Noise-induced tinnitus and hyperacusis are thought to correspond to a disrupted balance between excitation and inhibition in the central auditory system. Excitation and inhibition are often studied using pure tones; however, these responses do not reveal inhibition within the excitatory pass band. Therefore, we used a Wiener-kernel analysis, complemented with singular value decomposition (SVD), to investigate the immediate effects of acoustic trauma on excitation and inhibition in the inferior colliculus (IC). Neural responses were recorded from the IC of three anesthetized albino guinea pigs

Many people with Tourette syndrome are able to volitionally suppress tics, under certain circumstances. To understand better the neural mechanisms that underlie this ability, we used functional magnetic resonance neuroimaging to track regional brain activity during performance of an intentional inhibition task. On some trials, Tourette syndrome and comparison participants internally chose to make or withhold a motor action (a button press), while on other trials, they followed 'Go' and 'NoGo' instructions to make or withhold the same action. Using representational similarity analysis, a functi

In developing vertebrates, the neural tube forms from a sheet of neural ectoderm by complex cell movements and morphogenesis. Convergent extension movements and the apical constriction along with apical-basal elongation of cells in the neural ectoderm are thought to be essential for the neural tube closure (NTC) process. In addition, it is known that non-neural ectoderm also plays a crucial role in this process, as the neural tube fails to close in the absence of this tissue in chick and axolotl. However, the cellular and molecular mechanisms by which it functions in NTC are as yet unclear. We

OBJECTIVE: This study aimed to explore the relationship between corticomotor inhibition, assessed by transcranial magnetic stimulation (TMS), and balance performance in people with Parkinson's disease (PwPD).
METHODS: Thirty-nine PwPD, ranging from Hoehn and Yahr stages 1 - 3, underwent TMS to assess corticomotor inhibition-specifically GABAergic intracortical inhibition-under three postural conditions: lying, sitting, and standing. Balance performance was evaluated, including static, dynamic, and functional balance.
RESULTS: Participants' ages ranged from 52 to 76, and disease duration rang

Antisense oligonucleotides (ASOs) are an important therapeutic modality for neurological diseases. Some ASOs cause transient neurobehavioral responses acutely following intrathecal delivery to the central nervous system (CNS). We characterized a subset of these responses, including hypoactivity, spinal reflex loss, paresis, sedation, and ataxia, that are suggestive of neuronal inhibition in rodents and non-human primates. Across species, inhibition-like responses peaked ∼3 h post-ASO delivery, reversed within 24 h with no sequelae, and could be quantified using simple neurobehavioral scales. A

Motor learning relies on signals that instruct adaptive plasticity following errors. In the cerebellum, climbing fibers (CFs) provide these instructions to Purkinje cells (PCs). Yet CFs fire continuously, even without errors, requiring molecular layer interneuron (MLI) inhibition of PCs to counteract CF excitation and prevent maladaptive plasticity. Here, to identify how this regulatory inhibition is contextually suppressed to selectively permit error-driven learning in mice, we combined connectomics, functional recordings, computational modeling and behavioral manipulations. We discovered tha

BACKGROUND: Neural circuits throughout the CNS can exhibit rebound excitation following prolonged periods of inhibition. However, the potential to control this phenomenon and harness it for clinical applications remains largely unexplored.
OBJECTIVE: We investigate rebound excitatory responses evoked by spinal cord stimulation (SCS) that can generate functional motor output, providing a testable model of circuit-level rebound excitation relevant to neuromodulation across the CNS.
METHODS: Brief (5 s) electrical stimulation trains were delivered to the lumbar spinal cord in adult cats. We rec

BACKGROUND: Motor cortex disinhibition, as measured by impaired short-interval intracortical inhibition (SICI) using transcranial magnetic stimulation (TMS), is a well-established feature of Parkinson's disease (PD). However, its substantial variability among patients remains unexplained, prompting questions about its origin, clinical relevance, and connection to disease heterogeneity.
OBJECTIVE: Based on biological links between olfaction and motor function, we aimed to investigate the possible relationship between motor cortex disinhibition and olfactory dysfunction in PD.
METHODS: We asse

