Effect of bacteriocin produced by Lactococcus sp. HY 449 on skin-inflammatory bacteria.

This study was carried out to evaluate the effect of bacteriocin produced by Lactococcus sp.HY 449 against skin-inflammatory bacteria such as Staphylococcus epidermidis ATCC 12228, Staphylococcus aureus ATCC 65389, Streptococcus pyogenes ATCC 21059, and Propionibacterium acnes ATCC 6919. The spot-on-the-lawn method was used to determine the antimicrobial activity of bacteriocin against indicator strains on the human skin. The bacteriocin produced by Lactococcus sp. HY 449 inhibited the growth of S. epidermidis ATCC 12228, S. aureus ATCC 65389, Strep. pyogenes ATCC 21059, and P. acnes ATCC 6919…

MR imaging in an experimental model of brain tumor immunotherapy.

A murine model of implanted CNS neoplasia was used to study a new form of brain tumor immunotherapy with intralesional Corynebacterium parvum (C. parvum). Assessment of treatment protocols has been limited by the inability to assess, noninvasively, tumor burden and/or the inflammatory reaction induced in the murine brain by treatment with C. parvum. This study demonstrates that contrast-enhanced MR imaging can monitor in vivo tumor burden and the immune response to intracerebral C. parvum. KHT murine sarcoma was stereotaxically implanted into the right frontal lobe of C3H/HeN mice at doses of

Mycoplasmas and 'diphtheroids' in rheumatoid arthritis.

The failure to isolate conventional mycoplasmas from 88 synovial membranes and 119 synovial fluids from patients with proven rheumatoid arthritis using a variety of culture media and techniques agrees with the results of recent workers and suggests that these organisms are unlikely to play a role in the aetiology of the disease. In contrast, 'diphtheroid' organisms were isolated from 27 to 30% of rheumatoid specimens, but not from cultures of non-rheumatoid material. A significantly higher incidence of raised agglutination titres to C. acnes and to a 'diptheroid' isolated from a patient with r

Isolation and characterization of two novel oral bacteriophages with anti-biofilm activity against Cutibacterium acnes.

Bacteriophage therapy offers a promising solution to combat antibiotic-resistant infections, yet its potential against biofilm-associated pathogens in oral diseases remains underexplored. This study investigates the opportunistic bacterium Cutibacterium acnes, an overlooked contributor to dental implant and prosthetic joint infections. Biofilms formed by C. acnes are highly resilient and resistant to antibiotics, complicating treatment. Two novel lytic bacteriophages, Ristretto and Corretto, targeting C. acnes, were isolated from human saliva, with morphological analysis confirming their class

Myrtus communis and Celastrol enriched plant cell culture extracts control together the pivotal role of Cutibacterium acnes and inflammatory pathways in acne.

INTRODUCTION: Acne is a multifactorial inflammatory disease of the pilosebaceous unit in which Cutibacterium acnes is one of the main triggers. A strong predominance of C. acnes phylotype IA1 is present in acne skin with higher biofilm organization and virulence, promoting local immuno-inflammation, especially the Th17 pathway. OBJECTIVES: We evaluated the single and combined pharmacological properties of the plant extracts, Myrtus communis (Myrtacine®) and Celastrol enriched plant cell culture (CEE) extracts on the C. acnes/Th17 pathway. METHODS: The effect of Myrtacine® on the virulence of

New insights into the role of Cutibacterium acnes-derived extracellular vesicles in inflammatory skin disorders.

Cutibacterium acnes (C. acnes) is one of the most prevalent bacteria that forms the human skin microbiota. Specific phylotypes of C. acnes have been associated with the development of acne vulgaris, while other phylotypes have been linked to healthy skin. In this scenario, bacterial extracellular vesicles (EVs) play a role in the interkingdom communication role with the human host. The purpose of this study was to examine the impact of EVs generated by various phylotypes of C. acnes on inflammation and sebum production using different in vitro skin cell types. The main findings of this study r

Mode of action studies confirm on-target engagement of lysyl-tRNA synthetase inhibitor and lead to new selection marker for Cryptosporidium.

INTRODUCTION: Cryptosporidiosis is a leading cause of diarrheal-associated morbidity and mortality, predominantly affecting children under 5 years old in low-and-middle-income countries. There is no effective treatment and no vaccine. New therapeutics are emerging from drug discovery efforts. It is critical that mode of action studies are performed alongside drug discovery to ensure the best clinical outcomes. Unfortunately, technology to identify and validate drug targets for Cryptosporidium is severely lacking. METHODS: We used C. parvum lysyl-tRNA synthetase (CpKRS) and DDD01510706 as a ta

Effects of Prednisolone Derivative and Panaxydol: Biosurfactants on Cell Wall Integrity of Acne-Causing Resistant Bacteria.

