Acquisition of Host Cytosolic Protein by Toxoplasma gondii Bradyzoites.

Toxoplasma gondii is a protozoan parasite that persists in the central nervous system as intracellular chronic-stage bradyzoites that are encapsulated by a thick cyst wall. While the cyst wall separates bradyzoites from the host cytosol, it has been posited that small solutes can traverse the cyst wall to sustain bradyzoites. Recently, it was found that host cytosolic macromolecules can cross the parasitophorous vacuole and are ingested and digested by actively replicating acute-stage tachyzoites. However, the extent to which bradyzoites have an active ingestion pathway remained unknown. To in

Differential effects of noise exposure between substrains of CBA mice.

Noise trauma involves a plethora of mechanisms including reactive oxygen species, apoptosis, tissue damage, and inflammation. Recently, circadian mechanisms were also found to contribute to the vulnerability to noise trauma in mice, with greater damage occurring during their active phase (nighttime), when compared to similar noise exposures during their inactive phase (daytime). These effects seem to be regulated by mechanisms involving Bdnf responses to noise trauma and circulating levels of corticosterone (CORT). However, recent studies using different noise paradigms show contradicting resu

Lower level noise exposure that produces only TTS modulates the immune homeostasis of cochlear macrophages.

Noise exposure producing temporary threshold shifts (TTS) has been demonstrated to cause permanent changes to cochlear physiology and hearing function. Several explanations have been purported to underlie these long-term changes in cochlear function, such as damage to sensory cell stereocilia and synaptic connections between sensory cells and their innervation by spiral ganglion neurons, and demyelination of the auditory nerve. Though these structural defects have been implicated in hearing difficulty, cochlear responses to this stress damage remains poorly understood. Here, we report the acti

Enhanced autoimmunity associated with induction of tumor immunity in thyroiditis-susceptible mice.

BACKGROUND: Immunotherapeutic modalities to bolster tumor immunity by targeting specific sites of the immune network often result in immune dysregulation with adverse autoimmune sequelae. To understand the relative risk for opportunistic autoimmune disorders, we studied established breast cancer models in mice resistant to experimental autoimmune thyroiditis (EAT). EAT is a murine model of Hashimoto's thyroiditis, an autoimmune syndrome with established MHC class II control of susceptibility. The highly prevalent Hashimoto's thyroiditis is a prominent autoimmune sequela in immunotherapy, and i

A role for maternal physiological state in preserving auditory cortical plasticity for salient infant calls.

A growing interest in sensory system plasticity in the natural context of motherhood has created the need to investigate how intrinsic physiological state (e.g., hormonal, motivational, etc.) interacts with sensory experience to drive adaptive cortical plasticity for behaviorally relevant stimuli. Using a maternal mouse model of auditory cortical inhibitory plasticity for ultrasonic pup calls, we examined the role of pup care versus maternal physiological state in the long-term retention of this plasticity. Very recent experience caring for pups by Early Cocarers, which are virgins, produced s

Propionibacterium acnes CAMP factor and host acid sphingomyelinase contribute to bacterial virulence: potential targets for inflammatory acne treatment.

BACKGROUND: In the progression of acne vulgaris, the disruption of follicular epithelia by an over-growth of Propionibacterium acnes (P. acnes) permits the bacteria to spread and become in contact with various skin and immune cells. METHODOLOGY/PRINCIPAL FINDINGS: We have demonstrated in the present study that the Christie, Atkins, Munch-Peterson (CAMP) factor of P. acnes is a secretory protein with co-hemolytic activity with sphingomyelinase that can confer cytotoxicity to HaCaT keratinocytes and RAW264.7 macrophages. The CAMP factor from bacteria and acid sphingomyelinase (ASMase) from the

[Age-related characteristics of the melatonin-producing epiphyseal function in CBA mice immunized by T-dependent antigen].

