Adjuvants enhance both the magnitude and duration of immune responses, therefore representing a central component of vaccines. The nature of the adjuvant can determine the particular type of immune response, which may be skewed toward cytotoxic T cell (CTL) responses, antibody responses, or particular classes of T helper (Th) responses and antibody isotypes. Traditionally, adjuvants have been added to intrinsically poor immunogenic vaccines, such as those using whole killed organisms or subunit vaccines. Here, we have compared cellular immune responses induced by the immunogenic modified life-

Although dental caries is a global problem in modern times, no vaccines are available for preventing these diseases. Among the bacterial pathogens that cause dental caries, including Streptococcus mutans, S. sobrinus, and Lactobacillus acidophilus, and Actinomyces viscosus, S. mutans is the most prominent and prevalent species. During the past, much effort has been focused on developing vaccines against S. mutans. Early attempts used fixed whole cells of S. mutans, but later it was found that serological cross-reactivity between heart tissue antigens and Streptococcus antigens occurs in patien

Despite the success of global vaccination campaigns, vaccine access in low-resource settings is an ongoing challenge. Subunit vaccines are a well-established and clinically scalable intervention, yet they have achieved limited success for poorly immunogenic antigens such as those associated with SARS-CoV-2. Delivery strategies that promote gradual release of subunit vaccines from the injection site offer the potential to improve humoral immunity by enhancing lymph node exposure, however, clinical implementation of this strategy is challenging due to poor scalability and high costs. Here, we pr

Vaccination is a key strategy for control of tuberculosis (TB), and considerable progress has been made in the past 5 years to develop improved vaccines for humans and animals, differentiate vaccinated animals from those infected with Mycobacterium bovis and deliver vaccines to wildlife. Studies have moved from testing vaccines in small animal models to clinical trials in humans and from experimental challenge studies in cattle and wildlife to evaluation of vaccines in the field. Candidate vaccines undergoing testing in humans include live mycobacterial vaccines to replace bacille Calmette Gué

The authors outline the history of vaccination against anthrax in animals, from the end of the 19th century to the present time. The three main steps in the production of specific vaccines are described in detail: production of vaccines from live, encapsulated bacteria, followed by vaccines from live, unencapsulated bacteria and, finally, subunit vaccines. Advantages and disadvantages of these three types of vaccine, some of which are still in use today, are described and discussed.

Subunits of a proline-rich coccidioidal antigen (Ag2/PRA) of Coccidioides immitis were analyzed by comparison as vaccines in mice. The optimal dose of plasmid vaccine encoding full-length Ag2/PRA was determined to be between 10 and 100 microg. Mice vaccinated with plasmids encoding amino acids (aa) 1 to 106 were as protective as full-length Ag2/PRA (aa 1 to 194). The subunit from aa 27 to 106 was significantly but less protective. Plasmids encoding aa 90 to 151 or aa 90 to 194 were not protective. Analogous results were obtained with recombinant vaccines of the same amino acid sequences. In ad

Klebsiella pneumoniae causes severe respiratory-associated infections in healthy individuals, and widespread antibiotic resistance in K. pneumoniae poses a global health threat. Although great efforts have been taken to develop effective vaccines against K. pneumoniae, there are no licensed vaccines against K. pneumoniae available up to date. In the current study, we designed a potent subunit vaccine OmpW, which was obtained from an outer membrane protein (OmpW) to maximize host responses in mice. Antibody-mediated responses and protective effects were determined to compare the immunogenicity

Infectious coryza, caused by Avibacterium paragallinarum, is a respiratory disease that results in significant economic losses for the global poultry industry. Current inactivated whole-cell vaccines often fail to provide broad protection, largely due to the extensive antigenic diversity among circulating strains. To address this limitation, we applied a reverse vaccinology strategy to identify candidate antigens for rational subunit vaccine development. Using genomic and proteomic data from 95 Av. paragallinarum strains and a subtractive genomics-immunoinformatics pipeline, we prioritized pro

BACKGROUND: A coronavirus disease 2019 (COVID-19) subunit vaccine developed with the molecular clamp platform was among the first to enter clinical trials in 2020. While the vaccine was well suited to large-scale manufacture and demonstrated a favorable safety and immunogenicity profile, it did not progress into further clinical testing due to the human immunodeficiency virus 1 (HIV-1)-derived sequence in the molecular clamp, interfering with certain point-of-care HIV-1 diagnostic tests. A second-generation molecular clamp (MC2) has since been developed. Here, we describe a phase I clinical tr

