Identification of pyrrolo[2,3-d]pyrimidine-based dual MERTK and FLT3 inhibitor: Hit-to-lead, machine learning, modeling, synthesis, and biological evaluation.

Overexpression of MERTK and FLT3 plays a crucial role in activating signal transduction pathways in various human hematological malignancies. These signaling pathways have been extensively studied and have shown significant potential as a promising therapeutic target for the treatment of acute myeloid leukemia (AML). In this study, we employed a modern medicinal chemistry approach, hybridizing machine learning (ML) with a bioisosterism strategy, to design and synthesize a new series of pyrrolo[2,3-d] pyridine derivatives as potent dual inhibitors of MERTK and FLT3. Through successive structure

Discovery of [1,2,4]triazolo[1,5-a]pyrimidine-Imatinib Hybrids With Selective Cytotoxic Activity: A Mechanistically Divergent Series From Direct BCR-ABL1 Inhibition.

Chronic myeloid leukemia treatment faces the challenge of resistance to BCR-ABL1 tyrosine kinase inhibitors. To address this, we rationally designed six new imatinib-derived compounds 2(a-f) by replacing the quinoline moiety with a [1,2,4]triazolo[1,5-a]pyrimidine scaffold via classical bioisosterism. The compounds were efficiently synthesized and characterized. Biological evaluation revealed that compound 2a exhibited cytotoxic activity in BCR-ABL1-positive K562 cells (47% viability inhibition at 10 µM, IC50 = 9.7 µM). However, enzymatic assays demonstrated that 2a does not directly inhibit t

Anticancer evaluation of novel chiral tetrahydropyrazolo[1,5-a]pyrimidine derivatives: Inhibitory effects on proliferation and migration of colon cancer cells with high HER2 expression.

In colorectal cancer (CRC), oncogenic activation of the human epidermal growth factor receptor 2 (HER2) is strongly associated with tumor metastasis, poor prognosis, and resistance to anti-EGFR targeted therapies. Currently, there is a lack of tyrosine kinase inhibitors (TKIs) targeting HER2-high-expressing CRC. Current therapeutic approaches face challenges including intertumoral heterogeneity in treatment response and the rapid emergence of drug resistance. Therefore, the development of novel, potent, and selective TKIs for HER2-high-expressing CRC represents a critical unmet medical need. P

Design, Synthesis, Molecular Docking, ADME-T and In Vitro Anticancer Assessment of Phenyl-Substituted-Pyrimidin-Benzenesulfonamide Derivatives as Potential BRAFV600E /WT Inhibitors.

BRAF is one of the most commonly mutated oncogenes in human cancers. More than 90% of BRAF mutations are associated with malignant melanoma. Given the pivotal role of BRAFV600E/WT mutations in melanoma progression and therapy resistance, our study focused on the design of phenyl-substituted pyrimidin-benzenesulfonamide hybrids in the [αC-OUT/DFG-IN] conformation, inspired by previously synthesized molecules structurally related to FDA-approved BRAF inhibitors. A total of ten derivatives were synthesized, and their ADME-T properties, in silico binding affinities, in vitro cytotoxic activities a

Targeting p21-activated kinase 4 (PAK4) with pyrazolo[3,4-d]pyrimidine derivative SPA7012 attenuates hepatic ischaemia-reperfusion injury in mice.

p21-Activated kinase 4 (PAK4), one of the serine/threonine kinases activated by Rho-family GTPases, has been widely studied as an oncogenic protein that is overexpressed in many types of cancers. In our recent study, PAK4 upregulation was observed in mice exhibiting hepatic ischaemia-reperfusion (I/R) and in liver transplantation patients. Liver I/R injury was also attenuated in Pak4 KO mice. Herein, we report a novel series of pyrazolo[3,4-d]pyrimidine derivatives of type I ½ PAK4 inhibitors. The most potent compound SPA7012 was evaluated to determine the pharmacological potential of PAK4 inh

A diversity oriented, microwave assisted synthesis of N-substituted 2-hydro-4-amino-pyrido[2,3-d]pyrimidin-7(8H)-ones.

