Prolonged inhibition by X-rays of DNA synthesis in cells obtained by transformation of primary rat embryo fibroblasts with oncogenes H-ras and v-myc.

Transfection of primary rat embryo fibroblasts with the H-ras oncogene plus the cooperating oncogene v-myc results in the development of foci of morphologically altered tumorigenic cells. We examined radiation (X-rays) induced inhibition of DNA synthesis in cell lines derived from such transformed clones and compared the results to those obtained with the nontransformed parental cells, rat embryo fibroblasts, as well as with cells immortalized either spontaneously, or after transfection with nuclear oncogenes (v-myc, E1A). Inhibition by X-rays of DNA synthesis was higher and persisted for long…

Breaking adaptive oncogenic resistance in glioblastoma via targeting the EGFR-STAT3 axis.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults and remains refractory to current targeted therapies. Among the oncogenic pathways implicated in GBM pathogenesis, epidermal growth factor receptor (EGFR) signaling and the Janus kinase/signal transducer and activator of transcription 3 (JAK/STAT3) pathway have emerged as central drivers of tumor growth, immune evasion, and therapeutic resistance. Despite frequent EGFR amplification and mutation, clinical trials targeting EGFR have yielded disappointing outcomes, underscoring the existence of adaptive resistance mechanisms

Targeting Exportin-1 in Cancer: From Nuclear Export to Oncogenic Transcriptional Hubs.

Maintaining cellular homeostasis requires precise coordination of molecular trafficking between the nucleus and the cytoplasm. Central to this process is the nucleocytoplasmic transport system, which governs the spatial distribution of proteins and RNA. Exportin-1 (XPO1), also known as chromosome region maintenance 1 (CRM1), is the principal nuclear export receptor responsible for the transport of hundreds of cargo proteins bearing leucine-rich nuclear export signals. In a wide range of malignancies, XPO1 is frequently dysregulated through overexpression or recurrent somatic mutations. This dy

Engineered exosomes for CRISPR/Cas delivery to overcome oncogene-driven drug resistance.

Engineered exosomes are emerging as biocompatible nanocarriers for delivering CRISPR/Cas components to resistant tumor cells, enabling targeted disruption of oncogenic drivers and resistance-associated pathways. Engineered exosomes offer several delivery-platform advantages, including biocompatibility, membrane-mediated cargo protection, programmable tumor targeting, and potential tissue penetration. Selection of the CRISPR modality, Cas9 ribonucleoprotein, mRNA, base editor, or prime editor, depends on payload size, stability, editing duration, endosomal escape, and nuclear delivery requireme

Targeting Mouse Double Minute 2 Homolog (MDM2) Oncogene in Breast Adenocarcinoma.

INTRODUCTION: Breast adenocarcinoma (BAC) is a leading cause of cancer-dependent morbidity and mortality in females worldwide. Concerning critical genes that are implicated in its onset and progression in BAC, the tumor suppressor protein Tp53 (gene locus: 17p13.1) and its negative regulator the Mouse Double Minute 2 Homolog (MDM2) - an oncogene (gene locus: 12q14.3) - form a significant feedback loop. OBJECTIVE: The aim of the current molecular review was to investigate the MDM2 oncogene deregulation mechanisms in BAC and the corresponding targeted therapeutic approaches. MATERIAL AND METHO

The YAP1-NPM1 nuclear complex regulates MYC and reveals a targetable oncogenic node.

Yes-associated protein 1 (YAP1) and nucleophosmin 1 (NPM1) regulate diverse cellular functions; however, their cooperative roles in cancer remain unclear. Here, we demonstrate that YAP1 and NPM1 form a direct nuclear protein complex in prostate cancer cells and tissues, as revealed by proteomics, co-immunoprecipitation, proximity ligation, and GST-pulldown assays. Androgen signaling modulates the YAP1-NPM1 interaction in androgen receptor-positive cells. Genetic or pharmacologic inhibition of NPM1 disrupts YAP1-NPM1 protein assembly, decreases YAP1 and MYC expression, and suppresses cell proli

Dedifferentiation-driven oncogenic stemness promotes tumor-sustaining adaptability in the intestinal epithelium.

Intestinal tumorigenesis can occur via two distinct routes: bottom-up tumorigenesis occurs from mutations sustained in the Lgr5⁺ stem cells, whereas top-down tumorigenesis is driven by dedifferentiation of epithelial cells near the intestinal lumen. While sporadic human colon adenomas exhibit features of top-down tumorigenesis, their biological determinants remain elusive. Here, using a Smad4 loss-of-function and β-catenin gain-of-function (Smad4LOF:β-cateninGOF) mouse model, we demonstrate that dedifferentiation-derived oncogenic stem cells sustain tumorigenesis more effectively than endogeno

Lipid and Carbohydrate Modifications Enhance Potency of Antisense Oligonucleotides Targeting Oncogenic KRAS.

One prominent open question in the field of antisense oligonucleotide remains the intracellular delivery of active sequences. While different strategies have been developed for addressing this issue, including formulation and/or conjugation approaches, multiconjugations with both sugars and nucleolipids have not been investigated so far. In this contribution, we are interested in a new hypothesis that focuses on the double conjugation of oligonucleotides with both glucose-thymine and nucleolipid moieties. The lipid carbohydrate modified antisense oligonucleotides were the most efficient in dow

Oncogenic KRAS Promotes Extracellular DNA Clearance to Drive Immunosuppression in Colorectal Cancer.

