Lytic Cycle of Toxoplasma gondii: 15 Years Later.

Toxoplasmosis is the clinical and pathological consequence of acute infection with the obligate intracellular apicomplexan parasite Toxoplasma gondii. Symptoms result from tissue destruction that accompanies lytic parasite growth. This review updates current understanding of the host cell invasion, parasite replication, and eventual egress that constitute the lytic cycle, as well as the ways T. gondii manipulates host cells to ensure its survival. Since the publication of a previous iteration of this review 15 years ago, important advances have been made in our molecular understanding of paras

[RELATIONSHIPS IN THE "NORTHERN FULMAR (FULMAR US GLACIALIS)--TETRABOTHRIUS MINOR (CESTODA: TETRABOTHRIIDAE)" SYSTEM: PHYSIOLOGICAL ASPECTS].

Relationships between the northern fulmar (Fulmarus glacialis L., 1761) and cestodes Tetrabothrius minor Loennberg, 1893 (Cestoda: Tetrabothriidae) were studied. The results of calculation of the number of tapeworms in different parts (proximal, medial and distal) of bird intestines are represented. Parameters of protein metabolism in the northern fulmar and cestodes T. minor were investigated. Activity of proteases in different parts of northern fulmar intestine and in the strobila of T. minor was determined. Digestion processes occurring on digestive-absorptive surfaces of the intestine of t

From Fossil Parasitoids to Vectors: Insects as Parasites and Hosts.

Within Metazoa, it has been proposed that as many as two-thirds of all species are parasitic. This propensity towards parasitism is also reflected within insects, where several lineages independently evolved a parasitic lifestyle. Parasitic behaviour ranges from parasitic habits in the strict sense, but also includes parasitoid, phoretic or kleptoparasitic behaviour. Numerous insects are also the host for other parasitic insects or metazoans. Insects can also serve as vectors for numerous metazoan, protistan, bacterial and viral diseases. The fossil record can report this behaviour with direct

Basic and applied problems in developmental biology and immunobiology of cestode infections: Hymenolepis, Taenia and Echinococcus.

Differentiation and development of parasites, including longevity in host animals, are thought to be governed by host-parasite interactions. In this review, several topics on the developmental biology of cestode infections are discussed from immunobiological perspective with a focus on Hymenolepis, Taenia and Echinococcus infections. The basic premise of this review is that 'differentiation and development of cestodes' are somehow affected by host immune responses with an evolutionary history.

Disentangling the effects of exposure and susceptibility on transmission of the zoonotic parasite Schistosoma mansoni.

For all parasites, transmission is composed of two processes: host contact with parasites ('exposure') and risk of infection given such contact ('susceptibility'). Classic models, such as mass action (density-dependent) transmission, lump these processes together. However, separating these processes could enhance predictions for disease dynamics, especially for free-living parasites. Here, we outline three transmission models that partition exposure and susceptibility. Using data from a study of Schistosoma mansoni (trematode) infections in Biomphalaria glabrata snails, we competed these three

Fasciola hepatica in Cuba: compatibility of different isolates with two intermediate snail hosts, Galba cubensis and Pseudosuccinea columella.

In Cuba, only two lymnaeid snails, Galba cubensis and Pseudosuccinea columella, with different ecology and distribution patterns, are intermediate hosts for Fasciola hepatica. The compatibility of these two species as hosts was analysed through their rates of infection, the production of rediae and survivorship when exposed to F. hepatica miracidia. Ten populations of G. cubensis, eight of P. columella collected from various habitats and six isolates of F. hepatica sampled in slaughterhouses from different localities were tested. Our results clearly demonstrate that G. cubensis is a more compa

Scratching the itch: new tools to advance understanding of scabies.

Scabies remains a significant public health problem worldwide. Research into aspects of Sarcoptes scabiei biology and host-parasite interactions has been impeded by an inability to maintain mites in vitro and by limited access to parasite material and infected subjects. The generation of comprehensive expressed sequence tag libraries has enabled the initial characterisation of molecules of interest to diagnostics, vaccines, and drug resistance. The recent development and utilisation of animal models, combined with next-generation technologies, is anticipated to lead to new strategies to preven…

Wolbachia and its implications for the immunopathology of filariasis.

Filarial infections are characterized by immunopathological phenomena, that are responsible for the onset of often dramatic pathological outcomes, such as blindness (Onchocerca volvulus) and elephantiasis (W. bancrofti). In addition, the long-term survival (as long as 10 years) of these parasites in otherwise immunocompetent hosts indicates that these nematodes are capable of manipulating the host immune response. The ground-breaking discovery of the bacterial endosymbiont Wolbachia, which resides in most filarial nematodes causing disease, has led to increasing interest in the role it may pla

Hookworm virulence factors: making the most of the host.

Hookworm disease from Necator americanus and Ancylostoma duodenale affects approximately 700 million people, with N. americanus being the predominant species. Unlike other pathogens (e.g., bacterial infections), where "virulence" is described in regards to acute pathogenesis and case-fatality, hookworms are well-evolved, multicellular parasites that establish long-term infections in their human hosts with a subtle and chronic, but insidious, pathogenesis, usually in the form of iron deficiency anemia from parasite blood feeding that, over time, has devastating effects on the human host especia

Virulence of a malaria parasite, Plasmodium mexicanum, for its sand fly vectors, Lutzomyia vexator and Lutzomyia stewarti (Diptera: Psychodidae).

