Toxoplasma gondii rhoptry protein38 (TgROP38) affects parasite invasion, egress, and induces IL-18 secretion during early infection.

Toxoplasma rhoptry protein 38 (TgROP38) is a new active kinase that modulates host cell signal transduction and TgROP38 expression shows strain-specificity and stage-specificity in different isolates. In the present study, we overexpressed ROP38 in the RH and prugniaud (PRU) strain (RH+rop38II and PRU+rop38II), disrupted ROP38 (PRUΔROP38) in the PRU strain, complemented the ROP38 (PRUΔROP38comp+) in the PRUΔROP38 strain, and compared phenotypes of gene-edited and parental strains. We found that knockout of ROP38 led to increased proliferation (P < 0.01) and invasion (P < 0.01) ability of the p

Whole-genome analyses of speciation events in pathogenic Brucellae.

Despite their high DNA identity and a proposal to group classical Brucella species as biovars of Brucella melitensis, the commonly recognize … brane proteins suggests that these inactivations may play an important role in the establishment of host specificity and may have been a primary driver of speciation in the genus Brucella. Despite being nonmotile, the brucellae contain flagellum gene clusters and display species-specific flagellar gene inactivations, which lead to the putative generation of differ …

Langat Virus Biology and Infection.

Flaviviruses pose a major threat to global health and can cause severe disease in animals and humans. Among them, tick-borne encephalitis virus (TBEV) and related members of the tick-borne encephalitis (TBE) serocomplex are transmitted primarily by ticks and can lead to neurological disease, including encephalitis and death. Despite many years of research on the TBE-serocomplex, no specific antiviral treatment has been developed. Langat virus (LGTV), another member of this serocomplex typically causes asymptomatic infections in humans, but may cause severe neurological disease in rare cases. M

Suppression of Plant Defenses by Herbivorous Mites Is Not Associated with Adaptation to Host Plants.

Some herbivores suppress plant defenses, which may be viewed as a result of the coevolutionary arms race between plants and herbivores. However, this ability is usually studied in a one-herbivore-one-plant system, which hampers comparative studies that could corroborate this hypothesis. Here, we extend this paradigm and ask whether the herbivorous spider-mite Tetranychus evansi, which suppresses the jasmonic-acid pathway in tomato plants, is also able to suppress defenses in other host plants at different phylogenetic distances from tomatoes. We test this using different plants from the Solana

The genus Borrelia reloaded.

The genus Borrelia, originally described by Swellengrebel in 1907, contains tick- or louse-transmitted spirochetes belonging to the relapsing fever (RF) group of spirochetes, the Lyme borreliosis (LB) group of spirochetes and spirochetes that form intermittent clades. In 2014 it was proposed that the genus Borrelia should be separated into two genera; Borrelia Swellengrebel 1907 emend. Adeolu and Gupta 2014 containing RF spirochetes and Borreliella Adeolu and Gupta 2014 containing LB group of spirochetes. Alphavirus nsP3 as a multifunctional orchestrator of virus-host interplay.

Alphaviruses can cause polyarthralgia and encephalitis and pose an escalating global health threat to humans and animals. They are transmitted to vertebrates mainly by infected mosquitoes, leading to host alternation that drives an evolutionary arms race between the virus and host, during which the virus is selected to efficiently utilize the cellular factors of both hosts for replication. Alphaviruses encode the information necessary for replication in their genomic viral RNA, which is translated into nonstructural proteins, such as nonstructural protein 3 (nsP3). nsP3 is a remarkably multifu

Bat phylogeny and geographic location, rather than bat individual characteristics, explains the pattern of trypanosome infection in Europe.

Understanding the drivers of parasite susceptibility provides valuable information, such as how parasites spread, what conditions favour the … ctors rather than random encounters between bats and their trypanosome parasites. However, the high host specificity and absence of a strong cophylogenetic signal indicate that random host switching, rather than co-speciation, is the dominant mechanism shaping bat-trypanosome associations.

Host proteases: key regulators in viral infection and therapeutic targeting.

Viral infections pose a major threat to global health, causing severe pneumonia, meningitis, hepatitis, and fatal complications. Viruses are highly dependent on host cellular factors to complete their life cycle, and host proteases, as one of the core regulatory hubs, profoundly influence the progression of infection and pathogenicity. Viruses rely on specific host proteases (e.g., transmembrane serine proteases [TMPRSS family], furin, cathepsins, and others such as caspases and metalloproteases) to precisely cleave and activate viral surface glycoproteins and internal precursor proteins, ther

The Chlamydia pneumoniae effector SemD exploits its host's endocytic machinery by structural and functional mimicry.

To enter epithelial cells, the obligate intracellular pathogen Chlamydia pneumoniae secretes early effector proteins, which bind to and modulate the host-cell's plasma membrane and recruit several pivotal endocytic host proteins. Here, we present the high-resolution structure of an entry-related chlamydial effector protein, SemD. Co-crystallisation of SemD with its host binding partners demonstrates that SemD co-opts the Cdc42 binding site to activate the actin cytoskeleton regulator N-WASP, making active, GTP-bound Cdc42 superfluous. While SemD binds N-WASP much more strongly than Cdc42 does,

Interindividual Spread of Herpesviruses.

