Depolarization and electrical stimulation enhance in vitro and in vivo sensory axon growth after spinal cord injury.

Activity dependent plasticity is a key mechanism for the central nervous system (CNS) to adapt to its environment. Whether neuronal activity also influences axonal regeneration in the injured CNS, and whether electrical stimulation (ES) can activate regenerative programs in the injured CNS remains incompletely understood. Using KCl-induced depolarization, in vivo ES followed by ex-vivo neurite growth assays and ES after spinal cord lesions and cell grafting, we aimed to identify parameters important for ES-enhanced neurite growth and axonal regeneration. Using cultures of sensory neurons, neur

Normal glucose enhances neuronal regeneration after lidocaine-induced injury.

BACKGROUND: Local anaesthetics exhibit direct neurotoxic effects on neurones. Numerous studies have investigated the factors that may reverse this neuropathology, but the effects of glucose conditions on neuronal regeneration after lidocaine-induced injury have not been examined by observing living neurones. The present study investigated the effects of different glucose conditions on neurite length, growth cone regeneration, and cell death in dorsal root ganglia (DRG) neurones after lidocaine-induced injury in vitro. METHODS: DRG explants were isolated from chick embryos at embryonic day 8 a

Activation of taf7l promotes sensory nerve regeneration by enhancing conditioning effect.

Peripheral nervous system (PNS) neurons can regenerate after injury, yet functional recovery is often limited, especially following proximal lesions or delayed repair. Preconditioning nerve injury accelerates axonal growth by inducing regeneration associated genes (RAGs). To identify novel growth regulators, we screened nine genetically diverse Collaborative Cross (CC) mouse strains for strain-dependent differences in dorsal root ganglion (DRG) neurite outgrowth on Matrigel. All strains exhibited a ≥ 2-fold preconditioning effect, with DBA/2J showing the most pronounced growth. Genome-wide exp

Early White Matter Microstructure Alterations in Infants with Down Syndrome.

Down syndrome (DS), resulting from Trisomy 21, is the most prevalent chromosomal disorder and a leading cause of intellectual disability. Despite the significant impact of Trisomy 21 on brain development, research on white matter (WM) microstructure in infants with DS remains limited. While widespread reductions in WM integrity have been identified in children and young adults with DS, no study has examined WM microstructure in infancy. This study investigates early WM microstructure in infants with DS using diffusion tensor imaging (DTI) and neurite orientation dispersion and density imaging

Innervation of cervical carcinoma is mediated by cancer-derived exosomes.

OBJECTIVE: Recently, our laboratory identified sensory innervation within head and neck squamous cell carcinomas (HNSCCs) and subsequently d … ith cancer-derived exosomes, stained with the pan-neuronal marker (β-III tubulin) and the number of neurites quantified. To assess innervation in cervical cancer, we immunohistochemically stained cervical cancer patient samples for β-III tubulin and TRPV1 (sensory marker) and compared the staining to normal cervix. RESULTS: Here, we show the presence of s …

Wnt5a regulates midbrain dopaminergic axon growth and guidance.

During development, precise temporal and spatial gradients are responsible for guiding axons to their appropriate targets. Within the develo … M explants with Wnt5a-overexpressing cell aggregates revealed that Wnt5a is capable of repelling DA neurites. Antagonism experiments revealed that the effects of Wnt5a are mediated by the Frizzled receptors and by the small GTPase, Rac1 (a component of the non-canonical Wnt planar cell polarity pathway). Moreover, the effects were specific as they could be …

Isolated hepatic granulomas from mice infected with Schistosoma mansoni contain nerve growth factor.

Nerve growth factor (NGF) is a neurotrophic factor essential for the development and maintenance of specific neuronal cell populations. In addition, NGF has biological effects on inflammatory cells. The aim of this study was to determine whether NGF is present in chronic inflammation, using isolated hepatic granulomas from mice infected with Schistosoma mansoni as the model. The schistosome granuloma is a complex T-cell-mediated immune response to the egg. Intact granulomas were isolated from the livers of infected mice and examined for the presence of NGF. In homogenized granuloma samples, ra

Alterations in neuron morphology in mucopolysaccharidosis type I. A Golgi study.

Morphological changes in neurons with inborn defects of the lysosomal hydrolase, alpha-L-iduronidase, and with concomitant storage of glycosaminoglycans, were evaluated by Golgi staining in two animal models and compared to a similar study of a child with the same disease. Cortical pyramidal neurons in feline mucopolysaccharidosis type I often displayed axon hillock enlargements (meganeurites) and/or ectopic, secondary neuritic processes sprouting from this same region of the cell. The latter structures were prominent and often appeared longer than similar neurites reported in other neuronal s…

Relation of target encounter and neuronal death to nerve growth factor responsiveness in the developing mouse trigeminal ganglion.

An aim of this study was to define precisely the period in the developmental history of primary sensory neurons during which nerve growth fa … pheral fibers were first visible and increased in number until E13. Throughout this period in vitro neurites grew in the presence of anti-NGF. Peripheral fibers initially contacted the epithelium of the mandibular process by E10.5 and the maxillary process by E11. This coincided with the stage in vitro during which the magnitude of neurite outgrowth was si …

miR-196a enhances polymerization of neuronal microfilaments through suppressing IMP3 and upregulating IGF2 in Huntington's disease.