Cellular turnover and death result in shedding of DNA into the extracellular space. In the context of cancer, the constant turnover of cancer cells leads to the release of tumor-derived DNA (tDNA) into the circulation. Circulating tDNA (ctDNA) provides molecular insights into the genetic alterations present within cancer cells, which has been exploited to noninvasively detect and monitor tumors. In addition to its utility in cancer monitoring, tDNA has also been implicated in the formation of neutrophil extracellular traps that can promote oncogenesis and metastasis. Despite the appreciation t…

LNP-mediated silencing of oncogenic lncRNA PVT1 enhances chemotherapeutic efficacy in osteosarcoma.

Osteosarcoma is an aggressive bone malignancy characterized by early metastasis, chemoresistance, and poor prognosis, particularly in recurrent or metastatic cases. The long non-coding RNA PVT1 has been implicated as an oncogene in various cancers, but its therapeutic potential in osteosarcoma remains underexplored. This study investigates the feasibility of targeting PVT1 using lipid nanoparticle-encapsulated siRNA (LNP-siPVT1) as a standalone or combination therapy for osteosarcoma. PVT1 expression was found to be significantly upregulated in osteosarcoma cell lines (U2OS, Saos-2, MG-63), pa

[Summary of second-line treatment of stage IV NSCLC without oncogenic addiction].

Stage IV non-small cell lung cancer (NSCLC) without actionable mutations remains a major therapeutic challenge, especially following immunotherapy failure. The introduction of immune checkpoint inhibitors (anti-PD-1/PD-L1) has revolutionized first-line treatment, but second-line options remain largely dependent on older chemotherapies such as docetaxel or pemetrexed. A significant number of patients experience disease progression despite immunotherapy, highlighting tumor resistance mechanisms, whether primary (rapid failure) or acquired (after initial response). Resistance mechanisms include l

CRIP1 functions as an oncogene in acute myeloid leukemia by modulating glycolytic metabolic reprogramming.

Acute Myeloid leukemia (AML) is a stem cell-derived hematological malignancy associated with rapid progression and poor clinical outcome despite available treatments. CRIP1 exhibits tissue-specific activity, functioning as an oncogene in certain cancers while acting as a tumor suppressor in others. However, the role of CRIP1 and its association with metabolism in AML remains poorly investigated. To assess its metabolic role, a lactate release and a glucose consumption assay were performed, followed by gene expression analysis of LDHA, MCL1, and HK2 after CRIP1 knockdown. These results were con

Oncogenic overexpression of kallikrein-related peptidase 7 is associated with the progression of thyroid cancer.

Mounting evidence suggests that the kallikrein-related peptidase family is dysregulated in thyroid cancer. However, previous analyses of kallikrein-related peptidase 7 (KLK7) expression patterns have yielded inconsistent results. In this study, KLK7 expression was examined in different histological types of thyroid cancer using immunohistochemistry. We found no detectable KLK7 immunoreactivity in the follicular epithelium of normal thyroid tissue or follicular adenomas. Weak KLK7 expression was observed in papillary (mean H-score 47) and follicular carcinomas (mean H-score 29), with moderate e

An autochthonous CRISPR activation screening platform for characterizing tissue-specific oncogene selection.

Human adenocarcinomas exhibit tissue-specific mutation and copy-number patterns that suggest diverse selective pressures and distinct oncogene dependencies. Here, we use our FiCASCan platform to test whether in vivo CRISPR activation screening can recapitulate oncogene selection during tumor initiation. Using CRISPRa-competent PPKS mice and intranasal or intraductal delivery of pooled lentivirus, we screen frequently amplified and mutated genes in autochthonous lung and pancreas cancer models. We observe strong selection for Egfr, Myc, Sox2, and Pik3cb activation in lung tumors and near-comple

Oncogenic Signaling Activation and Potential Biomarkers in Congestive Hepatopathy Revealed by Proteomic Analysis.

AIM: Fontan surgery improves outcomes in univentricular heart disease; however, some patients develop severe Fontan-associated liver disease (FALD). Even during adolescence, hepatocellular carcinoma occurs in some patients with FALD, although the mechanism remains unclear. In this study, we conducted proteomic analyses of liver tissues obtained using laser capture microdissection (LCM) and serum samples to investigate FALD-related peripheral liver oncogenic factors and explore potential biomarkers. METHODS: Ten-week-old mice underwent sham surgery, and age-matched mice underwent partial ligat

RET fusion partners dictate oncogenic potential in undifferentiated spindle cell sarcomas.

BACKGROUND: Spindle cell tumors with RET rearrangements exhibit variable clinical behaviors, ranging from benign to highly aggressive malignancies. The underlying heterogeneity is suspected to be associated with distinct fusion partner genes. Understanding the impact of these fusion partners on oncogenic potential is crucial for precision therapy. METHODS: We report a case of a spindle cell tumor harboring an MYH10::RET fusion, which initially responded to anti-RET therapy but relapsed due to an NTRK1 fusion. Isogenic cell lines expressing MYH10::RET and CCDC6::RET were established. Functiona

Oncogenic Gα signaling requires AP-3-dependent recruitment to the endolysosomal compartment.

G protein-coupled receptors (GPCRs) constitute the largest superfamily of cell-surface receptors, yet the spatial constraints governing their signaling remain poorly defined. Here, we demonstrate that Gα, an essential downstream component of GPCR signaling, undergoes a spatial shift from the plasma membrane to the endolysosomal compartment upon its constitutive activation. Using a genome-wide CRISPR screen, we identify the adaptor protein AP-3 as the essential mediator of this trafficking event. Loss of AP-3-dependent recruitment impairs Gα-driven signaling and proliferation in uveal melanoma
