The control of Glossina morsitans morsitans (Diptera: Glossinidae) in a settled area in Petauke District (Eastern Province, Zambia) using odour-baited targets.

A trial to control G. m. morsitans with the use of 980 odour-baited, insecticide-impregnated targets was conducted in a 300 km2 area in the Eastern Province of Zambia between 1989 and 1991. The area is highly cultivated and cattle density is high (about 8 cattle/km2). Targets were deployed along roads and tracks. Deployment was restricted to suitable tsetse habitat. The effect of the targets on the tsetse population and on the transmission of tsetse-transmitted trypanosomosis was monitored by means of man-walked fly rounds and sentinel herds, respectively. The apparent density of tsetse in the

Trypanosome infections in warthogs (Phacochoerus aethiopicus) in the Gambia.

The prevalence of trypanosome infections in warthogs (Phacochoerus aethiopicus) in The Gambia was found to be 11% of a sample of 62 animals. All isolates were identified as Trypanosoma simiae. Serological evidence indicated a higher level of exposure to T. simiae, but results were inconclusive for the presence of Trypanosoma congolense. The course of T. simiae infection in warthog piglets showed a rapidly rising parasitaemia, with a concomitant fall in packed cell volume, and resulted in a prolonged period of low-level parasitaemia. The same infections killed domestic piglets.

Development of multiple drug resistance of Trypanosoma congolense in Zebu cattle under high natural tsetse fly challenge in the pastoral zone of Samorogouan, Burkina Faso.

Preliminary data from an ongoing epidemiological survey in the pastoral zone of Samorogouan (Kénédougou) indicate the occurrence of multiple-drug-resistant Trypanosoma congolense. Despite frequent trypanocidal drug treatments with diminazene aceturate (Berenil, Hoechst) at 7 mg/kg body weight (bw) at intervals of 2 to 4 weeks, no significant drop in the prevalence of African animal trypanosomosis (AAT) was observed. To examine a suspected drug resistance, 20 Zebu cattle, naturally infected with T. congolense and/or T. vivax, were transferred in December 1989 from Samorogouan into a fly-proof s

Parasite kinetics and cellular responses in goats infected and superinfected with Trypanosoma congolense transmitted by Glossina morsitans centralis.

Trypanosoma congolense infected tsetse were fed on the flanks of goats at sites drained by the prefemoral lymph node. The efferent lymphatic of this lymph node was surgically cannulated and the lymph was collected daily and examined for appearance of parasites, lymph flow and cells. Trypanosomes were detected in the lymph 4 days after infection, which was 2 days prior to the appearance of the local skin reaction or the presence of parasites in the blood. Once the animal became parasitaemic, trypanosomes were found to recirculate in the lymphatic system, appearing in the lymph of the contralate

High density lipoprotein levels in the serum of trypanosensitive and trypanoresistant cattle. Changes during Trypanosoma congolense infection.

Nonpermissiveness to trypanosome infection has been correlated in some instances with the presence of toxic serum factors, e.g. high density lipoproteins (HDL) of human serum can lyse T.b. brucei. The present study examines the possibility of a role for such factors in West African cattle that are resistant to trypanosomiasis. Cattle used in this study were previously selected as resistant or sensitive to trypanosomiasis under heavy natural Glossina challenge. - A comparison of the direct effect of serum from trypanoresistant and trypanosensitive Baoulé cattle on the development of pathogenic

Further studies of cyclical transmission and antigenic variation of the ILDar 1 serodeme of Trypanosoma vivax.

Antigenic variation in the ILDar 1 serodeme of the naturally rodent-infective stock of West African Trypanosoma vivax has been investigated following cyclical transmission. The immunofluorescent and immune lysis tests were employed with a panel of 39 variant-specific mouse antisera. When antigenically homogeneous, or mixed, populations were transmitted by tsetse flies to goats, the first peak parasitaemias arising in the goats were antigenic mixtures (up to 9 major, and several minor variants being recognized in some cases) from which the ingested variant was absent. Although first peak parasi…

Parasite development and host responses during the establishment of Trypanosoma brucei infection transmitted by tsetse fly.

