BACKGROUND: Maize (Zea mays L.) is a major cereal crop, with the United States accounting for over 40% of the worldwide production. Corn leaf aphid [CLA; Rhopalosiphum maidis (Fitch)] is an economically important pest of maize and several other monocot crops. In addition to feeding damage, CLA acts as a vector for viruses that cause devastating diseases in maize. We have shown previously that the maize inbred line Mp708, which was developed by classical plant breeding, provides heightened resistance to CLA. However, the transcriptomic variation conferring CLA resistance to Mp708 has not been i…

Common buckwheat (Fagopyrum esculentum Moench) is cultivated across diverse environments in which photoperiod and light quality influence plant development and productivity. This study evaluated the effects of short-day (10 h light/14 h dark) and long-day (16 h light/8 h dark) conditions applied during the first 25 days of growth on flowering, photosynthetic performance, and yield formation in the cultivar 'Panda'. Plants were grown in phytotron chambers under blue- and red-enriched LED illumination and contrasting photoperiods, while plants grown under greenhouse daylight served as a control.

Gain-of-function screens enable the discovery of gene functions and candidate targets for molecular breeding. Here we developed a protoplast-based dCas9-TV-mediated CRISPR activation screen in rice and identified OsTV1, an epidermal papillae-specific receptor kinase, as a positive regulator of fungal resistance. OsTV1 overexpression enhanced silicon deposition, leaf rigidity and basal defence gene expression, conferring broad-spectrum fungal resistance without compromising growth. This study establishes a high-throughput gain-of-function screen adaptable for investigating diverse cellular proc

While enhancing crop yield remains a critical breeding objective, such efforts often compromise nutritional quality and stress tolerance. The growing global population and increasing environmental stresses make combining stable crop yields with improved nutritional quality a vital objective for sustainable food security. Thiamine pyrophosphate (TPP), the active form of vitamin B1, regulates core metabolism to support both human health and plant stress tolerances. Here, we show that editing the rice TPP riboswitch elevates multiple micronutrients, simultaneously increasing grain yield, cold tol

The soil-borne Gram-negative beta-proteobacterium Ralstonia solanacearum species complex (RSSC) causes bacterial wilt, a devastating plant disease that threatens crop production and food security worldwide. In this review, we first summarize current knowledge of RSSC pathogenicity and virulence, focusing on the factors that determine its ability to infect and cause disease, including advances in resistance breeding and the genetic basis of host resistance to bacterial wilt. Next, we highlight the questions provided from the previous studies and describe our recent studies, which revealed that

This study identified a major QTL governing leaf shape, pinpointed a highly associated SNP, and developed a dCAPSmarker, providing a useful molecular tool to facilitate marker-assisted breeding for leaf shape in soybean. Optimizing plant architecture to enhance canopy light distribution is a central objective in modern crop breeding. Leaf shape is a critical determinant of canopy structure and yield potential, yet the natural genetic variation underlying this trait in soybean remains underexploited for marker-assisted improvement. Here, we performed a genome-wide association study (GWAS) of le

Advanced plant phenotyping technologies are vital for trait improvement and accelerating intelligent breeding. Due to the species diversity of plants, existing methods heavily rely on large-scale high-precision manually annotated data. For self-occluded objects at the grain level, unsupervised methods often prove ineffective. This study proposes IPENS, an interactive unsupervised multi-target point cloud extraction method. It utilizes radiance field information to lift 2D masks, segmented by SAM2 (Segment Anything Model 2), into 3D space for target point cloud extraction. A multi-target collab

Australia, the flattest, driest, and geologically oldest vegetated continent, has a uniquely high proportion of nutrient-poor soils. We develop a "Nutrient-Poverty/Intense-Fire Theory," which postulates that most anomalous features of organisms and ecosystems of Australia are the evolutionary consequences of adaptations to nutrient poverty, compounded by intense fire that tends to occur as a result of nutrient poverty. The fundamental tenet of the theory is that plants growing in environments with plentiful light and periodic adequate moisture, but on soils poor in phosphorus, zinc, and other

Brassinosteroid signaling kinases (BSKs) act as core signal transducers downstream of Brassinosteroid (BR) perception and integrate plant growth regulation with broad-spectrum biotic and abiotic stress tolerance. Despite well-established functional characterizations of BSK gene families in Arabidopsis thaliana and rice, comprehensive genome-wide profiling and salt response analysis of BSK homologs remain lacking in wheat. In this study, we systematically identified 18 TaBSK family members. Phylogenetic analysis separated wheat TaBSKs into three distinct evolutionary subgroups. The 18 TaBSK loc

BACKGROUND: Meloidogyne incognita is a major soil-borne pathogen in tobacco production, which severely restricts the sustainable development of the Chinese tobacco industry. At present, problems including the diminished disease resistance of mainstream tobacco cultivars and the scarcity of resistant germplasm resources have become increasingly pronounced, and thus the breeding of new tobacco varieties with high resistance to M. incognita has become an urgent priority. Previous studies have revealed that the tobacco germplasm SA1214 originating from South Africa displays extremely high resistan

