Insecticide resistance can arise from a variety of mechanisms, including changes to the target site, but is often associated with substantial fitness costs to insects. Here we describe two resistance-associated target-site mutations that have synergistic and compensatory effects that combine to produce high and persistent levels of resistance to fipronil, an insecticide targeting on γ-aminobytyric acid (GABA) receptors. In Nilaparvata lugens, a major pest of rice crops in many parts of Asia, we have identified a single point mutation (A302S) in the GABA receptor RDL that has been identified pr

The introduction of genetically modified plants into natural habitats represents a valuable means to determine organismic level functions of a gene and its effects on a plant's interaction with other organisms. Nicotiana attenuata, a wild tobacco species native of the southwestern USA that grows in the immediate postfire environment, is one of the important host plants for herbivore populations recolonizing recently burned habitats in the Great Basin Desert. Here, we provide detailed guidelines for the analysis, under field conditions, of jasmonate-dependent defense and its impact on the plant

The ribosomal protein S6 kinase (S6K) transduces the anabolic signals from nutrients and is involved in insect development and reproduction, however, its role in the regulation of the insecticide detoxification is still unclear. In this study, LsS6K was cloned and characterized from Laodelphax striatellus. RT-qPCR analysis revealed tissue-specific expression of LsS6K with the highest expression level in the fat body, and thiamethoxam exposure significantly down-regulated the expression of LsS6K at both the mRNA and protein levels. Functional analysis using RNA interference (RNAi) showed that k

Candidatus Liberibacter asiaticus (CLas) is a phloem-limited, gram-negative, fastidious bacterium that is associated with the development of citrus greening disease, also known as Huanglongbing (HLB). CLas is transmitted by the Asian citrus psyllid (ACP) Diaphorina citri, in a circulative manner. Two major barriers to transmission within the insect are the midgut and the salivary glands. We performed a thorough microscopic analysis within the insect midgut following exposure to CLas-infected citrus trees. We observed changes in nuclear architecture, including pyknosis and karyorrhexis as well

1. Masting events in the autumn provide a carbohydrate-rich pulse of resources that can influence the dynamics of small mammals and their natural enemies. Similar patterns are observed with the periodical cicada emergence which provides a protein-rich pulse in the spring, but comparisons are confounded by timing and food type. 2. We compared the influence of a naturally occurring spring pulse of cicadas with an experimental spring pulse of carbohydrate-rich seeds. We used a replicated population level field experiment and capture-mark-recapture techniques to record the vital rates, demographic

BACKGROUND: Triatomine bugs (Hemiptera, Reduviidae, Triatominae) are blood-sucking vectors of the protozoan parasite, Trypanosoma cruzi, which causes Chagas disease, a significant source of human morbidity and mortality in the Americas. Autochthonous transmission of Chagas disease in the United States is considered rare, despite evidence of Triatoma species harboring T. cruzi, invading homes, and biting occupants. In the southeastern United States, Triatoma sanguisuga is considered common, yet its distribution, host use, and T. cruzi infection are practically unknown in this region.

Social insect colonies constantly produce dead insects, which cause sanitary problems and potentially foster deadly pathogens and parasites. Hence, many social insects have evolved a variety of hygienic behaviors to remove cadavers from the colonies. To that end, they have to discriminate dead insects from live ones, where chemical cues should play important roles. In ants, bees and termites, such corpse recognition signals, also referred to as "death pheromones" or "necromones", have been identified as fatty acids, specifically oleic acid and/or linoleic acid. Meanwhile, there has been no suc

BACKGROUND: Triatoma infestans (Hemiptera: Reduviidae) is a hematophagous insect and the main vector of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae). In the present study, the authors investigated whether a serine protease activity from the saliva of T. infestans has a role in vasomotor modulation, and in the insect-blood feeding by cleaving and activating protease-activated receptors (PARs).
METHODS: T. infestans saliva was chromatographed as previously reported for purification of triapsin, a serine protease. The cleavage activity of triapsin on PAR peptides was investigated based o

Buggy Creek virus (family Togaviridae, genus Alphavirus, BCRV) is an alphavirus within the western equine encephalitis virus complex whose primary vector is the swallow bug, Oeciacus vicarius Horvath (Hemiptera: Cimicidae), an ectoparasite of the colonially nesting cliff swallow, Petrochelidon pyrrhonota, that is also a frequent host for the virus. We investigated ecological correlates of BCRV infection in 100-bug pools at 14 different swallow colony sites in southwestern Nebraska from summer 2004, by using plaque assay on Vero cells to identify cytopathic virus and reverse transcription-polym

