Biotoxicity of mercury to Chlorella vulgaris as influenced by amino acids.

The toxicity of mercury ion, on Chlorella vulgaris, is largely influenced by amino acids. Five amino acids, namely alanine, asparagine, glutamic acid, cysteine and histidine, were added separately to the medium containing static dose of mercury. Survival (%) of the alga was reduced with the increasing concentrations of mercury. Of these five amino acids, cysteine was found to be the most effective while alanine and glutamic acid were the least effective on reducing the toxic effect of mercury on the alga measured in terms of growth, chlorophyll and protein content. The order of detoxification

Neurotoxicity of paraquat and triphenyltin in the earthworm, Eisenia fetida Sav. A histo- and cytopathological study.

Neuropathological effects of the pesticides paraquat (PQ, a herbicide) and triphenyltin (TPT, a fungicide) were studied on the postclitellar segmental ganglia of juvenile E. fetida specimens using light and electron microscopic methods. Growth retardation of the worms in standard culture medium was used as quantitative marker of sublethal effect. There is a distinct neuron group in the segmental ganglia characterised by high sensitivity against PQ- and another one against TPT-toxication. However the marked histo- and neurocytopathological alterations were similar in PQ and TPT treated worms. B

Cryopreservation of semen from Japanese white rabbits for use in teratological studies.

Cryopreservation of semen from Japanese White rabbits was examined to reduce the number of their males for use in teratological studies. Semen was frozen with liquid nitrogen, preserved, thawed, and tested for motility according to the method of Chen et al. Even after cryopreservation an average of 52% of the thawed sperms were motile. In a previous study, frozen- thawed sperms with a motility of 40% resulted in a high conception rate (approximately 88%) on artificial insemination when New Zealand White rabbits were used [5]. These results indicate the possibility that cryopreservation of seme

Single acute and repeated subacute toxicity evaluations of Annona atemoya leaf extract with in vitro anti-inflammatory potential.

The nutraceutical and biological potential of Annona atemoya, a fruiting plant, has been reported. We and others have demonstrated that A. atemoya leaf extract (AAL) has various pharmacological properties, such as antioxidant, antimicrobial, and neuroprotective effects. However, knowledge about the safety and potential toxicity of AAL remains limited. We aimed to assess the potential toxicity of AAL using acute and repeated subacute oral toxicity tests in rats. In both acute and repeated subacute toxicity test, no AAL-related behavioral abnormalities or changes in mortality, food intake, body …

Optimizing Analog Identification and Evaluation in the OECD QSAR Toolbox to Enable Robust Read-Across.

Read-across is one of the most widely used New Approach Methods (NAMs) for evaluating the safety of a chemical with insufficient toxicological data. However, the identification of analogs suitable for read-across is not always straightforward as it requires understanding of critical chemical and biological features that drive toxicity. The OECD QSAR Toolbox is recommended as a valuable tool to support analog identification. Here, a case study is presented on Benzenesulfonic acid, comparing two different analog identification approaches within the Toolbox. The suitability of analogs identified

Behavioral toxicity of polyhydroxyalkanoate microplastics in Drosophila melanogaster larvae revealed by deep-learning pose estimation.

Polyhydroxyalkanoates (PHAs) are microbially synthesized polyesters developed as biodegradable alternatives to conventional plastics, but incomplete degradation can leave PHA-derived particles in the environment, and their biological effects remain poorly characterized. This study investigated whether 96-h dietary exposure to 5-µm PHA particles altered locomotor organization in third-instar Drosophila melanogaster larvae. Larvae were exposed to 0.25, 2.5, or 7.5 mg L⁻¹ PHA, and movement in the standardized 5-65 s window was quantified using SLEAP tracking, speed and angular-speed interval dist

A screening strategy for identifying the developmental and reproductive toxicity potential of botanicals.

CONTEXT: Botanicals are widely consumed as dietary supplements and traditional medicines, yet their developmental and reproductive toxicity (DART) potential is often inadequately characterized. Botanicals are chemically complex and variable, complicating traditional in vivo testing and interpretation. There is a need for fit-for-purpose new approach methodologies (NAMs) to evaluate DART hazards of complex mixtures while reducing reliance on mammalian models. OBJECTIVE: This review describes the Botanical Safety Consortium (BSC) DART Working Group strategy to evaluate a battery of NAMs for scr

Beyond insects: High sensitivity of the water fleas to insecticides and implications for ecotoxicological assessment.

