Bacterial viability determination in a dentinal tubule infection model by confocal laser scanning microscopy.

Dentinal tubule invasion protects bacteria from chemo-mechanical disinfection and frequently results in root canal treatment failures. Enterococcus faecalis is a primary causative agent, particularly in persistent, asymptomatic, and chronic apical periodontitis. In order to assess and compare the efficacies of endodontic antimicrobial agents and application strategies, we have developed a convenient and robust method to measure bacterial viability and assess distribution in an ex vivo tubule infection model. Following infection and antimicrobial treatment of prepared ex vivo roots, the tubule

Comparative Evaluation of Depth of Irrigant Penetration Using Different Irrigation Systems: An In Vivo Study.

INTRODUCTION: Effective irrigation is essential for successful root canal treatment because mechanical instrumentation alone cannot adequately clean the complex anatomy of the root canal system. Various irrigant activation techniques have been developed to enhance irrigant penetration into the apical third and anatomically inaccessible regions; however, clinical evidence comparing their penetration efficiency remains limited. The aim of our study was to compare the depth of irrigant penetration achieved using conventional manual irrigation, passive ultrasonic irrigation, sonic irrigation, air-

An unexpected positive hypersensitive reaction to eugenol.

Eugenol is an active, principal aromatic liquid responsible for several pharmacological activities. It is widely used in dental practice to relieve pain arising from various sources, such as pulpitis and dentinal hypersensitivity. As a primary irritant and sensitiser, it is known to cause contact urticaria as well as chronic urticaria. However, eugenol causes allergic contact dermatitis, possibly because it can react directly with proteins to form conjugate and reactive haptens. It is found that eugenol in various dental preparations-especially in the case of some zinc oxide-contains preparati

Detection of bacteria in histological sections of the dental pulp.

A series of deep cavities with and without traumatic pulpal exposures were prepared in the maxillary molars of albino rats. They were left untreated for various time intervals before killing. Demineralized sections were prepared in a mesiodistal plane and stained to demonstrate the presence of bacteria. In the cavities and coronal pulps that were grossly contaminated, bacteria were detected in the majority of sections. In the root canals and periapical tissues, bacteria were detected in only a minority of sections. It is considered that serial sections stained to demonstrate bacteria are neces

Evaluation of Antimicrobial Photodynamic Therapy Using Indocyanine Green and Near-Infrared Diode Laser Against Enterococcus faecalis in Infected Human Root Canals.

OBJECTIVE: Photodynamic therapy (PDT) is considered a promising adjunct to the currently available endodontic disinfection techniques leading to more effective reduction of intracanal bacteria. The present ex vivo study aimed to assess the antimicrobial effect of PDT using indocyanine green (ICG) as photosensitizer and a near-infrared (NIR) diode laser in root canals of human teeth infected with Enterococcus faecalis. MATERIALS AND METHODS: Ninety single-rooted teeth after chemomechanical preparation and sterilization were contaminated with an E. faecalis strain. The specimens were divided, r

In vitro comparison to study the antimicrobial effect of silver nanoparticles gel and its various combinants as an intracanal medicament against Enterococcus faecalis.

BACKGROUND: Endodontic infections have been clearly described as biofilm-mediated infections. Bacteria and their by-products have been known to cause these infections. With the introduction of new drugs and the use of nanoparticles in recent times, there has been a significant reduction in the bacterial load in endodontic infections. AIMS AND OBJECTIVES: The in vitro study focuses on checking the antibacterial efficacy of silver nanoparticles and its combination with other medicaments against the root canal pathogen - Enterococcus faecalis (E. faecalis). METHODOLOGY: In the present study, 14

Periodontal response to two intracanal medicaments in replanted monkey incisors.

Intracanal medicaments are recommended for use in replanted teeth to inhibit inflammatory root resorption. This study compared the effect of calcium hydroxide (Pulpdent) and a corticosteroid-antibiotic paste (Ledermix) on periodontal healing and root resorption following replantation. Incisors of eight Macaca fascicularis monkeys were extracted, stored dry for 15 min and replanted. After 11 days, root canals in two adjacent maxillary incisors were treated with one medicament and contralateral incisors with the other medicament, or left as untreated controls. Animals were sacrificed 8 weeks lat

Location of contact areas on rotary Profile instruments in relationship to the forces developed during mechanical preparation on extracted teeth.

AIM: The aim of this study was to locate the areas of direct instrument contact with dentine in the root canal system during rotary preparation and to analyse the relationship between these areas and the vertical forces and torque developed during the preparation. METHODOLOGY: Canal preparations were performed by endodontists either with the step-back (SB) or the crown-down (CD) technique. In order to locate the areas of contact, the instruments were coated with two layers of gold by electro-deposition. They were photographed before and after use, and a coding system, based on mm from the ins

Choosing the Right File for the Canal: Clinical Applications of Two Different Single-File Reciprocating Systems.

Reciprocating single-file nickel-titanium (NiTi) systems have simplified root canal shaping while offering improved resistance to cyclic fatigue, the predominant mode of instrument failure. This article presents two cases treated with two different reciprocating systems built on different heat-treated NiTi alloys with distinct mechanical profiles. In the first case, the reciprocating system's patented heat treatment provided the added cutting efficacy and stiffness needed to enable confident management of a four-canal maxillary first molar (tooth No. 14), including complete location and shapin
