Discriminant validity of the Ankylosing Spondylitis Disease Activity Score (ASDAS) in patients with non-radiographic axial spondyloarthritis and ankylosing spondylitis: a cohort study.

The aim of this study was to assess discriminant validity of Ankylosing Spondylitis Disease Activity Score (ASDAS)-C-reactive protein (-CRP) and ASDAS-erythrocyte sedimentation rate (-ESR) and to compare with The Bath Ankylosing Spondylitis Disease Activity Index (BASDAI) as clinical tools for the measurement of disease activity in patients with non-radiographic axial spondyloarthritis (nr-axSpA) and ankylosing spondylitis (AS). Also, the cut-off values for ASDAS-CRP in nr-axSpA and AS is revisited. Patients with axSpA were recruited from Erciyes Spondyloarthritis Cohort (ESPAC) and were asses…

vtRNA1-1/p62 regulates macrophages autophagy in ankylosing spondylitis.

This research aims to investigate the role and mechanisms of the vtRNA1-1/p62 molecular axis in the regulation of autophagy in AS, with the goal of identifying novel diagnostic and therapeutic targets for AS. Clinical sample analysis revealed that the transcription levels of vtRNA1-1 and p62 decreased in the peripheral blood mononuclear cells (PBMCs) of the AS group; conversely, the levels of canonical autophagy-related genes (ATG3, ATG5) and inflammatory factors (TNF-α) significantly increased. Correlation analysis revealed that vtRNA1-1 levels were positively associated with p62 but were inv

Case reports: Pulmonary destructive lesions caused by ankylosing spondylitis.

BACKGROUND: Ankylosing spondylitis (AS) frequently produces thoracic-cage deformities-kyphosis and rib fusion-that restrict chest expansion and impair ventilation, occasionally progressing to atelectasis or pulmonary fibrosis. Such pulmonary manifestations are often misdiagnosed as tuberculosis or fungal disease. CASE PRESENTATION: We describe two AS patients with bilateral upper-lobe destruction whose initial chest CT scans suggested tuberculosis. After thorough microbiological work-up remained negative, both were diagnosed with AS-associated lung injury (ASLI) and referred to rheumatology. …

Proteomics signatures associated with hip arthropathy in ankylosing spondylitis.

OBJECTIVE: Ankylosing spondylitis (AS) is a chronic inflammatory disease primarily affecting the axial skeleton and peripheral joints, with hip arthropathy representing a severe complication that critically impairs mobility. While persistent inflammation is a hallmark of AS, the molecular mechanisms driving hip involvement remain poorly characterized. This study aimed to identify and validate protein biomarkers associated with hip arthropathy progression in AS through integrated proteomic and functional analyses. METHODS: Liquid chromatography-mass spectrometry (LC-MS/MS) was employed to scre…

Multiomics Identifies Potential Biomarkers in Ankylosing Spondylitis Bone Formation.

Objective: Ankylosing spondylitis (AS) is a long-term inflammatory condition characterized by intricate pathogenesis and significant genetic predisposition. Current treatment methods cannot completely halt the progression of the disease. The purpose of this research is to discover possible therapeutic targets for AS by integrating Mendelian Randomization (MR), transcriptomics analysis, and machine learning, providing new options for the clinical treatment of AS. Methods: In this study, we initially pinpointed differentially expressed genes (DEGs) linked to AS from the GEO database and acquired…

Mendelian randomization studies in ankylosing spondylitis: A systematic review.

INTRODUCTION: Previous research has revealed connections between ankylosing spondylitis (AS) and comorbid conditions. However, observational studies frequently encounter difficulties in establishing definitive causal links between potential risk factors and AS. In contrast, Mendelian randomization (MR) offers a more robust approach to exploring causality by leveraging the random allocation of alleles at birth, thereby minimizing confounding factors. METHODS: The PubMed, Web of Science, and Cochrane databases were searched for MR studies related to AS with a time frame of database creation to …

Elevated BMPR2 expression amplifies osteoblast differentiation in ankylosing spondylitis.

OBJECTIVE: Bone morphogenetic protein receptor type 2 (BMPR2) has been associated with radiographic changes in ankylosing spondylitis (AS), but further characterization of the cellular signaling pathway in osteoprogenitor (OP) is not clearly understood. The aim of this study was to investigate the expression of BMPR2 and bone morphogenetic protein 2 (BMP2)-mediated responsibility in AS. METHODS: We collected 10 healthy control (HC) and 14 AS-OPs derived from facet joints. Subsequently, we then conducted RNA sequencing with two samples per group and selected BMP-related genes. Facet joint tiss…

Screening Key lncRNAs of Ankylosing Spondylitis Using Bioinformatics Analysis.

BACKGROUND: Long non-coding RNAs (lncRNAs) are important regulators in ankylosing spondylitis (AS). Few studies have examined the lncRNA-RNA binding protein (RBP) interaction in AS. This study performed bioinformatics analysis and clinical verification to identify key lncRNAs and propose their RBP interaction. METHODS: Three GEO datasets of AS were analyzed by differential expression analysis. The differentially expressed lncRNAs between the AS and control groups were screened out, and the intersecting lncRNAs were regarded as target lncRNAs. Functional was performed to identify target lncRNA…

Fulminant Herpes Pneumonia Postaortic Surgery with Known Ankylosing Spondylitis.

Herpes simplex virus (HSV) pneumonitis is rare after cardiac surgery. A 36-year-old gentleman with ankylosing spondylitis underwent emergency surgery for a complex aortic aneurysmal disease. Preoperative treatment of aortitis with antitumor necrosis factor and steroid medication and surgical stress including cardiopulmonary bypass potentially created an immunosuppressive state and reactivation of undiagnosed HSV. Rapid HSV pneumonia ensued, culminating in fulminant organ failure and mortality. HSV pneumonia should be considered postoperatively in patients with severe respiratory distress, espe…

Connection of COX-2 variants to ankylosing spondylitis susceptibility.

This study aimed to explore the associations of COX-2 polymorphisms rs5275, rs20417, and rs2745557 with the susceptibility to ankylosing spondylitis among Chinese Han people. Polymerase chain reaction-restriction fragment length polymorphism (PCR/RFLP) was adopted for genotyping COX-2 polymorphisms rs5275, rs20417, and rs2745557 among 109 AS patients and 122 healthy controls. Genotype distribution in the control group was examined for these three polymorphisms to test whether it conformed to Hardy-Weinberg equilibrium (HWE) expectation. A χ2 -test was employed to compare genotype and allele fr…

Neutrophil Extracellular Traps and Interleukin 17 in Ankylosing Spondylitis.

Ankylosing spondylitis (AS) is a chronic inflammatory disease traditionally regarded as mediated by T lymphocytes. Recent progress has identified that cells of innate immunity are also important for the processes of inflammation and new bone formation, a hallmark of AS. Moreover, interleukin-17 (IL-17) is a cytokine implicated in both processes. Neutrophils are increasingly recognized as mediators of autoinflammatory and autoimmune diseases through several mechanisms, one being the release of neutrophil extracellular traps (NETs). NETs are equipped with an array of bioactive molecules, such as…
