Cohort profile of the longitudinal Netherlands Study of Depression and Anxiety (NESDA) on etiology, course and consequences of depressive and anxiety disorders.

INTRODUCTION: The Netherlands Study of Depression and Anxiety (NESDA, www.nesda.nl) is a longitudinal, multi-site, naturalistic, case-control cohort study set up to examine the etiology, course and consequences of depressive and anxiety disorders. This paper presents a cohort profile of NESDA. METHODS AND RESULTS: The NESDA sample recruited initially 2329 persons with a remitted or current DSM-IV based depressive (major depressive disorder, dysthymia) and/or anxiety disorder (panic disorder, social phobia, agoraphobia, generalized anxiety disorder), 367 of their siblings and 652 healthy contr

Lower nasopharyngeal viral load during the latest phase of COVID-19 pandemic in a Northern Italy University Hospital.

OBJECTIVES: A milder clinical course of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has been anecdotally reported over the latest phase of COVID-19 pandemic in Italy. Several factors may contribute to this observation, including the effect of lockdown, social distancing, lower humidity, lower air pollution, and potential changes in the intrinsic pathogenicity of the virus. In this regard, the clinical severity of COVID-19 could be attenuated by mutations in SARS-CoV-2 genome that decrease its virulence, as well as by lower virus inocula. METHODS: In this pilot study

Severe childhood speech disorder: Gene discovery highlights transcriptional dysregulation.

OBJECTIVE: Determining the genetic basis of speech disorders provides insight into the neurobiology of human communication. Despite intensive investigation over the past 2 decades, the etiology of most speech disorders in children remains unexplained. To test the hypothesis that speech disorders have a genetic etiology, we performed genetic analysis of children with severe speech disorder, specifically childhood apraxia of speech (CAS). METHODS: Precise phenotyping together with research genome or exome analysis were performed on children referred with a primary diagnosis of CAS. Gene coexpre

Cross-disorder comparative analysis of comorbid conditions reveals novel autism candidate genes.

BACKGROUND: Numerous studies have highlighted the elevated degree of comorbidity associated with autism spectrum disorder (ASD). These comorbid conditions may add further impairments to individuals with autism and are substantially more prevalent compared to neurotypical populations. These high rates of comorbidity are not surprising taking into account the overlap of symptoms that ASD shares with other pathologies. From a research perspective, this suggests common molecular mechanisms involved in these conditions. Therefore, identifying crucial genes in the overlap between ASD and these comor

HLA-A, B, DRB1, DQA1, DQB1 alleles and haplotype frequencies in Dene and Cree cohorts in Manitoba, Canada.

BACKGROUND: First Nations in the Canadian province of Manitoba have disproportionately high rates of epidemic and endemic TB. Gene polymorphisms that modulate HLA Class I and II antigens are among the risk markers for TB, along with other biologic, and social determinants of health. HLA-A, B, DRB1, DQA1, DQB1 were typed in two Manitoba First Nation indigenous groups to identify and compare the frequency of gene polymorphisms that may influence susceptibility or resistance to TB. METHODS: Participants who self-identified as either Dene or Cree enrolled into the study from two First Nation comm

Syndromic autism spectrum disorders: moving from a clinically defined to a molecularly defined approach.

Autism spectrum disorder (ASD) encompasses a group of neurodevelopmental conditions diagnosed solely on the basis of behavioral assessments that reveal social deficits. Progress has been made in understanding its genetic underpinnings, but most ASD-associated genetic variants, which include copy number variants (CNVs) and mutations in ASD-risk genes, account for no more than 1 % of ASD cases. This high level of genetic heterogeneity leads to challenges obtaining and interpreting genetic testing in clinical settings. The traditional definition of syndromic ASD is a disorder with a clinically de

In twenty years, significant progress was made in the knowledge of viral hepatitis. Alphabet of hepatotropic viruses has extended, and C, D, E and G viruses have been added to hepatitis A and B viruses; their genomes have been characterized, allowing defining various types, subtypes and isolates with different pathological and therapeutic implications. Our knowledge regarding the epidemiology, virology and treatment of viral hepatitis is in constant evolution, allowing a better diagnostic and therapeutic approach of patients with acute and chronic hepatitis. Markers of infection and early vira

Cell-free fetal nucleic acid testing: a review of the technology and its applications.

