PaDef defensin from avocado (Persea americana var. drymifolia) is cytotoxic to K562 chronic myeloid leukemia cells through extrinsic apoptosis.

Plant defensins, a group of antimicrobial peptides, show selective cytotoxicity toward cancer cells. However, their mechanisms of action remain poorly understood. Here, we evaluated the cytotoxicity of PaDef defensin from avocado (Persea americana var. drymifolia) on K562 chronic myeloid leukemia cells and analyzed the pathway involved in the induction of cell death. The defensin PaDef was not cytotoxic against human PBMCs; however, it was cytotoxic for K562 cell line (IC50 = 97.3 μg/ml) activating apoptosis at 12 h. PaDef did not affect the mitochondrial membrane potential (ΔΨm), neither the

Analysis of Plant L-Cysteine Desulfhydrase (LCD) Isozymes by Non-denaturing Polyacrylamide Gel Electrophoresis.

Hydrogen sulfide (H2S) is a signaling molecule that achieves different regulatory functions in animal and plant cells. The cytosolic enzyme L-cysteine desulfhydrase (LCD; EC 4.4.1.28) catalyzes the conversion of cysteine (L-Cys) to pyruvate and ammonium with the concomitant generation of H2S, this enzyme being considered one of the main sources of H2S in higher plants. Using non-denaturing polyacrylamide gel electrophoresis (PAGE) in combination with a specific assay for LCD activity, the present protocol allows identifying diverse LCD isozymes present in different organs (roots, shoots, leave…

Role of salicylic acid in resistance to cadmium stress in plants.

We review and introduce the importance of salicylic acid in plants under cadmium stress, and provide insights into potential regulatory mechanisms for alleviating cadmium toxicity. Cadmium (Cd) is a widespread and potentially toxic environmental pollutant, originating mainly from rapid industrial processes, the application of fertilizers, manures and sewage sludge, and urban activities. It is easily taken up by plants, resulting in obvious toxicity symptoms, including growth retardation, leaf chlorosis, leaf and root necrosis, altered structures and ultrastructures, inhibition of photosynthesi

Multiple iron reduction by methoxylated phenolic lignin structures and the generation of reactive oxygen species by lignocellulose surfaces.

Chelator-mediated Fenton chemistry is capable of reducing non-stochiometric amounts of iron via hydroquinone oxidation. These types of reactions have previously been demonstrated to be promoted by some lignocellulose degrading fungi in generating hydroxyl radicals to permit lignified plant cell wall deconstruction. Here we demonstrate that lignocellulose surfaces, when exposed by chemical treatment or fragmentation, can promote a similar multi-oxidative mechanism in the presence of iron. Iron reduction by lignin surfaces permits the generation of hydroxyl radicals in the cell wall to help expl

Uptake and distribution of Zn, Cu, Cd, and Pb in an aquatic plant Potamogeton natans.

A better understanding of metal uptake and translocation by aquatic plants can be used to enhance the performance of constructed wetland systems for stormwater treatment. Specifically, this study examines whether the uptake of Zn, Cu, Cd, and Pb by Potamogeton natans is via the leaves, stems, or roots, and whether there is translocation from organs of uptake to other plant parts. Competition between the metals at uptake and at the level of the cell wall-bound part of the metals accumulated in stem and leaf tissue was also examined. The results show that Zn, Cu, Cd, and Pb were taken up by the

The Regulatory Army of Plant Defense: Transcription Factors in the War for Plant Immunity.

Plant diseases impose major constraints on global crop productivity and pose a major threat to food security. Here, we review transcription factors (TFs) as central orchestrators of plant defense, consolidating recent advances in how these regulators connect pathogen perception to immune signaling, transcriptional reprogramming, and durable defense responses. Initially, we combined a literature-based synthesis with a natural language processing (NLP) analysis of 1647 PubMed abstracts published between 2021 and 2026 to map dominant and underexplored TF families associated with plant immunity. Rheumatoid Arthritis and Secondary Plant Metabolites: An Analysis.

Chronic rheumatoid arthritis (RA) is a systemic inflammatory disease characterized by persistent joint inflammation, progressive joint destruction, and chronic pain. Although modern therapies like disease-modifying antirheumatic medications (DMARDs) can alleviate symptoms, they may also produce side effects. Because of their anti-inflammatory, antioxidant, and immunomodulatory qualities, plant secondary metabolites such as flavonoids, terpenoids, alkaloids, and phenolic acids have attracted attention as prospective RA treatment agents. This review discusses the pathogenesis of RA and provides

Light signaling-driven plant-microbiome interactions.

As sessile organisms, plants continuously perceive and adapt to dynamic light environments and complex microbial communities. Light serves not only as the energy source for photosynthesis, but also a key signal that regulates plant growth, development, and plant-microbe interactions. Through photoreceptor-mediated perception and downstream signaling networks, light modulates plant immunity and orchestrates beneficial microbial associations. This review summarizes how light signaling regulates plant-microbe interactions through three interconnected layers. First, photoreceptors and downstream f

Are plant factors a missing link in the evolution of endemic Burkitt's lymphoma?

Burkitt's lymphoma, an Epstein-Barr virus (EBV)-associated non-Hodgkin's malignant lymphoma is endemic in an area of Africa known as the Lymphoma Belt. This zone is demarcated by climatic requirements of temperature and rainfall. EBV-activating plant factors are among several co-factors which have been proposed for the development of epidemic Burkitt's Lymphoma (eBL). The distribution of Euphorbia tirucalli, a plant which possesses EBV-activating substances and can induce the characteristic 8:14 translocation of eBL in EBV-infected lymphoblastic cell lines in vitro, conforms closely to the cli

The use of soybean agglutinin (SBA) for bone marrow (BM) purging and hematopoietic progenitor cell enrichment in clinical bone-marrow transplantation.

Soybean agglutin (SBA) is a plant lectin that has been used to fractionate bone marrow cells. It binds bone marrow mononuclear cells, including mature myeloid, erythroid, and lymphoid cells, but has very low binding affinity and no toxic effect to the human hematopoietic cells. In this article we describe the possibilities of enriching bone-marrow-derived CD34+ hematopoietic progenitor cells using SBA. As the method is simple and elegant. SBA is of vast importance to the field of clinical bone marrow transplantation.

Exploring the significant contribution of silicon in regulation of cellular redox homeostasis for conferring stress tolerance in plants.

Silicon (Si), a bioactive metalloid is beneficial for plant growth and development. It also plays a key role in the amelioration of different abiotic and biotic stresses. Extensive studies have elucidated the morpho-physiological, biochemical and molecular background of Si-mediated stress tolerance in plants. However, the mechanism acquired by Si to enhance stress tolerance in plants is still unheeded. Present review summarized the prospective mechanisms of Si in acquisition of stress tolerance with emphasis on its interactions with secondary messengers. Silicon usually modulates the different
