Dynamic perturbation analysis: a modular rewiring framework for decoding cancer stage progression and identifying therapeutic vulnerabilities.

Disease progression is driven not only by transcriptional alterations but also by dynamic reorganization of molecular interaction networks. However, most existing approaches focus on differential gene expression and fail to capture modular-level reorganization. Here, we present dynamic perturbation analysis (DPA), a modular rewiring framework that quantifies dynamic perturbations across disease stages using a novel metric termed the degree of perturbation. Using gastric cancer (GC) progression as a model system, DPA revealed that extensive modular rewiring persists despite limited stage-specif

Functionality of platelets after in vitro testing of pulsatile total artificial hearts.

Hemolysis is a metric of hemocompatibility in total artificial hearts (TAHs) but provides limited insight into platelet biology and thrombogenicity. We demonstrated reduced hemolysis with Realheart® TAH (RH) compared to FDA-approved SynCardia TAH (Sync). Here, we evaluated platelet function following exposure to both devices in a whole-blood in vitro system using a standardized ASTM F1830 protocol. Human blood (n = 5) was circulated for 360 min under controlled conditions (5 L/min, 100 mmHg, 37 °C) in RH, Sync, while matched static controls were maintained under identical temperature condition

Development of a dual-energy CBCT calibration method for accurate electron density estimation in preclinical small-animal proton therapy dose planning.

In preclinical animal studies for proton therapy, Computer Tomography (CT) imaging is essential for planning and proton dose calculation, which requires accurate conversion of Hounsfield Unit (HU) to stopping power ratio (SPR). While single-energy CT (SECT) is traditionally used, dual-energy CT (DECT) offers improved accuracy in estimating SPR. This study-the first animal dual-energy cone-beam CT (CBCT) study for proton therapy-developed a DECT calibration method for a small-animal CBCT system using a CIRS 062M electron density phantom scanned at 40 and 60 kVp. SECT calibration applied a stoic

Molecular Insights into Adsorption Mechanisms of Micro- and Nanoplastics on Effective Adsorbent Materials.

Existing reviews on micro- and nanoplastic (MNP) removal from water rarely link adsorbent structural features to the molecular interactions governing removal performance. This review addresses this gap by examining MNP adsorption from a mechanism-oriented perspective, mapping six canonical interaction pathways across five adsorbent classes. Adsorption emerges as a system-dependent process governed by the interplay between polymer properties and surface chemistry rather than by the material alone. Interactions such as π-π stacking and hydrophobic affinity dominate for non-functionalized polymer

Eye-Tracking Based Measurement Framework For Quantifying Visual Attention In Screen-Based Interactive Digital Art Environments.

This article presents a gaze-enabled measurement framework designed to quantify visual attention during interactive digital art experiences and to link attention dynamics to experiential outcomes. Unlike traditional static art viewing, interactive digital art involves dynamic media, multi-source information, and continuous feedback loops, which challenge standard eye-tracking methodologies. The proposed system integrates eye-tracking acquisition, interaction logging, timestamp synchronization, dynamic Area of Interest (AOI) mapping, and metric computation into a unified pipeline. The protocol

Computer Vision-Assisted Semiautomatic Analysis of Zooplankton in a Longitudinal Study of the Ecological State of Lake Baikal.

Studying zooplankton in freshwater ecosystems is crucial for ecological research, providing insight into ecosystem health, biodiversity, and water quality. This study focuses on developing a hybrid approach for studying and analyzing zooplankton communities using machine learning and human expert analysis. The goal of the study was to automate the labor-intensive process of zooplankton analysis as part of a long-term Lake Baikal monitoring program (since 1945), while maintaining continuity with traditional methods. A software and algorithmic system were developed to automate the analysis: imag

Normalization of GC-MS Metabolomics Data in Adherent Cells: A Practical Comparison of Approaches.

Data compatibility remains a major challenge in metabolomics, as commonly used measures of biological material-such as sample weight or cell count-are often poorly reproducible. Here, we systematically evaluated practical normalization strategies for GC × GC-MS-based metabolomic profiling of two widely used model cell lines: human hepatoblastoma (HepG2) and mesenchymal stromal cells (MSCs). We compared orthogonal biomass estimates, including total protein and double-stranded DNA quantified either directly in aliquots of the cell suspension lysate aliquots or in the post-extraction cell precipi

Reducing CAUTI incidence: A multimodal and multidisciplinary approach for improved prevention and patient outcomes.

BACKGROUND: Catheter-associated urinary tract infections (CAUTIs) are common health care-associated infections and a key quality metric tied to patient safety. Although the standardized utilization ratio (SUR) declined, CAUTI rates remained above the target benchmark at a large academic trauma facility, prompting a multidisciplinary quality improvement initiative. METHODS: A multidisciplinary team implemented a multimodal approach targeting indwelling urinary catheter practices. Interventions included standardized urine culture protocols, education on dependent loop prevention, 2 person inser

Bioanalysis of monodisperse HO-PEG8-OH polymers in MCF-7 cells by UHPLC-MS/MS coupled with μ-SPE to improve greenness and sensitivity.

