Experimental vesicular stomatitis virus infection in horses: effect of route of inoculation and virus serotype.

Horses were inoculated with Vesicular stomatitis New Jersey and Indiana viruses by routes simulating contact and vector transmission. Clinical signs, lesions, antibody development, viral shedding and persistence, and viremia were monitored. Horses were infected with both viruses by all routes as confirmed by seroconversion. Salivation, primary lesions at inoculation sites, and secondary oral lesions were the most common clinical findings. Viral shedding was most often from the oral cavity, followed by the nasal cavity; titers were highest from oral cavity samples. Virus was rarely isolated fro…

Potential for contact and mechanical vector transmission of vesicular stomatitis virus New Jersey in pigs.

OBJECTIVE: To determine whether swine could be infected with vesicular stomatitis virus New Jersey (VSV-NJ) by routes simulating mechanical vector and contact transmission, and to document clinical response and measure source, duration, and extent of associated viral shedding. ANIMALS: 48 approximately 10-kg male pigs. PROCEDURE: 7 inoculation routes were tested at 3 virus concentrations (10(6), 10(4), and 10(2) median tissue culture-infective doses [TCID50]). Inoculation routes included intradermal (snout), topical application of virus to a scarified area (ear or mucous membrane of the lip)…

Oral findings, treatment and follow-up of a case with major aphthous stomatitis (Sutton's disease).

Major aphthous stomatitis (Sutton's disease) is a clinical variant of recurrent aphthous stomatitis, which is noted for its high morbidity. Since the etiology of the disease is not clear, many therapies have been attempted. However, the controversial results hinder the adoption of a single mode of management. We present a 13-year-old boy with Sutton's disease, who was successfully treated with a combination of burst systemic prednisone (1 mg/kg/day for five days, thereafter half dose on alternate days for one week) and topical triamcinolone (four rinses a day). He continued the mouth rinses wi…

Resolution of multiple complexes of phosphoprotein NS with nucleocapsid protein N of vesicular stomatitis virus.

The interaction of the nucleocapsid protein N and the phosphoprotein NS of vesicular stomatitis virus (VSV) was studied, free of other viral proteins, by transcription from SP6 vectors, followed by translation in a rabbit reticulocyte lysate. N-NS complex formation depended strongly on cotranslation of the two proteins; when N and NS were mixed following separate translation of each, very little complex formation occurred. Conditions were found under which at least six N-NS complexes were separated from each other by electrophoresis in a nondenaturing gel system, and the following findings wer…

A sensitive method for quantification of vesicular stomatitis virus defective interfering particles: focus forming assay.

A focus-forming assay for the quantification of defective interfering (DI) particles of vesicular stomatitis virus (VSV) is described. This assay is based on the procedures described for lymphocytic choriomeningitis (LCM) virus by Popescu et al. (1976). Under appropriate conditions this focus-forming assay can quantify fewer than 100 DI particles/ml in a preparation containing a large number of infectious particles.

Association of vesicular stomatitis virus glycoprotein with virion membrane: characterization of the lipophilic tail fragment.

The proteolytic enzyme, thermolysin, degraded the external segment of the membrane glycoprotein of intact vesicular stomatitis (VS) virions but left behind a small nonglycosylated fragment, presumably embedded in the virion membrane. Other proteases generated membrane-associated glycoprotein fragments differing somewhat in molecular weight. The thermolysin-resistant, virion-associated fragment, which can be selectively solubilized by either Triton X-100 or chloroform/methanol, has a molecular weight of 5,200. Amino acid analysis of the glycoprotein fragment reveals a preponderance of hydrophob…

Stomatal and leaf hydraulic conductivity responses to changing light and CO2 conditions in Phaseolus vulgaris.

Extreme conditions brought up by climate change have a negative impact on plant photosynthesis. However, characterizing and exploiting extant genetic variation in plants for structure and function could lead to the assembly of genotypes adapted to extreme environments. This could be accomplished by combining suitable structures and adaptive mechanisms. We selected two common bean genotypes from the Andean (Calima) and the Mesoamerican (Jamapa) gene pools and analyzed their leaf structural characteristics, leaf hydraulics, and stomatal responses to transient light and CO2. We used the patterns

Influence of vapour pressure deficit and CO2 on the thermal sensitivity of stomatal function in tropical trees.