Although many model species are widely used in toxicity testing, the sensitivity and ecological relevance of water fleas in the genus Simocephalus remain underexplored. We compiled acute toxicity data for Simocephalus, 73.1% involving insecticides, and compared their responses with those of other aquatic taxa. Simocephalus were generally more sensitive to insecticides than several major groups, including insects, their intended targets. Although they showed higher sensitivity than Daphnia magna, the differences from crustaceans overall were modest. Species sensitivity distributions for chlorpy

Fish models of obesity, test guidelines, and regulatory challenges of identifying non-EATS endocrine disruptors.

Endocrine disrupting chemicals (EDCs) that interfere with estrogenic, androgenic, thyroidal and steroidogenic (EATS) pathways, as well as non-EATS pathways, should be identified and classified under EU chemical regulations. Identifying and classifying obesogenic EDCs is particularly challenging, as they are typically regulated through non-EATS mechanisms and lack standardized test methods and guidance for evaluation. This review evaluates fish models of obesity, synthesizes current evidence on chemical effects on fish adipogenesis, and discusses the regulatory challenges associated with EDC id

Toxicity and Anti-inflammatory Effects of Eleutherine bulbosa (Miller) Urb, Ethanolic Extract, in Zebrafish (Danio rerio).

INTRODUCTION: Eleutherine bulbosa (Miller) Urb, popularly known as "marupazinho", is frequently used in traditional medicine for treating various diseases, including hypertension, ulcers, constipation, and intestinal infection. However, there is little scientific knowledge available regarding the pharmacological effects of this species. Thus in vivo and in silico phytochemical studies are required to establish whether this plant has these effects. Further tests were necessary to evaluate the pharmacological activity of the compounds found in this plant, and demonstrate results related to the a

A proposal of criteria to support the EU classification on endocrine disruption for the thyroid modality and their application to four data-rich case studies.

Recently, the European Commission has implemented hazard categories to classify substances as endocrine disruptors for human health, i.e. ED HH 1 or ED HH 2, depending on the weight-of-evidence. However, specific guidance on how to differentiate between the two is unavailable. This article presents the CropLife Europe (CLE) proposal for a structured approach to support the ED HH classification for the thyroid modality. Further, the Thyroid Function-Related Neurodevelopmental Toxicity Testing and Assessment Scheme (Thyroid-NDT-TAS) has been modified in view of the new classification. Applicatio

Zebrafish as a model organism in One Health Toxicology: Impact of solvents and exposure routes on the toxicity of platinum anticancer drugs.

Zebrafish are widely used not only as a model in ecotoxicology but also to study the potential impact of chemicals on human health. Typically, zebrafish are exposed to chemicals dissolved in egg water or other defined media, which is the standard routine for ecotoxicology testing. This straightforward exposure method is usually also employed to monitor adverse effects in zebrafish to predict potential hazards and modes of action in humans. Here, we compared different exposure media and studied the impact of salinity and solvents relevant to ecotoxicity testing. For comparison, toxicants also w

Assessment of phototoxicity potential of botanicals as cosmetic ingredients using the in vitro 3T3 neutral red uptake phototoxicity test.

Cosmetics and personal care products are frequently formulated with botanical ingredients due to their beneficial properties, the nature of their composition, and consumers' interest for products with more natural or organic profiles. Compounds that absorb light significantly and are in contact with the skin have potential to become phototoxic upon exposure to sunlight. Here we demonstrate that an in vitro test methodology, the 3T3 Neutral Red Uptake Phototoxicity Test (3T3 NRU PT), is an effective screening tool in evaluation of botanical ingredients that absorb light in the Ultraviolet/Visib

Exploring the use of fatty acid ethyl esters as a potential natural solution for the treatment of fish parasitic diseases.

Alternatives to conventional chemical treatments for parasitic diseases in fish are needed. Microalgal-sourced fatty acid ethyl esters (FAEEs) have shown an antiparasitic effect against Gyrodactylus turnbulli infection in guppies. Here, we tested a range of commercial FAEEs of various carbon chain lengths and unsaturation levels against two fish parasites. Guppies and barramundi infected with G. turnbulli and Trichodina sp., respectively, were used. The most effective FAEE, after excluding those toxic to fish, was ethyl laurate (12:0). For both parasites, the LD50 was 18.75 μM within 250 min o

Bioaccumulation mediated by water solubility leads to differences in the acute toxicity of organophosphorus insecticides to zebrafish (Danio rerio).

The use of some organophosphate insecticides is restricted or even banned in paddy fields due to their high toxicity to aquatic organisms. The aim of this study is to elucidate the main pathways and target organs of organophosphate insecticide toxicity to fish exposed via different routes by integrating histopathological and biochemical techniques. Using malathion as the model drug, when the dosage is 20-60 mg/L, the toxicity of whole body and head immersion drugs to zebrafish is much higher than that of trunk immersion drugs. A dose of 21.06-190.44 mg/kg of malathion feed was fed to adult zeb
