Localized cytomegalovirus encephalitis contiguous to metastatic nasopharyngeal carcinoma.

We report a case of focal infection of the brain with cytomegalovirus (CMV) in a 14-year-old boy with metastatic lymphoepithelioma (nasopharyngeal carcinoma). Cytomegalovirus-bearing subependymal glia were only seen in the fourth ventricle, in close proximity to tumor cells, these were associated with an intense inflammatory cell exudate. The latter was due to enterococcal meningitis. There was no evidence of systemic CMV infection, and the typical encephalitis with glial nodules seen in acquired forms of the disease was lacking. We postulate that the infection gained access to the brain eithe

Respiratory syncytial virus neutralizing activity in nasopharyngeal secretions.

Nasopharyngeal secretions were taken during the acute phase of illness from 30 children admitted to hospital with lower respiratory tract infections. The presence of neutralizing activity in secretions taken at the onset of illness was demonstrated in 11 out of 15 patients (73%) with bronchiolitis caused by R.S. virus, as compared with 4 out of 9 patients (44%) with R.S. virus infections other than bronchiolitis, and 1 out of 6 without R.S. virus infection. Second secretions were taken 7 days later from 10 of the children with R.S. virus infection. Eight of these paired secretions showed an in

Nasopharyngeal carcinoma with non-squamous phenotype may be a variant of nasopharyngeal squamous cell carcinoma after inhibition of EGFR/PI3K/AKT/mTOR pathway.

Nasopharyngeal carcinoma (NPC) is a cancerous tumor that develops in the nasopharynx epithelium and typically has squamous differentiation. The squamous phenotype is evident in immunohistochemistry, with diffuse nuclear positivity for p63 and p40. Nonetheless, a few NPCs have been identified by clinicopathological diagnosis that do not exhibit the squamous phenotype; these NPCs are currently referred to as non-squamous immunophenotype nasopharyngeal carcinomas (NSNPCs). In a previous work, we have revealed similarities between the histological appearance, etiology, and gene alterations of NSNP

Epstein-Barr virus mRNA profiles and viral DNA methylation status in nasopharyngeal brushings from nasopharyngeal carcinoma patients reflect tumor origin.

Undifferentiated nasopharyngeal carcinoma (NPC) is 100% associated with Epstein-Barr virus (EBV) as oncogenic driver. NPC is often diagnosed late due to initial vague complaints and obscured location. Prior studies suggest that measurement of EBV DNA load and RNA transcripts in nasopharyngeal (NP) brushings is useful for minimally invasive NPC diagnosis. However, whether these EBV markers relate to local virus replication or reflect tumor origin remains to be demonstrated. To resolve this, we analysed EBV-DNA characteristics and quantified latent and lytic viral RNA transcripts in NP brushings

[The ex vivo expansion of gamma delta T cells from the peripheral blood of patients with nasopharyngeal carcinoma and their cytotoxicity to nasopharyngeal carcinoma lines in vitro].

OBJECTIVE: gamma delta T cells from the peripheral blood of nasopharyngeal carcinoma(NPC) patients were expanded in vitro and their cytotoxic activity to NPC lines was analysed. A role of gamma delta T cell of immune mechanism in development of NPC was discussed. METHOD: The solid-phase monoclonal anti-TCR gamma delta McAb was used to expand the gamma delta T cells from peripheral blood of seven patients with NPC and six healthy donors. Then we detected the cytotoxicities of these gamma delta T cell against tumour cell lines including Daudi Burkitt's lymphoma and the cell lines of NPC, CNE1 a

Radiation-induced nasopharyngeal ulcers after re-irradiation with intensity-modulated radiotherapy in locoregional recurrent nasopharyngeal carcinoma patients: a dose-volume-outcome analysis.

OBJECTIVE: To analyze the interrelation between radiation dose and radiation-induced nasopharyngeal ulcer (RINU) in locoregional recurrent nasopharyngeal carcinoma (NPC) treated with intensity-modulated radiation therapy (IMRT). METHODS: Clinical data were collected from 363 patients with locoregional recurrent NPC who received re-irradiated with definitive IMRT from 2009 to 2017. Twenty-nine patients were diagnosed with RINU. Univariate and multivariate analyses were used to re-evaluate the first and second radiotherapy plans and to identify predictive dosimetric factors. RESULTS: All dosim

Irradiation-induced nasopharyngeal necrosis (INN) in newly diagnosed nasopharyngeal carcinoma treated by intensity-modulated radiation therapy: clinical characteristics and the influence of treatment strategies.

PURPOSE: To define the clinical characteristics of irradiation-induced nasopharyngeal necrosis (INN) after intensity-modulated radiotherapy (IMRT) and identify the influence of treatment strategies on INN in primary nasopharyngeal carcinoma (NPC) patients. PATIENTS AND METHODS: From 2008 to 2019, NPC patients pathologically diagnosed with INN after primary IMRT were reviewed. Those patients were matched with propensity scores for patients without INN in our center. The impact of treatment strategies on INN occurrence was assessed using univariate and multivariate logistic regression analysis.

[A basic study on the association of Epstein-Barr virus with nasopharyngeal carcinoma--detection and genotypic analysis of Epstein-Barr virus associated with nasopharyngeal carcinoma in Japanese patients].

