Varicella-zoster virus leukoencephalitis and cerebral vasculopathy.

Two patients with varicella-zoster virus leukoencephalitis and acquired immunodeficiency syndrome are described. Neither patient had cutaneous or disseminated varicella-zoster virus infection within the last six months of life. Demyelinated lesions resembling those of progressive multifocal leukoencephalopathy were seen in their brains at autopsy. Numerous cells with Cowdry type A intranuclear inclusions surrounded the lesions; these cells stained positively for varicella-zoster virus with immunohistochemistry and contained herpesvirus nucleocapsids by electron microscopy. Leptomeningeal vesse…

[Electrophoretic and sedimentation heterogeneity of the virion antigen of the tick-borne encephalitis virus].

Analysis of tick-borne encephalitis virus virion antigen by cross immunoelectrophoresis showed it to be separated into 2 subpopulations of virions differing by their mobility in the electric field. It was demonstrated that in immunoelectrophoresis the formation of precipitation bands typical of virion antigen occurred due to the movement of soluble immune complexes in the electric field. The immunoelectrophoretic characteristics of TBE virus virions are determined by the membrane E protein which was also found to be heterogeneous in its immunoelectrophoretic characteristics. The nucleocapsid o…

An In Vitro RNA Synthesis Assay for Rabies Virus Defines Ribonucleoprotein Interactions Critical for Polymerase Activity.

UNLABELLED: We report an in vitro RNA synthesis assay for the RNA-dependent RNA polymerase (RdRP) of rabies virus (RABV). We expressed RABV … RdRP. The L-P complex lacks full processivity, which we interpret to reflect the lack of the viral nucleocapsid protein (N) on the template. Using this assay, we define the requirements in P for stimulation of RdRP activity as residues 11 to 50 of P and formally demonstrate that ribavirin triphosphate (RTP) inhibits the RdRP. By comparing the properties of RA …

Selective inhibition of rabies virus gene expression by human miRNAs: a therapeutic approach using the 7mer-m8 model.

MicroRNAs, abbreviated as miRNAs, have a substantial impact on viral infections through their ability to control gene expression and influence the interactions between the host and the virus. This work investigates the capacity of miRNAs to selectively inhibit the expression of rabies virus genes, specifically Nucleoprotein N, Phosphoprotein M1 and M2, Transmembrane Glycoprotein G, and L protein. The 7mer-m8 model was utilized to identify human miRNAs that target these viral genes. The interactions between the miRNAs and the genes were then assessed based on binding energy, GC content, and sta

[EDP-938, a new antiviral with inhibitory activity against the nucleoprotein of the respiratory syncytial virus].

The absence of an effective vaccine against respiratory syncytial virus (RSV) has led to the development of various drugs with the ability to inhibit or block its replicative activity. The first generation, called fusion inhibitors, bind to the protein on the viral surface and prevent the virus from binding and entering the cell. However, its low efficacy has determined the start of studies with second-generation compounds capable of binding or blocking the nucleoprotein (N); most of these compounds are analogs of 1,4-benzodiazepines. EDP-938 has shown high efficacy against RSV. The first tria

Development and Evaluation of Two Double-Antibody Sandwich ELISAs for Detecting Severe Fever with Thrombocytopenia Syndrome Virus Infection.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is a newly emerging tick-borne virus with a fatality rate between 12% and 50%. Currently, effective vaccines or antiviral drugs are not available to treat SFTSV infection, and a diagnostic method for its detecting is urgently needed. Monoclonal (MAb) and polyclonal antibodies (PAbs) against SFTSV were prepared by immunizing animals with the SFTSV nucleocapsid protein (NP). We developed 2 double-antibody sandwich enzyme-linked immunosorbent assays (DAS-ELISAs) to detect the SFTSV NP, which was captured using the MAbs and PAbs generated. …

A paramyxovirus-like model for Ebola virus bipartite promoters.

