The Grimelius stain is a valuable histochemical method for detecting the specific intracytoplasmic secretory granules of carcinoid tumors. The usefulness of this argyrophilic technique for aspiration biopsy cytology (ABC) specimens was studied by the histologic staining procedure. Aspirates from a metastatic intra-abdominal neoplasm, morphologically suggestive of carcinoid tumor and from a control specimen were restained according to the Grimelius method. Diffuse cytoplasmic argyrophilia was observed in both. The application of Grimelius silver stain to cytology specimens enhances diagnostic a

We studied 309 constipated infants and children who underwent rectal manometry (n = 261) and/or punch rectal biopsy, in which acetylcholinesterase histochemistry (n = 124) was compared with routine histologic examination (n = 93) for diagnostic accuracy. A 99% diagnostic accuracy was achieved with acetylcholinesterase histochemistry compared with 61% accuracy with routine hematoxylin-eosin staining on punch rectal biopsy. We recommend screening cooperative, quiet patients with rectal manometry. Most patients with abnormal manometric studies or those on whom manometry cannot be performed should

Retrorenal extension of pancreatic effusion or phlegmon associated with pancreatitis has been described frequently and is generally considered to represent extension of these processes from the anterior into the posterior pararenal space. A series of meticulous cadaveric dissections (n = 8) and a review of a large number of patients with pancreatitis (n = 53) show that these posterior collections actually represent extension of pancreatitis from the anterior pararenal space to a potential space between the laminae of the posterior renal fascia. Of 40 patients with posterior extension of pancre

Golgi preparations of the anterior part of the truncus of the corpus callosum from 11 adult human brains were investigated. The vertical plane of section was situated symmetrically between the frontal and sagittal plane. The use of this oblique plane of section enabled easy identification of the neurons with dendrites intermingling with transcallosal fibers, what was not possible in standard frontal sections. 2 types of such neurons (with features of other interstitial neurons) were described: fusiform and multipolar. Both types of neurons were more frequently impregnated in areas adjacent to

Two distinct nuclear antigens, designated NSpI and NSpII, have been characterized and differentiated from the centromeric antigen that reacts with sera from patients with the CREST syndrome. Both NSpI and NSpII produce a speckled pattern of indirect immunofluorescence on HEp-2 cells that resembles the pattern seen with anticentromere antibodies (ACA). They are differentiated from the ACA staining pattern by the absence of metaphase chromatin staining by NSpI antisera and by the absence of a discrete speckled pattern of staining by NSpII. Further, both NSpI and NSpII stain predominantly the per

A case of Legionnaires' disease is presented in which gram staining of impression smears from lung biopsy specimens showed Legionella pneumophila while other laboratory findings remained negative. This case illustrates the usefulness of gram staining in this disease and in the management of therapy.

Following amputation of the posterior half of Tubifex, all the nerve cells stainable by paraldehyde fuchsin (NF) immediately discharged. Fifteen minutes later, some stained NFs reappeared. Their number increased, reached a peak far above the normal level, then slowly decreased. This peak occurred 8 hours after amputation in the brain, after 3-4 days in the ventral ganglia. A similar cycle took place during the anterior regeneration of Tubifex whose four anterior segments were previously cut away. Following a double amputation (behind the first or fourth segment and at half length), the stimula

A simple technique is described for counterstaining Golgi-Kopsch impregnations. The sections are first stabilized by the method of Geisert and Updyke and then stained in 0.15% gallocyanin and 5% chromium potassium sulfate for 45 minutes at 55-60 C. The sections are then rinsed, dehydrated to 70% ethanol, cleared in terpineol, mounted and coverslipped. This procedure results in a light to medium blue stain of those cells not impregnated by the silver chromate. The major advantages of this procedure over earlier methods are: (1) the counterstain does not fade and (2) since no differentiation is

The three-dimensional distribution of dendrites from the dorsal neck muscles biventer cervicis (BC) and complexus (CM) was examined in the adult cat using intracellular staining techniques. Motoneurons were electrophysiologically identified, stained with injection of horseradish peroxidase, and reconstructed from serial histological sections. The dendritic distributions of all motoneurons examined followed an orderly pattern. Many dendrites extended rostrally and caudally to form of complex parallel collection of dendrites in the ventromedial nucleus. Other dendrites projected dorsolaterally i

Olfactory axons of fishes can be traced after vital staining of the olfactory surface with the fluorescent dye Procion brilliant yellow M 4-RAN. Following selective accumulation by the receptor dendrites and perikarya the dye migrates intra-axonally over distances of more than 200 micrometer. Fluorescence microscope observation of formaldehyde fixed and paraffin sectioned preparations of the olfactory organ demonstrate clearly the system of intraepithelial axons and their first order bundles running toward the olfactory nerve.

The copper phthalocyanin dye astra blue has been used to stain differentially mast cells of the intestine; however; the procedure has not been used widely because of the difficulty in preparing and using the dye solution. Described here is a simple, reliable, and consistent method for selectively staining mast cells using a dye solution that may be prepared in any laboratory without the aid of sophisticated pH metering equipment. Astra blue is mixed with an alcoholic solution containing MgCl2-6H2O and the pH indicator pararosaniline hydrochloride. Concentrated hydrochloric acid is added dropwi

The acid mucopolysaccharides of brain tissues are disclosed by their metachromatic staining with toluidine blue following saponification with potassium hydroxide, presumably as a result of the liberation of acid groups previously esterified. Earlier histochemical studies had disclosed the presence of neutral mucopolysaccharides by staining with the periodic acid-Schiff technique, and such staining is intensified by prior saponification. Many biochemical studies have reported the presence of both acid and neutral mucopolysaccharides in brain tissues. Within the white matter following brain edem

Four types of chief cells can be discerned in the cat carotid body. These cells are referred to as types 1 to 4. They are characterized--in the given order--by decreasing amounts of noradrenaline (argentaffin reaction) and acidic protein(s) (HCl hydrolysis/basic dye methods). The concentrations of acidic protein roughly parallel that of noradrenaline, being highest in type 1 and lowest in type 4 cells. It is suggested that acidic protein and monoamines together are stored in specific granules and secreted by ekcytosis. The various cell types reflect functional conditions. Histochemical methods

The recognition of Cryptococcus neoformans in Gram-stained smears of purulent exudates may be hampered by the presence of the large gelatinous capsule which apparently prevents definitive staining of the yeast-like cells. In such stained preparations, C. neoformans may appear either as round cells with gram-positive granular inclusions impressed upon a pale lavender cytoplasmic background or as gram-negative lipoid bodies.