Avian infectious bronchitis virus (IBV) is the causative agent of infectious bronchitis, which results in considerable economic losses. It is imperative to develop safe and efficient candidate vaccines to control IBV infection. In the current study, recombinant baculoviruses co-expressing the S1 and N proteins and mono-expressing S1 or N proteins of the GX-YL5 strain of IBV were constructed and prepared into subunit vaccines rHBM-S1-N, rHBM-S1 and rHBM-N. The levels of immune protection of these subunit vaccines were evaluated by inoculating specific pathogen-free (SPF) chickens at 14 days of

Mink refractory diarrhea is a seasonal disease that occurs in many mink farms in China. Mink circovirus (MiCV) has been recognized as the causative agent of the disease. The aim of the study was to develop a subunit vaccine against mink refractory diarrhea. A recombinant baculovirus strain expressing the capsid protein was constructed using the baculovirus expression vector system (BEVS). A subunit vaccine was developed based on the capsid protein with appropriate adjuvant. Then, a field trial was carried out in two districts in order to evaluate the efficiency of the subunit vaccine. The fiel

Tuberculosis (TB) is a serious disease around the world. Bacillus Calmette-Guérin (BCG) is the only TB vaccine licensed for use in human beings, and is effective in protecting infants and children against severe miliary and meningeal TB. However, BCG's protective efficacy is variable in adults. Novel TB vaccine candidates being developed include whole-cell vaccines (recombinant BCG (rBCG), attenuated Mycobacterium tuberculosis, killed M. tuberculosis or Mycobacterium vaccae), adjuvanted protein subunit vaccines, viral vector-delivered subunit vaccines, plasmid DNA vaccines, RNA-based vaccines

Bovine viral diarrhea (BVD) poses a substantial economic threat to the cattle industry, necessitating vaccines with broad cross-protective immunity. Heterologous prime-boost immunization represents a promising strategy to broaden immune protection by engaging complementary immunological pathways across vaccine platforms. Here, we evaluated heterologous prime-boost regimens combining recombinant subunit, adenovirus-vectored, and DNA vaccines, all targeting the E2 glycoproteins of BVDV-1 (I E2) and BVDV-2 (II E2; I E2-II E2), in a BALB/c mouse model. The DNA/subunit and DNA/adenovirus regimens e

Zaire ebolavirus (EBOV), Sudan ebolavirus (SUDV), and Marburg marburgvirus (MARV) are the most prevalent and pathogenic species of filovirus. Previously, we showed that glycoprotein antigens from each virus could be lyophilized to create thermostable monovalent subunit vaccines. However, cross-protection is not expected from the monovalent vaccines and therefore developing a trivalent filovirus vaccine would be desirable. Subunit protein vaccines often require the addition of an adjuvant to sufficiently boost the immunogenicity. Typically, liquid suspensions or emulsions of adjuvants and lyoph

Foot-and-mouth disease (FMD) is an acute, febrile, and highly contagious infectious disease common in cloven-hoofed animals. Outbreaks and epidemics of FMD can result in major economic losses of livestock. Using ferritin nanoparticles as the scaffold for an antigen can enhance the immunogenicity of the subunit vaccine and provide possible protection against FMD. We used a baculovirus expression system to express four recombinant proteins (VP1, VP1-Ft, G-H loop-Ft, and ferritin) and the protective immunity of the FMD ferritin nanoparticle vaccines was evaluated in mice. The recombinant subunit

The respiratory mucosa is the primary portal of entry for numerous viruses such as the respiratory syncytial virus, the influenza virus and the parainfluenza virus. These pathogens initially infect the upper respiratory tract and then reach the lower respiratory tract, leading to diseases. Vaccination is an affordable way to control the pathogenicity of viruses and constitutes the strategy of choice to fight against infections, including those leading to pulmonary diseases. Conventional vaccines based on live-attenuated pathogens present a risk of reversion to pathogenic virulence while inacti

RESPIRATORY SYNCYTIAL VIRUS VACCINES. Respiratory syncytial virus (RSV) is responsible for lower respiratory infections, particularly in children under five years of age (acute infant bronchiolitis) and the elderly over 60. Monoclonal antibodies (palivizumab and nirsevimab) are used to prevent bronchiolitis. Four types of vaccine are currently under development: subunit vaccines composed of recombinant proteins or viral pseudoparticles, messenger RNA vaccines, recombinant vector vaccines and live attenuated vaccines. They are indicated for pregnant women to protect infants against bronchioliti

The failure of the BCG vaccine for tuberculosis in large, controlled clinical trials, coupled with the gradual consensus that it is mostly ineffective in preventing adult pulmonary disease in endemic areas, has led to a concerted effort to develop a new generation of vaccines. This work is ongoing in a variety of areas, including DNA vaccines, subunit vaccines, recombinant vaccines, and auxotrophic vaccines. Several such candidates are giving promising results in mouse and guinea pig, aerosol-challenge infection models and should move to clinical trials in the near future.
