Omega-6 Polyunsaturated Fatty Acids and Peripheral Artery Disease: An Individual Participant-Level Analysis of 9 International Prospective Studies.

BACKGROUND: Omega-6 polyunsaturated fatty acids (PUFAs) have previously been associated with several peripheral artery disease (PAD) risk factors. We studied the association between individual omega-6 PUFAs and incident PAD in a multicohort analysis. METHODS: This study was conducted among participants from 9 cohorts (N=137 704; PAD cases=2014) in the FORCE (Fatty Acids and Outcomes Research Consortium) who were free of clinical PAD at baseline. Omega-6 PUFAs were measured at baseline and with a median follow-up of 14 years. Cox proportional hazards regression analyses were conducted in indiv

Glucagon-like peptide-1 receptor agonists and gastric emptying time: a systematic review and meta-analysis of prospective studies.

PURPOSE: Current management of type 2 diabetes mellitus and obesity increasingly includes treatment with glucagon-like peptide-1 receptor agonists (GLP-1 RAs). In this systematic review and meta-analysis, we sought to characterize the effect of GLP-1 RAs on gastric emptying half-time (T½). METHODS: We conducted a systematic review of prospective studies reporting T½ with and without GLP-1 RA treatment. Inclusion criteria were 1) patients aged ≥ 18 yr taking a GLP-1 RA for diabetes mellitus and/or weight loss, 2) gastric emptying assessment reported as emptying T½, and 3) study design that was…

POEM vs laparoscopic Heller myotomy with fundoplication for achalasia: systematic review and meta-analysis of randomized and prospective studies.

BACKGROUND: Achalasia is definitively treated with peroral endoscopic myotomy (POEM) or laparoscopic Heller myotomy with fundoplication (LHM + PF), but reflux after POEM remains a key concern. We compared the safety and GERD outcomes of POEM and LHM + PF using randomized controlled trials and prospective comparative studies in treatment-naïve adults. METHODS: We searched the MEDLINE, Embase, CENTRAL, Web of Science, and Scopus databases on October 18, 2025. Eligible studies directly compared POEM and LHM + PF in adults. The primary outcome was adverse events, and the secondary outcomes were e

Dietary Stilbenes May Mitigate Risk of Lung Cancer: Evidence from Two Prospective Cohort Studies in the UK and Japan.

(1) Background: Polyphenols are believed to prevent the development of various diseases by counteracting inflammation and oxidative stress. However, polyphenols are difficult to quantify in foods and show substantial variability in intake and epidemiological evidence on their relationship with lung cancer remains limited. This study explored the association between dietary polyphenols and lung cancer in two prospective cohorts. (2) Methods: Data from the UK Biobank and the Japan Collaborative Cohort Study for Evaluation of Cancer Risk (JACC) Study were used. The Phenol-Explorer® database was u

Perioperative discontinuation of SGLT2-inhibitors and cardiac complications after noncardiac surgery: secondary analysis of two prospective observational cohort studies.

BACKGROUND: International clinical practice guidelines recommend discontinuing sodium-glucose cotransporter-2 inhibitors (SGLT2i) 3-4 days before surgery to prevent euglycemic diabetic ketoacidosis, chiefly on the basis of case reports/series. Whether SGLT2i discontinuation may increase the risk of postoperative cardiovascular complications is unclear. METHODS: In a secondary analysis of the Basel-PMI (NCT02573532) and PMI-Vital (NCT05866874) prospective cohort studies in major noncardiac surgery, the exposure of interest was continuing, or stopping, SGLT2i in participants receiving chronic S

Diagnostic test accuracies of 4AT items are consistent across the range of baseline cognition: results from two prospective studies.

BACKGROUND: Presenting features of delirium can be difficult to distinguish from co-existing dementia phenomenology. While the 4 'A's Test (4AT) is a well validated delirium screening tool across multiple patient populations and healthcare settings, how its diagnostic performance varies across different levels of baseline cognition is unclear. METHODS: This study harmonises data from two prospective cohorts: the Delirium and Population Health Informatics Cohort (DELPHIC) and Delirium and Cognitive Impact in Dementia (DECIDE). Both cohorts were stratified into tertiles according to baseline co

Gut microbiota and blood biomarkers as correlating factors in patients with postoperative delirium: analysis of three prospective observational studies.

Postoperative delirium is associated with both gut microbiota alterations and Tau phosphorylation; however, how these factors interact and jointly contribute to postoperative delirium remains poorly understood. This prospective observational cohort study screened 491 patients aged ≥65 years undergoing elective laminectomy or hip or knee replacement under general or spinal anesthesia at Massachusetts General Hospital (2016-2020). The study aimed to assess the correlation between plasma Tau protein levels, specific gut microbiota, and the gut microbiota-derived metabolite indole-3-propionic acid

Reproductive and Hormonal Factors and Thyroid Cancer Risk: Pooled Analysis of Prospective Cohort Studies in the Asia Cohort Consortium.

Given the female predominance of thyroid cancer, particularly in the reproductive age range, female sex hormones have been proposed as an etiology; however, previous epidemiological studies have shown conflicting results. We conducted a pooled analysis using individual data from nine prospective cohorts in the Asia Cohort Consortium to explore the association between 10 female reproductive and hormonal factors and thyroid cancer risk. Using Cox proportional hazards models, cohort-specific hazard ratios (HR) and 95% confidence intervals (CI) were estimated and then pooled using a random-effects

Examining the continuity of modifiable cancer-risk behaviors from youth into adulthood through prospective longitudinal studies: A scoping review.

OBJECTIVE: Modifiable health behaviors including tobacco and alcohol use, poor diet, and low physical activity increase risk for developing multiple cancers. Longitudinal research suggests that risky behaviors initiated in youth may persist into adulthood. This scoping review maps prospective longitudinal studies examining the continuity of these behaviors from youth into adulthood. METHODS: Four electronic databases were searched for prospective longitudinal studies on the continuity of tobacco use, alcohol consumption, poor diet, and low physical activity from youth (<18) into adulthood (≥1

Five-year outcomes after surgical versus transcatheter aortic valve replacement with new generation devices from the prospective OBSERVANT studies.

The efficacy and durability of transcatheter (TAVR) over surgical aortic valve replacement (SAVR) for severe aortic stenosis (AS) has been demonstrated in randomized studies, but these findings were not confirmed in several observational studies. This is an analysis of 5706 AS patients who underwent SAVR from 2010 and 2012, and 2989 AS patients who underwent TAVR from 2017 and 2018 from the prospective OBSERVANT I and II studies. TAVR procedures were performed with new-generation devices. Five-year all-cause mortality was the primary outcome of this analysis. Propensity score matching yielded
