Human antibody responses to latent membrane protein 1 (LMP1) in patients with Epstein-Barr virus (EBV)-related disease syndromes were analyzed in detail. Only by immunoblot analysis with purified recombinant LMP1 and by IFA on recombinant LMP1-expressing insect cells could human antibodies directed against LMP1 be detected. Low serum levels of LMP1-directed antibodies could be detected in 3 of 8 EBV-positive Hodgkin's disease patients, 3 of 40 nasopharyngeal carcinoma patients, 2 of 23 Burkitt's lymphoma patients, and 1 of 27 non-Burkitt's lymphoma patients. No LMP1-directed antibodies could b

Hormonal signaling pleiotropically regulates insect physiological processes, including development, reproduction, and immunity. Juvenile hormone (JH) and its receptor Methoprene-tolerant (Met) modulate immune responses across many insect species. Our previous studies demonstrated that mating behavior in Nilaparvata lugens induces the expression of defensin genes NlDfA/B, enhancing resistance to pathogens. Furthermore, the abundance of yeast-like symbionts (YLS) is closely linked to the reproduction-immunity trade-off. However, the regulatory mechanism underlying the dynamic changes in NlDfA/B

The superworm Zophobas morio, a coleopteran beetle, is widely distributed, adapted to environments with decaying organic material and a well-known secondary pest of stored commodities. The larval stage is found to inhabit in putrid conditions, and so are expected to possess distinct immune adaptations vital for their survival against microbial infections in such environments. Using transcriptome analysis of the entire gut tissue of Z. morio, this work systematically surveyed distinct transcripts involved in various pathways of the innate immune system on a comparative account. Olfactory learning in blood-feeding insects, such as mosquitoes, could play an important role in host preference and disease transmission. However, standardised protocols allowing testing of their learning abilities are currently lacking, and how different olfactory stimuli are learned by these insects remains unknown. Using a Pavlovian conditioning paradigm, we trained individuals and groups of Aedes aegypti mosquitoes to associate an odorant conditioned stimulus (CS) with a blood-reinforced thermal stimulus (unconditioned stimulus; US). Results showed, first, that mosquitoes could learn the

Hytrosaviridae is a proposed virus family encompassing viruses that cause salivary gland hypertrophy (SGH) syndrome in infected insects and reduce the fertility in their dipteran insect hosts. They contain a large, double stranded DNA genome of 120-190 kbp. To date, these viruses have been detected only in adult Diptera. These include hytrosaviruses detected in various tsetse fly species (Glossina spp.), the narcissus bulb fly Merodon equestris and the house fly Musca domestica. The limited number of hytrosaviruses reported to date may be a reflection of the frequent absence of external sympto

The aquaculture industry is exploring sustainable alternatives to fishmeal, with insect meals (IMs) emerging as promising options due to their high-quality proteins, lipids, and immunostimulatory compounds, such as chitin. This study evaluated how full-fat IMs affect the histomorphometry of Atlantic salmon (Salmo salar) mucosal tissues (intestine, skin, and gills), their barrier function, and immune gene expression to explore their immunomodulatory potential. Two insect species were tested: black soldier fly, Hermetia illucens (HI), and yellow mealworm, Tenebrio molitor (TM), formulating into

Plant-based diets have gained increasing attention in the management of chronic kidney disease (CKD), yet concerns persist regarding the quality of plant-derived proteins. Traditionally, animal-based proteins have been regarded as high biological value proteins, while plant proteins have been labeled as low biological value, contributing to misconceptions about their nutritional adequacy. In this narrative review, we challenge these assumptions by presenting emerging evidence on the nutritional quality of plant proteins, strategies to enhance their amino acid profiles, and their relevance in t

With rising demand for sustainable protein sources, insect proteins have gained attention for their nutritional value and environmental advantages. Silkworm pupae protein (SPP) is rich in essential amino acids and bioactives but has limited application due to poor solubility and high allergenicity. This study employed Lactiplantibacillus plantarum A2 (A2) to ferment SPP and evaluated structural, functional, and allergenic changes in vitro and in mice. Fermentation increased α-helix content, surface hydrophobicity, and sulfhydryl groups while reducing β-sheet content and fluorescence. Hydrolysi…

Transposable elements are abundant in host genomes but are generally considered to be confined to the cell in which they are expressed, with the notable exception of endogenous retroviruses. Here, we identify a group of LTR retrotransposons that infect the germline from somatic cells within the Drosophila ovary, despite lacking the fusogenic Envelope protein typically required for retroviral entry. Instead, these elements encode a short transmembrane protein, sORF2, with structural features reminiscent of viral cell-cell fusogens. Through genetics, imaging, and electron microscopy, we show tha

Odorant binding proteins (OBPs) play a critical role in insect survival by facilitating the detection of environmental odors. This study identified and characterized OcomOBP11 in the leaf beetle Ophraella communa Lesage. Sequence and phylogenetic analyses demonstrated that OcomOBP11 belongs to the Minus-C OBP family. Tissue-specific expression analysis revealed that OcomOBP11 showed the highest relative expression in the testis. Functional assays demonstrated that OcomOBP11 has strong binding affinity to 24 of the 26 volatiles from Ambrosia artemisiifolia L., a primary host plant. Molecular do

