RNAi Identified the Potential Functions of Actin-like Protein in the Growth Performance of Macrobrachium nipponense.

Macrobrachium nipponense is an important commercial freshwater prawn species in China. Since larger individuals command higher market value, there is a pressing need to identify growth-related genes and single-nucleotide polymorphisms (SNPs) to facilitate genetic improvement in this species. Previous studies have suggested a potentially regulatory role of an actin-like protein (ACTL) in the growth of M. nipponense. Therefore, the present study aimed to functionally characterize the role of ACTL in growth and identify growth-associated SNPs within this gene. The open reading frame of Mn-ACTL is

Ultrasound-initiated crosslinking between cod-rice dual-protein: Mechanisms underlying the development of dual-protein gel.

The combination of animal and plant proteins enables nutritional complementarity and functional synergies. This study systematically investigated the effects of different ultrasound time (0, 2, 4, 6, 8, and 10 min) on the physicochemical properties and structural characteristics of cod-rice dual-protein gel, and further explored the underlying mechanisms of gel formation. The results showed that the particle size of dual-protein system decreased from 1364.78 nm to 876.88 nm with the ultrasound treatment from 0 min (U0) to 8 min (U8), accompanied by the ζ-potential absolute value increasing. Nuclear pore pathology underlying multisystem proteinopathy type 3-related inclusion body myopathy.

OBJECTIVE: Multisystem proteinopathy type 3 (MSP3) is an inherited, pleiotropic degenerative disorder caused by a mutation in heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1), which can affect the muscle, bone, and/or nervous system. This study aimed to determine detailed histopathological features and transcriptomic profile of HNRNPA1-mutated skeletal muscles to reveal the core pathomechanism of hereditary inclusion body myopathy (hIBM), a predominant phenotype of MSP3. METHODS: Histopathological analyses and RNA sequencing of HNRNPA1-mutated skeletal muscles harboring a c.940G > A (p.D3

Statins Induce Locomotion and Muscular Phenotypes in Drosophila melanogaster That Are Reminiscent of Human Myopathy: Evidence for the Role of the Chloride Channel Inhibition in the Muscular Phenotypes.

The underlying mechanisms for statin-induced myopathy (SIM) are still equivocal. In this study, we employ Drosophila melanogaster to dissect possible underlying mechanisms for SIM. We observe that chronic fluvastatin treatment causes reduced general locomotion activity and climbing ability. In addition, transmission microscopy of dissected skeletal muscles of fluvastatin-treated flies reveals strong myofibrillar damage, including increased sarcomere lengths and Z-line streaming, which are reminiscent of myopathy, along with fragmented mitochondria of larger sizes, most of which are round-like

Attractin deficiency causes metabolic and morphological abnormalities in slow-twitch muscle.

Skeletal muscle fibers are classified as slow-twitch and fast-twitch fibers, which have different reactive oxygen species (ROS) metabolism and mitochondrial biogenesis. Recently, Attractin (Atrn), which encodes secreted (sAtrn) and transmembrane (mAtrn)-type proteins, has been shown to be involved in free radical scavenging. Although Atrn has been found in skeletal muscle, little is known about the expression levels and function of Atrn in each muscle fiber type. Therefore, we investigate sAtrn and mAtrn expression levels in the slow-twitch soleus (sol) and fast-twitch extensor digitorum longu

IUGR impairs cardiomyocyte growth and maturation in fetal sheep.

Placental insufficiency causes intrauterine growth restriction (IUGR), a common complication of pregnancy. In skeletal muscle, IUGR reduces fetal myofibril size, reduces myoblast proliferation and reduces expression of genes in cell cycle regulation clusters. The myocardium is striated like skeletal muscle, and IUGR also reduces cell cycle activity and maturation in cardiomyocytes, despite cardiac output preferentially directed to the coronary circulation. We hypothesized that cardiomyocyte growth restriction would be accompanied by similar changes in cell cycle regulation genes and would redu

Lamin A/C-Related Cardiac Disease: Late Onset With a Variable and Mild Phenotype in a Large Cohort of Patients With the Lamin A/C p.(Arg331Gln) Founder Mutation.

