Effect of injections of contrast media on regional uptake of (14C)-2-deoxyglucose by the rat brain.

Mechanisms contributing to the rare but consistent neurotoxicity of contrast media currently in clinical use for the radiological examination of the subarachnoid space remain to be isolated. We assessed, by means of the (14C)-2-deoxy-D-glucose (2-DG) autoradiographic method, the effect of three non-ionic, low-osmolar contrast media, namely metrizamide, iopamidol and iohexol, on the local cerebral glucose utilization in the rat brain after intracisternal application. A significant (-30%) global reduction of the brain's metabolic activity occurred following intracisternal metrizamide injection.

Increase in the blood pressure and decrease in the norepinephrine release in the ventrolateral medulla during intraventricular administration of hypertonic NaCl.

Norepinephrine (NE) release in the ventrolateral medulla (VLM) was serially measured in anesthetized male Wistar rats during the rise in the blood pressure (BP) produced by acute intraventricular (ICV) administration of hypertonic (1.5 M) NaCl. Catecholamine release was determined by a brain microdialysis method using high performance liquid chromatography and electrochemical detector. The release of NE in the VLM was significantly decreased after ICV 1.5 M NaCl. In another set of rats, the pressor response to acute ICV 1.5 M NaCl was attenuated by selective administration of NE to the VLM usi

Stimulation of central alpha-2 adrenoceptors inhibits gastric secretion in rats by releasing vasopressin.

The mechanism of the gastric antisecretory action of the stimulation of central alpha-2 adrenoceptors were studied in conscious, pylorus-ligated rats using intracerebroventricularly (i.c.v.) administered oxymetazoline as the model substance. I.c.v. administration of 10 micrograms of oxymetazoline strongly inhibited the secretion of acid, pepsin and fluid, whereas upon s.c. injection this dose was without any effect. Pretreatment with idazoxan abolished the antisecretory effect of i.c.v. administered oxymetazoline. I.c.v. injected oxymetazoline inhibited gastric secretion induced by carbachol i

Regional differentiation of sympathetic nerve activity during fever caused by intracerebroventricular injection of PGE2.

In urethane-anesthetized rabbits prostaglandin E2 (PGE2) injected into a lateral cerebral ventricle (icv) produced hyperthermia. During the phase of rising rectal temperature, renal sympathetic activity monitored by multi-unit recording was reduced while the drop of ear skin temperature indicated cutaneous sympathetic activation. These reciprocal changes in activity corresponded to those typical for cold stress as well as for the phase of rising body temperature in fever induced by endotoxic lipopolysaccharides (LPS). However, a slight early stimulation of the heart rate after icv PGE2, contra

Water drinking caused by intracerebroventricular infusions of hypertonic solutions in cattle.

Infusions into the lateral cerebral ventricle of hypertonic solutions of NaCl, mannitol or sucrose all induced water drinking in cattle. However, infusion of hypertonic NaCl caused a significantly greater water drinking response than did the infusions of mannitol or sucrose, despite the fact that CSF osmolality increase was similar. In contrast, hypertonic solutions of NaCl or mannitol had similar dipsogenic effects when infused intravenously. The intracerebroventricular infusions of hypertonic NaCl or mannitol did not affect the intakes of food or Na solution. The results are consistent with

Periventricular noradrenergic systems are critical for angiotensin-induced drinking and blood pressure responses.

Deficits in experimentally induced drinking and pressor responses after administration of 6-hydroxydopamine (6-OHDA) into the lateral cerebral ventricle (LCV) have been reported. Questions have arisen, however, as to whether these effects are due to non-specific actions of the neurotoxin and, if specific, whether the depletion of dopamine (DA) or of norepinephrine (NE) is the critical factor for producing the impairments. In the present report methods to deplete brain catecholamine (CA) differentially were employed in order to test the hypothesis that central 6-OHDA injections act on brain CA

Central effects of adenosine analogs on blood pressure and heart rate in the mouse.

Mice implanted with chronic indwelling cannulae were injected into the lateral cerebral ventricle with two adenosine analogs, NECA and L-PIA, and the effects on blood pressure and heart rate recorded. Both analogs produced dose-related reductions in blood pressure and heart rate. NECA exhibited approximately 10 fold more potency than L-PIA on mean arterial blood pressure. The effects of both drugs on blood pressure and heart rate were antagonized by parenteral injections of caffeine. These results show that injections of adenosine analogs into the lateral ventricle of mice can influence the ar

Nature of hypo- and hyperthermia induced by the calcium antagonist nicardipine.

The effects of nicardipine, a calcium channel blocking agent, injected into the cerebral ventricles, (i.c.v.), on the body temperature of unanaesthetized cats have been investigated. Nicardipine produced a biphasic effect on body temperature: a transient dose-dependent decline followed by a longlasting elevation. The fall, but not the rise, of body temperature was associated with a dose-dependent increase in respiration. Yohimbine, in small doses, but not prazosin and propranolol, when injected into the cerebral ventricles, attenuated the hypothermia evoked by i.c.v. nicardipine. However, all

Role of the central muscarinic receptor of prostaglandin I2 in cardiovascular function in rat.