Epstein-Barr virus (EBV) is known to be associated with two human malignant diseases, nasopharyngeal carcinoma (NPC) and endemic Burkitt's lymphoma. In this study, the genotypes of EBV in tissues from 13 NPC patients in Japan were analyzed by Southern blot hybridization using EBV genome fragment probes. Ten of the cases contained reiterated sequences (EBV BamHI-H, -B1*, -K fragments), showing that only one genotype was detected in each specimen. One of these had a BamHI fragment containing a fused sequence of BamHI-Y and -H. In all except one case, a single-sized EBV-joined terminus was observ

Imaging Immune Heterogeneity in Nasopharyngeal Carcinoma: A Multidimensional Radiomics Perspective.

RATIONALE AND OBJECTIVES: Nasopharyngeal carcinoma (NPC) is characterized by a distinctive virologic and immunologic profile, in which Epstein-Barr virus-driven immune infiltration coexists with immune escape. This complex interplay gives rise to pronounced immune heterogeneity across spatial, molecular, and temporal dimensions, which critically influences response to immunotherapy and clinical outcomes. However, conventional biopsy-based immune evaluation is limited by sampling bias, invasiveness, and restricted spatial representativeness, precluding comprehensive characterization of the glob

Molecular Functions and Drug Prediction of MMP1 and TGFBR2 in Nasopharyngeal Carcinoma.

BACKGROUND: Nasopharyngeal carcinoma (NPC) is partially driven by epithelial-mesenchymal transition (EMT). We identified potential EMT-related biomarkers and targeted drugs for NPC via computational and machine learning approaches. METHODS: Based on the NPC patient dataset derived from the public database, differentially expressed genes (DEGs) were screened using the limma package and intersected with the EMT gene set from the database of Epithelial-Mesenchymal Transition-associated genes (dbEMT). Subsequently, GO and KEGG enrichment analyses were performed, and a protein-protein interaction

Enhanced Radiotherapy Sensitivity of Nasopharyngeal Carcinoma by Stretchable Hyperthermia Patches.

Nasopharyngeal carcinoma (NPC) has a high incidence of cervical lymph node metastases, for which radiotherapy is a primary treatment. Hyperthermia is an important means to enhance the effectiveness of radiotherapy. However, the utility of conventional hyperthermia techniques in the clinic is compromised by limitations including bulky equipment, temperature inhomogeneity, and restricted treatment areas. This study developed a novel, soft, stretchable, and optically transparent hyperthermia patch based on a silver nanowire network to improve radiotherapy efficacy. The patch allowed precise tempe

Epstein-Barr virus reprograms immune escape in nasopharyngeal carcinoma.

Nasopharyngeal carcinoma (NPC) is an Epstein - Barr virus (EBV)-associated epithelial malignancy characterized by an immune-rich yet immunosuppressive microenvironment. EBV establishes an inflammatory but immune-tolerant ecosystem that underlies poor responses to immunotherapy. While prior studies have focused on single checkpoints such as PD-1/PD-L1, they often overlook the systemic immune reprogramming driven by EBV. Viral latent proteins, particularly LMP1, activate NF-κB and JAK - STAT signaling, inducing cytokine production, immune cell reeducation, and therapeutic resistance. This leads

Hierarchical dynamic model for risk-stratified screening of nasopharyngeal carcinoma.

Early detection of nasopharyngeal carcinoma through Epstein-Barr virus serology is hampered by a low positive predictive value. This study aims to develop a hierarchical dynamic model to refine risk stratification among individuals initially identified as medium- or high-risk by Epstein-Barr virus serology. By integrating longitudinal Epstein-Barr virus antibody data with age, sex, and family history, the model is trained using data from the PRO-NPC-001 program. In the validation cohort, the high-risk model using one-year data achieves a positive predictive value of 18.2% (about a fourfold inc

Tumor-infiltrating B cells inhibit nasopharyngeal carcinoma metastasis.

Nasopharyngeal carcinoma (NPC) in advanced stages often has a poor prognosis due to distant metastasis, yet the distribution characteristics and anti-metastatic mechanisms of tumor-infiltrating B cells (TIL-B) remain unclear. In this study, single-cell RNA sequencing combined with multiplex immunofluorescence revealed that B cells were highly enriched in non-metastatic NPC and formed complete tertiary lymphoid structures (TLS), whereas TLS was absent in metastatic NPC. Functional assays in vitro and in vivo showed that TIL-B and their conditioned medium significantly inhibited NPC cell prolife

Characterization of Nasopharyngeal Microbiota Dysbiosis in Children with Mycoplasma pneumoniae Pneumonia.

Mycoplasma pneumoniae pneumonia (MPP) is a leading cause of community-acquired pneumonia in children, yet little is known about the role of nasopharyngeal microbiota dysbiosis in susceptibility to infection and disease subtype. In this study, we performed 16S rRNA sequencing on nasopharyngeal samples from 102 pediatric MPP patients, 104 influenza A patients, and 103 healthy controls and compared the microbial diversity, composition, and functional profiles across groups. The MPP group exhibits an altered nasopharyngeal microbial composition, characterized by reduced microbial diversity and an
