Helicobacter pylori is a human pathogen that infects almost half of the population. Antibiotic resistance in H. pylori threatens health and increases the demand for prophylactic and therapeutic vaccines. Traditional oral vaccine research faces considerable challenges because of the epithelial barrier, potential enterotoxicity of adjuvants, and the challenging conditions of the gastric environment. We developed an intranasal influenza A virus (IAV) vector vaccine based on two live attenuated influenza viruses with modified acidic polymerase protein (PA) genes encoding the A subunit of H. pylori

Coccidiosis caused by Eimeria species poses a significant threat to the global poultry industry. Gametocyte antigens essential for oocyst wall biogenesis represent promising targets for transmission-blocking vaccine development. In this study, the E. necatrix gam82 gene (1650 bp) was cloned, encoding a 549-amino acid protein. A truncated fragment (amino acids 258-464) enriched in tyrosine‑serine-rich regions (amino acids 258-442) was expressed in Escherichia coli, yielding the recombinant protein rEnGAM82-T. Western blot analysis demonstrated that rEnGAM82-T exhibited strong immunoreactivity a

Vaccine design strategies against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are focused on the Spike protein or its subunits as the main antigen target of neutralizing antibodies. In this work, we propose rapid production methods of an extended segment of the Spike Receptor Binding Domain (RBD) in HEK293SF cells cultured in suspension, in serum-free media, as a major component of a COVID-19 subunit vaccine under development. The expression of RBD, engineered with a sortase-recognition motif for protein-based carrier coupling, was achieved at high yields by plasmid transient

Although inactivated vaccines against rabies have the advantage of high safety, effective protection against rabies virus (RABV) infection often requires multiple, high-dose immunization. Incorporating a molecular adjuvant into the viral particles has been found to be a useful strategy to promote the immune effectiveness of inactivated vaccines. In this study, we constructed a recombinant virus, rCVS11-LTB, which chimerically expresses a molecular adjuvant heat-labile enterotoxin B subunit (LTB) protein on the surface of the RABV particles. Immunogenicity in vivo was found to be promoted by rC

Streptococcus agalactiae, often referred to as group B streptococci (GBS), is a severe pathogen that can infect humans as well as other animals, including tilapia, which is extremely popular in commercial aquaculture. This pathogen causes enormous pecuniary loss, and typical symptoms of streptococcosis-the disease caused by S. agalactiae-include abnormal behavior, exophthalmos, and meningitis, among others. Multiple studies have examined virulence factors associated with S. agalactiae infection, and vaccines were explored, including studies of subunit vaccines. Known virulence factors include

Significant safety issues have emerged concerning the general use of DRYVAX vaccine. Vaccination with replication-defective recombinant adenovirus (rAd) vaccines may offer a safer and effective alternative to live vaccinia virus (VV) vaccination. Six individual poxvirus glycoproteins: A33R, A34R, A36R, B5R, A27L or L1R that are normally expressed on the surface of infectious vaccinia virus were encoded in rAd vaccines and tested in mice in this study. A single-shot intramuscular injection of rAd encoding A27L protected mice against a lethal intranasal challenge with VV at 4 weeks post-vaccinat

BACKGROUND: The prevailing public health threat posed by malaria, especially in developing countries, remains a serious concern despite the availability of preventive and control measures. While vaccination offers a powerful means of combating malaria, it has not been fully exploited due to previous unsuccessful attempts before the launch of the RTS,S vaccine. A major challenge in malaria vaccine development continues to be the identification of effective targets capable of eliciting robust immunity, given the complexity of the parasites' life cycle. Leveraging on the breakthrough of the newly

Salmonella Enteritidis (S. Enteritidis) is a major pathogen causing foodborne diseases worldwide, posing a serious threat to public health security and the sustainable development of the poultry industry. To overcome the limitations of conventional vaccines in terms of efficacy, safety, and cross-protection, a novel multi-epitope subunit vaccine (SEMV) was designed and constructed using an immunoinformatics-based strategy. Through systematic screening of epitopes from key immunogenic proteins of S. Enteritidis (SseB, SipC, OmpD and TolC), a total of 8 B-cell epitopes, 4 MHC-I-restricted epitop

Maternally derived antibodies (MDAs) are critical for offspring protection during the first days of life. However, due to their short half-l … le, Newcastle disease virus and infectious bursal disease virus, respectively. These may be used as protein subunit vaccines or live vaccines carried by a vector. This vaccination concept may overcome the gap in protection that occurs when MDAs decrease while self-immunity is still partial, to provide continuous protection at a young age.

