, selected on Mueller-Hinton agar supplemented with tetracycline 18 mg/L, were thoroughly characterized. In one clone, with an insert of 7,070 bp, it was found that the resistance to tetracycline was mediated by the tet(A) gene (1,200 bp) which encodes a tetracycline efflux pump. This gene was recovered together with tetR(A) (651 bp), the tet(A) repressor. Moreover, the partial sequence (2,008 bp) of a transposase

BACKGROUND: Streptococcus pneumoniae is an important human pathogen representing a major cause of morbidity and mortality worldwide. We sequenced the genome of a serotype 11A, ST62 S. pneumoniae invasive isolate (AP200), that was erythromycin-resistant due to the presence of the erm(TR) determinant, and carried out analysis of the genome organization and comparison with other pneumococcal genomes.
RESULTS: The genome sequence of S. pneumoniae AP200 is 2,130,580 base pair in length. The genome carries 2216 coding sequences (CDS), 56 tRNA, and 12 rRNA genes. Of the CDSs, 72.9% have a predicted

Wastewater treatment plants (WWTPs) are an understudied source of mercury methylating and demethylating microbes to downstream aquatic and terrestrial environments, where methylmercury production and subsequent bioaccumulation in the food web occur. To identify methylators and demethylators and evaluate their occurrence in WWTPs, metagenomic and metatranscriptomic analyses of raw sewage, activated sludge, and effluent samples from WWTPs across the globe were conducted. Results indicated that hgcA- and merB-carriers were widespread in WWTPs, with higher abundance in raw sewage and sludge compar

Horizontal gene transfer (HGT) is a driving force in microbial evolution that allows community members to rapidly evolve to cope with environmental stressors and competition. Despite the importance of HGT for the generation of genetic diversity, little is known about the specific mechanisms or dynamics of transfer in complex communities. Transductomics is a sequencing based technique which identifies potential HGT by bacteriophages (transduction) through sequencing of the transductome - the DNA carried by bacteriophages and other viruslike particles in a sample. We analyzed the murine gut tran

Oxidative stress induced by heavy metals and antibiotics in municipal sludge inhibits the gut development of black soldier fly larvae (BSFL), compromising pollutant-processing capacity of their gut microbiota. Herein, Deinococcus radiodurans was colonized into the BSFL gut and established an antioxidant cascade regulatory system in concert with the gut microbiota, synergistically reducing the enrichment of antibiotic resistance genes (ARGs) in municipal sludge. Results showed that bioaugmentation with Deinococcus radiodurans scavenges reactive oxygen species and upregulates the expression of G

Vibrio cholerae, the causative agent of cholera, is a model organism for studying virulence regulation, biofilm formation, horizontal gene transfer, and the cell-to-cell communication known as quorum sensing (QS). As in any research field, discrepancies between data from diverse laboratories are sometimes observed for V. cholerae. Such discrepancies are often caused by the use of diverse patient or environmental isolates. In this study, we investigated the inability of a few laboratories to reproduce high levels of natural transformation, a mode of horizontal gene transfer that is specifically

AbstractThe worldwide expansion of carbapenemase-producing hypervirulent K. pneumoniae (CP-hvKP) raises serious concerns about potentially untreatable invasive infections. ST23, an archetypal hvKP clone, has been highlighted due to the rising prevalence of carbapenem resistance largely driven by horizontal plasmid transfer. However, the factors facilitating carbapenemase acquisition in this lineage remained unclear. We characterized 1159 ST23 isolates across 49 countries, including nine clinical carbapenemase-producing strains from China's national bloodstream infection surveillance network. G

The ICEKp258.2 genomic island (GI) has been proposed as an important factor for the emergence and success of the globally spread carbapenem-resistant Klebsiella pneumoniae sequence type (ST) 258. However, a characterization of this horizontally acquired element is lacking. Using bioinformatic and experimental approaches, we found that ICEKp258.2 is not confined to ST258 and ST512, but also carried by ST3795 strains and emergent invasive multidrug-resistant pathogens from ST1519. We also identified several ICEKp258.2-like GIs spread among different K. pneumoniae STs, other Klebsiella species an

Edwardsiella piscicida is a significant pathogen that poses a particular threat to Japanese flounder (Paralichthys olivaceus) aquaculture in Japan and other countries. The damage is caused by the pathogen's ability to evade host immune defences and establish intracellular infections, intensified by its genomic plasticity and capacity for horizontal gene transfer. To investigate evolutionary adaptations between one older (2019) and four recent (2023) E. piscicida strains from the same geographical locations, we performed comparative genomic analysis of five isolates using high-quality hybrid ge