BACKGROUND: This study investigated the effect of sulfur nutrition, basalt rock aggregate (BA) application, with a carbon capture function, and speed breeding under light-emitting diode (LED) light, on the nutritional profile of potatoes and acrylamide formation in crisp production.
RESULTS: Taurus potatoes grown with sulfur showed reduced glucose, sucrose, and total amino acids, and increased asparagine. No difference in acrylamide content was observed in crisps from Taurus and Lady Claire cultivars, with either sulfur or BA application. Speed breeding under LED light reduced plant height in

Doubled haploid (DH) technology has become integral to maize breeding programs to expedite inbred line development and increase the efficiency of breeding operations. Unlike many other plant species that use in vitro methods, DH production in maize uses a relatively simple and efficient in vivo haploid induction method. However, it takes two complete crop cycles for DH line generation, one for haploid induction and the other one for chromosome doubling and seed production. Rescuing in vivo induced haploid embryos has the potential to reduce the time for DH line development and improve the effi

INTRODUCTION: Haemagogus are mosquitoes with diurnal habits that live preferentially in forest areas. In Brazil, they are considered the primary vectors of wild yellow fever.
METHODS: The ecological relationships between Haemagogus spegazzinii, the environment, and some of its activities in the semiarid region of Rio Grande do Norte were analyzed by collecting eggs with ovitraps, actively searching in tree holes, capturing adults in Shannon traps, and conducting an investigation for viral infections.
RESULTS: A total of 2420 eggs, 271 immature specimens (larvae and pupae), and 206 adults wer

The biological complexity of plants arises from highly coordinated cellular activities. We propose that a "cellular spatiotemporal dogma" governs the zygote's programmed development into a complete plant and its adaptation to various environmental stresses, representing the set of principles describing how gene expression, cell identity, and function are coordinated across physical spatial contexts and temporal developmental progressions. Historically, our understanding of this dogma remains limited because traditional bulk tissue sequencing provides only a homogenized average of gene expressi

Abiotic stresses-drought, salinity, extreme temperature, flooding, and heavy-metal toxicity-constrain rice (Oryza sativa L.) yield worldwide, and the cellular programmes underlying them are unevenly distributed across cell types that bulk-tissue assays average together. This review examines, from a methodological standpoint, what single-cell and spatial omics technologies can and cannot establish about rice abiotic stress biology. We first define the modality space: single-cell omics measures RNA, chromatin accessibility, DNA methylation, protein, or metabolite features at the resolution of in

Recent studies highlighted the influence of epigenetic marks in the variability of many complex traits, both in plants and animals. These studied focused only on specific sites of the genome having differentially methylated profiles among individuals and/or tissues. In contrast, we recently used the methylation rate of the entire genome as a unique measure considered as a novel quantitative phenotype in sheep. This phenotype named global DNA methylation rate (GDMR), measured by luminometric assay, integrates the methylation level of each CpG dinucleotide within the 6 million of CCGG sites alon

BACKGROUND: Understanding the ecology and behaviour of disease vectors, including the olfactory cues used to orient and select hosts and egg-laying sites, are essential for the development of novel, insecticide-free control tools. Selected graminoid plants have been shown to release volatile chemicals attracting malaria vectors; however, whether the attraction is selective to individual plants or more general across genera and families is still unclear.
METHODS: To contribute to the current evidence, we implemented bioassays in two-port airflow olfactometers and in large field cages with four

The Prunus mume species has undergone a long process of domestication and introduction in China, resulting in marked differences in freezing tolerance among populations across a latitudinal gradient of approximately 1,000 km. However, the introduction and domestication process is time-consuming, constraining the efficiency of species introduction. In this study, P. mume samples with the same genetic background spanning over 1,000 km from north to south were selected as experimental materials. The molecular mechanisms underlying cold-acclimation-mediated freeze tolerance in P. mume were investi

Starch is one of the primary products of photosynthesis, which accumulates in chloroplasts as discrete starch granules during the day and degrades throughout the night. It was found that Nicotiana tabacum L. showed a response to hypothermia by hyperaccumulation of starch in its leaves, whereas in Secale cereale L. cold acclimation was accompanied by a decrease of leaf starch. Transmission electron microscopy showed that the size of starch granules in tobacco chloroplasts during cold acclimation reached almost half of the chloroplast area, while in rye chloroplasts starch granules completely di

Variation in anthocyanin biosynthesis in pear fruit provides genetic germplasm resources for breeding, while dwarfing is an important agronomic trait, which is beneficial to reduce the management costs and allow for the implementation of high-density cultivation. Here, we combined bulked segregant analysis (BSA), quantitative trait loci (QTL), and structural variation (SV) analysis to identify a 14-bp deletion which caused a frame shift mutation and resulted in the premature translation termination of a B-box (BBX) family of zinc transcription factor, PyBBX24, and its allelic variation termed
