Research progress and prospect of β-carotene hydroxylase gene mediating plant carotenoid metabolism in response to abiotic stress.

Carotenoid metabolism plays a significant role in plants response to abiotic stress. The β-carotene hydroxylase gene (BCH) is an important gene located downstream of the carotenoid metabolic pathway. Its main function is to hydroxylate β-carotene to form zeaxanthin, which is the direct valve regulating the carotenoid xanthophyll cycle and also an important controller in the upstream of ABA synthesis. Most carotene components contain a molecular structure with conjugated double bonds, which determines that they have a powerful antioxidant function. When the BCH gene is overexpressed, Reactive o

Insights into E-Cadherin Impairment in CDH1-Unaltered Invasive Lobular Carcinoma: A Comprehensive Bioinformatic Study.

Invasive lobular carcinoma exhibits unique morphological features frequently associated with alterations in CDH1. Although some studies have identified abnormalities in adhesion factors other than E-cadherin, the molecular mechanisms underlying E-cadherin abnormalities in CDH1-unaltered invasive lobular carcinoma remain poorly understood. In this study, we investigated the molecular underpinnings of E-cadherin dysregulation in invasive lobular carcinoma in the absence of CDH1 gene alterations, using comprehensive bioinformatic analyses. We conducted a comparative study of CDH1-mutated and non-

The m6A reader YTHDC2 maintains visual function and retinal photoreceptor survival through modulating translation of PPEF2 and PDE6B.

Inherited retinal dystrophies (IRDs) are major causes of visual impairment and irreversible blindness worldwide, while the precise molecular and genetic mechanisms are still elusive. N6-methyladenosine (m6A) modification is the most prevalent internal modification in eukaryotic mRNA. YTH domain containing 2 (YTHDC2), an m6A reader protein, has recently been identified as a key player in germline development and human cancer. However, its contribution to retinal function remains unknown. Here, we explore the role of YTHDC2 in the visual function of retinal rod photoreceptors by generating rod-s

Therapeutic potential of pregnenolone and pregnenolone methyl ether on depressive and CDKL5 deficiency disorders: Focus on microtubule targeting.

Pregnenolone methyl-ether (PME) is a synthetic derivative of the endogenous neuroactive steroid pregnenolone (PREG), which is an important modulator of several brain functions. In addition to being the precursor of steroids, PREG acts directly on various targets including microtubules (MTs), the functioning of which is fundamental for the development and homeostasis of nervous system. The coordination of MT dynamics is supported by a plethora of MT-associated proteins (MAPs) and by a specific MT code that is defined by the post-translational modifications of tubulin. Defects associated with MA

Chikungunya E2 Protein Produced in E. coli and HEK293-T Cells-Comparison of Their Performances in ELISA.

Chikungunya virus (CHIKV) is a mosquito-borne pathogen that causes a disease characterized by the acute onset of fever accompanied by arthralgia and intense joint pain. Clinical similarities and cocirculation of this and other arboviruses in many tropical countries highlight the necessity for efficient and accessible diagnostic tools. CHIKV envelope proteins are highly conserved among alphaviruses and, particularly, the envelope 2 glycoprotein (CHIKV-E2) appears to be immunodominant and has a considerable serodiagnosis potential. Here, we investigate how glycosylation of CHIKV-E2 affects antig

From Translation Milestones to Clinical Breakthroughs: Decision-Making Hubs and Translational Strategies for RNA-Binding Proteins in the Immunotherapy of Haematological Malignancies.

Immunotherapy for haematological malignancies-including CAR-T and immune checkpoint inhibitors-is limited by resistance driven by rapid adaptive changes at the tumour-immune interface that transcriptional regulation cannot address. This review introduces 'translational checkpoints' as a synthesized working framework for overcoming resistance by targeting RNA-binding proteins (RBPs) that govern rapid protein synthesis in tumour and immune cells. We address three core questions: how tumours evade immune surveillance, how immune dysfunction arises, and how to develop targeted interventions. We co

Transcriptional profiling of a mouse model for Lafora disease reveals dysregulation of genes involved in the expression and modification of proteins.

Lafora's progressive myoclonus epilepsy (Lafora disease: LD) is caused by mutations in the EPM2A or NHLRC1 gene, but cellular mechanisms of the pathogenesis remain unclear. In an attempt to understand and elucidate the disease pathway, we have investigated the global gene expression profile in a mouse model for LD that developed a phenotype similar to that observed in human patients, including presence of Lafora bodies, neurodegeneration and profound neurological disturbances. We found 62 differentially expressed genes in the Epm2a knockout mice brains. These genes encode factors involved in p

The Role of HMGB1, a Nuclear Damage-Associated Molecular Pattern Molecule, in the Pathogenesis of Lung Diseases.