The effects of injection of prostaglandin (PG) I2 into the cerebral ventricle on cardiovascular responses and their modification by intracerebroventricular (i.c.v.) pretreatment with several drugs were studied in male Wistar rats under urethane anesthesia. When injected into the ventricle, PG I2 (50 nmol/kg) caused a decrease in blood pressure and an increase in heart rate which were of short duration and were significantly inhibited by i.c.v. pretreatment with drugs, such as atropine (0.4 mumol/kg) and diphenhydramine (1 mumol/kg), having an anticholinergic effect. The similar effects induced

Response of local blood flow in the caudate nucleus of the cat to intraventricular administration of carbachol.

The effect of perfusion of the cerebral ventricles with artificial cerebrospinal fluid containing carbachol on the blood flow in the caudate nucleus of the cat and the possibility to inhibit this effect by anticholinergic drugs was studied by means of the hydrogen clearance technique. After a control period during which both lateral ventricles were perfused with artificial CSF of identical composition, the drug under study was added on one side (experimental side) while the other side continued to be perfused with the control artificial CSF (control side). The blood flow on the experimental si

[Effect of 6-hydroxydopamine and 5,6-Dihydroxytryptophan on the luliberin and luteinizing hormone levels in female rats under constant illumination].

Administration of 5,6-oxytryptophan to female rats' 3rd cerebral ventricle, as opposed to 6-oxydopamine, prevents the decrease in the luteinizing hormone contents in the blood occurring under constant illumination. Administration of 6-oxydopamine against the background of desmethylimipramine caused a significant increase of the luliberine contents in the anterior hypothalamus while preserving its normal level in the medio-basal portion.

Sodium appetite elicited by intracerebroventricular infusion of angiotensin II in the rat: II. Synergistic interaction with systemic mineralocorticoids.

Angiotensin and mineralocorticoids, the hormones of sodium conservation, acted together to arouse a sodium appetite with shorter latency and greater magnitude than is produced by larger amounts of each acting alone. This potentiation was selective for sodium ingestion and occurred in the absence of significant changes in sodium balance. Therefore, because endogenous angiotensin and mineralocorticoids are concurrently elevated during sodium deficiency, sodium appetite may be aroused by a synergy of the peptide and the steroid.

Brain stem infusion of the gamma-aminobutyric acid antagonist bicuculline increases plasma insulin levels in the rat.

Previous neuroanatomical and physiological studies have indicated that nucleus ambiguus (Amb) is one source of vagal motoneurons in the brain stem that innervates the pancreas and which, when stimulated, increases insulin release. To investigate one of the neurotransmitter inputs to Amb neurons and its relation to insulin secretion, bicuculline, a specific gamma-aminobutyric acid (GABA) antagonist, was infused bilaterally into the Amb region as well as into a neighboring area, the rostral level of the lateral nucleus tractus solitarius (nts) of anesthetized male rats. Experiments were carried

The effects of paraoxon on blood pressure in the anaesthetized and in the conscious rat.

1 Intravenous administration of paraoxon (150-825 mug/kg) to anaesthetized rats induced long-lasting, dose-dependent pressor effects. Only after injection of 825 mug/kg paraoxon was the pressor response followed by a depressor effect and a bradycardia that could be blocked by N-methylatropine. Intracerebroventricular injection of paraoxon into anaesthetized rats also induced pressor effects.2 In order to elucidate the mechanism of the pressor action rats were given dexetimide, N-methylatropine, mecamylamine, phentolamine, prazosin, yohimbine, atenolol and metoprolol. If treatment with these dr

Muscimol induces gastric acid secretion after central administration.

Intracerebroventricular administration of the GABA agonist, muscimol, resulted in a dose-dependent increase in gastric acid secretion in pylorus ligated rats. The maximum increase in gastric acid secretion occurred in the second hour. The GABA antagonist, bicuculline methiodide, reversed the muscimol effect on gastric acid secretion. Intracerebroventricular administration of D-alanine methionine enkephalin, bombesin and calcitonin significantly suppressed the muscimol-induced gastric acid secretion. The present study suggests that the GABAergic system plays a role in the central control of gas

The role of the lymphatic system in drainage of cerebrospinal fluid and aqueous humour.

1. The jugular lymphatic trunks were cannulated in anaesthetized rabbits and cats. Over 6-8 hr, the mean lymph flow was 2.3 microliters min-1 in the rabbit (one side only) and 5.0 microliters min-1 in the cat (sum of both sides). 2. After a single injection of radio-iodinated albumin into a lateral cerebral ventricle without significant rise in pressure, a mean of 14.4% of the radioactivity was recovered in deep cervical lymph of one side in the rabbit and of 12.9% in that of both sides in the cat. 3. During slow infusion of [125I]albumin and fluorescent dextran of 150,000 mol. wt. into a late

Catecholaminergic alpha-receptors and shivering in the rat.

1. A rise or a fall in systemic blood pressure brought about by the I.V. infusion of peripherally acting drugs (adenosine, noradrenaline or methoxamine) inhibited shivering in the cold-exposed rat. 2. Since the injection of commonly used doses of noradrenaline (0.05-0.10 mumole) into a lateral cerebral ventricle of a rat was usually accompanied by a rise in blood pressure special precautious were required to determine whether noradrenaline had a specific central effect on shivering. 3.