It was investigated both melatonin blood level in the adult and old CBA-mice after immunization by sheep erythrocytes and the link between thymus and adrenal glands function in the old immunizated mice with high melatonin content in the pineal gland. It has been revealed that in the adult mice blood melatonin level was influenced by the phase changes in dynamic of immunization, namely raised significantly in 20 min with following suppression after 3 hrs and rehabilitation up to normal in 4 days. In the old mice fluctuations of melatonin level were impaired and characterized by gradually decrea

In vivo binding of complement regulator factor H by Streptococcus pneumoniae.

Pneumococcal surface protein C (PspC) binds to the complement regulatory protein factor H (FH), which inhibits alternative pathway activation. In the present study, using a mouse model of systemic infection and flow-cytometric analyses, we demonstrated an in vivo interaction between FH and pneumococci and showed differential FH binding during bacteremia. Flow-cytometric analyses of pneumococci harvested after intraperitoneal (ip) challenge demonstrated increased binding of FH, compared with that after intravenous (iv) challenge. Real-time polymerase chain reaction analyses of PspC mRNA showed

Protective effects of immunization with a recombinant cyst antigen in mouse models of infection with Toxoplasma gondii tissue cysts.

A portion of the major Toxoplasma gondii tissue cyst antigen (MAG1) was expressed in bacteria as a fusion to glutathione S-transferase (GST) and the purified fusion protein (rMAG1) was used to immunize mice in an attempt to induce protective immunity against challenge with live cysts from the T. gondii ME49 strain. Sixty percent of mice immunized with rMAG1 and challenged with 500 cysts of the T. gondii ME49 strain survived, while only 20% of mice immunized with GST alone survived, suggesting a protective effect specific to the MAG1 portion of the recombinant protein. In a model of chronic inf

In vivo virulence of Candida albicans isolates causing mucosal infections in people infected with the human immunodeficiency virus.

Mucosal candidiasis is common in human immunodeficiency virus (HIV) infection. Susceptibility to such infections may be attributed to reduced host defense mechanisms and/or virulence of the organism. In the present study, we compared the virulence of mucosal Candida albicans isolates from HIV-infected people, with and without fluconazole-refractory infection, in established murine models of systemic and vaginal candidiasis. Compared with the mortality rate ( approximately 70%) after intravenous challenge with 2 virulent reference isolates, challenge with most clinical isolates (66%-77%) result

Several different ion channel modulators abrogate contact hypersensitivity in mice.

A major obstacle in transdermal delivery of drugs is the development of adverse skin sensitization reactions. We tested the concept that ion channel modulators as a class of agents suppress contact hypersensitivity in a mouse model. Mice were sensitized to several contact sensitizing chemicals including dinitrochlorobenzene (DNCB) and a sensitizing drug, nadolol. We report our successful use of several ion channel modulators in suppressing contact hypersensitivity, including amiloride, ethacrynic acid (ECA), nifedipine and verapamil. For this purpose, Balb/c female mice were sensitized with DN

The use of a stereological method to estimate the volume of Schistosoma mansoni granulomas: the effect of zinc deficiency.

A quantitative, stereological method to assess the volume of Schistosoma mansoni granulomas is presented and its use demonstrated in a study of zinc-deficient animals. Twenty-six mice were divided into two groups, and given zinc-deficient or control food. After 4 weeks, the mean weight gain and thymus size were lower in the mice on the zinc-deficient diet. Eight mice in each group were then each infected with 60 S. mansoni cercariae and these mice were killed 12 weeks post-infection. There were then more eggs in the livers of the zinc-deficient mice than in those of the infected controls (7400

[Elucidation of functionally active domains in molecules of protective antigen bacillus anthracis's toxin].

Limited proteolysis has established that the protective antigen (PA) molecule consists of four functional-active domains. So, the shielding domain borrows an area in the linear structure of the PA molecule with NH2 of the end up to Lys 166 and plays a conducting role in the proteolytic activation of PA. The associative domain, engaging in the area Arg 167-Met266, plays a key role in the interaction with LF or EF at self-assembly toxic complexes LT or ET. The stabilizing domain borrows in the linear structure of the PA molecule are with Gly351 up to Met434. On the one hand, this area promotes f

Suppression of development of experimental autoimmune thyroiditis by oral administration of thyroglobulin.