Acne is one of the most common dermatological skin problem caused due to inflammation of the skin, leading to unfavorable growth of Propionibacterium acnes. It is a slow growing anaerobic, gram-positive bacterium that releases chemotactic factors and leads to the complex pathogenicity of acne. There are several acne treatments/therapies available, but topical therapy is usually the first choice for mild to moderate acne, and as the severity of the acne increases, the treatment modalities fail. There are many acne treatment options available, but topical therapy is best suited for mild - to - m…

Intracerebral abscess due to Cutibacterium acnes after lung transplantation.

Cutibacterium (C) acnes, a Gram-positive bacterium that is part of the commensal flora, is increasingly noticed as an opportunistic pathogen in serious infections in both immunocompromised and immunocompetent patients. The indolent character and often difficult identification because of its slow growth contribute to delayed diagnosis or underdiagnosis. This report highlights a unique case of a lung transplant recipient with a C acnes intracerebral abscess, and we recommend including this organism in such differential diagnosis. A 66-year-old woman, 2 years after bilateral lung transplantation

Systematic review of antibiotic resistance in acne: an increasing topical and oral threat.

Topical and oral antibiotics are routinely used to treat acne. However, antibiotic resistance is increasing, with many countries reporting that more than 50% of Propionibacterium acnes strains are resistant to topical macrolides, making them less effective. We reviewed the current scientific literature to enable proposal of recommendations for antibiotic use in acne treatment. References were identified through PubMed searches for articles published from January, 1954, to March 7, 2015, using four multiword searches. Ideally, benzoyl peroxide in combination with a topical retinoid should be us…

Effects of the removal of the primary tumor and immunotherapy with Corynebacterium parvum on metastatic tumor proliferation.

The effects of surgical intervention by removal of the primary focus, and the effectiveness of an immunomodulator, Corynebacterium parvum (Cp), on the proliferation of metastatic tumor tissue were investigated by following the postoperative changes in the 3H-thymidine labelling rate of metastatic tissue in an experimental model of metastasis in mice. In addition, the delayed type hypersensitivity reaction (DTH) was studied to investigate the immune capacity of the host. The labelling rate of mice that had the primary focus removed remained high with little variation, while that of the mice not

Heterologous immunity between piroplasms and malaria parasites: the simultaneous elimination of Plasmodium vinckei and Babesia microti from the blood of doubly infected mice.

Mice which have recovered from infections with the avirulent piroplasm Babesia microti are also resistant to challenge with the virulent malaria parasite Plasmodium vinckei. In mice infected with P. vinckei before the peak of the B. microti infection the numbers of malaria parasites in the blood increase until that peak and are then eliminated at the same time as the piroplasms. In mice infected with P. vinckei at or after the peak there is no apparent multiplication and the malaria parasites begin to disappear from the blood immediately. The malaria parasites in doubly infected mice show sign

Effect of BCG and C. parvum on in vivo Listeria clearance and tumor growth. Comparative studies in normal and congenitally athymic (nude) mice.

Clinically, it is important to know whether agents used for immunostimulation require the presence of functional thymus-derived (T) cells. Previous studies showed that both BCG and C. parvum induced a macrophage-dependent enhanced in vivo listeria clearance and in vivo inhibition of a fibrosarcoma induced by 20-methylcholanthrene. These two models were re-evaluated in congenitally thymusless (nude or nu/nu) B10LP mice lacking functional T cells. In nu/nu mice, (1) BCG failed to enhance listeria clearance, and when given simultaneously with tumor cells, had no effect on tumor growth; (2) C. par

Sinonasal Squamous Cell Carcinoma Mimicking a Brain Abscess: Report of a Unique Case.

Sinonasal cancers are rare tumors with squamous cell carcinoma (SCC) being one of the more common histological subtypes. These carcinomas typically invade the sinus cavity from which they originate, progressively eroding surrounding bony structures and extending to adjacent anatomical regions. In rare instances, they may breach the posterior or superior walls to invade the anterior cranial fossa (ACF) and frontal lobes. The normal flora of the nasal cavity and paranasal sinuses includes Staphylococcus aureus, Staphylococcus epidermidis, α- and γ-streptococci, Propionibacterium acnes, and aerob…
