Neurosymptomatic Cerebrospinal Fluid Escape Is a Central Nervous System-Focused Form of Antiretroviral Therapy Treatment Failure.

BACKGROUND: During antiretroviral therapy, people with HIV-1 (PWH) occasionally present with new symptoms and signs of central nervous system (CNS) injury accompanied by higher HIV-1 RNA levels in cerebrospinal fluid (CSF) than in blood, a condition defined as neurosymptomatic CSF escape (NSE). PWH meeting these general criteria but with plasma HIV-1 RNA levels >500 copies/mL have typically been assumed to be experiencing systemic treatment failure without special consideration for HIV-1 dynamics in the CNS. Thus, people with CNS-focused treatment failure may receive suboptimal treatment, targ

Neonatal hemolysis and risk of bilirubin-induced neurologic dysfunction.

The pathologic phenotype of severe hyperbilirubinemia in the newborn infant is primarily due to excessive bilirubin production and/or impaired conjugation, resulting in an increased bilirubin load. This may, in turn, increase an infant's risk for the development of bilirubin-induced neurologic dysfunction (BIND). The highest-risk infants are those with increased bilirubin production rates due to hemolysis. Several immune and non-immune conditions have been found to cause severe hemolysis, and these are often exacerbated in those infants with perinatal sepsis and genetic predispositions. Theref

Epigenetic mechanisms in Alzheimer's disease: implications for pathogenesis and therapy.

The vast majority of Alzheimer's disease (AD) are late-onset forms (LOAD) likely due to the interplay of environmental influences and individual genetic susceptibility. Epigenetic mechanisms, including DNA methylation, histone modifications and non-coding RNAs, constitute dynamic intracellular processes for translating environmental stimuli into modifications in gene expression. Over the past decade it has become increasingly clear that epigenetic mechanisms play a pivotal role in aging the pathogenesis of AD. Here, we provide a review of the major mechanisms for epigenetic modification and ho

Hearts and bones: shared regulatory mechanisms in heart valve, cartilage, tendon, and bone development.

The mature heart valves are dynamic structures composed of highly organized cell lineages and extracellular matrices. The discrete architecture of connective tissue within valve leaflets and supporting structures allows the valve to withstand life-long functional demands and changes in hemodynamic forces and load. The dysregulation of ECM organization is a common feature of heart valve disease and can often be linked to genetic defects in matrix protein structure or developmental regulation. Recent studies have identified specific regulatory pathways that are active in the developing valve str

[Interrelationship between purine, carbohydrate, lipid and aminoacid metabolism, in hyperuricaemia and gout (author's transl)].

The AA, have studied the literature concerning the interrelationship between purine, carbohydrate, lipid and aminoacid metabolism, in hyperuricaemia and gout. The behaviour of uricaemia and purine metabolism in the glycogen storage disease type I, in the glutathione reductase variant, after ethanol ingestion, after fructose load, the levels of lipoproteins and triglycerides in gouty patients and the reason for their increase, have been taken into special consideration. The AA. underline the evidence that gout and hyperuricaemia are metabolic defects which are not limited to purine biosynthesis

Two distinct polymorphisms in the basic region of Meq protein of marek's disease virus alter pathological progression and clinical manifestations.

BACKGROUND: Marek's disease virus (MDV) causes Marek's disease (MD) in chickens, which is characterized by malignant lymphomas and neurological disorders. Although MD is currently controlled using live vaccines, the virulence of field strains has continuously increased in recent decades. Polymorphisms in the MDV-encoded oncoprotein Meq are shared among field strains according to their virulence. In particular, very virulent MDV strains harbor characteristic amino acid changes in the basic region of Meq at positions 77 and 80; however, the contribution of these polymorphisms to virulence remain

Anti-tembusu virus of capsid-targeted viral inactivation delivered by lentiviral vector in vivo.

