Influence of tick and mammalian physiological temperatures on Borrelia burgdorferi biofilms.

The spirochaete bacterium Borrelia burgdorferisensu lato is the aetiologic agent of Lyme disease. Borrelia is transmitted to mammals through tick bite and is adapted to survive at tick and mammalian physiological temperatures. We have previously shown that B. burgdorferi can exist in different morphological forms, including the antibiotic-resistant biofilm form, in vitro and in vivo. B. burgdorferi forms aggregates in ticks as well as in humans, indicating potential of biofilm formation at both 23 and 37 °C. However, the role of various environmental factors that influence Borrelia biofilm for

Neural space integrity of the lower cervical spine: effect of normal range of motion.

STUDY DESIGN: An experimental investigation of intervertebral foramen and spinal canal neural space integrity was performed throughout physiologic range of motion of the lower cervical spine in intact human cadaver specimens. OBJECTIVE: To investigate cervical positions that might place the neural tissues of the spine in heightened risk of injury. To meet this objective the following hypotheses were tested: 1) spinal canal integrity varies with specific normal range of motion positions of the lower cervical spine, and 2) intervertebral foramen integrity is dependent on and unique for differen

Environmental Modulation of Physiological Parameters in the Paediatric Intensive Care Unit After Extracorporeal Cardiac Surgery.

BACKGROUND: The paediatric intensive care unit exposes critically ill children to non-physiological environmental stimuli, such as noise, artificial light and thermal instability, which may interfere with physiological regulation and circadian organization. AIM: To examine how environmental conditions in the paediatric intensive care unit (PICU), including noise, lighting and ambient temperature, affect cardiovascular, respiratory and thermoregulatory physiology in paediatric patients admitted after extracorporeal cardiac surgery. STUDY DESIGN: This prospective cohort study used continuous e

Blood buffers: The viewpoint of a biochemist.

Mammalian blood is a very complex system whose multiple physiological roles require that its pH is maintained constant, in spite of the necessity of carrying over 15 moles of CO2 a day from the tissues to the lungs. The blood pH is maintained constant by several buffers, whose interplay is complex. The study of blood buffers is over a century old and has crossed major reinterpretations of the nature of acids and bases, from Arrhenius to Bronsted and Lowry, as well as an enormous evolution of our knowledge of protein structure, proteins being the most relevant among blood buffers. As a conseque

The inhibition of mitochondrial cytochrome oxidase by the gases carbon monoxide, nitric oxide, hydrogen cyanide and hydrogen sulfide: chemical mechanism and physiological significance.

The four gases, nitric oxide (NO), carbon monoxide (CO), hydrogen sulfide (H(2)S) and hydrogen cyanide (HCN) all readily inhibit oxygen consumption by mitochondrial cytochrome oxidase. This inhibition is responsible for much of their toxicity when they are applied externally to the body. However, recently these gases have all been implicated, to greater or lesser extents, in normal cellular signalling events. In this review we analyse the chemistry of this inhibition, comparing and contrasting mechanism and discussing physiological consequences. The inhibition by NO and CO is dependent on oxyg

Congenital deficiency of histidine-rich glycoprotein: failure to identify abnormalities in routine laboratory assays of hemostatic function, immunologic function, and trace elements.

Histidine-rich glycoprotein (HRGP) has many biologic activities, but its physiologic function is still unclear. To show the physiologic function of HRGP, we studied five patients with congenital HRGP deficiency. Hemostatic screening tests, activities of natural anticoagulants and fibrinolytic proteins, markers of thrombin and plasmin generation, plasma levels of platelet-specific proteins, thrombin times with various concentrations of bovine thrombin, prolongation of thrombin time after addition of heparin or demartan sulfate, and contact activation of blood coagulation were normal or nearly n

Deconstructing cardiovascular and coagulation-related traits links dietary ecology to multi-functional snake venom specificity.

Animal venoms vary greatly in compositional complexity, where complex venoms are hypothesized to be maintained by greater dietary breadth. Beyond explaining venom composition, the dietary breadth hypothesis predicts that these more complex venoms should show greater functional breadth in terms of overall toxicity and by disrupting multiple prey physiological processes. We evaluate these predictions with six distinct physiological assays of cardiac and blood clotting functions and compared the effects of venoms from snake species with a range of taxonomic dietary diversity levels. We compared t

Comparison of Physiological Stress Indices in Anesthetized and Manually Restrained Leopard Sharks, Triakis semifasciata.

