Despite extensive research on insect oils, their application in food systems remains limited. In this study, roasting pretreatment was integrated with ultrasound-assisted extraction to enhance nutritional and sensory properties of insect oils. Optimal roasting conditions were 160 °C for 25 min at 60 rpm for Protaetia brevitarsis larvae and 155 °C for 35 min at 75 rpm for Gryllus bimaculatus. Incorporating these oils into bone broth significantly increased oleic, linoleic, and palmitic acids, resulting in a fatty acid profile similar to that of beef. G. bimaculatus oil contained elevated levels…

This study investigated how cream origin and fermentation time impact sour cream flavor by integrating microbial dynamics, fatty acid profiles, and volatile formation under pilot-scale conditions. Korean sour cream (KRS; produced from in-house cream) and Italian sour cream (ITS; produced from frozen cream) samples were fermented with Lactobacillus delbrueckii and Streptococcus thermophilus for 24 h. KRS exhibited greater lipid degradation, with palmitic acid decreasing by 7.8 %, and showed more dynamic volatile shifts, particularly at 12-18 h. Ethanol increased 1.35-fold and butanoic acid was

This study provides a targeted bioactive lipid and nutritional profile of the under-investigated leaves of Vitex agnus-castus L. An absolute methanolic extract of shade-dried leaves was analysed using gas chromatography/mass spectrometry (GC/MS) after derivatization, alongside essential mineral quantification via atomic absorption spectroscopy (AAS) and proximate analysis. Rigorous data validation excluded analytical artefacts. GC/MS tentatively identified ten validated bioactive compounds, predominantly glycerol (45.21%, a lipid hydrolysis artefact), oleic acid (9.89%), and palmitic acid (9.1

The tumor microenvironment (TME) domesticates macrophage function by decreasing chromatin accessibility. The activation and nuclear translocation of ATP-citrate lyase (ACLY) convert citrate to acetyl-CoA, providing a substrate necessary for histone acetylation. However, the underlying mechanisms in macrophage remodeling are poorly understood. Here, we found that saturated fatty acids (sFA), especially palmitic acid (PA), were lower in TAMs of patients with hepatocellular carcinoma (HCC). Scd1 knockout promoted PA accumulation, resulting in both primary and metastatic liver cancer retardation a

The overall response rate to immunotherapy is modest in hepatocellular carcinoma (HCC), and immunotherapy resistance mechanisms are incompletely understood. We report that the E3 ubiquitin ligase Riplet is universally silenced by promoter hypermethylation in HCC. Loss of Riplet modulates fatty acid metabolism to promote terminal exhaustion of CD8 T cells. Riplet loss impedes K48-linked polyubiquitination of fatty acid synthase (FASN), consequently accelerating fatty acid production in HCC. Tumor cell-derived free fatty acids, especially palmitic acid (PA/C16:0), activate STAT3 (signal transduc

Nine CYRS-FA complexes were prepared by resistant starch in Chinese yam (CYRS) and nine fatty acids (FAs) with different chain lengths and degrees of unsaturation. CYRS-myristic acid and CYRS-palmitic acid showed higher complexing index (CI) and relative crystallinity (RC); CYRS-myristic acid and CYRS-oleic acid exhibited lower estimated glycemic index (eGI). Chain lengths of FAs showed significantly positive correlations with CI and contact angle (CA), and yet, unsaturation degree of FAs was negative correlated with both CI and CA. The eGI exhibited positive relations with solubility, and neg

In this study, we continue our research on Rubus ulmifolius Schott extracts obtained by supercritical fluid extraction (SFE) using CO2 as solvent. Extraction experiments were carried out at pressures of 300 bar and a temperature of 40 °C, SFE [300:40], on mature fruits of R. ulmifolius. The fatty acid (FA) profile of extracts was assessed by reversed-phase HPLC-DAD/ELSD analysis. The most represented fatty acids in the extract were linoleic acid (386 mg/g), oleic acid (210 mg/g), linolenic acid (112 mg/g), palmitic acid (48 mg/g), and stearic acid (40 mg/g). SFE extraction showed a total FA am

Lipolysis and flavor development during fermentation of sour cream were studied by evaluating the physicochemical changes, sensory differences and volatile components. The fermentation caused significant changes in pH, viable count and sensory evaluation. The peroxide value (POV) decreased after reaching the maximum value of 1.07 meq/kg at 15 h, while thiobarbituric acid reactive substances (TBARS) increased continuously with the accumulation of secondary oxidation products. The Free fatty acids (FFAs) in sour cream were mainly myristic, palmitic and stearic. GC-IMS was used to identify the fl

Quinolinic acid (QUIN), one of the end metabolites in the kynurenine pathway, plays an important role in the pathogenesis of several diseases. Serum QUIN concentration rises in patients with renal dysfunction, liver cirrhosis, and many other inflammatory diseases. In the present study, osmotic minipumps containing QUIN (0.3 and 1mg/day) were implanted intraperitoneally into rats for 28days. Then, the physiological and toxicological variables were evaluated and LC-QTOF-MS serum metabolic fingerprinting was performed. QUIN significantly decreased the serum concentrations of several amino acids (

