Vitiligo is an acquired depigmentation disease with an unknown etiology. Several theories on its etiology have been proposed, including autoimmune mechanisms, oxidative stress, genetic factors, environmental factors, and lack of melanocyte growth factor. Recent studies have demonstrated a possible correlation between microbial infections and the development or consequences of vitiligo. This study aims to investigate the potential association between microbial infections and vitiligo, focusing on identifying any explainable correlations between these two. A systematic search was performed on al…

Vitiligo is a cutaneous autoimmune disease characterized by the destruction of epidermal melanocytes leading to white patches with a global prevalence of about 0.5-2%, and patients' quality of life are greatly affected by the change in appearance and social discrimination caused by the disease. Most of the key cytokines in the pathogenesis of vitiligo act through the Janus kinase/signal transducer and activator of transcription signaling pathway, which is an effective therapeutic target. The first generation Janus kinase inhibitors, i.e., tofacitinib and ruxolitinib, inhibit a variety of Janus…

INTRODUCTION: Vitiligo is a chronic autoimmune skin disorder characterized by progressive melanocyte loss, resulting in well-demarcated depigmented patches that significantly impact quality of life. Affecting 0.5-2% of the global population, vitiligo poses both psychosocial and therapeutic challenges. Despite multiple conventional therapies such as corticosteroids, calcineurin inhibitors, and phototherapy, durable repigmentation remains difficult to achieve.
AREAS COVERED: This review summarizes recent advances in understanding the emerging medical treatment of vitiligo. A targeted literature…

Vitiligo is a chronic autoimmune depigmenting disorder affecting 0.5%-2% of the global population, characterized by bidirectional interplay between psychological stress and disease progression, with accumulating evidence highlighting the central role and translational relevance of the neuro-endocrine-immune-cutaneous axis in its pathogenesis. Epidemiological data indicate over half of patients experience significant psychological stress prior to disease onset, while visible depigmentation markedly elevates the burden of depression and anxiety, establishing a self-amplifying pathogenic loop. Me…

BACKGROUND: Vitiligo is an autoimmune skin disorder primarily characterized by the absence of melanocytes, leading to the development of white patches on the patient's skin. Narrowband Ultraviolet B (NB-UVB) therapy is among the most effective approaches for stimulating the reformation of hyperpigmentation. This treatment utilizes a narrow spectrum of NBUVB wavelengths ranging from 311 to 313 nm to irradiate the affected area, thereby preventing the destruction of migrating and proliferating melanocytes. Nevertheless, the molecular alterations occurring in both the hair follicle and the interf…

Vitiligo and alopecia areata are chronic autoimmune dermatoses that are relatively common in the pediatric population. Vitiligo is a pigmentation disorder and alopecia areata is a non-scarring alopecia. Both conditions can be highly stigmatizing and have a significant impact on the mental health and quality of life of young patients and their families. There are no large-scale prospective therapeutic studies in the pediatric population. As a result, the expertise of the pediatric dermatologist, along with shared decision-making with the patient and his/her parents, is critical for effective ma…

Vitiligo is characterized by the autoimmune destruction of melanocytes, which most visibly affect skin pigmentation, but are also present in the choroid and retinal pigmented epithelium of the eye raising the possibility of underappreciated vision loss in affected populations. To inform whether this vulnerable population should be more rigorously screened and offered early intervention, we performed a multi-center retrospective analysis of optotype recognition of 248 pediatric vitiligo patients compared with 496 age- and sex-matched healthy subjects between 2000 and 2023; we found a significan…

Vitiligo is a chronic autoimmune disorder characterized by the selective destruction of melanocytes, leading to depigmented patches of skin. Whilst its pathogenesis is not fully understood, genetic predisposition, environmental triggers, oxidative stress, metabolic dysfunction and impaired cell adhesion are all implicated. Vitiligo occurs in two primary forms - non-segmental and segmental - and affects approximately 0.5-2% of the global population. Beyond its physical manifestations, vitiligo imposes a significant psychosocial burden on patients. Current treatments include topical corticostero…

The imbalance of the skin redox system is regarded as a crucial factor contributing to the loss of melanocytes in vitiligo. However, it remains unclear whether alterations in signal transmission between melanocytes and other cells impact the homeostasis of the skin microenvironment. Hence, leveraging single-cell sequencing and microarray data, we investigated the role of cell-cell and ligand receptor interactions in the pathogenesis of vitiligo. We discovered that the granulin-sortilin 1 ligand-receptor serves as an essential bridge for communication between melanocytes and other skin cells in…

Vitiligo is a multifactorial chronic depigmentary disorder characterised by the loss of functional melanocytes. Its pathogenesis involves complex interactions among genetic predisposition, environmental factors, and autoimmune dysregulation. In recent years, extensive research has highlighted the critical role of epigenetic regulation in vitiligo development, including aberrant DNA methylation, dysregulated histone modifications, and non-coding RNA (ncRNA) expression disturbances. These abnormal epigenetic modifications contribute to disease progression by disrupting melanogenesis, promoting o…