Tembusu virus (TMUV) is a member of genus flavivirus, which mainly causes decrease in production in egg ducks and neurological symptom in meatducks, causing serious economic losses to the poultry industry. Recently, the commercialized TMUV vaccines are mainly the WF100 live vaccine and the attenuated live vaccine (FX2010-180P), so it is particularly important to find new methods to combat TMUV. The capsid-targeted viral inactivation (CTVI) strategy is based on a viral core protein and an exogenous factor that can destroy viral DNA or RNA. Lentivirus vectors are an effective tool for transferri

Epigenetic landscape in the kick-and-kill therapeutic vaccine BCN02 clinical trial is associated with antiretroviral treatment interruption (ATI) outcome.

BACKGROUND: The BCN02-trial combined therapeutic vaccination with a viral latency reversing agent (romidepsin, RMD) in HIV-1-infected individuals and included a monitored antiretroviral pause (MAP) as an efficacy read-out identifying individuals with an early or late (< or > 4weeks) viral-rebound. Integrated -omics analyses were applied prior treatment interruption to identify markers of virus control during MAP. METHODS: PBMC, whole-genome DNA methylation and transcriptomics were assessed in 14 BCN02 participants, including 8 Early and 4 Late viral-rebound individuals. Chromatin state, histo

Secreted protein kinases regulate cyst burden during chronic toxoplasmosis.

Toxoplasma gondii is an apicomplexan parasite that secretes a large number of protein kinases and pseudokinases from its rhoptry organelles. Although some rhoptry kinases (ROPKs) act as virulence factors, many remain uncharacterized. In this study, predicted ROPKs were assessed for bradyzoite expression then prioritized for a reverse genetic analysis in the type II strain Pru that is amenable to targeted disruption. Using CRISPR/Cas9, we engineered C-terminally epitope tagged ROP21 and ROP27 and demonstrated their localization to the parasitophorous vacuole and cyst matrix. ROP21 and ROP27 wer

Evidence for determining parasitic factors in addition to host genetics and immune status in the outcome of murine Leishmania infantum visceral leishmaniasis.

C.B-17 SCID and congenic BALB/C mice were used to examine Leishmania infantum strain pathogenicity independently of host genetic factors. While parasite loads were significantly higher in immunodeficient mice than in immunocompetent mice, the kinetics of infection during a long-term follow-up were similar, suggesting that intrinsic parasitic factors also influence the outcome of L. infantum infection.

Familial factors in SARS-CoV-2 susceptibility and COVID-19 severity: a nationwide Danish study of adult twins and their spouses.

BACKGROUND: Transmission of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) often clusters in families, suggesting role of shared environmental and genetic factors. We performed a nationwide Danish study of twins, their spouses, and matched population controls with the aim of disentangling the host genetic and environmental mechanisms underlying the familial clustering of SARS-CoV-2 susceptibility and coronavirus disease 2019 (COVID-19) severity. METHODS: This registry-linkage study included 83 902 twins and 32 719 spouses, matched to 839 020 population controls and 327 190 spous

Genetic association study between rs2234253 (p.T96K) variant of TREM2 and Alzheimer's disease in a Tunisian population.

BACKGROUND: Alzheimer's disease (AD) is the leading cause of major neurodegenerative cognitive impairment. The risk of developing AD is influenced by a complex interaction of genetic predisposition and environmental factors. Among the genetic risk factors, the APOE ɛ4 allele is the most significant, while variants in the TREM2 (Triggering Receptor Expressed on Myeloid Cells 2) and ABCA7 (ATP-binding cassette transporter A7) genes have also been associated with an increased risk of AD. OBJECTIVE: This study aimed to investigate the association of APOE ɛ4, TREM2 gene variants (rs75932628 [p.R47

Right Ventricular Remodeling and Function in Hypoplastic Left Heart Syndrome.