BACKGROUND: Leopard sharks (Triakis semifasciata) are abundant, coastal, eastern Pacific, mesopredatory sharks and are frequently managed in aquariums and zoos. Medical examinations are a routine part of good husbandry practices, but the handling protocols vary greatly between facilities. In this study, we compared the physiological stress responses of T. semifasciata associated with manual restraint and sedated handling under parallel holding and handling procedures in a 10 min interval. METHODS: Blood was collected and analyzed for the hematocrit (Hct), lactate, glucose, beta-hydroxybutyrat

Evaluation of long-term dermal exposure to soil contaminated with spent engine oil in male Wistar rats: An experimental approach.

Continuous occupational exposure to spent engine oil (SEO) poses physiological risks to humans, especially to automobile mechanics. This study investigated the physiological effects of SEO-contaminated soil in a male Wistar rat model. Animals were dermally exposed to soil contaminated with SEO for 120 consecutive days and compared with rats exposed to uncontaminated soil (negative control). Heavy metal (lead (Pb), nickel (Ni), zinc, and cadmium (Cd)) accumulations, hematology, biochemical (aspartate aminotransferase (AST), alanine aminotransferase, alkaline phosphatase (ALP), urea, and creatin

Etanercept provides a more physiological approach in the treatment of psoriasis.

Psoriasis is a common chronic inflammatory disease affecting the skin and joints. Moderate to severe psoriasis is traditionally treated with systemic treatments, which can be effective but are often associated with relevant adverse effects, even when administered intermittently or rotationally. Biologic therapies may provide high and consistent efficacy over time, long-term safety, and simple administration schedules compared with nonbiologic therapies, and can be used in patients intolerant and/or resistant to these therapies. TNF-antagonists have a definite advantage over other biologic agen

A single-center before-and-after study of physiology-first algorithmic surveillance versus standard observation for arteriovenous fistula maturation.

OBJECTIVE: To evaluate whether a structured, physiology-first surveillance protocol improves arteriovenous fistula clinical maturation rates and reduces time to cannulation readiness compared with standard postoperative observation. METHODS: Two consecutive cohorts of surgically created arteriovenous fistulas by a single surgeon at a community-based center were compared: a standard observation cohort (January-May 2025; 64 created, 58 analyzable) and a physiology-first cohort (June 2025-March 2026; 52 created, all analyzable) managed with a structured protocol featuring standardized 2.7-mm ana

Inhibition of 2-Oxoglutarate Dehydrogenase as a Chemical Model of Acute Hypobaric Hypoxia.

Both hypoxia and inhibition of 2-oxoglutarate dehydrogenase complex (OGDHC) are known to change cellular amino acid pools, but the quantitative comparison of the metabolic and physiological outcomes has not been done. We hypothesize that OGDHC inhibition models metabolic changes caused by hypoxia, as both perturb the respiratory chain function, limiting either the NADH (OGDHC inhibition) or oxygen (hypoxia) supply. In the current study, we quantify the changes in the amino acid metabolism after OGDHC inhibition in the highly sensitive to hypoxia cerebellum and compare them to the earlier chara

Larger-Caliber Alternative Devices for Decompression of Tension Hemopneumothorax in the Setting of Hemorrhagic Shock.

BACKGROUND: The 14-gauge (14G) angiocatheter (AC) has an unacceptably high failure rate in treatment of tension pneumothorax (tPTX). Little is known regarding the interplay among hemorrhage, hemothorax (HTX), and tPTX. We hypothesized that increased hemorrhage predisposes tension physiology and that needle decompression fails more often with increased HTX. METHODS: This is a planned secondary analysis of data from our recent comparison of 14G AC with 10-gauge (10G) AC, modified 14G Veress needle, and 3mm laparoscopic trocar conducted in a positive pressure ventilation tension hemopneumothorax

Dynamic vertebral artery elongation during frontal and side impacts.

