Multi-omics analysis unravels dysregulated lysosomal function and lipid metabolism involved in sub-chronic particulate matter-induced pulmonary injury.

Particulate matter (PM) is a huge environmental threat and is of major public concern. Oxidative stress and systemic inflammation are known factors that contribute to PM- related damage; however, a systematic understanding of the deleterious pulmonary effects of PM using multi-omics analysis is lacking. In this study, we performed transcriptomic, proteomic, and metabolomic analyses in a mouse model exposed to PM for three months to identify molecular changes in lung tissues. We identified 1690 genes, 326 proteins, and 67 metabolites exhibiting significant differences between PM-challenged and …

Evidence for Assimilatory Nitrate Reduction as a Previously Overlooked Pathway of Reactive Nitrogen Transformation in Estuarine Suspended Particulate Matter.

Suspended particulate matter (SPM) contributes to the loss of reactive nitrogen (Nr) in estuarine ecosystems. Although denitrification and anaerobic ammonium oxidation in SPM compensate for the current imbalance of global nitrogen (N) inputs and sinks, it is largely unclear whether other pathways for Nr transformation exist in SPM. Here, we combined stable isotope measurements with metagenomics and metatranscriptomics to verify the occurrence of dissimilatory nitrate reduction to ammonium (DNRA) in the SPM of the Pearl River Estuary (PRE). Surprisingly, the conventional functional genes of DNR…

Nucleotide Excision Repair Pathway Activity Is Inhibited by Airborne Particulate Matter (PM10) through XPA Deregulation in Lung Epithelial Cells.

Airborne particulate matter with a diameter size of ≤10 µm (PM10) is a carcinogen that contains polycyclic aromatic hydrocarbons (PAH), which form PAH-DNA adducts. However, the way in which these adducts are managed by DNA repair pathways in cells exposed to PM10 has been partially described. We evaluated the effect of PM10 on nucleotide excision repair (NER) activity and on the levels of different proteins of this pathway that eliminate bulky DNA adducts. Our results showed that human lung epithelial cells (A549) exposed to 10 µg/cm2 of PM10 exhibited PAH-DNA adducts as well as an increase in…

Prolonged exposure to traffic-related particulate matter and gaseous pollutants implicate distinct molecular mechanisms of lung injury in rats.

BACKGROUND: Exposure to air pollution exerts direct effects on respiratory organs; however, molecular alterations underlying air pollution-induced pulmonary injury remain unclear. In this study, we investigated the effect of air pollution on the lung tissues of Sprague-Dawley rats with whole-body exposure to traffic-related PM1 (particulate matter < 1 μm in aerodynamic diameter) pollutants and compared it with that in rats exposed to high-efficiency particulate air-filtered gaseous pollutants and clean air controls for 3 and 6 months. Lung function and histological examinations were performed …

Contribution of tailpipe and non-tailpipe traffic sources to quasi-ultrafine, fine and coarse particulate matter in southern California.

Exposure to traffic-related air pollution (TRAP) in the near-roadway environment is associated with multiple adverse health effects. To characterize the relative contribution of tailpipe and non-tailpipe TRAP sources to particulate matter (PM) in the quasi-ultrafine (PM0.2), fine (PM2.5) and coarse (PM2.5-10) size fractions and identify their spatial determinants in southern California (CA). Month-long integrated PM0.2, PM2.5 and PM2.5-10 samples (n = 461, 265 and 298, respectively) were collected across cool and warm seasons in 8 southern CA communities (2008-9). Concentrations of PM mass, el…

The benzo[b]fluoranthene in the atmospheric fine particulate matter induces mouse glomerular podocytes injury via inhibition of autophagy.

BACKGROUND: Evidence shows that individuals who are under long-term exposure to environmental PM2.5 are at increased risk of chronic kidney disease. Various laboratory experiments also suggest several mechanistic links between PM2.5 exposure and kidney injury. Polycyclic aromatic hydrocarbons (PAHs) are common organic chemicals existing in PM2.5. However, whether benzo [b]fluoranthene (BbF), the most potent carcinogens and the highest content of PAHs, plays an important role in podocyte injury via reducing autophagy, have not been reported. METHODS: Podocytes were exposed to different concent

Effects of Placenta-Derived Mesenchymal Stem Cells on the Particulate Matter-Induced Damages in Human Middle Ear Epithelial Cells.