Huntington's disease (HD) is one of the inheritable neurodegenerative diseases, and these diseases share several similar pathological characteristics, such as abnormal neuronal morphology. miR-196a is a potential target to provide neuroprotective functions, and has been reported to enhance polymerization of neuronal microtubules in HD. While microtubules and microfilaments are two important components of the neuronal cytoskeleton, whether miR-196a improves neuronal microfilaments is still unknown. Here, we identify insulin-like growth factor 2 mRNA binding protein 3 (IMP3), and show that miR-1

Implication of Endoplasmic Reticulum Stress in Autism Spectrum Disorder.

Autism spectrum disorder (ASD) is categorized as a neurodevelopmental disorder according to the Diagnostic and Statistical Manual of Disorders, Fifth Edition and is defined as a congenital impairment of the central nervous system. ASD may be caused by a chromosomal abnormality or gene mutation. However, these etiologies are insufficient to account for the pathogenesis of ASD. Therefore, we propose that the etiology and pathogenesis of ASD are related to the stress of the endoplasmic reticulum (ER). ER stress, induced by valproic acid, increased in ASD mouse model, characterized by an unfolded

[Leprosy tests: diagnostic problems (apropos of 2 cases)].

Two cases of reactional leprosy leading to wrong diagnosis are reported. The first one concerns a reversal reaction predominantly neuritic, initially taken for polyarthritis. The second one concerns an erythema nodosum leprosum with extensive cutaneous necrosis (Lucio's phenomenon or ulcerative lazarine leprosy). Main aspects and mechanisms of leprosy reactional states are reviewed. It is emphasized that errors or delays in diagnosis are often caused by failing to recognize cutaneous or neuritic symptoms.

Ubiquitin immunoreactivity in kuru plaques in Creutzfeldt-Jakob disease.

Cerebellar kuru plaques in 2 cases of Creutzfeldt-Jakob disease were studied immunohistochemically. Similar to cerebellar senile plaques in Alzheimer's disease, many kuru plaques contained ubiquitin-positive, tau-negative small granular elements, presumably representing dystrophic neurites. Our results suggest that similar mechanisms are involved in neuritic changes in cerebellar plaques in Creutzfeldt-Jakob and Alzheimer's diseases despite differences of amyloid proteins in the plaques.

Detection of apolipoprotein E/dimeric soluble amyloid beta complexes in Alzheimer's disease brain supernatants.

The inheritance of the apolipoprotein (apo) E4 allele is an important risk factor for late-onset Alzheimer's disease (AD). A major component of the Alzheimer's disease neuritic plaques is amyloid beta (A beta). We previously identified apoE/A beta complexes within neuritic plaques (1). It was not known if this interaction takes place before or after A beta peptides become incorporated into neuritic plaques. To address this question we sought evidence of apoE complexes with brain soluble A beta peptides in AD and control patients. In addition, numerous proteins have been shown to bind A beta pe

Laminin "A" chain fragment of Mr 43 kilodalton contains PC12 cell attachment promoting site.

In the present study, the ability of a 43 K fragment, originating from the long arm of the 400 K laminin "A" chain (Rao and Kefalides, manuscript in press), to promote the attachment and de novo neurite outgrowth by the pheochromocytoma cell line, (PC12) was studied. Unprimed PC12 cells (not treated with nerve growth factor, NGF), just as the primed cells (treated with NGF), rapidly and efficiently attach to the laminin fragment but less efficiently to the intact laminin coated bacteriological plastic surfaces. The neurite outgrowth was continuous from primed cells attached to both laminin and

Neuroinvasion by herpes simplex virus. An in vitro model for characterization of neurovirulent strains.

A cell culture technique with rat dorsal root ganglion neurons cultured in a two-chamber system allowing exposure of neuritic extensions or neuronal cell soma to herpes simplex virus (HSV) infection was exploited for studies of viral neuroinvasiveness. Four groups of HSV strains were investigated. Type 1 strains were isolated from cases of either encephalitis or mucocutaneous infections and type 2 strains were from cases of meningitis or genital infections. The neuroinvasiveness of the type 1 encephalitis strains was significantly greater than the neuroinvasiveness calculated for the type 1 st

Ultrastructural diversity and subcellular organization of nigral Lewy pathology in Parkinson's disease.

Lewy bodies, the defining pathological feature of Parkinson's disease, are intraneuronal inclusions enriched in aggregated alpha-synuclein (αSyn). We used correlative light and electron microscopy to selectively investigate phosphorylated αSyn (αSynpS129)-positive inclusions in the substantia nigra of end-stage postmortem Parkinson's disease brain. Here we show that somatic αSynpS129 inclusions in nigral dopaminergic neurons are consistently fibrillar, whereas the membranous-type inclusions are restricted to neuritic processes. These neuritic inclusions displayed marked ultrastructural heterog
