Effects of acetylcholine on thyrotropin-releasing hormone release from the rat caecum in vitro.

Effects of acetylcholine on the release of thyrotropin-releasing hormone (TRH) from the rat caecum in vitro were studied. The rat caecum was incubated in medium 199 with 1.0 mg/ml of bacitracin and 100 micrograms/ml of ascorbic acid (pH 7.4) (medium). The amount of TRH release into the medium was measured by radioimmunoassay. The immunoreactive TRH (ir-TRH) release from the rat caecum was enhanced significantly in a dose-related manner with the addition of acetylcholine, but not changed with atropine. The stimulatory effect of acetylcholine on ir-TRH release from the rat caecum was blocked wit…

In vitro electrophysiological effects of cimetidine, prostaglandin E2 and acetylcholine on the rabbit gastric mucosa.

The electrophysiological effects of cimetidine, cytoprotective dose of prostaglandin E2 (PGE2) and acetylcholine were determined in parallel in Ussing-chambered rabbit fundic and antral mucosal preparations. In the fundic mucosal preparations both cimetidine and PGE2 caused an increase in transmucosal potential difference (PD) and in short-circuit current (ISC); the transepithelial resistance (Rt) was essentially unchanged. Addition of acetylcholine to the pretreated fundic preparations produced further gradual increases in PD and ISC; cimetidine pretreatment delayed this effect of acetylcholi…

[Induction of coronary arterial spasm by intracoronary administration of acetylcholine in patients with vasospastic angina].

To examine whether intracoronary injections of acetylcholine induce coronary artery spasm in patients with vasospastic angina, incremental doses (20, 30 and 50 micrograms) were injected directly into the coronary arteries in 12 patients with variant angina (Group A: rest angina with electrocardiographic ST-segment elevation during attacks), 19 with vasospastic angina (Group B: rest angina and/or effort angina with variable threshold in the treadmill exercise stress test), 11 with organic coronary artery stenosis but without angina (Group C), and 14 without coronary artery disease (Group D). A …

Antagonism by tubocurarine and verapamil of the regenerative acetylcholine release from mouse motor nerve.

The role of presynaptic acetylcholine receptors and Ca2+ channels in the regenerative acetylcholine release was studied in the cut muscle preparation of mouse phrenic nerve hemidiaphragm. The regenerative release shown as a prolonged endplate depolarization was evoked by stimulation of the nerve with a train of pulse at 75-300 Hz when acetylcholinesterase activity was depressed with neostigmine or by lowering temperature. Tubocurarine, cobratoxin, verapamil, diltiazem and nifedipine at low concentrations, which had a negligible effect on the endplate potential, shortened the duration of regene…

Effects of acetylcholine, isoprenaline and forskolin on electrolyte and protein composition of rabbit mandibular saliva.

1. The major purpose of this study was to investigate cellular regulation of the ductal transport processes in salivary glands which act to … l and as individual components after their separation by high-performance liquid chromatography. 2. Acetylcholine was used as a 'Ca2+-mobilizing' agonist (i.e. one which both elevates intracellular Ca2+ concentration and activates protein kinase C). Isoprenaline was initially used to elevate intracellular cyclic AMP concentration but was subsequently abandoned …

Endothelium-derived relaxing factor and the effects of acetylcholine and histamine on resistance blood vessels.

1. The role of endothelium-derived relaxing factor (EDRF) in the action of vasodilator (acetylcholine, histamine, nitroprusside) and vasoconstrictor (noradrenaline, vasopressin) drugs on vascular resistance in the isolated perfused kidney and mesentery of the rat was studied. 2. Acetylcholine (EC50 = 0.18 +/- 0.05 nmol and 3.1 +/- 0.06 nmol, n = 8) and histamine (EC50 = 31.2 +/- 4.9 nmol and 46.2 +/- 3.9 nmol, n = 8) produced dose-related vasodilatation in noradrenaline-preconstricted (i.e. 'high tone') rat renal and mesenteric blood vessels. The response to both vasodilators (but not nitropru…

Acetylcholine content in rat brain is elevated by status epilepticus induced by lithium and pilocarpine.

The effects of status epilepticus on the concentration, synthesis, release, and subcellular localization of acetylcholine, the concentration of choline, and the activity of acetylcholinesterase in rat brain regions were studied. Generalized convulsive status epilepticus was induced by the administration of pilocarpine to lithium-treated rats. The concentration of acetylcholine in the cortex, hippocampus, and striatum decreased prior to the onset of spike activity or status epilepticus. Once status epilepticus began, the concentration of acetylcholine increased over time in the cortex and hippo…

Acetylcholine receptor antibodies in myasthenia gravis: use of a qualitative assay for diagnostic purposes.

