The effect of xylazine epidural anaesthesia on blood gas and acid-base parameters in rams.

The blood gas and acid-base effects of epidural xylazine were studied in rams. Xylazine at a dose of 0.4 mg kg-1 led to a metabolic alkalosis with a significant increase of pH, plasma bicarbonate and base excess and respiratory depression. Epidural administration of xylazine provided prolonged and very effective analgesia during abdominal and hindlimb surgery in rams.

Step trajectory analysis of spinal cord injured rats trained with neuromuscular electrical stimulation coordinated with robotic treadmill training.

Applying neuromuscular electrical stimulation (NMES) during treadmill training (TT) has been shown to improve functional outcomes, such as gait speed and walking distance, in spinal cord injury (SCI) patients. However, ways to improve this combined NMES+TT therapy have not been investigated. We have developed NMES system for a rodent model of SCI to investigate whether and how more precisely timing the stimulation to robotically assisted hindlimb position might achieve rehabilitation of independent stepping after SCI. In our therapy (NMES+RTT), rodent ankle flexor muscles are stimulated while …

Targeted misexpression of constitutively active BMP receptor-IB causes bifurcation, duplication, and posterior transformation of digit in mouse limb.

Members of bone morphogenetic proteins (BMPs) play important roles in many aspects of vertebrate embryogenesis. In developing limbs, BMPs ha … sterior mesenchyme of the forelimb bud, the lateral plate mesoderm, and the whole mesenchyme of the hindlimb bud. While the forelimbs appeared normal, the transgenic hindlimbs exhibited several phenotypes, including bifurcation, preaxial polydactyly, and posterior transformation of the anterior digit. However, the size of bones in the transgenic limbs seem …

Trifluoperazine inhibition of insulin-induced increase in skeletal muscle glucose uptake.

The ability of trifluoperazine, a calmodulin blocker, to prevent increases in skeletal muscle glucose uptake induced by insulin was tested. Insulin significantly increased glucose uptake by the rat hindlimb and trifluoperazine (10(-6) M) completely inhibited this increase. We conclude that calmodulin is a factor involved in the regulation of insulin-mediated glucose transport in muscle cells.

Mutation profiling of a limbless pig reveals genome-wide regulation of RNA processing related to bone development.

Mutation is the basis of phenotypic changes and serves as the source of natural selection. The development of limbs has been the milestone in vertebrate evolution. Several limb and bone-related genes were verified experimentally, but other indirect and regulatory factors of limb development remained untested, especially very few cases were observed in natural environment. We report a naturally born serpentized pig without hindlimbs. Whole genome sequencing followed by comparative genomic analysis revealed multiple interesting patterns on the handicapped pig-specific mutations. Although the bon…

Qualitative and quantitative analysis of talons of diurnal bird of prey.

The aim of this work is to establish qualitative and quantitative differences and similarities among talons lacking the keratin sheath in diurnal raptors. We set out to find a tool that allows for the determination of toe (I-IV) and hindlimb (left or right) identity. Different methodologies were used, such as classical anatomical descriptions and Fourier analysis of the medial contour of the ungual phalanx. A series of measurements and ratios were obtained and analysed using t-tests and principal components analysis. The qualitative descriptors are more important than quantitative analyses for…

Somatotopically arranged inputs from putamen and subthalamic nucleus to primary motor cortex.

Employing retrograde transsynaptic transport of rabies virus, we investigated the organization of basal ganglia inputs to hindlimb, proximal and distal forelimb, and orofacial representations of the macaque primary motor cortex (MI). Four days after rabies injections into these MI regions, neuronal labeling occurred in the striatum and the subthalamic nucleus (STN) through the cortico-basal ganglia loop circuits. In the striatum, two distinct sets of the labeling were observed: one in the dorsal putamen, and the other in the ventral striatum (ventromedial putamen and nucleus accumbens). The do…

Sirenomelia in Bmp7 and Tsg compound mutant mice: requirement for Bmp signaling in the development of ventral posterior mesoderm.

Sirenomelia or mermaid-like phenotype is one of the principal human congenital malformations that can be traced back to the stage of gastrulation. Sirenomelia is characterized by the fusion of the two hindlimbs into a single one. In the mouse, sirens have been observed in crosses between specific strains and as the consequence of mutations that increase retinoic acid levels. We report that the loss of bone morphogenetic protein 7 (Bmp7) in combination with a half dose or complete loss of twisted gastrulation (Tsg) causes sirenomelia in the mouse. Tsg is a Bmp- and chordin-binding protein that …

Lidocaine in the rostroventromedial medulla and the periaqueductal gray attenuates allodynia in neuropathic rats.

In the present study we attempted to find out if the rostroventromedial medulla (RVM) or the periaqueductal gray (PAG) might contribute to chronic allodynia induced by unilateral ligation of two spinal nerves in the rat. Lidocaine was microinjected in the RVM or PAG and allodynia was quantitatively determined by measuring the hindlimb withdrawal thresholds to mechanical stimulation of the paw. For comparison, lidocaine was also injected systemically (s.c.). Lidocaine in the RVM produced a dose-related (20 and 40 micrograms) antiallodynic effect. Lidocaine (20 micrograms) in the PAG produced id…

Prostaglandins in inflammatory bone pathology: mechanism and therapeutic benefit of etodolac.

To investigate the role of PGE2 in the development of bone and joint pathology in rat adjuvant arthritis, hindlimb paws were evaluated by calcified tissue histologic techniques focusing on histochemical visualization of cartilage and bone lesions. Case studies of hindlimbs from normal, adjuvant arthritic, and etodolac-treated arthritic rats demonstrated the association of disease severity with inflammation, chondromalacia, replacement of adipose bone marrow with a fibroid marrow, osteoclastic bone resorption, synovial cysts, and pannus formation within the joints. Extensive periosteal intramem…

Osteoarticular sporotrichosis in a dog.

Osteoarticular sporotrichosis was diagnosed in a dog referred for evaluation of hindlimb lameness. There was radiographic evidence of osteopenia of the fourth tarsal and proximal aspects of the metatarsal bones. The diagnosis was based on histologic findings and results of physical examination, radiography, fungal culturing, and serologic tests. The dog was treated successfully with ketoconazole for 3 1/2 months.

Sensory-evoked pocket scratch motor patterns in the in vitro turtle spinal cord: reduction of excitability by an N-methyl-D-aspartate antagonist.

1. In intact turtles, tactile stimulation of the body surface in the "shell pocket" region surrounding the hindlimb elicits a pocket scratch reflex, in which the hindlimb reaches toward and rhythmically rubs the stimulated site. In the present study, we utilized reduced in vitro preparations of the turtle spinal cord with attached peripheral nerves to investigate the time course and pharmacology of sensory-evoked excitability in the pocket scratch neural network. Fictive pocket scratch motor output was elicited by electrically stimulating either the ventral-posterior pocket (VPP) cutaneous ner…

A Feasibility Study to Determine Whether Neuromuscular Adaptations to Equine Water Treadmill Exercise Can Be Detected Using Synchronous Surface Electromyography and Kinematic Data.

Despite growing evidence on the adaptive movement patterns that horses adopt during water treadmill (WT) exercise, underlying adaptations in muscle activity remain uninvestigated. This feasibility study aimed to develop a method for the synchronous measurement of muscle activity and movement of horses during WT exercise. Combined surface electromyography (sEMG) (2000 Hz) from selected hindlimb (biceps femoris, gluteus medius, tensor fasciae latae) and epaxial (longissimus dorsi) muscles, and three-dimensional kinematic (200 Hz) data from the back and pelvis of one (n = 1) horse were collected …
