Studies on the relation of a prior immune response to immunogenicity.

(1) Two polymers of D-glutamic acid, one synthetic (poly-α-D-glutamic) and the other the capsular polypeptide of Bacillus anthracis (poly-γ-D-glutamic), which did not induce detectable antibody when injected in saline, elicited precipitating antibody in rabbits when complexed with methylated bovine serum albumin (MBSA). (2) Animals that had made an immune response to either of the polypeptides did not produce detectable anti-hapten antibodies when subsequently immunized with the homologus polypeptide conjugated with p-amino-ṕ-dimethylaminoazobenzene (PDA) groups. Neither was there an observabl…

Induction of transient proteinuria, hematuria, and glucosuria by ethanol consumption in Japanese alcoholics.

Urinalysis was carried out in 231 inpatients with alcohol dependence syndrome (215 males and 16 females). Fifty-nine patients (25.5%) showed proteinuria, 97 (42.0%) showed glucosuria, and 62 patients (26.8%) showed hematuria on admission. A total of 135 out of 231 patients (58.4%) showed abnormal urinalysis. Proteinuria was related to high blood pressure, high serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase, lactate dehydrogenase, uric acid, and triglyceride levels, and high urinary amylase concentration. Glucosuria was related to high serum glutamic-oxaloacetic transami

Mediator substances of the pineal neuronal network of mammals.

In addition to receptor-type pinealocytes, the mammalian pineal organ contains small and large neurons and ependymal/glial cells as well. Axons of pinealocytes form synaptic ribbon-containing axo-dendritic synapses on large secondary pineal neurons and/or terminate as neurohormonal endings on the basal lamina of the vascular surface of the organ. The small pineal neurons were found to be gamma-aminobutyric acid (GABA)-immunoreactive, while large secondary neurons and pinealocytes contained immunoreactive amino acids (glutamate and aspartate). Glutamate accumulated presynaptically in pinealocyt

Frontal cortical and left temporal glutamatergic dysfunction in schizophrenia.

Glutamatergic mechanisms have been investigated in postmortem brain samples from schizophrenics and controls. D-[3H]Aspartate binding to glutamate uptake sites was used as a marker for glutamatergic neurones, and [3H]kainate binding for a subclass of postsynaptic glutamate receptors. There were highly significant increases in the binding of both ligands to membranes from orbital frontal cortex on both the left and right sides of schizophrenic brains. The changes are unlikely to be due to antemortem neuroleptic drug treatment, because no similar changes were recorded in other areas. A predicted

Fractionation of scorpion (Mesobuthus tamulus) venom and the impact of specific fractions on glutamate metabolism.

The fractionation of the venom of Indian red scorpion (Mesobuthus tamulus) was performed using CM-52 (carboxy methyl cellulose) ion-exchange chromatography. The toxic potential of these fractions were tested on lepidoptera larvae and mice. The CM-52 fractions, which showed toxicity were further used to study their impact on glutamate metabolism. The glutamate content increased and glutamine synthetase and glutamate decarboxylase activity levels were elevated in envenomated animals. The present results reveal that glutamine is rapidly metabolized to glutamate and gamma-aminiobutyric acid indica

Nitrogenous compounds stimulate glucose-derived acid production by oral Streptococcus and Actinomyces.

Both Streptococcus and Actinomyces can produce acids from dietary sugars and are frequently found in caries lesions. In the oral cavity, nitrogenous compounds, such as peptides and amino acids, are provided continuously by saliva and crevicular gingival fluid. Given that these bacteria can also utilize nitrogen compounds for their growth, it was hypothesized that nitrogenous compounds may influence their acid production; however, no previous studies have examined this topic. Therefore, the present study aimed to assess the effects of nitrogenous compounds (tryptone and glutamate) on glucose-de

Glutamate, Immune Cells and Alzheimer's Disease: An Attempt to Link the Triad.

Glutamate (Glu), although the main excitatory neurotransmitter (NT) of the central nervous system (CNS), evidence recognizes its role in influencing immune cell function and activation, including cytokine production, cell proliferation, migration, and survival. Importantly, dysregulation of Glu homeostasis has been implicated in the pathogenesis of Alzheimer's disease (AD), a neurodegenerative disorder characterized by progressive cognitive decline, neuronal loss and neuroinflammation. This review synthesizes current evidence on the biochemistry of the glutamatergic synapse, as well as the exp

Effect of trimethoprim-sulphamethoxazole combination on folate metabolism in malnourished children.

The effect of a 10-day course of trimethoprim-sulphamethoxazole on folate metabolism was studied in 10 children with severe protein-calorie malnutrition. There was a significant increase in formimino-glutamic acid (FIGLU) excretion after a histidine load in children treated with trimethoprim-sulphamethoxazole, when compared with a control group, but no haematological evidence of folate deficiency.

Amino acids in neurotransmission and disease.

BACKGROUND: The amino acids, glutamate, GABA and glycine are involved in normal cellular function as well as in neuro-transmission. Glutamate in particular may be linked to neuron development and an oversupply may be the major cause of many acute and chronic neurological diseases. Such diseases include retinopathy of prematurity, glaucoma, hypertension, diabetes and ischemic damage secondary to vessel occlusion. CONCLUSIONS: This paper provides a review of the localization, proposed function and the mechanisms of manufacture and deactivation of the amino acid neurotransmitters. Current mechan

The glutamate/GABA system in the retina of male rats: effects of aging, neurodegeneration, and supplementation with melatonin and antioxidant SkQ1.