Paramyxo- and filovirus nucleocapsids (NCs) have bipartite promoters at their 3' ends to initiate RNA synthesis. The 2 elements, promoter element 1 (PE1) and promoter element 2 (PE2), are separated by a spacer region that must be exactly a multiple of 6 nucleotides (nt) long. Paramyxovirus NCs have 13 nucleoprotein (NP) subunits/turn, such that PE1 and PE2 are juxtaposed on the same face of the NC helix, for concerted recognition by the viral polymerase. Ebola virus (EBOV) NCs, in contrast, have 25 to 28 subunits/turn, meaning that PE1 and PE2 cannot be juxtaposed. However, there is evidence t…

A molecular epidemiological study of rabies in Cuba.

To investigate the emergence and current situation of terrestrial rabies in Cuba, a collection of rabies virus specimens was employed for genetic characterization. These data supported the monophyletic nature of all terrestrial rabies viruses presently circulating in Cuba but additionally delineated several distinct variants exhibiting limited spatial distribution which may reflect the history of rabies spread on the island. The strain of rabies currently circulating in Cuba, which emerged on the island in the early 20th century, has very close evolutionary ties to the Mexican dog type and is

Molecular characterization of rabies virus isolates in China during 2004.

Human rabies cases have been on the rise during the past few years in China and a total of 2651 cases were reported in 2004. To better understand the current rabies epidemics in China, we isolated rabies viruses from dogs and humans from five provinces and characterized these isolates genetically by sequencing the entire nucleoprotein (N) gene. Comparison of the N genes among these isolates revealed 86.6-99.9% homology and these viruses can be grouped into three lineages. Phylogenetic analysis indicates that all the Chinese isolates have a close relationship with viruses circulating in Asian c

A recombinant adenovirus-vectored PEDV vaccine co-expressing S1 and N proteins enhances mucosal immunity and confers protection in piglets.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious enteric coronavirus that causes severe diarrhea and high mortality in neonatal piglets. Effective control requires systemic, mucosal, and T cell-mediated immune responses, highlighting the need for a safe, effective, and versatile vaccine platform. In this study, a recombinant adenovirus type 5-based vaccine co-expressing the PEDV S1 and N proteins (rAd5-S1-N) was developed, and its immunogenicity and protective efficacy were evaluated in mice and piglets. Intranasal immunization of BALB/c mice with rAd5-S1-N induced strong and sust

4-Diketopyrrole-2-carboxamide derivatives affecting HBV capsid assembly: structure-activity relationship around the GLP-26 scaffold.

Chronic hepatitis B virus (HBV) infection remains a major global health burden with limited curative options. Capsid assembly modulators (CAMs) have emerged as a promising antiviral strategy by disrupting nucleocapsid formation and suppressing cccDNA amplification. In this study, we discuss the structure-activity relationship (SAR) around the 4-diketopyrrole-2-carboxamide scaffold of GLP-26, focusing on modifying the aniline moiety and the oxoacetamide region, and introducing alternative acetamide groups. Compounds were synthesized through efficient coupling strategies, including microwave-ass…

AI-discovered protein fragments as generalizable regulators of biomolecular condensates.

Biomolecular condensates are a major driver of cellular organization; however, we lack a predictable and systematic approach to modulate their underlying multivalent interactions. Here, we demonstrate a generalizable AI-driven method for designing protein fragments to control condensate formation, applying this approach across G3BP1, SARS-CoV-2 nucleocapsid, TDP-43, and focal adhesion kinase (FAK). Computationally screening 2,235 fragments, we selected 18 for experimental investigation, attaining a 50% success rate. Furthermore, predicted fragment binding modes align with their activities, rev…

The regulatory mechanisms of SARS-CoV-2 N protein helicase and its annealing activity.

The nucleocapsid (N) protein of SARS-CoV-2 performs multiple functions essential for viral replication and host adaptation. Here, we identify ionic strength as a critical determinant of its dual nucleic acid-manipulating activities. Under low-salt conditions (≤100 mM NaCl), SARS-CoV-2 N primarily promotes strand annealing, whereas at high ionic strength (≥300 mM NaCl), its helicase activity is additively enhanced. SARS-CoV-2 N also preferentially acts on DNA rather than RNA substrates and modulates the helicase activity of Nsp13 in an ion-dependent manner-inhibiting it under low-salt condition…
