Limited association between Wolbachia and Plasmodium falciparum infections in natural populations of the major malaria mosquito Anopheles moucheti.

Since the discovery of natural malaria vector populations infected by the endosymbiont bacterium Wolbachia, a renewed interest has arisen for using this bacterium as an alternative for malaria control. Among naturally infected mosquitoes, Anopheles moucheti, a major malaria mosquito in Central Africa, exhibits one of the highest prevalences of Wolbachia infection. To better understand whether this maternally inherited bacterium could be used for malaria control, we investigated Wolbachia influence in An. moucheti populations naturally infected by the malaria parasite Plasmodium falciparum. Bio-products from Serratia marcescens isolated from Ghanaian Anopheles gambiae reduce Plasmodium falciparum burden in vector mosquitoes.

Novel ideas for control of mosquito-borne disease include the use of bacterial symbionts to reduce transmission. Bacteria belonging to the family Enterobacteriaceae isolated from mosquito midgut have shown promise in limiting Plasmodium intensity in the Anopheles vector. However, the mechanism of interaction between bacteria and parasite remains unclear. This study aimed at screening bio-products of two bacteria candidates for their anti-Plasmodial effects on mosquito stages of P. falciparum. Enterobacter cloacae and Serratia marcescens were isolated from field-caught Anopheles gambiae s.l. Sp

Development of CS-TPP-dsRNA nanoparticles to enhance RNAi efficiency in the yellow fever mosquito, Aedes aegypti.

Mosquito-borne diseases are a major threat to human health and are responsible for millions of deaths globally each year. Vector control is one of the most important approaches used in reducing the incidence of these diseases. However, increasing mosquito resistance to chemical insecticides presents challenges to this approach. Therefore, new strategies are necessary to develop the next generation vector control methods. Because of the target specificity of dsRNA, RNAi-based control measures are an attractive alternative to current insecticides used to control disease vectors. In this study, C

Susceptibility of Culex quinquefasciatus (Diptera: Culicidae) in Southern Louisiana to Larval Insecticides.

Mosquito control districts conduct rigorous insecticide treatments against both larval and adult Culex quinquefasciatus Say (Diptera: Culicidae), the primary vector of West Nile virus in the southern United States. However, the development of resistant populations of Cx. quinquefasciatus in response to extensive larvicide or adulticide applications has been demonstrated repeatedly across the world. Examining changes in insecticide susceptibility in treated field areas can help inform mosquito control districts as to whether or not their treatments remain effective. We hypothesized that frequen…

[Use of insecticide-treated cattle to control Rift Valley fever and West Nile virus vectors in Senegal].

Rift Valley Fever (RVF) and West Nile fever (WN) viruses are transmitted by several mosquito species and share the same vectors in Northern Senegal (West Africa). In absence of an effective treatment and vaccines, vector control remains an alternative method of prevention and control of these vector-borne diseases. The methods targeting adults' pest mosquitoes and malaria vectors which are currently used by the population in the Barkedji area (insecticides treated nets, bombs and copper coil) would not be effective against these vectors because of their exophagic and zoophilic behavior. Thus,

Mosquito repellency mediated by skin commensal Bacillus species via volatile metabolite reprogramming.

Individual variation in human attractiveness to mosquitoes critically influences vector-borne disease transmission, and this phenomenon arises from multiple underlying factors. This study integrated human skin microbiome profiling, non-human primate in situ intervention, and volatilomics to uncover a microbial-mediated mechanism underlying host-specific mosquito repellency. Bacillus species were identified as a microbial signature associated with reduced mosquito attraction in humans. Targeted Intervention of B. licheniformis and B. paralicheniformis on primate skin was sufficient to significa

Plasmodium falciparum Gametocyte Culture and Mosquito Infection Through Artificial Membrane Feeding.