Glutamate and -aminobutyric acid (GABA) are the most abundant amino acids in the retina. An imbalance of the glutamate/GABA system is involved in the pathogenesis of various neurodegenerative disorders. Here we for the first time analyzed alterations of expression of glutamate- and GABA-synthesizing enzymes, transporters, and relevant receptors in the retina with age in Wistar rats and in senescence-accelerated OXYS rats who develop AMD-like retinopathy. We noted consistent age-dependent expression changes of GABAergic-system proteins (GAD67, GABA-T, and GAT1) in OXYS and Wistar rats: upregula

Reciprocal regulation of nitric oxide and glutamate in the nucleus tractus solitarii of rats.

Nitric oxide (NO) and glutamate are both important mediators of the central cardiovascular regulation in the nucleus tractus solitarii. Our previous studies revealed that the central cardiovascular effects of NO in the nucleus tractus solitarii could be inhibited by glutamate receptor blockade. On the other hand, nitric oxide synthase (NOS) inhibitor attenuated the cardiovascular effects of glutamate. Thus, NO and glutamatergic systems appear to interact in central cardiovascular regulation. The present study examined whether NO and glutamate may affect each other's release/production in the n

Implications for soluble iron accumulation, oxidative stress, and glial glutamate release in motor neuron death associated with sporadic amyotrophic lateral sclerosis.

Oxidative stress in sporadic amyotrophic lateral sclerosis (ALS) has been evidenced by accumulation of oxidatively modified products of nucleic acids, lipids, sugars, and proteins in the motor neuron system of brains and spinal cords obtained at autopsy from the patients. We recently demonstrated soluble iron accumulation in activated microglia of sporadic ALS spinal cords. This finding could indicate that iron-mediated Fenton reaction is most likely to be responsible for oxidative stress associated with this disease. The excitatory amino acid neurotoxicity hypothesis for sporadic ALS has been

Tyrianthinic acids from Ipomoea tyrianthina and their antimycobacterial activity, cytotoxicity, and effects on the central nervous system.

Four new partially acylated tetrasaccharides of 11-hydroxyhexadecanoic acid (1-4) were isolated from a methanolic extract of Ipomoea tyrianthina. The structures of these compounds were elucidated by spectroscopic and chemical methods. The resin glycoside composition of I. tyrianthina varied with the location of growth in Mexico. Compounds 1-4 showed antimycobacterial activity, were cytotoxic against the KB cell line, and, in a mouse model, exhibited potentiation of hypnosis induced by pentobarbital, protected against seizures induced by pentylenetetrazole, and released GABA and glutamic acid.

Morphological and GAD immunocytochemical properties of the dorsal lateral geniculate nucleus in a reptile.

Morphological and glutamic acid decarboxylase (GAD) immunocytochemical properties of the dorsal part of the lateral geniculate nucleus were investigated in reptiles, Caiman crocodilus. GAD immunoreactive neurons, although sparse, were found in the dorsal part of the lateral geniculate nucleus. In addition, puncta and fibers immunoreactive for GAD were also observed in this same nucleus. These results in Caiman utilizing GAD are similar to observations on the dorsal part of the lateral geniculate nucleus of other reptiles in which antisera to gamma amino butyric acid (GABA) were used.

Depressed convulsions by diazepam and its effects on brain monoamines and amino acids in E1 mice.

The effect of diazepam on inbred mutant E1 mice, which develop convulsive seizures after repeated sessions of being tossed up, was examined. Acute administration of diazepam (32 mg/kg, i.p.) completely inhibited the convulsions. At that time, the dopamine level was increased in the cortex and hippocampus, and the norepinephrine level in the cerebellum was decreased. 5-Hydroxytryptamine levels were not changed. As for amino acids, the glutamine level increased and the levels of GABA, glutamic acid, aspartic acid, alanine and other amino acids were not changed.

Cleavage of human serum transferrin with N-bromosuccinimide.

Human serum transferrin was fragmented by N-bromosuccinimide and reduction-alkylation. It was observed that there were at least two each of tryptophanyl-serine and tryptophanyl-aspartic acid, and one each of tryptophanyl-alanine and tryptophanyl-glutamic acid bonds. The size of fragments detected by polyacrylamide gel electrophoresis ranged from 8,000 to 70,000 daltons. Several of the fragments were isolated in a homogeneous form with respect to molecular weight, but were shown to be mixtures of at least five molecular species each by end group analysis.

A comparative analysis of fruit fly and human glutamate dehydrogenases in Drosophila melanogaster sperm development.

Glutamate dehydrogenases are enzymes that take part in both amino acid and energy metabolism. Their role is clear in many biological processes, from neuronal function to cancer development. The putative testis-specific Drosophila glutamate dehydrogenase, Bb8, is required for male fertility and the development of mitochondrial derivatives in spermatids. Testis-specific genes are less conserved and could gain new functions, thus raising a question whether Bb8 has retained its original enzymatic activity. We show that while Bb8 displays glutamate dehydrogenase activity, there are significant func

Intrasubthalamic nucleus metabotropic glutamate receptor activation: a behavioral, Fos immunohistochemical and [14C]2-deoxyglucose autoradiographic study.

Metabotropic glutamate receptors are a major class of excitatory amino acid receptors. Eight metabotropic glutamate receptor subtypes have been cloned, and are classified into three groups (I, II and III) based on amino acid sequence identity, effector systems and pharmacological profile. Previous results have shown that unilateral stimulation of metabotropic glutamate receptors in the subthalamic nucleus with the non-subtype-selective metabotropic glutamate receptor agonist 1S,3R-1-amino-1,3-cyclopentane dicarboxylate results in contralateral rotation in rats and Fos expression in the subthal