This study was aimed at investigating the effects of placenta-derived mesenchymal stem cells (PL-MSCs) on particulate matter- (PM-) exposed human middle ear epithelial cells (HMEECs). HMEECs were treated with 300 μg/ml PM for 24 hours. The PL-MSCs were cocultured with PM-treated HMEECs. Cells were harvested on days 0, 1, and 4, and the expression of the inflammatory genes TNFα, COX2, IL1β, IL6, and MUC5B in HMEECs and anti-inflammatory genes PTGES, TGFβ, and VEGF in PL-MSCs was examined by qRT-PCR. The culture media were collected to measure the secreted PGE2 level using an enzyme-linked immun…

Particulate matter, nitrogen oxides, ozone, and select volatile organic compounds during a winter sampling period in Logan, Utah, USA.

Particulate matter mass (PM), trace gaseous pollutants, and select volatile organic compounds (VOCs) with meteorological variables were measured in Logan, Utah (Cache Valley), for >4 weeks during winter 2017 as part of the Utah Winter Fine Particle Study (UWFPS). Higher PM levels for short time periods and lower ozone (O3) levels were present due to meteorological and mountain valley conditions. Nitrogenous pollutants were relatively strongly correlated with PM variables. Diurnal cycles of NOx, O3, and fine PM(PM 2.5) (aerodynamic diameter <2.5 μm [PM2.5]) suggested formation from NOx. O3 leve…

Effects of vitamin D on inflammatory and oxidative stress responses of human bronchial epithelial cells exposed to particulate matter.

BACKGROUND: Particulate matter (PM) pollutant exposure, which induces oxidative stress and inflammation, and vitamin D insufficiency, which compromises immune regulation, are detrimental in asthma. OBJECTIVES: Mechanistic cell culture experiments were undertaken to ascertain whether vitamin D abrogates PM-induced inflammatory responses of human bronchial epithelial cells (HBECs) through enhancement of antioxidant pathways. METHODS: Transcriptome analysis, PCR and ELISA were undertaken to delineate markers of inflammation and oxidative stress; with comparison of expression in primary HBECs fr…

Resveratrol relieves particulate matter (mean diameter < 2.5 μm)-induced oxidative injury of lung cells through attenuation of autophagy deregulation.

Oxidative stress and inflammation are critically implicated in ambient fine particulate matter (mean diameter < 2.5 μm; PM2.5 )-induced lung injury. Autophagy, playing a crucial role in various physiopathological conditions, modulates cellular homeostasis and stress adaptation. Resveratrol is a phytoalexin that exerts potent antioxidant effects on cardiopulmonary diseases. To date, the mechanisms by which resveratrol protects against PM2.5 remain to be elucidated. In the present study, we investigated the effect of resveratrol on PM2.5 -induced oxidative injury. The potential role of nuclear f…

Exposure to particulate matter 2.5 (PM2.5) induced macrophage-dependent inflammation, characterized by increased Th1/Th17 cytokine secretion and cytotoxicity.

Particulate matter PM2.5 is a class of airborne particles and droplets with sustained high levels in many developing countries. Epidemiological studies have shown the association between sustained high level of PM2.5 and the risk of many diseases in the respiratory system, including lung cancer. However, the precise mechanisms through which PM2.5 induces respiratory diseases are still unclear. In this study, we demonstrated that CD4+ and CD8+ T cells following PM2.5 treatment demonstrated significantly elevated mRNA and protein levels of interferon (IFN)-γ, interleukin (IL)-10, IL-17, and IL-2…

Exposure to fine particulate matter during pregnancy and risk of term low birth weight in Jinan, China, 2014-2016.