Evolutionary theory predicts that virulence of parasites for mobile vector insects will be low for natural parasite-host associations that have coevolved. I determined virulence of the malaria parasite of lizards, Plasmodium mexicanum, for its vectors, two species of sand fly (Diptera: Psychodidae), Lutzomyia vexator (Coquillett 1907) and Lutzomyia stewarti (Mangabeira Fo & Galindo 1944), by measuring several life history traits. Developmental rate from egg to eclosion differed for the two species when noninfected. For both sand fly species, developmental rate for each stage (egg to larval hat

Polyamine homoeostasis as a drug target in pathogenic protozoa: peculiarities and possibilities.

New drugs are urgently needed for the treatment of tropical and subtropical parasitic diseases, such as African sleeping sickness, Chagas' disease, leishmaniasis and malaria. Enzymes in polyamine biosynthesis and thiol metabolism, as well as polyamine transporters, are potential drug targets within these organisms. In the present review, the current knowledge of unique properties of polyamine metabolism in these parasites is outlined. These properties include prozyme regulation of AdoMetDC (S-adenosylmethionine decarboxylase) activity in trypanosomatids, co-expression of ODC (ornithine decarbo

Morphological features of Cloacotaenia megalops (Nitzsch in Creplin, 1829) (Cestoda, Hymenolepididae) from different hosts.

The tapeworm species Cloacotaenia megalops (Nitzsch in Creplin, 1829) is characterized by a very specific morphology. A particularly distinct feature is a large almost square scolex in which four fleshy suction cups are embedded and a rudimentary rostellum. During standard studies of wild duck cestode fauna in northwestern Poland, some morphological differences were observed among specimens, mainly in the shape of scolices and hermaphroditic proglottids of C. megalops isolated from different bird species. This paper attempts to demonstrate the impact of the host species on morphology of the pa

[Impact of host factors on the schistosome-killing process induced by praziquantel].

Experimental studies indicated that the killing process of schistosomes induced by praziquantel comprises two aspects, i.e. the direct effect of praziquantel on schistosomes and the host immune reaction. The former one appears in stimulation of worm activity, spasmodic contraction of worm musculatures and severe damage to the tegument, which results in hepatic shift of schistosomes, influence on the nutrition absorption, excretion/secretion and defense function of the tegument, followed by the secondary interference with the worm metabolism. While the latter one involves the destruction of the

Immunogenomic approaches to study host immunity to enteric pathogens.

With increasing consumer demands for safe poultry products, effective control of disease-causing pathogens is becoming a major challenge to the poultry industry. Many chicken pathogens enter the host through the gastrointestinal tract, and over the past few decades, in-feed antibiotics and active vaccination have been the 2 main mechanisms of disease control. However, increasing public concerns are prompting government regulations on the use of growth-promoting drugs in animal production, and the ability of current vaccines to protect against emerging hypervirulent strains of pathogens is beco

Kinetic modeling of Toxoplasma gondii invasion.

The phylum Apicomplexa includes parasites responsible for global scourges such as malaria, cryptosporidiosis, and toxoplasmosis. Parasites in this phylum reproduce inside the cells of their hosts, making invasion of host cells an essential step of their life cycle. Characterizing the stages of host-cell invasion, has traditionally involved tedious microscopic observations of individual parasites over time. As an alternative, we introduce the use of compartment models for interpreting data collected from snapshots of synchronized populations of invading parasites. Parameters of the model are es

Ectoparasites and age-dependent survival in a desert rodent.

Host age is one of the key factors in host-parasite relationships as it possibly affects infestation levels, parasite-induced mortality of a host, and parasite distribution among host individuals. We tested two alternative hypotheses about infestation pattern and survival under parasitism in relation to host age. The first hypothesis assumes that parasites are recruited faster than they die and, thus, suggests that adult hosts will show higher infestation levels than juveniles because the former have more time to accumulate parasites. The second hypothesis assumes that parasites die faster tha

Biology and genome of Trichinella spiralis.

Clade I nematode species in the genus Trichinella can cause infections in humans that lead to mortality and serious morbidity. There are currently eight recognized species or genotypes that comprise this genus. The species display diverse biological characteristics, the evolutionary significance of which recently has been extensively clarified. Some of that diversity translates into variable importance as zoonotic pathogens, with T. spiralis having the highest significance. Trichinellosis has re-emerged as an important zoonotic infection in various parts of the world, reminding us that control

A note on Hyalomma nitidum, its distribution and its hosts.

The paper summarises the morphological characteristics, the known geographical distribution and hosts of Hyalomma nitidum, a tick which has never been properly characterised. H. nitidum was first described in Cameroon in 1919, long considered as a synonym of Hyalomma truncatum and recognised as a separate and valid species only since the early 1970s. H. nitidum occurs from Senegal to the Central African Republic in more humid conditions than H. truncatum, roughly to the south of the isohyet of 900 mm. Adults are parasitic on various wild and domestic ungulates, on which immatures have not been

Olfactory regulation of mosquito-host interactions.

Mosquitoes that act as disease vectors rely upon olfactory cues to direct several important behaviors that are fundamentally involved in establishing their overall vectorial capacity. Of these, the propensity to select humans for blood feeding is arguably the most important of these olfactory driven behaviors in so far as it significantly contributes to the ability of these mosquitoes to transmit pathogens that cause diseases such as dengue, yellow fever and most significantly human malaria. Here, we review significant advances in behavioral, physiological and molecular investigations into mos