Interindividual spread of herpesviruses is essential for the virus life cycle and maintenance in host populations. For most herpesviruses, the virus-host relationship is close, having coevolved over millions of years resulting in comparatively high species specificity. The mechanisms governing interindividual spread or horizontal transmission are very complex, involving conserved herpesviral and cellular proteins during the attachment, entry, replication, and egress processes of infection. Also likely, specific herpesviruses have evolved unique viral and cellular interactions during cospeciati

Prospective monitoring for alloimmunization in cord blood transplantation: "virtual crossmatch" can be used to demonstrate donor-directed antibodies.

Preformed host antibodies may contribute to graft rejection after hematopoietic stem-cell transplantation. In cord blood transplantation (CBT), donor-directed host antibodies may be particularly relevant because patients are often markedly mismatched to donors, and limited donor cells preclude cross-matching. The recent development of single human leukocyte antigen (HLA) microbead array assays allows characterization of host alloreactivity to individual HLA antigens with sufficient sensitivity and specificity to allow consideration of "virtual crossmatch" testing as a surrogate for conventiona

Draft Genome Sequences of Six Ruminant Coxiella burnetii Isolates of European Origin.

Coxiella burnetii is responsible for Q fever, a worldwide zoonosis attributed to the inhalation of aerosols contaminated by livestock birth products. Six draft genome sequences of European C. burnetii isolates from ruminants are presented here. The availability of these genomes will help in understanding the potential host specificity and pathogenicity and in identifying pertinent markers for surveillance and tracing.

Microsatellite markers for the human nematode parasite Ascaris lumbricoides: development and assessment of utility.

We describe 35 microsatellite markers from the human parasitic nematode Ascaris lumbricoides. We found 7 sex-linked markers and demonstrate that 26 autosomal loci can be scored reliably. These markers have high genetic variability and provide the tools to address multiple questions concerning the epidemiology, fine-scale genetic structure, host specificity, and mating systems of this parasite.

[Toxocariasis--a current clinical and diagnostic problem].

Toxocariasis is zoonotic disease caused by worms Toxocara canis and Toxocara cati. Human is its paratenic host. There are some clinical forms: Visceral larva migrans, cerebral larva migrans, ocular larva migrans and covert toxocariasis. At the present time diagnostics of toxocariasis is very difficult due to polymorphism of clinical symptoms and low specificity of laboratory tests.

Development of an in-House TaqMan Real-Time PCR-Based Method to Detect Residual Host Cell DNA in HBV Vaccine.

Biological therapeutic products such as recombinant hepatitis B virus (HBV) vaccine, produced by microbial fermentation in complex media, should be evaluated for host cell DNA contamination in purification steps. Eliminating these contaminations increases the efficacy of the vaccine and decreases its side effects. The objective of the present study is to trace the residual host cell DNA (HCD) in recombinant HBV vaccine by developing a TaqMan Real-Time PCR method which is more sensitive, specific, and reproducible than traditional methods such as Picogreen analysis and Threshold DNA assay. Use of recombinant Entamoeba histolytica cysteine proteinase 1 to identify a potent inhibitor of amebic invasion in a human colonic model.

Cysteine proteinases are key virulence factors of the protozoan parasite Entamoeba histolytica. We have shown that cysteine proteinases play a central role in tissue invasion and disruption of host defenses by digesting components of the extracellular matrix, immunoglobulins, complement, and cytokines. Analysis of the E. histolytica genome project has revealed more than 40 genes encoding cysteine proteinases. We have focused on E. histolytica cysteine proteinase 1 (EhCP1) because it is one of two cysteine proteinases unique to invasive E. histolytica and is highly expressed and released. Recom

Seasonal immune dynamics and host-microbiota interactions in the mussel Mytilus galloprovincialis under a warming Mediterranean Sea.

Marine bivalves are increasingly threatened by disease outbreaks and mass mortality events driven by climate change and polymicrobial infections. Mytilus galloprovincialis is the main farmed bivalve in the Mediterranean, one of the world's fastest-warming marine regions, with the Ligurian Sea experiencing particularly rapid increases in temperature and heatwave frequencies and intensities. Bivalve hemocytes integrate environmental and microbial cues, making their seasonal dynamics a key determinant of host resilience under changing climate conditions. Previous studies identified surface seawat

Exploring bacteriophages as potential tools for combating multi drug-resistant Klebsiella pneumoniae: Isolation, characterization, genomic insights, and in vitro evaluation.

Bacteriophages are promising alternatives to antibiotics for combating multidrug-resistant (MDR) pathogens such as Klebsiella pneumoniae, a major cause of hospital-acquired infections. In this study, two K. pneumoniae-specific phages, vB_Kpn_RAH1 and vB_Kpn_RAH2, were isolated from sewage using an MDR clinical isolate (KP26) as the host. Their morphological, physicochemical, and genomic characteristics were analyzed. Both phages exhibited Siphovirus-like morphology, and the genome sizes of RAH1 and RAH2 were 46,784 bp and 59,380 bp, respectively. The phages were stable at 25-37 °C and pH 7-9 b