BACKGROUND CONTEXT: Elongation-induced vertebral artery (VA) injury has been hypothesized to occur during nonphysiological coupled head motions during automobile impacts. Although previous work has investigated VA elongation during head-turned and head-forward rear impacts, no studies have performed similar investigations for frontal or side impacts. PURPOSE: The present study quantified dynamic VA elongations during simulated frontal and side automotive collisions, and compared these data with corresponding physiological limits. STUDY DESIGN/SETTING: In vitro biomechanical study of dynamic

Coupling of cerebral blood flow and oxygen consumption during physiological activation and deactivation measured with fMRI.

The physiological basis of the blood oxygenation level dependent (BOLD) signal and its dependence on baseline cerebral blood flow (CBF) were investigated by comparing responses to a visual stimulus after physiological changes of the baseline. Eight human subjects were imaged with 3 and 4 T MRI scanners, and both BOLD signal and CBF were simultaneously measured. Subjects viewed a flickering radial checkerboard in a block design experiment, alternating between eyes open or closed during the off periods. Compared to a baseline state with eyes open in a darkened room, substantial deactivation (ave

Beyond CABG vs. PCI: Contemporary and Future Coronary Revascularisation from Historical Evolution to Artificial Intelligence, Robotics, and Hybrid Strategies.

Coronary artery bypass grafting (CABG) and percutaneous coronary intervention (PCI) are the two dominant revascularisation strategies for obstructive coronary artery disease, yet their relative roles continue to shift because they address coronary pathophysiology differently with ever-evolving techniques. PCI has advanced through iterative improvements, including balloon angioplasty, bare-metal stents, and drug-eluting stents, with contemporary outcomes increasingly driven by procedural optimisation using intracoronary imaging and physiology-guided lesion selection rather than device category

Physiological-model-based neural network for modeling the metabolic-heart rate relationship during physical activities.

BACKGROUND AND OBJECTIVE: Heart failure (HF) poses a significant global health challenge, with early detection offering opportunities for improved outcomes. Abnormalities in heart rate (HR), particularly during daily activities, may serve as early indicators of HF risk. However, existing HR monitoring tools for HF detection are limited by their reliability on population-based averages. The estimation of individualized HR serves as a dynamic digital twin, enabling precise tracking of cardiac health biomarkers. METHODS: This study introduces a novel physiological-model-based neural network (PMB

Comprehensive S-acylation profiling of the porcine epididymis and exosomes reveals a role in cargo sorting and long-distance trafficking.

The epididymis orchestrates sperm maturation through microenvironmental regulation and epididymosome-mediated cargo delivery. Despite emerging evidence implicating protein S-acylation in vesicular trafficking, its compartment-specific dynamics and functional implications in epididymal physiology remain poorly characterized. Here, we employed acyl-biotin exchange-based 4D proteomics to decode the S-acylation proteomic of porcine caput/cauda epididymidis and their exosomes. Comparative analysis identified 2780 and 2084 S-acylated proteins in caput and cauda tissues, respectively, with 317 upregu

Elevated plasma glucose links intrafollicular bile acids to altered embryological outcomes in IVF patients.

BACKGROUND: Human reproduction is intricately linked to systemic metabolic regulation, with disturbances in glucose metabolism adversely affecting fertility outcomes. Beyond their classical role in lipid digestion, bile acids (BAs), cholesterol-derived catabolites, have emerged as bioactive signaling molecules influencing glucose homeostasis and reproductive physiology. Notably, enhanced glycemic control observed after bariatric surgery, BA administration, or BA sequestrant therapy underscores the potential of BAs as regulators of glucose metabolism and as promising biomarkers or therapeutic t

STIM1 Reduction Prevents Tubular Aggregate Formation and Compromises Muscle Performance in Ageing Mice.

BACKGROUND: Ageing is an irreversible process involving the gradual decline of cellular functions in all tissues. In male mice, age-related loss of muscle force is accompanied by the formation of tubular aggregates, which are honeycomb-like structures composed of membrane tubules, proteins and Ca2+ deposits. Tubular aggregates are also found in tubular aggregate myopathy (TAM) and Stormorken syndrome (STRMK), two clinically overlapping human disorders affecting skeletal muscle, bones, skin, spleen and platelets. TAM/STRMK is caused by gain-of-function mutations in the ubiquitously expressed Ca
