VU0810464, a selective GIRK channel activator, improves hippocampal-dependent synaptic plasticity and memory disrupted by amyloid-β oligomers.

Selective pharmacological activation of G protein-gated inwardly rectifying K⁺ (GIRK) channels -a key physiological determinant of neuronal excitability control- has proven highly effective in counteracting amyloid-β oligomer (oAβ)-induced hyperexcitability and hippocampal dysfunction in early Alzheimer's disease (AD). However, GIRK gain-of-function in healthy animals disrupts learning, memory and underlying synaptic plasticity, greatly limiting its therapeutic potential in preclinical asymptomatic AD patients. Therefore, GIRK-based pharmacological treatment needs further investigation to over

In the here and now: Short term memory predictions are preserved in Alzheimer's disease.

According to neuropsychological models of anosognosia, there is a failure to transfer on-line awareness of dysfunction into a more generalised long term belief about memory function in Alzheimer's disease. This failure results in specific metamemory deficits for global predictions: patients overestimate their performance before the task but are able to monitor their memory performance after having experienced the task. However, after a delay, they are still not able to make accurate predictions. As previous work has mainly focused on long-term memory, the present study investigates this issue

The neuroscience of positive memory deficits in depression.

Adults with unipolar depression typically show poor episodic memory for positive material, but the neuroscientific mechanisms responsible for this deficit have not been characterized. I suggest a simple hypothesis: weak memory for positive material in depression reflects disrupted communication between the mesolimbic dopamine pathway and medial temporal lobe (MTL) memory systems during encoding. This proposal draws on basic research showing that dopamine release in the hippocampus is critical for the transition from early- to late-phase long-term potentiation (LTP) that marks the conversion of

Evidence for superior encoding of detailed visual memories in deaf signers.

Recent evidence shows that deaf signers outperform hearing non-signers in some tests of visual attention and discrimination. Furthermore, they can retain visual information better over short periods, i.e., seconds. However, it is unknown if deaf signers' retention of detailed visual information is superior following more extended periods. We report a study investigating this possibility. Our data revealed that deaf individuals outperformed hearing people in a visual long-term memory test that probed the fine detail of new memories. Deaf individuals also performed better in a scene-discriminati

Classify head-turning in walking: Multi-segment IMU sensing with recurrent neural networks.

BACKGROUND: Head movement is a fundamental component of gait control and environmental orientation during locomotion. Wearable inertial measurement units (IMUs) enable real-time inference of upcoming movement, and early detection of impending head turns during walking is critical for enhancing interactive systems. RESEARCH QUESTIONS: The study aimed to investigate whether multi-segment IMU data reliably detect and classify upcoming head turns and their direction before and during head turns while walking, and to hypothesize that neural networks effectively capture the temporal integration pat

Multimodal deep learning for objective skill assessment in robot-assisted vesico-urethral anastomosis.

This study aimed to classify robot-assisted surgery (RAS) skill levels (inexperienced, competent, and experienced) during performance of vesico-urethral anastomosis (VUA) using multimodal physiological signals. We trained Convolutional Neural Networks (CNNs) and Long Short-Term Memory (LSTM) models on data collected from 23 RAS (RAS trainees and experienced surgeons) performing two VUAs on animal tissue. The dataset included 116-channel electroencephalogram (EEG) and 20 eye-tracking signals recorded during two VUA subtasks: (1) needle grasping, positioning, and entry; and (2) needle driving wi

Cytisine inhibits the protective activity of various classical and novel antiepileptic drugs against 6 Hz-induced psychomotor seizures in mice.

BACKGROUND: Cytisine (CYT) is a partial agonist of brain α4β2 nicotinic acetylcholine receptors widely used in Central/Eastern Europe for smoking cessation. OBJECTIVES: This study evaluated the effect of CYT on the ability of classical and novel antiepileptic drugs to prevent seizures evoked by the 6-Hz test, a model of psychomotor seizures in mice thought as a model of drug-resistant seizures. RESULTS: CYT administered intraperitoneally (i.p.) in a dose of 2 mg kg-1 significantly inhibited the anticonvulsant activity of lacosamide, levetiracetam, and pregabalin, increasing their median effe

Genetic risk for Alzheimer's disease alters the five-year trajectory of semantic memory activation in cognitively intact elders.

Healthy aging is associated with cognitive declines typically accompanied by increased task-related brain activity in comparison to younger counterparts. The Scaffolding Theory of Aging and Cognition (STAC) (Park and Reuter-Lorenz, 2009; Reuter-Lorenz and Park, 2014) posits that compensatory brain processes are responsible for maintaining normal cognitive performance in older adults, despite accumulation of aging-related neural damage. Cross-sectional studies indicate that cognitively intact elders at genetic risk for Alzheimer's disease (AD) demonstrate patterns of increased brain activity co

HIV- and FIV-derived gp120 alter spatial memory, LTP, and sleep in rats.

Human immunodeficiency virus (HIV)-associated dementia (HAD) has been detected in 20-30% of patients suffering AIDS. The envelope glycoprotein 120 (gp120) derived from HIV seems to play a critical role in the pathophysiology of this dementia. Likewise, the feline immunodeficiency virus (FIV)-derived gp120 causes neurological and electrophysiological abnormalitites in cats. We have studied the effects of gp120 derived from HIV or FIV on learning and memory processing, hippocampal long-term potentiation (LTP), hippocampal neuronal cAMP production, the sleep-waking cycle, and locomotor activity a

Emergent properties of neural systems: how focal molecular neurobiological alterations can affect behavior.