Infection with multiple drug resistant (MDR) Escherichia coli poses a life threat to immunocompromised pediatric cancer patients. Our aim is to genotypically characterize the plasmids harbored in MDR E. coli isolates recovered from bacteremic patients of Children's Cancer Hospital in Egypt 57357 (CCHE 57357). In this study, 21 carbapenem-resistant E. coli (CRE) isolates were selected that exhibit Quinolones and Aminoglycosides resistance. Plasmid shot-gun sequencing was performed using Illumina next- generation sequencing platform. Isolates demonstrated resistant to all beta-lactams, carbapene

A plasmid-encoded ClpK protein was recently identified as a predictor of a heat-resistant phenotype in the opportunistic pathogen Klebsiella pneumoniae. This study was undertaken to evaluate the presence of the clpK gene in extended-spectrum β-lactamase (ESBL)-producing K. pneumoniae and to assess the probable co-transfer of multi-resistance with the heat resistance phenotype. A Danish collection of 80 ESBL-producing K. pneumoniae bloodstream infection isolates was screened for clpK by colony hybridization. Nineteen isolates (24%) were positive for clpK; some of them representing major clones

The spread of carbapenem-resistant Klebsiella pneumoniae has become a significant problem for public health in recent years. In this study, we reported a New Delhi metallo-β-lactamase 1 (NDM-1)-producing K. pneumoniae strain KP14003 from a neonate in Beijing, China. Whole-genome sequencing was performed. The strain belonged to sequence type ST719. Coexistence of blaNDM-1 and blaSHV-12 was found on a self-transferable plasmid, which had a typical IncX3 backbone. The horizontal transfer of blaNDM-1 was associated with Tn125 followed by possible transposition events. Other class A extended-spectr

Escherichia coli is the most common cause of complicated as well as uncomplicated urinary tract infections (UTIs). Most of these uropathogenic E. coli (UPEC) strains exhibit certain virulence factors (VFs), including adhesins, iron uptake systems, synthesis of cytotoxins, and specific O:K:H serotypes. Molecular epidemiological studies of UPEC have contributed to the discovery of uropathogenic VFs, to an understanding of the pathogenesis of UTIs as ascending infections, and to the clarification of genetic linkages between different virulence genes such as pathogenicity islands (PAIs), which are

OBJECTIVES: Escherichia coli is a major pathogen, and the emergence of carbapenem-resistant E. coli (CREC) significantly restricts clinical treatment options. Polymyxins are considered the last-line treatment for CREC infections; however, the occurrence of polymyxin-resistant CREC, particularly following reports of plasmid-mediated colistin resistance (mcr), poses an increasing challenge.
METHODS: In this study, we identified a polymyxin-resistant CREC strain isolated from the rectal swab of a patient hospitalized in a hematology ward. Antimicrobial susceptibility testings, S1-PFGE, Southern

Salmonella genomic island 1 (SGI1) is a multidrug resistance integrative mobilizable element that harbors a great diversity of antimicrobial resistance gene clusters described in numerous Salmonella enterica serovars and also in Proteus mirabilis. A serious threat to public health was revealed in the recent description in P. mirabilis of a SGI1-derivative multidrug resistance island named PGI1 (Proteus genomic island 1) carrying extended-spectrum-β-lactamase (ESBL) and metallo-β-lactamase resistance genes, blaVEB-6 and blaNDM-1, respectively. Here, we report the first description of Salmonella

Diverse antibiotic resistance genes (ARGs) present in sewage sludge are difficult to be eliminated using conventional sludge treatment processes. To date, little remains known on the fate of the ARGs during vermicomposting of sludge. This study aimed to investigate the effect of earthworms on the fate of tetracycline and fluoroquinolone resistance genes, and integrons during vermicomposting of sewage sludge through contrasting two systems of sludge stabilization with and without earthworms. Compared to the control without earthworms, vermicomposting significantly (p < 0.05) decreased the abund

Enzymes capable of inactivating tetracycline are paradoxically rare compared with enzymes that inactivate other natural-product antibiotics. We describe a family of flavoenzymes, previously unrecognizable as resistance genes, which are capable of degrading tetracycline antibiotics. From soil functional metagenomic selections, we discovered nine genes that confer high-level tetracycline resistance by enzymatic inactivation. We also demonstrate that a tenth enzyme, an uncharacterized homolog in the human pathogen Legionella longbeachae, similarly inactivates tetracycline. These enzymes catalyze

The stable structure of biofilms and the characteristics of the bacteria within them make biofilms an important barrier for bacteria to resist external stress, and a key factor contributing to the difficulty of eradicating clinical infections. This article reviews the multi-stage formation process of biofilms, the various mechanisms of antibiotic tolerance and resistance (such as physical barriers, metabolic adaptations, horizontal gene transfer, etc.), as well as the integrated regulatory roles of molecular networks like quorum sensing (QS) and cyclic diguanosine monophosphate (c-di-GMP).