Oculocutaneous albinism type 1 (OCA1) is an autosomal recessive disorder caused by mutations in the tyrosinase gene. Two subtypes of OCA1 have been described: severe OCA1A with complete absence of tyrosinase activity and less severe OCA1B with residual tyrosinase activity. Here, we characterize the recombinant human tyrosinase intramelanosomal domain and mutant variants, which mimic genetic changes in both subtypes of OCA1 patients. Proteins were prepared using site-directed mutagenesis, expressed in insect larvae, purified by chromatography, and characterized by enzymatic activities, tryptoph

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus that causes severe arthralgia. The envelope of CHIKV is composed of 240 copies of two glycoproteins: E1 and E2. In this work, we have characterized the N-glycosylation patterns of CHIKV virus-like particles (VLPs), containing both E1 and E2 proteins, derived from mammalian and insect cells using hydrophilic interaction liquid chromatography (HILIC) with fluorescence (FL) and mass spectrometry (MS) detection. While HEK293 derived CHIKV VLPs contain oligomannose, hybrid and complex glycans, VLPs derived from SfBasic predominantly contain o

Somatic reproductive tissues, such as the male accessory glands in insects, are critical for insect fertility. However, the signaling pathways that maintain accessory gland homeostasis remain poorly understood. Using tissue-specific knockdown or overexpression in Drosophila, we modulated insulin signaling pathway genes in male accessory glands and examined their effects on male fertility. The impact of FOXO overexpression on gland structure and function was studied by analyzing cellular integrity, expression of genes encoding accessory gland proteins, and autophagy markers. Additionally, we ex

Most problems caused by the use of common pesticides can be avoided or reduced by the use of well-established, safe, unique, and selective organic compounds. For example, juvenile hormone analogues exhibit promise as insect growth regulators (IGRs) due to their effective mechanism of action against pests and lower toxicity to children compared to conventional pesticides. Thus, as analogues of insect growth regulators, a special class of pure 1-(1-naphthalenyloxy)-2-propanol derivatives has been created. Through elemental and modern spectroscopy investigations (infrared (IR), ultraviolet (UV),

Tsetse are the insects responsible for transmitting African trypanosomes, which cause sleeping sickness in humans and animal trypanosomiasis in wildlife and livestock. Knowing the age of these flies is important when assessing the effectiveness of vector control programs and modelling disease risk. Current methods to assess fly age are, however, labour-intensive, slow, and often inaccurate as skilled personnel are in short supply. Mid-infrared spectroscopy (MIRS), a fast and cost-effective tool to accurately estimate several biological traits of insects, offers a promising alternative. This is

Insect CAPA-PVK (periviscerokinin) and pyrokinin (PK) neuropeptides belong to the PRX family peptides and are produced from capa and pyrokinin genes. We identified and characterised the two genes from the western flower thrips, Frankliniella occidentalis. The capa gene transcribes three splice variants, capa-a, -b, and -c, encoding two CAPA-PVKs (EVQGLFPFPRVamide; QGLIPFPRVamide) and two PKs (ASWMPSSSPRLamide; DSASFTPRLamide). The pyrokinin mRNA encodes three PKs: DLVTQVLQPGQTGMWFGPRLamide, SEGNLVNFTPRLamide, and ESGEQPEDLEGSMGGAATSRQLRTDSEPTWGFSPRLamide, the most extended pheromone biosynthes

BACKGROUND: Andean lupin (Lupinus mutabilis Sweet) is an important leguminous crop from South America with a high protein content. In Ecuador, lupin yields are severely affected by the infestation of Delia platura larvae on germinating seeds. The application of elicitor molecules with activity against herbivorous insects to control D. platura infestation constitutes an unexplored and promising alternative for chemical insecticides. In this study, methyl jasmonate (MeJA), hexanoic acid, menadione sodium bisulfite, and DL-β-aminobutyric acid were evaluated for their ability to induce resistance

Spinosyns, including spinosad and spinetoram, act on the insect central nervous system, gradually paralyzing or destroying the target insect. Spinosad resistance is associated with loss-of-function mutations in the nicotinic acetylcholine receptor (nAChR) α6 subunit in a number of agricultural pests. Using gene editing, nAChR α6 has been verified as a target for spinosyns in five insect species. Recently, a point mutation (G275E) in exon 9 of nAChR α6 was identified in spinosad-resistant strains of Thrips palmi and Tuta absoluta. To date, no in vivo functional evidence has been obtained to sup

BACKGROUND: Adipokinetic hormones (AKHs) are metabolic regulators in insects, and many are still unknown. Bioinformatic analysis can assist entomologic research with predictions based on sequence similarity.
METHODS: AKHs are short peptides with specific sequence features, which were used together with the rules for preprohormone processing to interrogate National Center for Biotechnology Information (NCBI) genomic/transcriptomic data for novel putative AKH sequences by sequence alignment and similarity search. The structures of the signal peptide processing site and the dibasic processing si

Most alphaviruses are mosquito borne and exhibit a broad host range, infecting many different vertebrates, including birds, rodents, equids, humans, and nonhuman primates. Recently, a host-restricted, mosquito-borne alphavirus, Eilat virus (EILV), was described with an inability to infect vertebrate cells based on defective attachment and/or entry, as well as a lack of genomic RNA replication. We investigated the utilization of EILV recombinant technology as a vaccine platform against eastern (EEEV) and Venezuelan equine encephalitis viruses (VEEV), two important pathogens of humans and domest