UNLABELLED: Cell-free fetal nucleic acids circulating in the blood of pregnant women afford the opportunity for early, noninvasive prenatal genetic testing. The predominance of admixed maternal genetic material in circulation demands innovative means for identification and analysis of cell-free fetal DNA and RNA. Techniques using polymerase chain reaction, mass spectrometry, and sequencing have been developed for the purposes of detecting fetal-specific sequences, such as paternally inherited or de novo mutations, or determining allelic balance or chromosome dosage. Clinical applications of th

The core FOXG1 syndrome phenotype consists of postnatal microcephaly, severe mental retardation, absent language, dyskinesia, and corpus callosum hypogenesis.

BACKGROUND: Submicroscopic deletions in 14q12 spanning FOXG1 or intragenic mutations have been reported in patients with a developmental disorder described as a congenital variant of Rett syndrome. This study aimed to further characterise and delineate the phenotype of FOXG1 mutation positive patients. METHOD: The study mapped the breakpoints of a 2;14 translocation by fluorescence in situ hybridisation and analysed three chromosome rearrangements in 14q12 by cytogenetic analysis and/or array comparative genomic hybridisation. The FOXG1 gene was sequenced in 210 patients, including 129 patien

Genetics of recurrent early-onset depression (GenRED): design and preliminary clinical characteristics of a repository sample for genetic linkage studies.

This is an initial report on a six-site collaborative project, Genetics of Recurrent Early-Onset Depression (GenRED). This is a study of a large sample of families with recurrent major depressive disorder (DSM-IV) beginning by the age 30 in probands or 40 in relatives. Evidence suggests that early onset and recurrence of depressive episodes predict substantially increased risk of depression in first-degree relatives compared with the general population, suggesting that susceptibility genes might be mapped with this phenotype. The projected sample of 800-1,000 affected sibling pairs (ASPs) and

Mutations of the 5alpha-steroid reductase type 2 gene in six Turkish patients from unrelated families and a large pedigree of an isolated Turkish village.

We describe six Turkish patients with 5alpha-steroid reductase type 2 deficiency from unrelated Turkish families and a large pedigree of one of these patients who reside north-west of Anatolia. Patients NA, KS, BD and SY presented for evaluation of bilateral inguinal masses with female phenotypes. Patient ABE had penoscrotal hypospadias with male phenotype. Homozygous mutation of the 5alphaSR2 gene was identified in five of these patients by genomic DNA analysis. These mutations were Leu55Gln in exon 1 (in patients FG, BD and ABE), deltaMet157 in exon 3 (in patient NA), and splice junction abn

Attention-Deficit/Hyperactivity Disorder and Late-Life Cognitive Impairment.

IMPORTANCE: Attention-deficit/hyperactivity disorder (ADHD) affects tens to hundreds of millions of adults worldwide, and preliminary evidence suggests it may be associated with increased risk of mild cognitive impairment (MCI) and dementia. However, concerns regarding diagnostic ascertainment and residual confounding remain. OBJECTIVE: To investigate the association of ADHD with incident cognitive impairment, defined as MCI or dementia, using electronic health record (EHR) diagnoses and genetic liability measured using genome-wide ADHD polygenic risk score (ADHD PRS) in the same individuals.

Developing Trustworthy Artificial Intelligence Models to Predict Vascular Disease Progression: the VASCUL-AID-RETRO Study Protocol.

INTRODUCTION: Abdominal aortic aneurysms (AAAs) and peripheral artery disease (PAD) are two vascular diseases with a significant risk of major adverse cardiovascular events and mortality. A challenge in current disease management is the unpredictable disease progression in individual patients. The VASCUL-AID-RETRO study aims to develop trustworthy multimodal predictive artificial intelligence (AI) models for multiple tasks including risk stratification of disease progression and cardiovascular events in patients with AAA and PAD. METHODS: The VASCUL-AID-RETRO study will collect data from 5000

Capturing Perceived Utility from Qualitative Studies of Genomic Sequencing: Translating a Conceptual Model into a Working Codebook.