BACKGROUND: Existing studies exploring the association between low birth weight (LBW) and maternal fine particulate matter (aerodynamic diameter<2.5μm, PM2.5) exposure have presented equivocal results, and one of the possible reasons for this finding might be due to relatively low maternal exposures. In addition, relatively narrow maternal exposure windows to PM2.5 have not been well established for LBW. METHODS: We employed a nested matched case-control design among 43,855 term births in a large maternity and child care hospital in Jinan, China. A total of 369 cases were identified, and four…

Pulmonary toxicity following acute coexposures to diesel particulate matter and α-quartz crystalline silica in the Sprague-Dawley rat.

The effects of acute pulmonary coexposures to silica and diesel particulate matter (DPM), which may occur in various mining operations, were investigated in vivo. Rats were exposed by intratracheal instillation (IT) to silica (50 or 233 µg), DPM (7.89 or 50 µg) or silica and DPM combined in phosphate-buffered saline (PBS) or to PBS alone (control). At one day, one week, one month, two months and three months postexposure bronchoalveolar lavage and histopathology were performed to assess lung injury, inflammation and immune response. While higher doses of silica caused inflammation and injury a…

Indoor particulate matter exposure is associated with increased black carbon content in airway macrophages of former smokers with COPD.

INTRODUCTION: Exposure to fine particulate matter (PM2.5) is associated with worse morbidity in individuals with COPD. Inhaled PM is phagocytosed by airway macrophages (AM), and black carbon measured in AM may serve as a biomarker of air pollution exposure. As there is little data on how indoor PM exposure may influence AM black carbon content in those with respiratory disease, we investigated the association of indoor PM2.5 concentration to AM black carbon content in adults with COPD. METHODS: Former smokers (>10 pack-years smoking history, quit date >1 year prior to enrollment) older than 4…

The effect of aged litter materials on polyatomic ion concentrations in fractionated suspended particulate matter from a broiler house.

UNLABELLED: Inorganic emissions from livestock production and subsequent deposition of these ions can be a major source of pollution, causing nitrogen enrichment, eutrophication, acidification of soils and surface waters, and aerosol formation. In the poultry house, ammonia and hydrogen sulfide emissions can also adversely affect the health, performance, and welfare of both animals and human operators. The persistence and long life expectancy of ammonia, odors and toxic pollutants from poultry houses may be due to the ability of suspended particulate matter (SPM) to serve as carriers for odoro…

The Exposure Uncertainty Analysis: The Association between Birth Weight and Trimester Specific Exposure to Particulate Matter (PM2.5 vs. PM10).

Often spatiotemporal resolution/scale of environmental and health data do not align. Therefore, researchers compute exposure by interpolation or by aggregating data to coarse spatiotemporal scales. The latter is often preferred because of sparse geographic coverage of environmental monitoring, as interpolation method cannot reliably compute exposure using the small sample of sparse data points. This paper presents a methodology of diagnosing the levels of uncertainty in exposure at a given distance and time interval, and examines the effects of particulate matter (PM) ≤2.5 µm and ≤10 µm in dia…

Land use regression modeling of ultrafine particles, ozone, nitrogen oxides and markers of particulate matter pollution in Augsburg, Germany.

Important health relevance has been suggested for ultrafine particles (UFP) and ozone, but studies on long-term effects are scarce, mainly due to the lack of appropriate spatial exposure models. We designed a measurement campaign to develop land use regression (LUR) models to predict the spatial variability focusing on particle number concentration (PNC) as indicator for UFP, ozone and several other air pollutants in the Augsburg region, Southern Germany. Three bi-weekly measurements of PNC, ozone, particulate matter (PM10, PM2.5), soot (PM2.5abs) and nitrogen oxides (NOx, NO2) were performed …

Short-term exposure to noise, fine particulate matter and nitrogen oxides on ambulatory blood pressure: A repeated-measure study.

Exposure to road traffic noise, fine particulate matter (aerodynamic diameter ≤2.5 μm; PM2.5) and nitrogen oxides (NOx) has been associated with transient changes in blood pressure, but whether an interaction exists remains unclear. This panel study investigated whether noise, PM2.5 and NOx exposure were independently associated with changes in 24-h ambulatory blood pressure. We recruited 33 males and 33 females aged 18-32 years as study subjects. Personal noise exposure and ambulatory blood pressure were monitored simultaneously in 2007. During the data collection periods, 24-h data on PM2.5 …
