A spontaneous, nonacute, age-dependent prostatitis was found in a high incidence in the lateral prostatic lobes of Lewis rats. Such rats were treated with methylprednisolone, indometacin , testosterone, hexyloxyphenylproprionate , polyestradiol phosphate, various antibiotics, or were caged together with female rats. The effect of the different treatment modalities was evaluated microscopically by blind observation of the degree of inflammatory reaction in the lateral prostate. Our main objective was to describe the metabolism of dithiobiuret (DTB) in the adult, male rat. Based on the thin-layer chromatographic analysis of urine from animals treated ip with 1 mg/kg of [14C] or [35S] labeled DTB, two pathways for metabolism are proposed. One pathway is reversible and involves the oxidation of DTB to thiuret and the reduction of thiuret back to DTB. The other pathway consists of the desulfuration of DTB to monothiobiuret. The liver appears to desulfurate DTB because DTB-derived [35S] was eliminated from the liver more rapidly (T1/2 = 10 hr) than [14C] (T1/2 = 15 hr). T

The metabolism of 1-(3-butoxy-2-carbamoyloxypropyl)-5-ethyl-5-phenyl-(1H,3H,5H)-pyrimidine-2,4,6- trione (febarbamate) in the rat has been studied after oral administration. Using high performance liquid chromatography, fifteen metabolites were isolated from urine, purified and identified by MS and NMR spectrometry. Febarbamate was extensively metabolized and only traces of the unchanged compound were found. A method for removal of the thymus in the newborn rat employing measures aimed at reducing the mortality incurred by the surgical procedure, cannibalism and infection is described in detail. Tranquilisation of the dam, sterile technique, magnification of the operative site for thymus aspiration, warming to 37 degrees C during the recovery phase, and application of pheromones to the offspring prior to their reunion with the dam, all proved important in increasing the survival rate.
