Simultaneous detection of West Nile and Japanese encephalitis virus RNA by duplex TaqMan RT-PCR.

West Nile virus (WNV) and Japanese encephalitis virus (JEV) are important mosquito-borne viruses of the Flaviviridae family, associated with encephalitis, mainly in humans and horses. WNV is also pathogen for many bird species. The incidence of human and animal WNV infections in Europe has risen, mostly in recent years, and JEV was detected in 2011 in mosquitoes collected in Italy and may emerge in Europe in the same way as other flaviviruses had emerged recently (USUTU and Bagaza virus) and should be regarded as a potential threat to public health. Prompt identification and discrimination bet

A new molecular approach to help conclude drowning as a cause of death: simultaneous detection of eight bacterioplankton species using real-time PCR assays with TaqMan probes.

We developed a novel tool for concluding drowning as a cause of death. We designed nine primer pairs to detect representative freshwater or marine bacterioplankton (aquatic bacteria) and then used real-time PCR with TaqMan probes to rapidly and specifically detect them. We previously cultured the genus Aeromonas, which is a representative freshwater bacterial species, in blood samples from 94% of victims who drowned in freshwater and the genera Vibrio and/or Photobacterium that are representative marine bacteria in 88% of victims who drowned in seawater. Based on these results, we simultaneous

How reliable is the next generation of multiplex-PCR for diagnosing prosthetic joint infection compared to the MSIS criteria? Still missing the ideal test.

INTRODUCTION: Identification of the pathogen in case of a periprosthetic joint infection (PJI) remains 1 of the greatest challenges in septic surgery. Rapid germ identification enables timely, specific, antimicrobial therapy. The first multiplex PCR (polymerase chain reaction) generation (Unyvero-i60) enables germ detection within 5 hours with a sensitivity of 78.8% and a specificity of 100%. The aim of this study is to investigate the performance of the new generation of cartridges (Unyvero-ITI) of multiplex PCR in the case of a PJI. METHODS: In a prospective study, intraoperatively aspirate

Detection of gastroenteritis viruses among pediatric patients in Hiroshima Prefecture, Japan, between 2006 and 2013 using multiplex reverse transcription PCR-based assays involving fluorescent dye-labeled primers.

Multiplex reverse transcription (RT)-polymerase chain reaction (PCR)-based assays involving fluorescent dye-labeled primers were modified to detect 10 types of gastroenteritis viruses by adding two further assays to a previously developed assay. Then, these assays were applied to clinical samples, which were collected between January 2006 and December 2013. All 10 types of viruses were effectively detected in the multiplex RT-PCR-based assays. In addition, various viral parameters, such as the detection rates and age distributions of each viral type, were examined. The frequency and types of m

An integrated microfluidic sample-to-answer system for automated detection of common deletional α-thalassemia mutations.

Thalassemia is a prevalent hereditary hematological disorder, and rapid, automated genetic screening is essential for large-scale prevention and control, particularly in primary healthcare settings. To address the limitations of conventional molecular diagnostic methods, we developed an integrated microfluidic sample-to-answer system for simultaneous detection of four common deletional α-thalassemia mutations (-α3.7, -α4.2, --SEA, and --THAI), together with the HBA2 wild-type locus, enabling accurate discrimination between heterozygous and homozygous deletion genotypes. The system directly use

One-step multiplex PCR assay for identification of Mycobacterium kansasii complex species.

UNLABELLED: Currently, the Mycobacterium kansasii complex (MKC) species identification involves PCR amplification, followed by sequencing or digestion of the amplicons. The purpose of this study was to develop a one-step multiplex PCR assay allowing for a fast and accurate identification of MKC species. A total of 158 Mycobacterium sp. genomes were searched for sites that would yield easily detectable amplicons of different sizes among the MKC species while producing no amplicons for other-than-MKC mycobacterial species. Three primer sets were designed and tested in vitro on seven reference st

Clinical and cost implications of Biofire FilmArray® meningitis / encephalitis panel testing: a systematic review.

BACKGROUND: Meningitis and encephalitis are diseases with high case fatality rates and serious long-term sequalae. A significant percentage do not receive an aetiological diagnosis, in part due to limitations of conventional testing methods. The Biofire FilmArray® Meningitis / Encephalitis Panel (MEP) is the first commercially approved multiplex PCR panel for diagnosis of infectious meningoencephalitis. It provides rapid results and has the potential to reduce empiric antimicrobial use and length of hospitalisation when compared to conventional testing. METHODS: We conducted a systematic revi

One Assay, Nine Targets: Advancing Viral Surveillance with Multiplex RT-ddPCR.

Viral infections continue to pose a major global health challenge, driven by factors such as population growth, migration, and environmental change, all of which contribute to the emergence and reemergence of infectious viruses. Advances in technology now enable the detection of multiple targets from a limited sample volume; however, few studies have fully leveraged these capabilities. In this study, we developed and analytically validated a highly sensitive and specific 9-plex one-step RT-ddPCR assay for the detection of high-risk viruses, including SARS-CoV-2 (N1 and N2 genes), Influenza A a

Development of cell combos micromethod to isolate respiratory viruses not detected by molecular techniques.

Virological diagnosis of respiratory infections relies mainly on molecular methods but can be overlooked in cases of unexpected or unknown emerging viruses. In this case, modern approaches use mainly metagenomics without a priori. The other approach to detecting new viruses is based on their isolation in cell culture using virus-specific cells and culture conditions. Unfortunately, inoculation on several lines and metagenomics requires a large volume of clinical sample, while the initial sample has already been largely used to carry out multiplexed RT-PCR. Herein, we aimed to develop micro-met