Malaria remains one of the most important public health problems, causing significant morbidity and mortality. Malaria is a mosquito borne disease transmitted through an infectious bite from the female Anopheles mosquito. Malaria control will eventually rely on a multitude of approaches, which includes ways to block transmission to, through and from mosquitoes. To study mosquito stages of malaria parasites in the laboratory, we have optimized a protocol to culture highly infectious Plasmodium falciparum gametocytes, a parasite stage required for transmission from the human host to the mosquito

Habitat-based larval interventions: a new perspective for malaria control.

Interest in environmental management of mosquito larval habitats has been rekindled due to deterioration of malaria in tropical Africa. Environmental management programs were typically implemented as "all-out" campaigns by treating all potential breeding habitats. In contrast, targeted environmental management is based on a sound understanding of the heterogeneity in mosquito productivity. However, deficiencies in field methodology for measuring productivity hamper our progress in understanding of mosquito productivity. To address these issues, we develop a framework of habitat-based intervent

Spatial Exposure Responses of Malaria Vectors to Eucalyptus grandis (W. Hill ex Maiden) and Cymbopogon citratus (DC.) Stapf Essential Oils.

Background: Recently, the use of volatile compounds as spatial repellents have received special attention as a promising strategy for adult An. gambiae s.l control. Anopheles gambiae s.l is a primary vector of malaria, an arthropod-borne disease of global significance. Current strategies for controlling mosquitoes heavily rely on vector control methods. Understanding the responses of these vectors to volatile compounds will be helpful in the formulation of repellants or attractants for control vector populations. This study was conducted in Nwoya district, Uganda, one of the high-malaria-trans

A global assessment of surveillance methods for dominant malaria vectors.

The epidemiology of human malaria differs considerably between and within geographic regions due, in part, to variability in mosquito species behaviours. Recently, the WHO emphasised stratifying interventions using local surveillance data to reduce malaria. The usefulness of vector surveillance is entirely dependent on the biases inherent in the sampling methods deployed to monitor mosquito populations. To understand and interpret mosquito surveillance data, the frequency of use of malaria vector collection methods was analysed from a georeferenced vector dataset (> 10,000 data records), extra

Upscaling irradiation protocols of Aedes albopictus pupae within an SIT program in Reunion Island.

The implementation of the sterile insect technique against Aedes albopictus relies on many parameters, in particular on the success of the sterilization of males to be released into the target area in overflooding numbers to mate with wild females. Achieving consistent sterility levels requires efficient and standardized irradiation protocols. Here, we assessed the effects of exposure environment, density of pupae, irradiation dose, quantity of water and location in the canister on the induced sterility of male pupae. We found that the irradiation of 2000 pupae in 130 ml of water and with a do

Dengue Suppression by Male Wolbachia-Infected Mosquitoes.

BACKGROUND: Wild-type female Aedes aegypti mosquitoes that mate with male A. aegypti mosquitoes that have been infected with the wAlbB strain of Wolbachia pipientis bacteria produce nonviable offspring owing to cytoplasmic incompatibility. Repeated releases of wolbachia-infected males can potentially suppress wild-type mosquito populations and reduce the risk of dengue virus infection. METHODS: We conducted a trial involving the release of male A. aegypti mosquitoes infected with the wAlbB strain of wolbachia bacteria for the control of dengue in Singapore, a tropical city-state. In this clus

Boric acid toxic sugar bait suppresses male Aedes aegypti (Diptera: Culicidae): wing beat frequency and amplitude, flight activity, fecundity, insemination, and mate-finding Allee effect.

BACKGROUND: Controlling the spread of arboviral diseases remains a considerable challenge due to the rapid development of insecticide resistance in Aedes mosquitoes. This study evaluated the effects of boric acid-containing toxic sugar bait (TSB) on field populations of resistant Aedes aegypti mosquitoes. In addition, this study examined the flight activity and wing beat frequency and amplitude of males and the flight activity, fecundity, and insemination of females after pairing with males exposed to TSB. The population dynamics of Aedes mosquitoes under imbalanced sex ratios were examined to

Scaling up of Eco-Bio-Social Strategy to Control Aedes aegypti in Highly Vulnerable Areas in Fortaleza, Brazil: A Cluster, Non-Randomized Controlled Trial Protocol.