Significance: High-mobility group protein box 1 (HMGB1), a ubiquitous nuclear protein, regulates chromatin structure and modulates the expression of many genes involved in the pathogenesis of lung cancer and many other lung diseases, including those that regulate cell cycle control, cell death, and DNA replication and repair. Extracellular HMGB1, whether passively released or actively secreted, is a danger signal that elicits proinflammatory responses, impairs macrophage phagocytosis and efferocytosis, and alters vascular remodeling. This can result in excessive pulmonary inflammation and comp

RNA modifications and metabolism in cancer: mechanisms, cross-talk and therapeutic implications.

RNA modifications, particularly in the realm of epitranscriptomics, have emerged as crucial regulatory factors in cancer progression, influencing cancer cell behavior by reprogramming metabolic pathways. These modifications, such as N⁶-methyladenosine (m⁶A), 5-methylcytosine (m⁵C), N¹-methyladenosine (m¹A), N⁷-methylguanosine (m⁷G), N⁴-acetylcytidine (ac⁴C) and pseudouridine (Ψ), modulate mRNA stability, splicing, translation efficiency, and non-coding RNA function, thereby fine-tuning the expression of metabolic enzymes and transporters. Importantly, these marks converge on shared metabolic e

Post-Translational Modifications in Pancreatic Cancer: Mechanisms to Clinical Applications.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies, characterized by an extremely low 5-year survival rate. Its aggressiveness is attributed to early metastasis, rapid progression, and resistance to conventional therapies. Post-translational modifications (PTMs), which involve covalent modifications of amino acid residues, dynamically regulate protein activity, stability, and interactions. Increasing evidence has identified PTMs as key drivers in the pathogenesis of PDAC. PTMs encompass classical modifications such as phosphorylation, acetylation, glycosylation, ubi

Proteomic approaches to quantify cysteine reversible modifications in aging and neurodegenerative diseases.

Cysteine is a highly reactive amino acid and is subject to a variety of reversible post-translational modifications (PTMs), including nitrosylation, glutathionylation, palmitoylation, as well as formation of sulfenic acid and disulfides. These modifications are not only involved in normal biological activities, such as enzymatic catalysis, redox signaling, and cellular homeostasis, but can also be the result of oxidative damage. Especially in aging and neurodegenerative diseases, oxidative stress leads to aberrant cysteine oxidations that affect protein structure and function leading to neurod

Comprehensive review on alcohol-induced protein modifications in hepatic mitochondrial membranes and their functional implications.

Chronic alcohol consumption induces significant structural and functional impairments in hepatic mitochondria, primarily mediated through increased production of reactive oxygen and nitrogen species (ROS/RNS), leading to oxidative and nitrosative damage. The mitochondrial electron transport chain (ETC), especially complexes I, II, IV, and V, is a major target of such damage, resulting in diminished catalytic activities as demonstrated by decreased NADH dehydrogenase and cytochrome c oxidase activities in ethanol-fed rat models. Notably, succinate dehydrogenase (complex II) remains largely unaf

[Research progress on the involvement of N6-methyladenosine modification in the regulation of immune response in sepsis].

Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection, its exact pathogenesis has not yet been fully elucidated, and the clinical mortality rate remains high. N6-methyladenosine (m6A) modification is a common form of post-transcriptional chemical modification in eukaryotes. By influencing RNA splicing, nuclear-cytoplasmic transport, and translation, it plays a role in regulating various pathophysiological processes. Recent studies have revealed that dynamic changes in m6A modifications are associated with damage caused by the immune response in seps

Interplay Between Motor Proteins and Microtubule Stability: Involvement of Tubulin PTMs and MAPs.

Microtubules and motor proteins are components of a cytoskeleton that influence each other to maintain cellular functions. Microtubules act as tracks for motor protein-driven intracellular transport of organelles, vesicles, and macromolecules; therefore, their structural organization and stability are important. Kinesin and dynein motor proteins function not only as cargo transporters but also as contributors to the regulation of microtubule organization, stability, and interactions. The interaction of motor proteins and microtubules is essential for cellular homeostasis, and dysregulations ca

Evaluation of CAAX prenyl protease II of Leishmania donovani as potential drug target: Infectivity and growth of the parasite is significantly lowered after the gene knockout.

Prenylation pathway is responsible for post translational modification of various signal proteins, including proteins of Ras superfamily. CAAX prenyl proteases are known to be key players in prenylation pathway. In the current study, we have evaluated CAAX prenyl protease II as a possible drug target against Leishmania donovani parasite, the causative agent of visceral leishmaniasis. Gene knockout strategy was employed to target CAAX prenyl protease II and subsequent effects were studied. CAAX prenyl protease II knockout resulted in significant decrease in growth and infectivity.

Mapping phosphoproteins in Neisseria meningitidis serogroup A.

To investigate the phosphorylation capability of serogroup A Neisseria meningitidis (MenA) and to implement our knowledge in meningococcal biology and in bacterial post-translational modifications, cell extracts were separated by 2-DE and 51 novel phosphoproteins were revealed by the use of the highly specific Ser/Thr/Tyr-phosphorylated proteins staining by Pro-Q Diamond and identified by MALDI-ToF/MS. Our results indicate that phosphorylation in MenA is comparable to that of other bacterial species. A first functional characterization of the identified modified proteins was also given, in ord