Stomatal conductance to water vapour (gs) is typically considered to operate in coordination with photosynthesis (A) to balance carbon gain and water loss in an optimal manner. However, at high temperatures that suppress A, gs has been seen to increase or remain high - reducing the predictive accuracy of some stomatal conductance models. We investigated the temperature sensitivity of leaf-level gas exchange and evaluated a temperature-dependent modification of a widely used stomatal conductance model across a temperature gradient (30 to 40°C) in three tropical tree species (Mallotus philippens

Bewitching the host: Striga disrupts host stomatal ABA signaling by impairing the tryptophan lock.

Parasitic plants of the Orobanchaceae family, particularly Striga hermonthica, rank among the world's most devastating agricultural weeds (Jamil et al., 2021). They inflict massive yield losses on staple cereal crops across Africa, including sorghum, millet, and rice, as well as globally on other essential crops such as maize, rapeseed, tomato, and legumes (Parker et al., 2009; Bouwmeester, 2018). These root parasites have evolved a specialized organ called the 'haustorium' that connects to the vascular system of the host to siphon water, minerals, and nutrients (Yang et al., 2015). A critical

The combined effect of decreased stomatal density and aperture increases water use efficiency in maize.

Stomata play a crucial role in balancing carbon dioxide uptake and water vapor loss, thereby regulating plant water use efficiency (WUE). Enhancing WUE is important for sustainable agriculture and food security, particularly for crops such as maize (Zea mays L.), as climate change and growing global food demand exacerbate limitations on water availability. Genetic factors controlling stomatal density and levels of the plant hormone abscisic acid (ABA) in leaves, which affect stomatal aperture, are key determinants of stomatal conductance (gs) and intrinsic WUE (iWUE). In this study, we demonst

Can Oxygen Isotopes in Tree Rings Be Used to Detect Stomatal Responses to Global Change?

Stomatal conductance (gs) regulates CO2 and water fluxes of plants. Although experiments have shown that gs decreases with elevated CO2, it is unclear how gs is responding in situ to long-term exposures to rising CO2 and a changing climate. Tree ring isotope analysis provides a unique method to assess tree ecophysiological responses to long-term exposures of slowly changing environmental conditions. In particular, it has been suggested that changes in gs can potentially be inferred from tree ring stable oxygen isotope ratios (δ18Otrc). Several studies have indeed used δ18Otrc trends to conclud

H2S modulates BrSDH1-1 alternative splicing to induce stomatal closure in Chinese cabbage.

Hydrogen sulfide (H2S), a gasotransmitter molecule, plays critical roles in stomatal closure and cellular bioenergetics. Alternative splicing (AS) is a key regulatory mechanism during plant development and stress responses; however, the interplay between H2S signaling and AS in drought tolerance remains unexplored in Chinese cabbage. In this study, we found that the mitochondrial inner membrane enzyme succinate dehydrogenase (SDH) responds to H2S signaling during stomatal closure. Silencing of BrSDH1-1 impaired the effects of H2S on stomatal closure, SDH activity, and ATP production. RNA-Seq a

Limited Regulation of Canopy Water Use Efficiency by Stomatal Behavior Under Drought Propagation.

Water use efficiency (WUE) is a critical ecosystem function and a key indicator of vegetation responses to drought, yet its temporal trajectories and underlying drivers during drought propagation remain insufficiently understood. Here, we examined the trajectories, interdependencies and drivers of multidimensional WUE metrics and their components (gross primary production (GPP), evapotranspiration, transpiration (T), and canopy conductance (Gc)) using a conceptual drought propagation framework. We found that even though the carbon assimilation efficiency per stomata increases during drought, t

US expert Delphi consensus on the prevention and management of stomatitis in patients treated with datopotamab deruxtecan.

BACKGROUND: Datopotamab deruxtecan (Dato-DXd) is an investigational antibody-drug conjugate that has shown promising results in phase III clinical trials of lung and breast cancers. Stomatitis is a common adverse event associated with Dato-DXd in these trials, and there are currently no formalized guidelines for its prevention and management. An expert consensus was needed to establish these guidelines. METHODS: Fifteen experts participated in a modified three-round Delphi to achieve consensus on the prevention and management of Dato-DXd-related stomatitis in lung and breast cancer patients. …
