Complete Genome Sequencing of Acinetobacter baumannii AC1633 and Acinetobacter nosocomialis AC1530 Unveils a Large Multidrug-Resistant Plasmid Encoding the NDM-1 and OXA-58 Carbapenemases.

Carbapenem-resistant Acinetobacter spp. are considered priority drug-resistant human-pathogenic bacteria. The genomes of two carbapenem-resistant Acinetobacter spp. clinical isolates obtained from the same tertiary hospital in Terengganu, Malaysia, namely, A. baumannii AC1633 and A. nosocomialis AC1530, were sequenced. Both isolates were found to harbor the carbapenemase genes blaNDM-1 and blaOXA-58 in a large (ca. 170 kb) plasmid designated pAC1633-1 and pAC1530, respectively, that also encodes genes that confer resistance to aminoglycosides, sulfonamides, and macrolides. The two plasmids wer

In vitro activities of zosurabalpin and comparator antimicrobials against clinical isolates of carbapenem-resistant Acinetobacter baumannii.

UNLABELLED: Carbapenem-resistant Acinetobacter baumannii (CRAB) poses a major therapeutic challenge due to the limited availability of effective treatment options and is associated with substantially increased mortality in healthcare-associated infections. Zosurabalpin (ZAB, RG6006) is a novel macrocyclic peptide (MCP) antibiotic that exerts its antibacterial activity by inhibiting lipopolysaccharide transport, ultimately leading to bacterial cell death. This study evaluated the in vitro activities of ZAB and other antimicrobial agents against clinical CRAB isolates. A total of 206 clinical A.…

Biofilm Formation in Clinical Acinetobacter baumannii Is Influenced by Isolate Source and Is Inversely Correlated With Antibiotic Resistance.

Acinetobacter baumannii is a troublesome bacterium that is highly prevalent in hospital settings, particularly in intensive care units (ICUs). Biofilm is one of the main virulence factors that makes A. baumannii a successful pathogen, enabling it to survive the harshest environments. This study aimed to corrolate the biofilm-forming capacity of clinical A. baumannii isolates with their antibiotic resistance phenotypes and isolation sources. A total of 327 clinical isolates originated from different hospitals, were recovered from diverse clinical specimens collected from patients admitted to th…

Internalization of antibiotics by the multi-resistant bacteria Acinetobacter baumannii through the CarO outer membrane porin.

Antibiotic resistance represents an emerging global health threat. In this contribution, we report all-atom molecular dynamics simulations of a bacterial protein in Acinetobacter baumanii. By using enhanced sampling we show the favorable internalization of the antibiotic via the CarO porin. This result correlates with the knockout or the mutation of the porin as a resistance mechanism.

Acinetobacter baumannii's lifestyle includes soil-dwelling colonization of decaying plant material and airborne spread.

Acinetobacter baumannii is a Gram-negative nosocomial pathogen that is notorious for its rapid development of antibiotic resistance. However, its ecology and evolution outside hospital settings remain poorly defined. Here, we demonstrate that the natural lifestyle of A. baumannii includes soil-dwelling and airborne dissemination, which helps explaining its adaptability and tolerance to desiccation, radiation and antibiotics, and thus its predisposition to establish within hospitals. Starting from white stork nestlings previously discovered as a reservoir, we studied food chains and associated …

Effect of polymyxin resistance evolution on the fitness costs of Acinetobacter baumannii and the underlying mechanisms of polymyxin resistance.

The increasing incidence and emergence of polymyxin-resistant Acinetobacter baumannii pose a significant threat to global public health. The presence of sub-minimal inhibitory concentration (sub-MIC) antibiotics during clinical treatment is inevitable and may drive the development of antibiotic resistance. Therefore, understanding the fitness costs and underlying mechanisms of resistance evolution under sub-MIC polymyxin B (PMB) is crucial for controlling the spread of PMB-resistant strains. In this study, PMB resistance evolved rapidly within 1-2 days in different A. baumannii strains and acq…

Regulation of drug resistance and virulence of Acinetobacter baumannii by quorum sensing system under antibiotic pressure.

INTRODUCTION: Acinetobacter baumannii is a formidable pathogen renowned for its role in hospital-acquired infections. In recent years, largely due to antibiotic abuse and other reasons, bacteria are frequently exposed to sub-minimum inhibitory concentration (sub-MIC) levels of antibiotics. Accumulating evidence suggests that sub-MIC antibiotic pressure serves as a critical driver of bacterial resistance evolution and virulence adaptation. However, the regulatory mechanisms underlying antibiotic stress adaptation in A. baumannii remains poorly understood. The quorum sensing (QS) system is a key…

Mobilization of the AbGRI4 resistance island in Acinetobacter baumannii: IS26 action and homologous recombination both contribute.

UNLABELLED: Carbapenem-resistant Acinetobacter baumannii isolate HUMC1 (USA, 2009), which is hypervirulent and extensively antibiotic resistant, has been used in many experimental studies. The complete sequence of the chromosome and two plasmids was determined, and the causes of the resistance phenotype were investigated. HUMC1 (ST2:ST1839:KL22:OCL3) is an unusual global clone 2 (GC2) isolate that lacks an AbGRI1 type resistance island interrupting the comM gene. However, a version of AbGRI2, which is present together with AbGRI1 in the oldest known GC2 isolate, and versions of AbGRI3 and the …

A requirement for Acinetobacter baumannii purine biosynthesis during lung infection is exacerbated by host zinc deficiency.

Acinetobacter baumannii is a leading cause of hospital-acquired infections, including ventilator-associated pneumonia. Dietary zinc deficiency is a major risk factor for pneumonia, and hospitalized patients at risk for A. baumannii infection have increased rates of zinc deficiency. We previously showed that dietary zinc deficiency enhanced A. baumannii pneumonia pathogenesis via an IL-13-dependent mechanism in mice. Here, we identified A. baumannii genes required for proliferation in the lungs of zinc-deficient mice using a genome-wide transposon sequencing (Tn-seq) screen. In zinc-deficient m…