Following inoculation of Trypanosoma brucei into large mammals by the tsetse fly a local skin reaction, the 'chancre', develops due to trypanosome proliferation. We have cannulated the afferent and efferent lymphatics of the draining lymph node in goats and examined the onset of a cellular reaction, the emigration of the parasite from the chancre and the development of both antigenic variation and the specific immune response. The chancre first became detectable by day 3 post-infection, peaked by day 6 and then subsided. Lymphocyte output increased 6- to 8-fold by day 10 and the number of lymp

Failure of trypanosomal membrane antigens to induce protection against tsetse-transmitted Trypanosoma vivax or T. brucei in goats and rabbits.

A purified protein, relative molecular weight 83 kilodalton (kD), and plasma membranes from Trypanosoma brucei were tested as potential vaccines against tsetse-transmitted T. vivax and T. brucei in goats and rabbits. The 83 kD protein was found in lysates of all clones of T. brucei examined, as well as in lysates of T. vivax, T. congolense and T. rhodesiense. Rabbits and goats were immunized with various amounts of antigen in Freund's complete adjuvant and boosted twice with antigen in Freund's incomplete adjuvant. Two weeks after the last inoculation, the goats were challenged with T. vivax-i

Spiroplasma endosymbiont reduction of host lipid synthesis and Stomoxyn-like peptide contribute to trypanosome resistance in the tsetse fly Glossina fuscipes.

Tsetse flies (Glossina spp.) vector African trypanosomes that cause devastating diseases in humans and domestic animals. Within the Glossina genus, species in the Palpalis subgroup exhibit greater resistance to trypanosome infections compared to those in the Morsitans subgroup. Varying microbiota composition and species-specific genetic traits can significantly influence the efficiency of parasite transmission. Notably, infections with the endosymbiotic bacterium Spiroplasma have been documented in several Palpalis subgroup species, including Glossina fuscipes fuscipes (Gff). While Spiroplasma…

Livestock, pathogens, vectors, and their environment: A causal inference-based approach to estimating the pathway-specific effect of livestock on human African trypanosomiasis risk.

Livestock are important reservoirs for many zoonotic diseases, however the effects of livestock on human and environmental health extend wel … here livestock keeping activities modify the environment in such a way that habitat suitability for tsetse flies, and in turn human infection risk, is reduced. We conducted this study in four high burden countries: at the point level in Uganda, Malawi, and Democratic Republic of Congo (DRC), and at the county level in South Sudan. Our results indicate cattle a …

Heterogeneity in the prevalence and intensity of bovine trypanosomiasis in the districts of Amuru and Nwoya, Northern Uganda.

BACKGROUND: Livestock trypanosomiasis, transmitted mainly by tsetse flies of the genus Glossina is a major constraint to livestock health and productivity in the sub-Saharan Africa. Knowledge of the prevalence and intensity of trypanosomiasis is important in understanding the epidemiology of the disease. The objectives of this study were to (a) assess the prevalence and intensity of trypanosome infections in cattle, and (b) to investigate the reasons for the heterogeneity of the disease in the tsetse infested districts of Amuru and Nwoya, northern Uganda. METHODS: A cross-sectional study was …

The rate of antigenic variation in fly-transmitted and syringe-passaged infections of Trypanosoma brucei.

Rates of antigenic variation were measured in vivo in several populations of cloned lines of Trypanosoma brucei before and after cyclical transmission through tsetse flies. Two cloned lines were adapted for use in laboratory conditions by extensive syringe passaging and rates of antigenic switching/cell/generation were less than 3 x 10(-6) and 1 x 10(-4) in each line. Rates of switching were then determined after fly transmission of the first line and generated per capita rate values of greater than 2 x 10(-3) in three of four populations examined. In the fourth population the switch rate was …

Susceptibility of African buffalo and Boran cattle to Trypanosoma congolense transmitted by Glossina morsitans centralis.

Four African buffalo (Syncerus caffer) and four Boran cattle (Bos indicus) were each exposed to the bites of 10 tsetse flies infected with Trypanosoma congolense. Although both groups of animals became infected, the buffalo showed no clinical signs of trypanosomiasis while the cattle suffered from the disease characterized by pronounced skin reactions, high parasitaemia and severe anaemia. The prepatent periods in the buffalo varied from 18 to 27 days in comparison with 11 to 14 days in the cattle. In the buffalo, skin reactions were only detectable by histological examination of skin biopsies…