A 24-hr pretreatment of cultured human fibroblasts with trifluoperazine induced a marked increase in incorporation of saturated (stearic, palmitic) and unsaturated (oleic, arachidonic) fatty acids into phospholipids (1.5- to 2-fold for 5.10(-5) M trifluoperazine). Concomitantly, incorporation into cholesteryl esters was strongly inhibited (20% of control for 5.10(-5) M trifluoperazine). The drug did not change the phospholipid composition of treated cells. The effect of trifluoperazine on oleic acid incorporation into phospholipids was time-dependent and reached a maximum after a six-hr preinc

The effect of free fatty acids (FFAs) on the binding of antibiotics to human serum proteins was studied by addition of palmitic acid to albumin solutions and normal pooled serum. FFA reduced the binding of dicloxacillin, cefamandole, and sulfamethoxazole at molar ratios of FFA to albumin of greater than 2.0. In contrast, FFA enhanced the binding of benzylpenicillin, cephalothin, and cefoxitin at physiologic molar ratios of FFA to albumin. Elevated levels of FFAs induced in vivo by heparin produced similar results with dicloxacillin and benzylpenicillin. Addition of FFA to serum, but not to alb

Determination of fatty acid compositions and contents in Chinese mitten crabs is of great significance to evaluate its nutritional value and quality. However, in the face of a wide range of fatty acid extraction and methyl esterification reagents, the measurement results are uneven, and it is difficult to accurately quantify the rich fatty acids in Chinese mitten crabs. In this paper, four kinds of oil extraction reagents and two kinds of methylating reagents, were investigated. Chloroform-methanol (1∶1, v/v) was used as the extraction solvent, and methanol containing 2% sulfuric acid was used

Aromatase inhibitors (AI) are first-line therapy for postmenopausal women with estrogen receptor-expressing (ER+) breast cancer (BC). AI therapy effectively reduces recurrence and extends lifespan for patients with ER+ BC through long-term estrogen deprivation (LTED) resulting from inhibition of the enzyme aromatase that converts androgens to estrogens. However, up to 50% of ER+ BC recurs as AI-resistant metastatic disease within 10 years of diagnosis. AI-resistant BC upregulates androgen receptors (AR) and mitochondrial oxidative phosphorylation (OXPHOS) and requires OXPHOS and fatty acid oxi

A series of glycolipids (glycopyranosides) was prepared by coupling glucosamine or galactosamine (at the C2 position) with myristic (tetradecanoic) or palmitic (hexadecanoic) acid. The conformation and self-assembly were examined in aqueous solutions containing 10% methanol. Circular dichroism and FTIR spectroscopy reveal notable differences in the chiral ordering and conformation comparing analogues with glucose and galactose "headgroups". A glucose derivative (to palmitic acid) was also prepared with a different (C1-) substitution position of the lipid chain, and this was found to significan

The pathogenesis of non-alcoholic fatty liver disease (NAFLD) is influenced by a number of variables, including endoplasmic reticulum stress (ER). Thioredoxin domain-containing 5 (TXNDC5) is a member of the protein disulfide isomerase family and acts as an endoplasmic reticulum (ER) chaperone. Nevertheless, the function of TXNDC5 in hepatocytes under ER stress remains largely uncharacterized. In order to identify the role of TXNDC5 in hepatic wild-type (WT) and TXNDC5-deficient (KO) AML12 cell lines, tunicamycin, palmitic acid, and thapsigargin were employed as stressors. Cell viability, mRNA,

INTRODUCTION AND OBJECTIVES: The incidence of non-alcoholic fatty liver disease (NAFLD) is increasing. Previous studies indicated that Liraglutide, glucagon-like peptide-1 analogue, could regulate glucose homeostasis as a valuable treatment for Type 2 Diabetes. However, the precise effect of Liraglutide on NAFLD model in rats and the mechanism remains unknown. In this study, we investigated the molecular mechanism by which Liraglutide ameliorates hepatic steatosis in a high-fat diet (HFD)-induced rat model of NAFLD in vivo and in vitro.
MATERIALS AND METHODS: NALFD rat models and hepatocyte s

BACKGROUND & AIMS: Our previous studies have provided evidence that p38 mitogen-activated protein kinase (MAPK) is involved in total parenteral nutrition (TPN)-associated complications, but its exact effects and mechanisms have not been fully understood. This study aimed to evaluate the roles of p38 MAPK inhibitor SB203580 in the TPN-induced loss of intestinal barrier function and liver disease.
METHODS: A rodent model of TPN was used to analyze the roles of SB203580 in TPN-associated complications.Intestinal barrier function was evaluated by transepithelial electrical resistance (TER) and pa