A novel double-stranded RNA (dsRNA) virus designated Psammotettix alienus reovirus (PARV) was found in the leafhopper Psammotettix alienus in China. Spherical particles approximately 70 nm in diameter arranged in a crystalline array were observed in the salivary gland tissues of infected leafhoppers by transmission electron microscopy. Some viral particles were also encased in tubules, similar to those of previously described reoviruses. Whole-genome sequencing revealed that the dsRNA genome of PARV consists of 29 569 nucleotides (nt) divided into 10 segments ranging from 4403 to 1476 nt, with

Ultraviolet (UV) radiation directly regulates a multitude of herbivore life processes, in addition to indirectly affecting insect success via changes in plant chemistry and morphogenesis. Here we looked at plant and insect (aphid and whitefly) exposure to supplemental UV-A radiation in the glasshouse environment and investigated effects on insect population growth. Glasshouse grown peppers and eggplants were grown from seed inside cages covered by novel plastic filters, one transparent and the other opaque to UV-A radiation. At a 10-true leaf stage for peppers (53 days) and 4-true leaf stage f

The EFSA Panel on Plant Health performed a pest categorisation of Matsucoccus matsumurae (Hemiptera: Matsucoccidae), the Massonian pine bast scale, for the EU territory. This pest categorisation was initiated following the commodity risk assessment of artificially dwarfed plants from China consisting of Pinus parviflora (Japanese white pine) grafted on P. thunbergii (Japanese black pine) performed by EFSA, in which M. matsumurae was identified as a pest of possible concern. However, its identity is not firmly established due to uncertainty regarding its taxonomic relationship with Matsucoccus

Phloem-inhabiting unculturable bacterial pathogens are persistently transmitted by insect vectors. However, how they evade insect immune responses to ensure persistent transmission remains unknown. The important melanization immune response in insects is triggered by cleavage of prophenoloxidase (PPO) into active phenoloxidase (PO) via clip-domain serine proteases (CLIPs). Here, we demonstrate that infection of Candidatus Liberibacter asiaticus (CLas) in psyllid vectors activates the peptidoglycan recognition protein (PGRP)-CLIP1-CLIP4-PPO-PO signaling cascade to induce a mild melanization res

The whitebacked planthopper (WBPH), has become a devastating pest for rice crops, causes serious yield losses each year, and urgently needs biological control. Here, we developed a WBPH-resistant rice cultivar by overexpressing the OsF3H gene. A genetic functional analysis of the OsF3H gene confirmed its role in facilitating flavonoid contents and have indicated that the expression of the OsF3H gene is involved in regulation of the downstream genes (OsDFR and OsFLS) of the flavonoid pathway and genes (OsSLR1 and OsWRKY13) involved in other physiological pathways. OxF3H (OsF3H transgenic) plant

BACKGROUND: Asian citrus psyllid (ACP), Diaphorina citri Kuwayama, is an important pest of citrus worldwide because it transmits the bacteria causing huanglongbing (HLB). We investigated the effects and persistence of two soil application rates of thiamethoxam on ACP populations in two flushing seasons in the field. Thiamethoxam and clothianidin residues in the fruit were detected to evaluate food safety.
RESULTS: Soil application of 50% thiamethoxam water-dispersible granules at concentrations of 4 and 10 g tree-1 significantly decreased ACP populations, and there was a positive correlation

Chagas disease is a complex vector borne parasitic disease involving blood feeding Triatominae (Hemiptera: Reduviidae) insects, also known as kissing bugs, and the vertebrates they feed on. This disease has tremendous impacts on millions of people and is a global health problem. The etiological agent of Chagas disease, Trypanosoma cruzi (Kinetoplastea: Trypanosomatida: Trypanosomatidae), is deposited on the mammalian host in the insect's feces during a blood meal, and enters the host's blood stream through mucous membranes or a break in the skin. Identifying the blood meal sources of triatomin

The whitefly, Bemisia tabaci, as a globally significant agricultural pest, poses a serious threat to various food crops and thus causes enormous economic losses to global agricultural production. In recent decades, with the long-term and extensive use of chemical pesticides, B. tabaci has developed high levels resistance to thiamethoxam. Therefore, an in-depth analysis of the molecular mechanisms involved in insecticide detoxification within B. tabaci is of great theoretical and practical significance. Neuropeptide FF receptor 2 (NPFF2), a G protein-coupled receptor (GPCR), plays a key role in

Vector-borne diseases cause over 700,000 deaths annually and represent 17% of all infectious illnesses worldwide. This public health menace highlights the importance of understanding how arthropod vectors, microbes and their mammalian hosts interact. Currently, an emphasis of the scientific enterprise is at the vector-host interface where human pathogens are acquired and transmitted. At this spatial junction, arthropod effector molecules are secreted, enabling microbial pathogenesis and disease. Extracellular vesicles manipulate signaling networks by carrying proteins, lipids, carbohydrates an