A protocol for the synthesis of N-substituted 2-hydro-4-amino-pyrido[2,3-d]pyrimidin-7(8H)-ones (11) is described. Thus, the formylation of a 2-aminopyridone 12 in 85% formic acid/Ac(2)O, proceeding via in situ cyclization to the intermediate formamide 13, affords the corresponding 2-hydro-4-oxo-pyridopyrimidine 14, which is converted to a 4-chloro-pyridopyrimidine 15 upon treatment with POCl(3). The subsequent transformation to the title compounds is carried by treatment with the corresponding amine in MeOH under microwave irradiation conditions.

Repeated exposures of human skin equivalent to low doses of ultraviolet-B radiation lead to changes in cellular functions and accumulation of cyclobutane pyrimidine dimers.

Chronic exposure to sunlight may induce skin damage such as photoaging and photocarcinogenesis. These harmful effects are mostly caused by ultraviolet-B (UVB) rays. Yet, less is known about the contribution of low UVB doses to skin damage. The aim of this study was to determine the tissue changes induced by repeated exposure to a suberythemal dose of UVB radiation. Human keratinocytes in monolayer cultures and in skin equivalent were irradiated daily with 8 mJ/cm2 of UVB. Then structural, ultrastructural, and biochemical alterations were evaluated. The results show that exposure to UVB led to

Evidence for defective repair of cyclobutane pyrimidine dimers with normal repair of other DNA photoproducts in a transcriptionally active gene transfected into Cockayne syndrome cells.

Cockayne syndrome (CS) and xeroderma pigmentosum (XP), autosomal recessive diseases with clinical and cellular hypersensitivity to UV radiation, differ in ability to repair UV DNA photoproducts in their overall genome: normal repair in CS, defective repair in XP. In order to characterize a DNA repair defect in an active gene in CS, we measured the capacity of cells from patients with CS and XP to reactivate 2 major types of UV-induced DNA damage, photoreactivatable (i.e., cyclobutane pyrimidine dimers) and non-photoreactivatable (primarily pyrimidine-(6-4)pyrimidone photoproducts), in the acti

Discovery of Dihydropyrimido-Pyrimidine-Based Bone Marrow Tyrosine Kinase Gene in Chromosome X Protein Proteolysis-Targeting Chimeras for the Treatment of Prostate Cancer.

Bone marrow tyrosine kinase gene in chromosome X protein (BMX) plays a critical role in the initiation, progression, and development of castration-resistant prostate cancer. While several ATP-competitive BMX inhibitors have been developed as cellular chemical tools to investigate the function of BMX, they are unable to target the nonenzymatic roles of BMX. Proteolysis-targeting chimeras (PROTACs) represent an emerging technology that enables the rapid and complete degradation of target proteins. Here, we describe the design, synthesis, and characterization of BMX PROTACs by conjugating the BMX

Design, Synthesis, In Vitro and In Vivo Biological Evaluation of Novel Pyrazolo[3,4-d]Pyrimidine-Based Derivatives as Promising Multitarget Candidates for Alzheimer's Disease.

Multi-target directed ligands (MTDLs) advocate for a novel therapeutic framework for Alzheimer's disease (AD) owing to its complicated multifactorial nature. Therefore, the present study focuses on designing and synthesizing pyrazolo[3,4-d]pyrimidine derivatives, followed by biological evaluation as MTDLs for AD. Most of the obtained derivatives showed potent AChE inhibitory efficacy, significant suppression of Aβ, and pronounced antioxidant activity in vitro. Compound 8b (IC50 = 0.346 μM) and 9a (IC50 = 0.168 μM) are the most potent inhibitors of AChE relative to donepezil (IC50 = 0.213 μM).