The right ventricle (RV) in hypoplastic left heart syndrome (HLHS) becomes the systemic ventricle pumping against systemic afterload. It also has to adapt to an initially increased volume load followed by a decrease in volume load after Fontan completion. Anatomical HLHS subtype, therapeutic strategy, tricuspid valve regurgitation, recoarctation, and genetics influence RV size and function. The resulting remodeling process can be maladaptive and lead to ventricular systolic and diastolic dysfunction. While systolic dysfunction is a strong predictor for mortality before Fontan, there is increas

Bio-inspired virus-like nanovesicle for effective vaccination.

Developing effective vaccines is of vital importance for protecting public health by preventing potential pandemics or by controlling ongoing ones. However, there is a threshold of rapidly design and develop effective vaccines to prevent virus infection. Inspired by the natural budding processes associated with cell membrane scission when enveloped viruses invade host cells and replicate themselves, a similar strategy was applied to achieve virus-mimetic nanovesicles (VMVs). This strategy loaded genetically engineered viral antigens onto mammalian cell membranes to produce antigen-loaded vesic

Recombinant D. radiodurans cells for bioremediation of heavy metals from acidic/neutral aqueous wastes.

The stability and superior metal bioremediation ability of genetically engineered Deinococcus radiodurans cells, expressing a non-specific acid phosphatase, PhoN in high radiation environment has already been established. The lyophilized recombinant DrPhoN cells retained PhoN activity and uranium precipitation ability. Such cells also displayed an extended shelf life of 6 months during storage at room temperature and showed surface associated precipitation of uranium as well as other metals like cadmium. Lyophilized cells, immobilized in polyacrylamide gels could be used for uranium bioprecipi

Tetrahydrobiopterin sensitivity in German patients with mild phenylalanine hydroxylase deficiency.

We report the results of tetrahydrobiopterin (BH4) loading tests in 10 German patients with mild phenylketonuria. A significant decline of phenylalanine values after application of BH4 was observed in all but one patients. Molecular genetic analyses revealed a range of different PAH gene mutations. Re-testing of one patient previously reported as non-responsive to BH4 loading showed a moderate response with a higher dose of BH4. Nevertheless, there appear to be kinetic differences in phenylalanine hydroxylation in patients with the same genotype. Non-responsiveness to 20 mg/kg BH4 was observed

Cerebral Amyloidosis in Individuals with Subjective Cognitive Decline: From Genetic Predisposition to Actual Cerebrospinal Fluid Measurements.

The possible relationship between Subjective Cognitive Decline (SCD) and dementia needs further investigation. In the present study, we explored the association between specific biomarkers of Alzheimer's Disease (AD), amyloid-beta 42 (Aβ42) and Tau with the odds of SCD using data from two ongoing studies. In total, 849 cognitively normal (CN) individuals were included in our analyses. Among the participants, 107 had available results regarding cerebrospinal fluid (CSF) Aβ42 and Tau, while 742 had available genetic data to construct polygenic risk scores (PRSs) reflecting their genetic predispo

A Genetically Engineered Biomimetic Nanodecoy for the Treatment of Liver Fibrosis.

Liver fibrosis, arising from factors such as viral infections or metabolic disorders, represents an ongoing global health challenge and is a major risk factor for hepatocellular carcinoma. Unfortunately, there are no clinically approved drugs available for its treatment. Recent studies have illuminated the pivotal role of macrophage recruitment in the pathogenesis of liver fibrosis, presenting a potential therapeutic target. Therefore, it holds great promise to develop novel anti-fibrotic therapies capable of inhibiting this process. Herein, a drug-loaded biomimetic nanodecoy (CNV-C) is develo

[Stress analysis of the molar with the all-ceramic crown prosthesis based on centric occlusal optimization].

Stress distribution of denture is an important criterion to evaluate the reasonableness of technological parameters, and the bite force derived from the antagonist is the critical load condition for the calculation of stress distribution. In order to improve the accuracy of stress distribution as much as possible, all-ceramic crown of the mandibular first molar with centric occlusion was taken as the research object, and a bite force loading method reflecting the actual occlusal situation was adopted. Firstly, raster scanning and three dimensional reconstruction of the occlusal surface of mola