Lumazine synthase from Brucella spp. (BLS) is a highly immunogenic decameric protein. It is possible to insert foreign peptides or proteins at its ten-amino acid termini. These chimeras elicit systemic and oral immunity without adjuvants, which are commonly needed in the formulation of subunit-based vaccines. Here, we show that BLS induces the cross presentation of a covalently attached peptide OVA(257-264) and a specific cytotoxic response to this peptide in the absence of adjuvants. Unlike other subunit-based vaccines, this chimera induces rapid activation of CTLs and a specific cytotoxic re

Current dengue vaccines remain limited by serotype-dependent efficacy and interference from preexisting anti-dengue immunity. We developed a novel multivalent fusion protein vaccine composed of three engineered dengue virus (DENV) components: a C-terminal truncated nonstructural protein 1 (ΔcNS1) to block NS1-mediated pathologic effects without harmful cross-reactivity, a consensus envelope protein domain III (cEDIII) to induce broad neutralizing antibodies, and an N-terminal truncated NS3 (ΔnNS3) to enhance cellular immune responses. In a murine dengue disease model, three-dose immunization w

Myeloid cells are capable of releasing web-like extracellular structures composed of DNA and granular proteins, known as extracellular traps (ETs), which function to eliminate invading pathogens and modulate immune responses. Intraperitoneal vaccine administration has been demonstrated to induce ETs, but their roles in vaccine-mediated immune responses remain largely unknown. In this study, we show that a protein subunit vaccine triggers the release of ETs in the peritoneal cavity of teleost fish. Pharmacological inhibition of ET formation by 4-aminobenzoic acid hydrazide (ABAH) or enzymatic d

Brucellosis, a zoonosis caused by Brucella, is highly detrimental to both humans and animals. Most existing vaccines are live attenuated vaccines with safety flaws for people and animals. Therefore, it is advantageous to design a multi-epitope subunit vaccine (MEV) to prevent Brucella infection. To this end, we applied a reverse vaccinology approach. Six cytotoxic T cell (CTL) epitopes, seven T helper cell (HTL) epitopes, and four linear B cell epitopes from CU/ZN-SOD, Omp31, and BP26 were obtained. We linked the CTL, HTL, B-cell epitopes, the appropriate CTB molecular adjuvant, and the univer

The need to simultaneously target infections with epidemiological overlap in the population with a single vaccine provides the basis for developing combination vaccines. Vibrio cholerae ghosts (rVCG) offer an attractive approach for developing vaccines against a number of human and animal pathogens. In this study, we constructed a multisubunit vaccine candidate co-expressing the serovar D-derived Porin B and polymorphic membrane protein-D proteins of Chlamydia trachomatis and evaluated its ability to simultaneously induce broad-based chlamydial immunity and elicit a vibriocidal antibody respon

BACKGROUND: Neisseria gonorrhoeae, causative agent of the human sexually transmitted infection gonorrhea, is a significant global health concern because of increasing antimicrobial resistance and the lack of an effective vaccine. Recent ecological analyses have shown a reduced incidence of gonorrhea in recipients of detergent-extracted outer membrane vesicle (OMV)-containing meningococcal vaccine, which has contributed to identification of shared, protective antigens. Previously, our group has developed an immunobioinformatics-based pipeline (CASS, Candidate Antigen Selection Strategy) for ide

Typhoid fever caused by Salmonella enterica serovar Typhi (S.Typhi) continues to be a major problem, especially in developing countries. Due to the rapid emergence of multi-drug-resistant (MDR) strains, which limits the efficacy of conventional antibiotics as well as problems associated with the existing vaccines, efforts are being made to develop effective prophylactic agents. CdtB subunit of typhoid toxin was selected for assessing its vaccine potential due to its high conservation throughout the Typhi strains. In-vitro assessment of DNase activity of cloned and purified CdtB protein showed

The hepatitis B virus (HBV) core protein (HBc) has formed the building block for virus-like particle (VLP) production for more than 30 years. The ease of production of the protein, the robust ability of the core monomers to dimerize and assemble into intact core particles, and the strong immune responses they elicit when presenting antigenic epitopes all demonstrate its promise for vaccine development (reviewed in Pumpens and Grens (Intervirology 44: 98-114, 2001)). HBc has been modified in a number of ways in attempts to expand its potential as a novel vaccine platform. The HBc protein is pre

Infectious bursal disease virus (IBDV) is one of the most important immunosuppressive viruses in poultry, causing the global spread of infectious bursal disease (IBD). It poses a significant threat to the healthy development of the poultry industry. Vaccination is an effective approach for controlling IBDV infection. Therefore, reliable immune monitoring for IBDV is critical for maintaining poultry health. The enzyme-linked immunosorbent assay (ELISA) is a common technique used to detect specific antibodies in clinical serum testing and for the serological evaluation of IBDV vaccines. Among th

Improving vaccine immunogenicity by targeting antigens to dendritic cells has recently emerged as a new design strategy in vaccine development. In this study, the VP1 gene of foot-and-mouth disease virus (FMDV) serotype A was fused with the gene encoding human immunodeficiency virus (HIV) membrane glycoprotein gp120 or C2-V3 domain of hepatitis C virus (HCV) envelope glycoprotein E2, both of which are DC-SIGN-binding glycoproteins. After codon optimization, the VP1 protein and the two recombinant VP1-gp120 and VP1-E2 fusion proteins were expressed in Sf9 insect cells using the insect cell-bacu

Proteus mirabilis is a gram-negative bacterium particularly known for its unique swarming ability. The swarming gives the bacteria ability to enhance adherence to the catheter surface and epithelium cells of the urethra to cause catheter associated urinary tract infections. P. mirabilis has evolved resistant to antibiotics. Additionally, there is an approved vaccine against P. mirabilis, thus demanding for identification of new vaccine targets. This gram-negative bacterium consists of 19,502 core proteins, out of which 19,063 are redundant proteins and remaining 439 are non-redundant proteins.