BACKGROUND: Interpretation of missense variants can be especially difficult when the variant is also found in control populations. This is what we encountered for the LMNA c.992G>A (p.(Arg331Gln)) variant. Therefore, to evaluate the effect of this variant, we combined an evaluation of clinical data with functional experiments and morphological studies. METHODS AND RESULTS: Clinical data of 23 probands and 35 family members carrying this variant were retrospectively collected. A time-to-event analysis was performed to compare the course of the disease with carriers of other LMNA mutations. Myo

Atrophy responses to muscle inactivity. I. Cellular markers of protein deficits.

The goal of this study was to use the model of spinal cord isolation (SI), which blocks nearly all neuromuscular activity while leaving the motoneuron muscle-fiber connections intact, to characterize the cellular processes linked to marked muscle atrophy. Rats randomly assigned to normal control and SI groups were studied at 0, 2, 4, 8, and 15 days after SI surgery. The slow soleus muscle atrophied by approximately 50%, with the greatest degree of loss occurring during the first 8 days. Throughout the SI duration, muscle protein concentration was maintained at the control level, whereas myofib

Arrhythmogenic right ventricular cardiomyopathy: clinicopathologic correlation based on a revised definition of pathologic patterns.

Different morphologic features of arrhythmogenic right ventricular cardiomyopathy (ARVC) have been described. However, it is still unclear whether they correspond to distinct forms of the same disease. A pathologic study was performed on a series of ARVC (15 from heart transplant and 12 from autopsy) from 2 Italian referral university hospitals. Based on both myocellular features and the nature of myocardial replacement, hearts were divided into 2 groups: infiltrative, with a lacelike pattern of transmural fatty infiltration and strands of normal residual cardiomyocytes (n = 11); and cardiomyo

Klhl41b orchestrates sarcomere assembly and growth to drive skeletal muscle hypertrophy in vivo.

BACKGROUND: Nemaline myopathy (NM) is a clinically and genetically heterogeneous disease caused by mutations in sarcomere-associated genes, with congenital forms representing the most severe end of the spectrum. Recessive mutations in KLHL41 underlie a severe congenital NM characterized by sarcomeric disorganization and impaired muscle growth. The in vivo mechanisms by which KLHL41 governs sarcomere assembly and growth remain poorly understood. METHODS: We generated zebrafish models of KLHL41 deficiency by the CRISPR-Cas9 approach to examine the temporal and molecular basis of sarcomere assem

Effect of fermentation-driven acid-mediated protein degradation on chicken and its gel properties.

This study systematically examines the impact of protein breakdown on the gel properties and texture of chicken under fermentation-induced acidic conditions. Chicken mince samples with different degrees of protein degradation were prepared by controlling fermentation time, followed by comprehensive analyses of physicochemical characteristics, protein structure, microstructure, and gel performance. Results showed that as degradation progressed, the pH value diminished from 5.86 in the control group to 4.19 at 50 days. Under acidic conditions, the surface hydrophobicity of myofibrillar protein w

miR-29b-3p promotes the recovery of alkali-burned cornea by targeting Ddx3x.

Corneal alkali burns often cause vision impairment by triggering excessive inflammation and fibrosis, which compromise corneal transparency. Given miR-29b-3p's anti-inflammatory and anti-fibrotic properties, this study evaluated its therapeutic potential in corneal alkali injury. A murine corneal alkali burn model was established, and differentially expressed proteins were analyzed using liquid chromatography-mass spectrometry (LC-MS) at 24 h and 1-week post-injury. The results showed corneas exhibited both continuous and phase-specific pathway alterations during different stages of alkali bur

Ultrastructural Signs of High Functional Activity of Neuromuscular Synapses in Aging Rats After Photobiomodulation.

Aging is characterized by progressive degeneration of neuromuscular junctions (NMJs), which significantly contributes to muscle weakness and the development of sarcopenia. Photobiomodulation (PBM), a non-invasive therapeutic method based on the use of low-intensity light, has shown promising results in mitigating muscle degeneration in both experimental and clinical studies. The aim of this study was to evaluate the ultrastructural effects of photobiomodulation on neuromuscular junctions and skeletal muscle fibers in the m. vastus lateralis muscle of aged rats using light and transmission elec