Down syndrome (DS) is a developmental genetic disorder that is associated with an accelerated aging profile and high probability of early incidence Alzheimer's disease like symptoms. It is well established that there are morphological differences in the brains of adults with DS, but the net impact of the genetic disruption on cortical function remains poorly understood. To address this knowledge gap, we used magnetoencephalographic (MEG) brain imaging to assess the somatosensory cortical activity elicited by a paired-pulse electrical stimulation of the right median nerve of adults with DS (N =

Dexterous motor skills, like those needed for playing musical instruments and sports, require the somatosensory system to accurately and rapidly process somatosensory information from multiple body parts. This is challenging due to the convergence of afferent inputs from different body parts into a single neuron and the overlapping representation of neighboring body parts in the somatosensory cortices. How do trained individuals, such as pianists and athletes, manage this? Here, a series of five experiments with pianists and nonmusicians (female and male) shows that pianists have enhanced inhi

AIMS: Epilepsy, frequently comorbid with depression, easily develops drug resistance. Here, we investigated how dorsal raphe (DR) and its 5-HTergic neurons are implicated in epilepsy.
METHODS: In mouse hippocampal kindling model, using immunochemistry, calcium fiber photometry, and optogenetics, we investigated the causal role of DR 5-HTergic neurons in seizure of temporal lobe epilepsy (TLE). Further, deep brain stimulation (DBS) of the DR with different frequencies was applied to test its effect on hippocampal seizure and depressive-like behavior.
RESULTS: Number of c-fos+ neurons in the D

Autism spectrum disorder (ASD) is a developmental disability that is poorly understood. ASD can influence communication, social interaction, and behavior. Children with ASD often have sensory hypersensitivities, including auditory hypersensitivity (hyperacusis). In adults with hyperacusis who are otherwise neurotypical, the medial olivocochlear (MOC) efferent reflex is stronger than usual. In children with ASD, the MOC reflex has been measured, but without also assessing hyperacusis. We assessed the MOC reflex in children with ASD by measuring the strength of MOC-induced inhibition of transien

Minimal hepatic encephalopathy (MHE) is the earliest form of hepatic encephalopathy (HE) and affects up to 80 % of patients with liver cirrhosis. By definition, MHE is characterized by psychomotor slowing and subtle cognitive deficits, but obvious clinical manifestations are lacking. Given its covert nature, MHE is often underdiagnosed. This study was aimed at detecting neurophysiological changes, as assessed by means of transcranial magnetic stimulation (TMS), involved in the early pathogenesis of the HE. We investigated motor cortex excitability in 15 patients with MHE and in 15 age-matched

Human brain imaging has revealed that acute pain results from activation of a network of brain regions, including the somatosensory, insular, prefrontal, and cingulate cortices. In contrast, many investigations report little or no alteration in brain activity associated with chronic pain, particularly neuropathic pain. It has been hypothesized that neuropathic pain results from misinterpretation of thalamocortical activity, and recent evidence has revealed altered thalamocortical rhythm in individuals with neuropathic pain. Indeed, it was suggested nearly four decades ago that neuropathic pain

Headaches affect 90% of the population sometime during their life. Most are benign and fleeting, some are serious and life-threatening, and others require ongoing medical consultation and treatment. A careful history and physical is necessary to establish a differential diagnosis and to guide the choice of testing to make an accurate diagnosis. The most common types of headaches are discussed in this review. They are divided into primary and secondary headache disorders as classified by the International Headache Society. Primary headache disorders include migraine without and with aura, clust

Recent studies in normal subjects suggested that callosal motor fibers pass through the posterior body of the corpus callosum (CC), but this has not been tested in patients with callosal infarction. The objective of this study is to define the pathways involved in transcallosal inhibition by examining patients with infarctions in different subregions of the CC. We hypothesized that patients with lesions in the posterior one-half of the CC would have greater reduction in transcallosal inhibition between the motor cortices. Twenty-six patients with callosal infarction and 14 healthy subjects wer

OBJECTIVE: To investigate whether hand muscle repetitive use reduces surround inhibition (SI) as observed in patients with focal hand dystonia, we performed a transcranial magnetic stimulation (TMS) study in 15 healthy right-handed volunteers.
METHODS: TMS was set to be triggered by self-initiated flexion of the index finger at 3ms after movement onset. Motor evoked potentials (MEPs) of the abductor digiti minimi (ADM) were measured before and at 0, 10, 20 and 30min after 'single' (little finger abduction) and 'dual' (both index finger flexion and little finger abduction) exercise at 0.5Hz fo