INTRODUCTION: As genomic sequencing (GS) is integrated into clinical medicine, understanding how patients and families perceive its utility is critical. Conceptual models for perceived utility have been developed, however, direct application of such models to independently-collected qualitative data remains rare. To fill this gap, we translated a comprehensive model of perceived utility (Smith et al. 2022) into a codebook and revised it via application to a qualitative dataset capturing parent-reported utility of GS. METHODS: Using the domains and subdomains from Smith et al.'s conceptual mod

Adult ADHD with comorbid major depression shows a distinguishable polygenic pattern and negative cognitive style.

Attention-deficit/hyperactivity disorder (ADHD) is a highly heritable neurodevelopmental disorder, and comorbidity with other mental diseases is common. Specifically, adult ADHD (aADHD) is highly comorbid with major depressive disorder (MDD). A genetic correlation between ADHD and MDD might underlie the risk of comorbidity of both disorders. However, whether patients with ADHD and comorbid MDD differ genetically from those without comorbid MDD is currently unclear. We therefore studied the genetic background of an aADHD cohort including 352 patients with lifetime MDD and 349 patients with no h

Cestode infection is linked to transcriptional shifts in neuropeptide signalling and caste-specific ageing pathways in a social insect.

Social insects are key models for phenotypic plasticity, as queens and workers show striking differences in behaviour and lifespan despite sharing the same genome. Neuropeptides are central regulators of behavioural plasticity, and caste-specific ageing is governed by the insulin/insulin-like growth factor 1 signalling-target of rapamycin-juvenile hormone network. Parasites that manipulate host behaviour and lifespan may exploit these regulators by targeting conserved neuropeptides or by altering ageing pathways. Here, we examine a host-parasite system in which the cestode Anomotaenia brevis e

Genetic and Ecological Divergence Between Northwest Atlantic Killer Whale Populations.

Killer whales (Orcinus orca) exhibit substantial genetic and ecological variation across their global distribution. Differentiation between neighboring or sympatric populations is thought to be driven by foraging specialization and social organization, which can lead to reproductive isolation and facilitate the emergence of distinct ecotypes or morphotypes. Here, we use whole-genome resequencing and compound-specific stable isotope analysis of amino acids to investigate links between genetic and ecological differentiation in two genetically distinct killer whale populations in the northwest At

Bidirectional association between immune-mediated diseases and major depressive disorder: evidence from cohort, genome-wide pleiotropic, and experimental studies.

Although immune-mediated diseases (IMDs) and major depressive disorder (MDD) commonly co-occur, the bidirectional relationship between them remains to be fully elucidated. Using data from the prospective UK Biobank cohort, we evaluated the bidirectional associations by time-varying Cox proportional hazards regression models and assessed shared genetic architecture using genome-wide association study summary statistics. Additionally, we employed collagen-induced arthritis (CIA) and chronic social defeat stress (CSDS) mouse models to investigate the relationship between rheumatoid arthritis (RA)

Salmonella Infections: Global Trends and Emerging Challenges.

Salmonella remains a leading cause of foodborne illness worldwide, with non-typhoidal Salmonella (NTS) responsible for an estimated 93.8 million infections and substantial global morbidity and mortality. This review synthesizes current evidence on the epidemiology, molecular pathogenesis, and prevention of both typhoidal and nontyphoidal Salmonella, with emphasis on emerging challenges in disease control. We highlight key virulence mechanisms, including Salmonella pathogenicity islands and Type III secretion systems, that mediate host cell invasion, intracellular survival, and immune modulatio

Role of vitamin D receptor genetic variants (ApaI and FokI) in association with nephropathy stages in a group of Egyptian patients with type 2 diabetes mellitus.

Diabetic nephropathy (DN) is one of the most worrisome complications of diabetes, causing significant social and economic impacts. Genetic polymorphisms in vitamin D receptor (VDR) gene may lead to genomic instability and increase susceptibility to end-stage renal disease (ESRD). In this research, we aimed to identify the association of genetic variants: ApaI "rs7975232" and FokI "rs10735810" in the VDR gene with nephropathy stages in diabetic patients. This case-control hospital-based study included 200 Egyptian participants divided into a group of 150 patients with type 2 diabetes mellitus (