Aedes aegypti is a cosmopolitan vector for arboviruses dengue, Zika and chikungunya, disseminated in all Brazilian states. The Eco-Bio-Social (EBS) strategy is vital in Aedes aegypti control as it mobilizes stakeholders (government, professionals, society, and academics) to promote healthy environments. This paper describes the rationale and methods of expanding the EBS strategy for Aedes aegypti control in Fortaleza, Northeast Brazil. A cluster, non-randomized controlled clinical trial was developed to analyze the strategy's effectiveness in vulnerable territories (high incidence of dengue an

Trapping of Rift Valley Fever (RVF) vectors using light emitting diode (LED) CDC traps in two arboviral disease hot spots in Kenya.

BACKGROUND: Mosquitoes' response to artificial lights including color has been exploited in trap designs for improved sampling of mosquito vectors. Earlier studies suggest that mosquitoes are attracted to specific wavelengths of light and thus the need to refine techniques to increase mosquito captures following the development of super-bright light-emitting diodes (LEDs) which emit narrow wavelengths of light or very specific colors. Therefore, we investigated if LEDs can be effective substitutes for incandescent lamps used in CDC light traps for mosquito surveillance, and if so, determine th

Management of insecticide resistance in the major Aedes vectors of arboviruses: Advances and challenges.

BACKGROUND: The landscape of mosquito-borne disease risk has changed dramatically in recent decades, due to the emergence and reemergence of urban transmission cycles driven by invasive Aedes aegypti and Ae. albopictus. Insecticide resistance is already widespread in the yellow fever mosquito, Ae. Aegypti; is emerging in the Asian tiger mosquito Ae. Albopictus; and is now threatening the global fight against human arboviral diseases such as dengue, yellow fever, chikungunya, and Zika. Because the panel of insecticides available for public health is limited, it is of primary importance to prese

Attracting, trapping and killing disease-transmitting mosquitoes using odor-baited stations - The Ifakara Odor-Baited Stations.

BACKGROUND: To accelerate efforts towards control and possibly elimination of mosquito-borne diseases such as malaria and lymphatic filariasis, optimally located outdoor interventions could be used to complement existing intradomicilliary vector control methods such as house spraying with insecticides and insecticidal bednets. METHODS: We describe a new odor-baited station for trapping, contaminating and killing disease-transmitting mosquitoes. This device, named the 'Ifakara Odor-baited Station' (Ifakara OBS), is a 4 m3 hut-shaped canvas box with seven openings, two of which may be fitted wi

Mosquito bite hypersensitivity in children: Clinical features outweigh specific IgE in diagnosis.

Background: Mosquito bite hypersensitivity results from abnormal immune responses to salivary proteins with potent immunomodulatory properties. In children, both type I (immunoglobulin E [IgE] mediated) and type IV (cell mediated) mechanisms contribute to the reaction spectrum, ranging from localized erythema to extensive inflammatory lesions. Although mosquito-specific IgE assays are commercially available, their diagnostic utility and correlation with clinical severity remain unclear. Objective: The objective was to describe the clinical and immunologic characteristics of children with mosqu

Genetic modification of Anopheles stephensi for resistance to multiple Plasmodium falciparum strains does not influence susceptibility to o'nyong'nyong virus or insecticides, or Wolbachia-mediated resistance to the malaria parasite.

Mosquitoes that have been genetically engineered for resistance to human pathogens are a potential new tool for controlling vector-borne disease. However, genetic modification may have unintended off-target effects that could affect the mosquitoes' utility for disease control. We measured the resistance of five genetically modified Plasmodium-suppressing Anopheles stephensi lines to o'nyong'nyong virus, four classes of insecticides, and diverse Plasmodium falciparum field isolates and characterized the interactions between our genetic modifications and infection with the bacterium Wolbachia.