Whereas the basic wiring diagram of the mammalian central nervous system (CNS) is genetically preprogramed, its fine tuning throughout different phases of infancy, childhood, and adulthood are highly experience dependent. The potential neurobiological mechanisms and behavioral effects of such experience-dependent neuroplasticity as a function of stage of development are outlined. A basic thesis of this paper is that mechanisms involved in neuronal learning and memory, such as long-term potentiation (LTP) and long-term depression (LTD), are used and reused not only in the sculpting of the CNS i

Hippocampal Protein Kinase C Signaling Mediates the Short-Term Memory Impairment Induced by Delta9-Tetrahydrocannabinol.

Cannabis affects cognitive performance through the activation of the endocannabinoid system, and the molecular mechanisms involved in this process are poorly understood. Using the novel object-recognition memory test in mice, we found that the main psychoactive component of cannabis, delta9-tetrahydrocannabinol (THC), alters short-term object-recognition memory specifically involving protein kinase C (PKC)-dependent signaling. Indeed, the systemic or intra-hippocampal pre-treatment with the PKC inhibitors prevented the short-term, but not the long-term, memory impairment induced by THC. In con

Are frontal and temporal lobe epilepsy dissociable in their memory functioning?

There is controversy in the literature as to how dissociable frontal lobe epilepsy (FLE) and temporal lobe epilepsy (TLE) are in terms of memory deficits. Some researchers have demonstrated that FLE is associated with greater executive dysfunction including working memory, whereas TLE is associated with greater memory impairment. Others have found the two groups to be comparable in memory functioning. Hence, we examined this question in children with FLE and TLE versus typically developing controls. We found most of the expected effects when the groups with focal onset epilepsy were compared t

Human antibody and memory B and T-cell responses after primary and booster immunisation against Neisseria meningitidis B.

Vaccination against disease aims at the induction of long-lasting cellular and humoral immune responses. Few studies have addressed the mechanisms by which meningococcal vaccines generate and sustain immunological memory. The goal of this study was to investigate the development of long-term humoral and cellular memory to Neisseria meningitidis serogroup B (MenB) in health subjects after immunisation with the Cuban outer membrane protein (OMP) vaccine (VA-MENGOC-BC). The results showed that three doses of vaccine were necessary to induce a detectable memory B-cell response (mean of 0.46%) whic

Graph Neural Networks and sequential architectures for autism detection from eye-tracking biomarkers: a multi-site study.

BACKGROUND: Autism Spectrum Disorder (ASD) affects 1 in 100 children globally, yet early detection remains challenging due to reliance on subjective behavioral assessments and limited specialist availability, particularly in resource-constrained settings. Eye-tracking biomarkers offer objective, quantitative measurements of visual attention patterns, but existing Neural Network approaches lack systematic evaluation across diverse neural network architectures and multi-site data. This study provides a comprehensive comparison of Graph Neural Networks, Long Short-Term Memory networks, and Multil

Hypergraph-Based Audio-Visual Fusion for Obstructive Sleep Apnea Severity Estimation During Wakefulness.

Obstructive sleep apnea (OSA) is associated with psychophysiological impairments, and recent studies have shown the feasibility of using speech and craniofacial images during wakefulness for severity estimation. However, the inherent limitations of unimodal data constrain the performance of current methods. To address this, we proposed a novel hypergraph-based multimodal fusion framework (HMFusion) that integrates psychophysiological information from audio-visual data. Specifically, we employ long short-term memory (LSTM)-based encoders to extract modality-specific temporal dynamics from pre-t

Integrating deep-learning methods and web-based data sources for surveillance, forecasting and early warning of avian influenza.

Highly pathogenic avian influenza (HPAI), especially the H5N1 subtype has caused repeated global outbreaks, primarily affecting birds, but occasionally spreading between humans. These events pose serious public health and economic risks, demanding enhanced surveillance. This study evaluates novel web-based data for predicting HPAI outbreaks using machine learning models in Canada as a case study. Seven web-based sources, Google Trends, Google News, Global Database of Events, Language, and Tone (GDELT), Reddit, Facebook, minimum temperature and air quality (UV index and CO levels), were automat

Early detection of mental health on social media using a hybrid Bi-LSTM-XGBoost model: a comparative study.

The case of mental health disorders has been a main topic in the clinical and psychological field. The advancement of computing studies, especially in Natural Language Processing (NLP)-a subset of Machine Learning, created a system of detection that can detect the mental health state of a person in early stage to prevent the eventuality of the worst case. This is crucial since there has been a lot of case of mental health disorder-such as depression and suicide, remains undetected and untreated-especially when the internet usage is more prevalent than ever even among the most vulnerable users,

Time Distributed Classification of Alzheimer's Disease on MRI Scans.

The diagnosis of Alzheimer's disease (AD) has progressively depended on sophisticated neuroimaging methods alongside cognitive assessments. This study combines volumetric feature analysis with computational modeling techniques, focusing on spatial and temporal analysis, to categorize individuals as cognitively normal (CN), mild cognitive impairment (MCI), or AD using magnetic resonance imaging (MRI) data. In the initial phase, volumetric changes, comprising cortical thickness, white matter, grey matter, cerebrospinal fluid, and total intracranial volume, were derived from the Alzheimer's Disea