Experimental autoimmune thyroiditis (EAT), which to some extent represents an experimental model of human chronic lymphocytic thyroiditis, is an organ-specific autoimmune disease characterized by autoantibody production to thyroid antigens (Ag) and mononuclear infiltration of the thyroid gland. EAT induced by immunization with human thyroglobulin (hTG) with Freund's adjuvant in CBA/J (H-2K) mice is associated with prominent B and T cell responses. We report that oral administration of hTG effectively reduces the immune responses in EAT in mice in an Ag-specific manner. Both cellular and humora

Immune attrition of adult schistosomes.

Mouse infection models are described that demonstrate reduction in the rate of egg production in Schistosoma haematobium worms 6-10 weeks after the onset of oviposition and loss of Schistosoma bovis worms around 10 weeks after infection. Neither phenomenon has been shown in Schistosoma mansoni- or Schistosoma japonicum-infected mice. The immunological basis for these anti-adult responses was inferred by comparison with infections in T-cell deprived mice and by transference of the ability to reduce a S. bovis worm burden in immuno-compromised hosts with immune serum. Vaccination with irradiatio

In vivo beneficial effects of cyclosporin A and 1,25-dihydroxyvitamin D3 on the induction of experimental autoimmune thyroiditis.

In a recent work, we provided evidence that the in vitro inhibitory effect of cyclosporin A (CsA) was potentiated by the addition of another immunosuppressive molecule, the 1,25-dihydroxyvitamin D3 (1,25(OH)2D3). In the present study, we investigated the in vivo influence of the association of both drugs administered at infratherapeutic doses, using an experimental model of autoimmune thyroiditis in CBA mice. Treatment regimen of the animals was initiated at priming with thyroglobulin (Tg) and consisted of daily administration of CsA (10 and 20 mg/kg/day, intragastrically) and/or 1,25(OH)2D3 (

[The effect of the protective antigen of Bacillus anthracis on the formation of immunity under the action of live anthrax vaccines].

The preparation of low-molecular protective antigen (PA) isolated from strain 34F2 (Sterne) and having a molecular weight of 34 and 51 kD, unlike the preparation of high-molecular PA with a molecular weight of 87 kD, suppressed the formation of acquired resistance to anthrax when introduced into guinea pigs in mixture with live spores of strains of STI, 34F2 and new vaccine strain 228/8; this phenomenon was mainly accompanied by a decrease in the level of antibodies to lethal factor (LF) nad in the antitoxic activity of blood serum. The immunosuppressing action of low-molecular PA depended on

Development of polyclonal antibody against polyestradiol and its practical use in detection of estrogen receptors in situ.

Polymerization of 17 beta-estradiol was performed. The final product with molecular weight of about 30,000 was used as antigen to immunization of mice CBA x C 57 B1 10/F1. The obtained antiserum was tested for the presence of antibodies to polyestradiol. It was applied to immunohistochemical detection of free estrogen receptors in breast carcinoma samples fixed with formaldehyde and embedded in paraffin. The advantages and disadvantages of this method and other perspectives of its application to routine examination of estrogen receptors for clinical practice are discussed.

The contribution of B-cell proliferation to spleen enlargement in Babesia microti-infected mice.

Flow cytofluorimetric analysis showed that B-cell proliferation makes a major contribution to the enlargement and increased cellularity of the spleen, which are characteristic of Babesia microti infections in mice. Expansion of the B-cell population was accompanied by modulation of the cell surface, which affected most B lymphocytes, and which was detected as a reduction in the density of surface immunoglobulin. This effect was noted as early as Day 7, shortly after the appearance of parasites in the circulation and the onset of gross spleen changes. In contrast to the results for B cells, the