We have modified the techniques of Lindstrom and of Tindall to measure serum acetylcholine receptor antibody using human antigen bound to 125I-alpha Bungarotoxin. By using 10 microliters of serum and precipitating antigen-antibody complexes with an excess of staph A, we found that only one out of 43 patients with clinically diagnosed active generalized Myasthenia Gravis had no antibodies. In pooling these results with the results of tests done for diagnostic purposes we found positive results in 54/55 generalized active MG, 8/21 MG in remission, 16/37 ocular MG and 0/55 healthy controls. Two o…

Loss of acetylcholine receptor clusters induced by treatment of cultured rat myotubes with carbachol.

Prolonged exposure to carbachol disrupts the acetylcholine receptor (AChR) clusters of cultured rat myotubes without causing myotube loss. The effect is reversible, and is dependent on temperature. Half-maximal cluster loss is achieved at 3 microM carbachol. Cluster loss is also caused by other agonists of the AChR and is blocked by receptor antagonists. QX314 (a lidocaine derivative), meproadifen, and fluphenazine also completely block cluster loss caused by carbachol. These results are consistent with the idea that cluster loss caused by carbachol and other receptor agonists results from the…

In vivo effects of beta-bungarotoxin on the acetylcholine system in different brain areas of the rat.

The in vivo effects of beta-bungarotoxin (beta-BT) on the acetylcholine (ACh) system were studied in the whole cerebrum and in different brain regions. The effect of beta-BT on cerebral ACh and choline (Ch) contents was time-dependent. The results show that a single intracerebroventricular injection of 1 microgram toxin increased both the ACh and Ch contents in the cortex, hippocampus, and cerebellum, while in the striatum the ACh level was decreased. Ten nanograms of toxin injected into the lateral ventricle twice, on the first and third days, led to a reduced ACh level 2 days after the last …

Increased release of [3H]acetylcholine in vitro from the mouse hippocampus by a convulsant barbiturate.

The effects of the convulsant barbiturate, 5-(2-cyclohexylidene-ethyl)-5-ethyl barbituric acid (CHEB), on the spontaneous release of [3H]acetylcholine (ACh) from mouse hippocampal slices in an in vitro superfusion system have been evaluated. The pattern of the release of [3H]ACh by a single treatment with CHEB or an elevated potassium concentration was similar, with the peak release occurring in the same fraction. A maximally effective concentration of CHEB (500 microM) caused a 177% stimulation of spontaneous release of [3H]ACh, while 50 mM KCl increased the release by 2100% above the baselin…

Comparison of the release of endogenous and of 3H-acetylcholine from slices of rat cerebral cortex.

A comparison of the release of endogenous and of tritiated acetylcholine (3H-ACh) was performed in eserinized slices of rat cerebral cortex labelled in vitro in the presence of 0.43 microM tityustoxin (TsTX), a scorpion venom known to increase the overflow of endogenous acetylcholine. The endogenous and the radioactive ACh were spontaneously released into the organ bath. At the moment when the spontaneous efflux had levelled off, the endogenous ACh collected during 1 min accounted for 1.03 +/- 0.14% of the tissue content and a similar value (0.80 +/- 0.26%) was found for 3H-ACh. The basal outf…

alpha-Bungarotoxin-acetylcholine receptors in the chick ciliary ganglion: effects of deafferentation and axotomy.

alpha-Bungarotoxin (alpha-BuTX) binds in a saturable and practically irreversible fashion to membrane-associated receptors in the ciliary ganglion of the adult chick. The binding of toxin to receptors is competitively inhibited by nicotinic cholinergic ligands, and for these properties the receptors are regarded as acetylcholine receptors of the nicotinic type (alpha-buTX-AChRs). The rate constant of association (K1) and dissociation (K-1) of the toxin-receptor reaction has been estimated to be K1 = 7.4 x 104 M(-1) sec(-1) and K-1 = 9.6 X 10(-6) sec(-1), respectively. Light autoradiography sho…

[A histochemical study of brain tissue in experimental convulsive seizures (componetns of acetylcholine metabolism)].

The author studied substances related to acetylcholine metabolism (acetylcholinesterase, non-specific acetylcholinesterase, SH, SS-groups, histidine, Ca) using histochemical methods. There was an increase in the intensity of reactions on SH-groups and Ca, and a rise in the staining degree on SS-groups and acetylcholinesterase in the subconvulsive phase of the stroke. These shifts were more significant in the brain stem. A generalization of convulsive strokes intensifies the given changes and causes an increase of the intensity of the reactions on histidine after the end of the stroke (after 1,…