Monodisperse polyethylene glycol (PEG) derivatives offer significant advantages over conventional polydisperse PEGs for biomedical applications due to their precisely defined molecular structures. This study establishes an eco-efficient, selective, and sensitive analytical assay integrating microscale solid-phase extraction (μ-SPE) with ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) to investigate the cellular uptake of HO-PEG8-OH polymers in MCF-7 cells. The method achieved greater than 91% recovery from 20 μL lysates using M-PEG6-OH as the internal standard, wi

Vertical Jump Height Estimation Using Low-Sampling IMU in Countermovement Jumps: A Feasible Alternative to Motion Capture and Force Platforms.

Vertical jump height from a countermovement jump is a widespread metric to assess the lower limb functionality. Motion capture systems and force platforms are considered gold standards to estimate vertical jump height; however, their use in ecological settings is limited. This study aimed to evaluate the feasibility of low-sampling-rate inertial measurement units as an alternative to the gold standard systems. The validity of three computational methods for IMU-based data-numerical double integration, takeoff velocity, and flight time-was assessed using data from 18 healthy participants who pe

Classification of Breast Lymphedema in a Racially Diverse Cohort.

Breast lymphedema is a common sequela of breast conservation that delays healing and reduces quality of life. No rigorous classification system exists for this condition. We explored approaches for classifying breast lymph-edema based on breast ultrasound, physical exam, and patient-reported outcomes. We enrolled 80 patients from two institutions. Each site enrolled 30 invasive breast cancer patients treated with breast conservation and radiotherapy, and 10 control patients evaluated for benign breast complaints. All patients underwent bilateral breast ultrasound to measure dermal thickness an

A foundation model for clinician-centered drug repurposing.

Drug repurposing-identifying new therapeutic uses for approved drugs-is often a serendipitous and opportunistic endeavour to expand the use of drugs for new diseases. The clinical utility of drug-repurposing artificial intelligence (AI) models remains limited because these models focus narrowly on diseases for which some drugs already exist. Here we introduce TxGNN, a graph foundation model for zero-shot drug repurposing, identifying therapeutic candidates even for diseases with limited treatment options or no existing drugs. Trained on a medical knowledge graph, TxGNN uses a graph neural netw

Poor ovarian response classification systems in the clinical setting - time for an update?

PURPOSE OF REVIEW: Poor ovarian response (POR) remains a key challenge to the success of assisted reproductive technology. Here, we offer a comprehensive review of the two main classification systems for POR, discussing their promises and pitfalls, evaluating their performance, and exploring potential avenues for improving upon these definitions of POR. RECENT FINDINGS: The Bologna criteria represented the first meaningful attempt to create a universal POR definition. Subsequently, the POSEIDON classification system was published to provide a more nuanced view of POR, classifying patients int

Beyond Correlation: Incorporating Counterfactual Guidance to Better Support Exploratory Visual Analysis.

Providing effective guidance for users has long been an important and challenging task for efficient exploratory visual analytics, especially when selecting variables for visualization in high-dimensional datasets. Correlation is the most widely applied metric for guidance in statistical and analytical tools, however a reliance on correlation may lead users towards false positives when interpreting causal relations in the data. In this work, inspired by prior insights on the benefits of counterfactual visualization in supporting visual causal inference, we propose a novel, simple, and efficien

Automated detection of COVID-19 through convolutional neural network using chest x-ray images.

The COVID-19 epidemic has a catastrophic impact on global well-being and public health. More than 27 million confirmed cases have been reported worldwide until now. Due to the growing number of confirmed cases, and challenges to the variations of the COVID-19, timely and accurate classification of healthy and infected patients is essential to control and treat COVID-19. We aim to develop a deep learning-based system for the persuasive classification and reliable detection of COVID-19 using chest radiography. Firstly, we evaluate the performance of various state-of-the-art convolutional neural

Development and Intensification of the Ethylene Process Utilizing a Catalytic Membrane Reactor.

Ethylene is considered the most important petrochemical constituent in the world today. It is currently produced via the thermal cracking process, which is generally expensive. Ethane dehydrogenation (EDH) is endothermic, and the thermodynamic equilibrium limits its conversion. The present study explores the viability of using a catalytic membrane reactor (MR) for ethylene production from EDH. The removal of hydrogen from the reaction zone using a palladium-silver (Pd-Ag) membrane has led to a high shift in the equilibrium conversion. The effects of operating conditions and reactor configurati

Fetal Age Assessment From Primary Teeth and Long Bones.

This study evaluated traditional and expedited methods for assessing the age of fetal remains. Because of their rare occurrence, the discovery of fresh, decomposing, disfigured, or skeletal fetuses engenders heightened awareness by forensic pathologists primarily tasked with age estimation in relation to viability. With decomposed complete or isolated fetal remains, dentists focus on primary molar mineralization, whereas anthropologists perform long bone measurements along with discernment of other indicators of skeletal maturity to obtain an age estimation. The results of this study are 4-fold

Surgical Guidance for Removal of Cholesteatoma Using a Multispectral 3D-Endoscope.

We develop a stereo-multispectral endoscopic prototype in which a filter-wheel is used for surgical guidance to remove cholesteatoma tissue in the middle ear. Cholesteatoma is a destructive proliferating tissue. The only treatment for this disease is surgery. Removal is a very demanding task, even for experienced surgeons. It is very difficult to distinguish between bone and cholesteatoma. In addition, it can even reoccur if not all tissue particles of the cholesteatoma are removed, which leads to undesirable follow-up operations. Therefore, we propose an image-based method that combines multi
